1.Exploration of Theory of Kidney Having Its Manifestations in Hair Through Observing Effect of Hushen No.域on Hair Amino Acids Contents in Patients with Chronic Kidney Disease at Stage 1-4

Objective To investigate the changes of hair amino acids contents in patients with chronic kidney disease at stage 1-4 (CKD 1-4) and the intervention effect of Hushen No.Ⅱ, thus to provide modern medical evidences for the theory of kidney having its manifestations in hair. Methods One hundred cases of patients suffering from CKD 1-4 were recruited and were randomly divided into control group and treatment group, 50 cases in each group. And twenty healthy volunteers were included in normal group. The treatment group and control group received conventional treatment, and the treatment group were additionally treated with Hushen No. Ⅱ orally, the treatment lasting 12 months. And then high performance liquid chromatography was used to test the changes of hair amino acids of the three groups before and after treatment. Results Compared with the normal group, 14 kinds of amino acids in the hair of the control group and the treatment group were significantly low ( P<0.05) before treatment. After treatment, 14 kinds of amino acids in the hair of the control group and the treatment group were significantly increased ( P<0.05) as compared with those before treatment, the increase of the treatment group being superior to the control group (P<0.05). Conclusion The abnormal change of 14 kinds of amino acids in the hair of patients with abnormal renal function may contribute to the modern medical material basis of the theory of kidney having its manifestations in hair.
2.DbcAMP inhibitted the proliferation of FTC-133 cell

Objective To study the effect of dualdi butyryl cyclic AMP (dbcAMP)on the proliferation of FTC-133 cell line. Methods FTC-133 cells were normally cultured and divided into control group,dbcAMP treatment group (0.5,1.0,2.0 mmol/L). After FTC-133 cells were treated with dbcAMP (0.5,1.0,2.0 mmol/L)for 24 h or 48 h,the growth activity and growth curve was detected by MTT.Changes of the cell cycle were detected by flow cytometry.The mRNA and protein expression of Raf1 were measured by RT-qPCR and Western blotting.Results Compared with control group,the growth activity of FTC-133 cells was re-duced by different levels of dbcAMP in a dose-time dependence manner.The number of FTC-133 cells was decreased in the S phase and increased in the G2/M phase.The mRNA and protein expression of Raf1 of treatment group were both reduced compared with control group.Conclusion dbcAMP significantly reduced FTC-133 cells proliferation and promoted apoptosis,and which might be involoved by ERK MAPK signalling.

4.Clinicopathological analysis of primary rhabdomyosarcoma of the ear

Objective To investigate the clinicopathological change in primary rhabdomyosarcoma of the ear, and its differential diagnosis with infantile rhabdomyosarcoma, primary neuro-ectodermoma, and malignant rhabdomyoma. Methods Gross and microscopic examination and immunohistochemical staining were performed in 8 cases of primary rhabdomyosarcoma of ear. Results Seven male and one female patients with mean age of 7.4 years were found. The tumor was diagnosed as embryonal rhabdomyosarcoma in 7 patients, and botryoid rhabdomyosarcoma in 1 patient. Immunostaining revealed that vimentin and myoglobin were 100% positive, desmin in 75% and actin in 82%. Conclusions Although the histological appearance of primary ear rhabdomyosarcoma does not differ from those found in other soft tissues, its clinicopathological diagnosis may be difficult in some cases, and further immunohistochemical analyses, may be necessary for a correct diagnosis. Its differential diagnosis includes infantile rhabdomyofibrosarcoma, primary neuroectodermal tumor and malignant rhabdoid tumor.
5.Research on NICU HD video mobile visitation system design and application

