1.Protective Effects of Defatted Dabai Peel Extracts in Hypercholesterolemic Rabbits Based on Histopathological Methods

Defatted dabai peel contains a high amount of anthocyanin. Anthocyanins are known to prevent several
types of disease, including cardiovascular-related complications. This study aimed to describe the
effects of different doses of defatted dabai peel extract by histopathological analyses on lesions in the
liver, kidney, heart and aorta. Histopathology methods were applied to determine the protective effects
of defatted dabai peel extracts against hypercholesterolemia-induced oxidative damages to animal
organs. Haematoxylin and eosin staining was applied for histopathology examination for liver, kidney,
heart and aorta. Data showed that a high dose of defatted dabai extract (3000 mg per day) applied to
hypercholesterolemic rabbits for eight weeks had mild protective effect, especially reducing the severity
of hepatic fibrosis and steatosis of the renal medulla. The high dose of extract supplementation also
reduced inflammation of aorta and formation of atherosclerosis plaque in the cell wall of right ventricle
of the heart. The high dose of defatted dabai peel extract could be a protective agent against oxidative
stress.

Background and Objectives:
During infection, Reactive oxygen species (ROS) signaling is activated to protect the cells from invading microorganisms. However, a high level of ROS may also damage the host tissue. The anthocyanin delphinidin is known to have a strong antioxidant activity that protects cells from oxidative damage. This study explored the potential of crude anthocyanin extract from the fruit of Solanum melongena (Eggplant) and Delphinidin-3-glucoside in enhancing the innate immunity in Caenorhabditis elegans against Staphylococcus aureus and Klebsiella pneumoniae.
Methodology:
Caenorhabditis elegans was used to study innate immune response because it lacks adaptive immunity. First, the sublethal concentration of S. melongena crude anthocyanin extract (SMCAE) and Delphinidin-3-glucoside (D3G) in C. elegans was determined. The sublethal concentration of SMCAE and D3G was used to supplement the nematodes during its exposure to S. aureus and K. pneumoniae. The survival rate
was then observed until day five post-L4. SMCAE and D3G were also tested for probable antimicrobial activity against Staphylococcus aureus and Klebsiella pneumoniae.
Results and Conclusion
This study found that both SMCAE and D3G showed no inhibitory effect on the growth of the bacteria. However, both SMCAE and D3G enhanced the survival of the nematode when exposed to S. aureus and K. pneumoniae. Overall, this study indicates that the anthocyanin delphinidin in S. melongenacrude extract protected the C. elegans against S. aureus and K. pneumoniaeinfection through its antioxidant activity.

3.Cytoprotective Mechanism of Cyanidin and Delphinidin against Oxidative Stress-Induced Tenofibroblast Death.

Age-related rotator cuff tendon degeneration is related to tenofibroblast apoptosis. Anthocyanins reduce oxidative stress-induced apoptotic cell death in tenofibroblasts. The current study investigated the presence of cell protective effects in cyanidin and delphinidin, the most common aglycon forms of anthocyanins. We determined whether these anthocyanidins have antiapoptotic and antinecrotic effects in tenofibroblasts exposed to H₂O₂, and evaluated their biomolecular mechanisms. Both cyanidin and delphinidin inhibited H₂O₂-induced apoptosis in a dose-dependent manner. However, at concentrations of 100 μg/ml or greater, delphinidin showed cytotoxicity against tenofibroblasts and a decreased antinecrotic effect. Cyanidin and delphinidin both showed inhibitory effects on the H₂O₂-induced increase in intracellular ROS formation and the activation of ERK1/2 and JNK. In conclusion, both cyanidin and delphinidin have cytoprotective effects on cultured tenofibroblasts exposed to H₂O₂. These results suggest that cyanidin and delphinidin are both beneficial for the treatment of oxidative stress-mediated tenofibroblast cell death, but their working concentrations are different.

