Objective To investigate the effects of focal mild hypothermia on apoptotic neurons and the expression of glutamate transporter(GLT-1)and metabotropic glutamate receptor(mGluR2/3)after cerebral ischemia in rats.Methods Eighty-eight adult rats were randomly divided into mild hypothermia group,normothermia group and control group.Unilateral permanent middle cerebral artery occlusion(pMCAO)was induced.In mild hypothermia group,the brain temperature of occlusive side was decreased and maintained for 3 h at(33?0.5)℃ with Sticking Focal Mild Hypothermia Instrument.The body temperature of normothermia group was maintained at(37?0.5)℃.3 h,6 h,12 h,24 h,3 d,7 d after cerebral ischemia,the expression of GLT-1 and mGluR2/3 around infarction cortex were determined by Western Blot.6 h,24 h,3 d,7 d after cerebral ischemia,TUNEL method was performed to label neuronal apoptosis.Results 24 h,3 d after cerebral ischemia of mild hypothermia group,24 h,3 d,7 d after cerebral ischemia of normothermia group,the apoptotic neurons around infarction core markedly increased comparing with 6 h after cerebral ischemia(all P

Objective:To explore the feasibility and stability of using oleic acid to establish acute respiratory distress syndrome(ARDS) model in Beagle dogs.Methods:A total of 25 Beagle dogs were injected oleic acid with a speed of 0.25?0.03ml/Kg through jugular vein after being anesthetized.The gas ventilation index,oxygen metabolism,histopathological changes,chest-ray scan and stability of each index after establishing the ARDS situation were continuously monitored and evaluated.Results:All 25 dogs were reached the diagnosis criteria of ARDS in 4.16?0.92 hours after injection of oleic acid,The arterial PaCO2 of the animals was 27.98?8.25mmHg,PaO2 was 65.40?11.48mmHg,and PaO2/FiO2 was 182.3?29.6.After mechanical ventilation treatment,PaO2/FiO2≤200.Pulmonary mesenchyme,and alveolus edema,bleeding,shrinkage and transparent formation were seen under microscope.There was darkness or coarse shadow in the lung by the X-ray scan.Conclusions:Through injection of oleic acid via jugular vein,a Beagle dog ARDS model may be quickly established with stable changes of hemodynamics,pulmonary mechanics and histology.
3.Pathological changes of rabbit sciatic nerve caused by exposure to high intensity ultrasound

Objective To observe the pathologic changes of the rabbit sciatic nerves after the exposure of high intensity ultrasound(HIU) (7.1 MHz,45.86 W/cm2)and to establish the experimental base for protection of the nerves near the organs and tissues treated with HIU. Methods The rabbit sciatic nerves were exposed to HIU of different doses as defined by the action time of the ultrsound(0, 15, 25 and 50 seconds, respectively); The Pathological changes of the nerves were observed at different times after exposed to HIU(1 week,2 weeks,1 month,3 months and 6 months). Results In 0 s (45.86 W/cm2?0 s)group, no significant pathological changes were observed. In 15 s(45.86 W/cm2?15 s) group, slight deformation of the myelin sheath were observed and recovery took place at 2 weeks. In 25 s (45.86 W /cm2?25 s) group, the nerves became broken partly and recovered at 3 months. In 50 s(45.86 W/cm2?50 s) group, great part of the nerves got coagulation necrosis and had not recovered at 6 months. Conclusion The larger the dosage of HIU, the higher degree the injury of the sciatic nerve, the more difficult the nerves to get repaired.

5.Evaluation of supine fulcrum pressurized radiograph for predicting spinal flexibility in adolescent idiopathic scoliosis

Objective To examine supine fulcrum pressurized radiograph to predict spinal flexibility in adolescent idiopathic scoliosis (AIS). Methods 1. Empirical study: put two points (A and B) on one side of the plastic stick and one point on the other side ( C), pressurize on the plastic stick when point C is in the middle of A and B, and then measure the projection shifting of point A. Repeat the pressure test when the distance between A and C doubled. Analyze the effect of increasing distance on diminishing pressure. 2.Clinical study :45 patients ( 16 males and 29 females) underwent standing anteroposterior radiograph ,fulcrum lateral flexion radiograph, supine lateral flexion radiograph. The Cobb's angles were measured and the flexibility ratio was determined on preoperative fulcrum radiograph. Results (1) If the length of A and C was 25 cm, pressurize on point C, when pressure quantitative scale number located 1 scale and 2 scale of the rule, projection shifting of point A were 5 cm and 10 cm. If the length of A and C was 50 cm, pressurize on point C, when pressure quantitative scale number located 1 scale and 2 scale of the rule, projection shifting of point A were 8 cm and 15 cm. The longer the distance, the lesser the pressure force of plastics stick. (2)The Cobb angles and curve flexibility provided by fulcrum lateral flexion radiograph had significant difference from that provided by supine lateral flexion radiograph in male group and female group. Cobb angles of male group was 28. 6°± 4. 1 °, 16. 7°± 4. 6° respectively (t= 7. 438, P < 0. 01 )and curve flexibility was 47. 6% ±8. 1% ,69. 4% ± 8. 5% respectively( t = 7. 438, P < 0. 01 ). Cobb angles of female group was 24.5°± 2. 7°,12. 6°±2. 4° respectively(t = 17. 540,P <0. 01 )and curve flexibility was 53.4% ±5.2% ,76. 0% ±4. 7% respectively(t = 17. 54, P < 0. 01 ). The flexibility of spinal main curve of female patients was obviously higher than male patients. Conclusions Supine fulcrum pressurized technique can make up for the insufficiency of supine lateral flexion technique, and provide a convenient, safe, precise and useful method for evaluation of spinal flexibility in AIS.