Objective: To explore a proper way of building hospital's NICU visit system. Methods: Compare and analyze existing NICU visit systems, put forward the approach of HD mobile video visit system in NICU. Design and develop a visit system consists of visiting cart, HD video capturing subsystem, image acquisition and transmission software, wireless HDMI receiving and monitor system and intercom system. Apply the method and analyze the effect of the visit system. Results: The mobile video visit system can avoid direct contact between parents and neonatal to prevent cross-infection, in the same time, the system has shown the hospital's humanistic care for parents, and that will help to establish a harmonious doctor-patient relationship. Conclusion:The HD mobile video visit system is a new effective choice for hospital's NICU visit systems, and it will be good clinical significance.
6.Induction of BAG2 in proteasome inhibitor-induced apoptosis in human thyroid carcinoma cells

Background and purpose: For neoplasms with different sources, proteasome inhibitors can inhibit their growth and promote the cell apoptosis. Its mechanism may be associated with Bcl-2-associated athanogene 2 (BAG2). We aimed to investigate the involvement of in thyroid cancer cell death induced by proteasome inhibitors. Methods:A panel of thyroid cancer cells (ARO, FRO, KTC1, KTC2, KTC3, 8305C and 8505C) were treated with vehicle or proteasome inhibitor MG132;BAG2 mRNA and protein levels were analyzed using quantitative real-time polymerase chain reaction (qRT-PCR) and Western blot, respectively. Results:MTT results indicated that FRO and KTC2 cell lines were the most sensitive to proteasome inhibitors. MG132 induced BAG2 mRNA and protein expression in the panel of thyroid cancer cells with various degree (P<0.01), for FRO and KTC2 cell line, the BAG2 mRNA level was increased in 20 to 25 times, meanwhile the protein expression level also increased signiifcantly;Sensitive FRO cells demonstrated quicker induction of BAG2 compared with insensitive ARO cells. Moreover, the extents of BAG2 induction in FRO cells were higher than that in ARO cells. Conclusion: BAG2 is a novel molecule induced by proteasome inhibitor, which might function as a proapoptotic molecule in thyroid cancer cell death mediated by proteasome inhibitor.
7.Expression of soluble H1N1 haemegglutinin in two distinct insect baculovirus expression systems

Objective To express functional haemegglutinin(HA)protein in two different bacularvirus expression systems.Methods The whole open reading frame of A/Sichuan/1/2009(H1N1)HA was obtained by synthesis,and the HA protein were expressed in insect cells by two different bacularvius expression systems:BaculoGold system and Bac-to-Bac system. Soluble HA protein was identified by Western blot and haemegglutination test. Results The correct full length of HA gene was obtained and cloned into pAcGP67B and pFAST Bacl vectors,respectively.After 3 rounds of virus amplifyjng by re-infection of Sf9 cells,the HA protein was detected in supematant of BaculoGold system and in intracellular of Bac-to-Bac system which is much better than the former.Purified HA protein was positive not only identified by Western blot,but also detected by haemegglutinin test. Conclusion Functional HA protein was successfully expressed in two distinct bacularvirus expression systems,of which the Bac-to-Bac bacularvirus expression system is more suitable for expression of A/Sichuan/1/2009(H1N1)HA protein.
8.Risk factor analysis of feeding intolerance in very low birth weight infants

Objective To investigate the risk factors associated with feeding intolerance in very low birth weight infants (VLBWI) and to investigate the beneficial factors for preventing feeding intolerance.Methods Retrospectively analysis was carried out in 151 VLBWI cases in our hospital from January 2005 to November 2010.Logistic regression analysis was performed to find the risk factors of feeding intolerance.Results The overall rate of feeding intolerance in VLBWI was 47.7 % (72/151).Dysporia and infection were associated with feeding intolerance in VLBWI (Wald x2 =7.748 and 4.555,P =0.05 and 0.033 respectively).Gender,multifetation were not associated with feeding intolerance(Ps >0.05).Conclusion The feeding intolerance is associated with dysporia and infection in VLBWI.
9.Effect of NR2B antagonist on behavior and related signal proteins in a rat model of levodopa-induced motor complications