4.The Safety and Pharmacokinetics of Cyanidin-3-Glucoside after 2-Week Administration of Black Bean Seed Coat Extract in Healthy Subjects.

We analyzed the pharmacokinetics of C3G on data from twelve subjects, after 2-week multiple dosing of black bean (Phaseolus vulgaris, Cheongjakong-3-ho) seed coat extract, using the mixed effect analysis method (NONMEM, Ver. 6.2), as well as the conventional non-compartmental method. We also examined the safety and tolerability. The PK analysis used plasma concentrations of the C3G on day 1 and 14. There was no observed accumulation of C3G after 2-week multiple dosing of black bean seed coat extract. The typical point estimates of PK were CL (clearance)=3,420 l/h, V (volume)=7,280 L, Ka (absorption constant)=9.94 h(-1), ALAG (lag time)=0.217 h. The black bean seed coat extract was well tolerated and there were no serious adverse events. In this study, we confirmed that a significant amount of C3G was absorbed in human after given the black bean seed coat extract.

5.Antioxidant activities and determination of phenolic compounds isolated from oriental plums (Soldam, Oishiwase and Formosa).

The purposes of this study were to determine phenolic compounds and to evaluate antioxidant activities of plums (Soldam, Oishiwase and Formosa). Soldam contains the highest amount of total phenolics among cultivars (Formosa: 4.0%, Oishiwase: 3.3%, Soldam: 6.4% for total phenolic) as well as the total flavonoids of which constituents were mainly myricetin and anthocyanidin. The antioxidant activities were measured by DPPH, ABTS radical scavenging, and SOD-like activities. The DPPH radical scavenging activity of Korean plum extracts (200 microg/mL) showed more than 43%, and the Soldam turned out to be the highest : ID50 value: 160-177 microg/mL for Formosa and Oishiwase; 58-64 microg/mL for Soldam. The ABTS radical scavenging activity of Korean plum extracts (200 microg/mL) was found to be more than 50%. The SOD-like activity of Korean plum extracts (200 microg/mL) showed more than 70%. Among three kinds of cultivars, Soldam had the highest antioxidant activity. The nitrite scavenging activity of Soldam was 61.5%, which is the highest, compared with that of the other cultivars, about 50%. From these results, Korean plums turned out to be phytochemical rich fruit as well as to show high antioxidant activities.

6.Research progress on chemical constituents and pharmacological activities of Morus alba.

Morus alba, a traditional economic crop, is also a significant medicinal plant. The branches(Mori Ramulus), leaves(Mori Folium), roots and barks(Mori Cortex), and fruits(Mori Fructus) of M. alba are rich in chemical components, such as alkaloids, flavonoids, flavanols, anthocyanins, benzofurans, phenolic acids, and polysaccharides, and possess hypoglycemic, hypolipidemic, anti-inflammatory, anti-tumor, anti-microbial, liver protective, immunoregulatory, and other pharmacological activities. This study analyzed the sources, classification, and functions of the main chemical components in M. alba and systematically summarized the latest research results of essential active components in M. alba and their pharmacological effects to provide references for in-depth research and further development as well as utilization of active components in M. alba.

7.Adsorption dynamics study on interactions of pigments and human whole saliva.

OBJECTIVEIn situ, real timely and dynamically monitoring the processes of theaflavin (TF), curcumin (Cur) and cyanidin (Cy) binding to human whole saliva (WS) surface has been investigated by surface plasmon resonance (SPR) technique at the molecular level.
METHODSThe affinity between pigments and WS, association rate constant (k(a)), dissociation rate constant (k(d)), association equilibrium constant (K(A)) and dissociation equilibrium constant (K(D)) of pigments binding on WS surface had been determined by SPR and the Langmuir model as well as the Freundlich model. The data were analyzed by one-way ANOVA and SNK-q test.
RESULTSThere were significant differences among TF, Cur and Cy in k(a), k(d), K(A) and K(D) (P<0.05). Our results showed that the adsorption isotherm of pigments on WS surface could be better described by the Freundlich model than the Langmuir model. The pigments adsorption on WS surface was dominant by specific interactions, such as hydrogen bonding. The affinity of pigments to WS were TF> Cur>Cy (P<0.05), as evidenced by the rate constants and equilibrium constants.
CONCLUSIONCompared with Cur and Cy, TF shows much higher adsorption capacity on WS surface, suggesting the importance of the hydroxyl group in pigment/protein interactions.