Both dosage and volume of local anesthetics taken for extradural anesthesia in ESWL were studied in 112 cases. The results showed that small dose of coxylacaine (1.33% of xylocaine and 0.16% Bupivacaine in 3-5ml) given extradurally is not only safe but also suitable for ESWL without obvious side effect
7.Drug resistance characteristics and epidemic trend of Escherichia coli in Anning, Yunnan

Objective To investigate the drug resistance characteristics and epidemic trend of Escherichia coli in Anning, Yunnan, and to provide a reference for clinical rational use of antibiotics. Methods A total of 376 strains of Escherichia coli were isolated from June 2018 to June 2019 from Kungang Hospital of Yunnan Province. The ESBLs producing strains were screened by double disk method and the sensitivity of ESBLs to antibiotics was detected by K-B method. The genotyping of ESBLs strains and the class I, II and III integrase genes in integron 3' and 5' conservative regions was conducted using polymerase chain reaction (PCR). Results The detection rate of ESBLs producing Escherichia coli was 53.19% (200/376). ESBLs-producing Escherichia coli had a low resistance rate to carbapenem antibacterial drugs with a relatively high sensitivity. The resistance rates to amikacin and piperacillin were 7.50% and 12.50% respectively, which had good antibacterial activity. The resistance rate to other antibiotics was high. There were 74.00% (148/ 200) of ESBLs-producing Escherichia coli carrying CTX-M gene, 34.50% (69/200) carrying TEM genes, and 1.00% (2/200) of strains carrying SHV genes. In addition, there were 9.00% (18/200) of strains carrying both CTX-M and TEM genes, and 0.50% (1/200) of strains carrying both CTX-M and SHV genes. The detection rate of integrons accounted for 37.00% (74/200), all of which were class I integrons. Conclusion The prevalence of ESBLs-producing Escherichia coli was relatively high in Anning, Yunnan. ESBLs-producing Escherichia coli showed a trend of multi-drug resistance. The genotype was mainly CTX-M, and the integron was classified as class I.

Objective:
To detect the expression of nuclear transcription factor OCT-4 in laryngeal squamous cell carcinoma (LSCC) and its relationship with CD44, and to explore the relationship between OCT-4 and the cancer stem cells and its significance in tumors.
Methods:
Sixty-nine specimens of laryngeal surgical resection were selected from LSCC patients from January 2008 to December 2012 in Lanzhou General Hospital of Lanzhou Military Region. Another 18 cases of normal laryngeal mucosa tissues were selected as control. OCT-4, CD44 and OCT-4+ CD44+ cells were observed by using tissue microarray, immunohistochemistry staining and immunohistochemical double staining techniques, and the relationship between their pathological features, such as expression intensity, distribution, cell morphology and clinical parameters were analyzed to explore the clinicopathological significance of CD44+ OCT-4+ cells.
Results:
The percentage of CD44-positive cells in LSCC was significantly higher than that in normal laryngeal mucosa tissues [(63.62±8.76)% vs. (13.61±5.81)%,

10.Influence of antiviral therapy with telbivudine on serum soluble complement receptor type 1 in patients with chronic hepatitis B or liver cirrhosis

ObjectiveTo investigate the changes in serum soluble complement receptor type 1 (sCR1) at 3 and 6 months after antiviral therapy with telbivudine in patients with chronic hepatitis B (CHB) or liver cirrhosis and the influence of telbivudine on serum sCR1 level. MethodsA total of 57 patients with HBeAg-positive CHB or liver cirrhosis were enrolled and given the antiviral therapy with telbivudine. Venous blood was collected before treatment and at 3 and 6 months after treatment, and enzyme-linked immunosorbent assay was used to measure serum sCR1 level. The paired t-test was used for comparison within one group. ResultsAt 3 and 6 months after the antiviral treatment with telbivudine, the serum sCR1 level changed significantly compared with corresponding baseline values (t=4.864 and 6.238, both P<0.05). The patients with CHB or liver cirrhosis showed significant changes in the serum sCR1 level at 3 and 6 months after treatment compared with corresponding baseline values (t=3425,5468,4047,7378 all P<0.05). The patients with CHB had a lower serum sCR1 level at baseline and at 3 and 6 months after treatment than those with liver cirrhosis, but the serum sCR1 level at each time point showed no significant differences between the two groups (all P>005). ConclusionIn patients with HBeAg-positive CHB or liver cirrhosis, serum sCR1 level is reduced significantly after antiviral therapy with telbivudine.