ObjectiveTo investigate the role of N-methyl-D-aspartate receptor subunit 2B (NR2B) antagonist CP-101.606 in behaviour and expression of related signal proteins in a rat model of levodopa-induced motor complications.MethodsThe hemi-parkinsonian rat model was produced by injecting stereotaxically 6-OHDA to right medial forebrain bundle.Then,rats were intraperitoneally treated with levodopa (50 mg/kg with benserazide 12.5 mg/kg,twice daily) for 22 days.On 23th day,rats received CP-101.606 before levodopa administration.Rotational duration was estimated.After sacrificed,phosphorylated NR2B and Ca2+/calmodulin-dependent protein kinase Ⅱ (CaMKⅡ) and glutamate receptor 1 ( GluR1S831 ) were observed by western blot.ResultsThe results showed that NR2B antagonist CP-101.606 reversed the levodopa-induced shortened rotational duration.Chronic levodopa treatment increased abundance of the phosphorylated NR2B and downstream related signal proteins CaMKⅡ and GluR1S831 to (145.3±6.5)% and (132.5±5.7)% and (105.6±6.3)%,respectively.Moreover,CP-101.606 could reduce hyperphosphorylation of NR2B and CaMKⅡ and GluR1 S831 to (102 ± 4.9 )%,(98.4±3.9)% and (49.5 ± 4.2 )%,respectively.ConclusionsThese results indicate that the enhancement of N-methyl-D-aspartate (NMDA) receptor function mediated by NR2B phosphorylation contribute to development of motor complications,through a mechanism that involved the downstream signal mediators of NMDA receptor overactivation.Pharmaceuticals which act to inhibit NR2B may be useful in the treatment of the motor complications in parkinsonian patients.
10.Effects of neural stem cells transplantation on apoptosis-related genes expression in the motor cortex of rats subjected to spinal cord transection

BACKGROUND: Many studies showed that neural stem cells (NSC) transplantation can promote functional improvements in rats subjected to spinal cord injury. However, the underlying molecular mechanisms remain poorly understood.OBJECTIVE: To observe the effects of NSC transplantation on expressions of Bax, Bcl-2 and Caspase-3 in the motor cortex in rats subjected to spinal cord transection.DESIGN, TIME AND SETTING: The randomized controlled animal experiment was performed at Institute of Neuroscience,Kunming Medical College from July 2007 to December 2008.MATERIALS: Five green fluorescent protein transgenic mice with 14-15 embryonic days were prepared for NSC. Additionally 88 adult female Sprague-Dawley (SD) rats were randomly divided into sham operation group (n=8), model group (n=40) and NSC transplantation group (n=40).METHODS: Model of spinal cord transection was established by cut transversely Sprague-Dawley (SD) rats T_9 segment. Rats in the sham operation group were subjected to laminectomy at T_8, without spinal cord injury. After the spinal cord was exposed at the lesion site, a small piece of 2 mm~3 gel foam soaked with 3×10~5 NSC were implanted into the gap to fill the lesion site of the T_9 level in rats of the NSC transplantation group. The measurements of relative indexes were performed at the days 3, 7,14, 21 and 28 after transplantation.MAIN OUTCOME MEASURES: Changes of Bax, Bcl-2 and Caspase-3 expressions were detected by RT-PCR.RESULTS: Compared to the sham operation group, the Bax expression in the model group had no significant difference (P >0.05), the expression of Bcl-2 was decreased obviously at days 14 and 28 after operation (P < 0.05), while a significant increase on the expression of Caspase-3 at day 3 (P < 0.05). Compared to the model group, the expression of Bax was dramatically decreased in the NSC transplantation group at day 3 (P < 0.05), with a notably increased Bcl-2 expression at days 14 and 21 after transplantation (P < 0.05), but the expression of Caspase-3 presented a significant decrease at days 3 and 7 after transplantation (P < 0.05).CONCLUSION: NSC transplantation probably regulates the expression of apoptosis genes (Bax, Bcl-2 and Caspase-3 mRNA) to promote neurological function recovery in rats subjected to cord transection.