8.Effect of homogeneity on cell growth and anthocyanin biosynthesis in suspension cultures of Vitis vinifera.

The instability of secondary metabolite production is a ubiquitous problem in plant cell culture. To understand the instability, the investigation of anthocyanin accumulation in suspension cultures of Vitis vinifera, as a model system, has been initiated in our laboratory. Suspension culture of a relatively homogeneous cell line E of V. vinifera, was established by long-term cell line selection by anthocyanin content differentiation. The aggregate size of E was smaller than that of other cell lines obtained by routine screening method. The variation coefficients of anthocyanin content in suspension cultures of E were 8.7% in long-term subcultures and 5% in repeated flasks, respectively. The effects of elicitor, precursor feeding and light irridiation on biomass and anthocyanin accumulation in suspension cultures of E had been investigated and the results showed that all the variation coefficients were lower than 12% and this indicated the importance of homogeneity on stable production in plant cell culture. With the combination treatment of 30micromol/L phenylalanine and 218micromol/L methyl jasmonate in the dark in suspension cultures of E, the anthocyanin content and production in suspension culture of E was 5.89-fold and 4.30-fold of the controls, respectively, and all the variation coefficients of biomass and anthocyanin accumulation were lower than those of the controls in 5 successive subcultures.

9.Colored Foods and Diabetes.

Brightly colored foods are a rich source of phytochemicals that may act against hyperglycemic agents. The major concern in diabetes is increased oxidative stress. Some studies on phytochemicals have revealed their antioxidant activities and beneficial effects for diabetes. Some well-known phytochemicals are carotinoids and flavonids such as lycopene, beta-carotin, and anthocyanin. Many studies suggest that phytochemical-rich foods might have therapeutic uses. However, the ingestion of improper amounts of these chemicals may be harmful. The most important message of this study is that a balanced diet has favorable effects on oxidative stress status in type 2 diabetes patients.

10.Cloning and temporal-spatial expression analysis of dfr gene from Scutellaria baicalensis with different colors.

Dihydroflavanol-4-reductase (Dfr) is a key enzyme that regulates the synthesis of anthocyanin and proanthocyanidin in the flavonoid biosynthesis pathway. To investigate the difference of dfr gene in Scutellaria baicalensis Georgi with different colors in the same ecological environment, three complete full-length sequences of dfr gene were cloned from the cDNA of S. baicalensis with white, purple-red and purple colors using homologous cloning and RACE techniques. The three genes were named Sbdfr1, Sbdfr2 and Sbdfr3, respectively, and their corresponding structures were analyzed. The results showed that all three Dfr proteins have highly conserved NADPH binding sites and substrate-specific binding sites. Phylogenetic analysis showed that they are closely related to that of the known S. viscidula (ACV49882.1). Analysis of key structural domains and 3D models revealed differences in the catalytically active regions on the surface of all three Dfr proteins, and their unique structural characteristics may provide favorable conditions for studying the substrate specificity of different Dfr proteins. qRT-PCR analysis shows that dfr was expressed at different level in all tissues except the roots of S. baicalensis in full-bloom. During floral development, the expression level of dfr in white and purple-flowered Scutellaria showed an overall upward trend. In purple-red-flowered Scutellaria, the expression first slowly increased, followed by a decrease, and then rapidly increased to the maximum. This research provides a theoretical basis for further exploring the mechanism and function of Dfr substrate selectivity, and are of great scientific value for elucidating the molecular mechanism of floral color variation in S. baicalensis.
