2.Effect of water temperature, stocking density and feeding cycle on growth of juvenile Poecilobdella manillensis.

The effect of water temperature, stocking density and feeding cycle on the growth of Poecilobdella manillensis juvenile was conducted P. manillensis was conducted respectively under different conditions: water temperatures(18, 22, 26, 30,34, 38 degrees C and CT), stocking density (75, 125, 200, 275, 350 individual/L) and feeding cycle(2, 4, 6, 8, 12, 16 d). After 30 days, survival rate, weight gain rate, specific growth rate were measured. There was a significant correlation between water temperature and specific growth rate (γ = -0.066x2 + 3.543 1x -38.09, R2 = 0.837 9). Based on the regression equation, the specific growth rate of P. manillensis achieved the maximum (9.461 4) at 26.84 degrees C. And the most optimal water temperature was 26-30 degrees C. Meanwhile, the survival rates of P. manillensis was 0 at 38 degrees C in 3 d. There was significant negative correlation between density and specific growth rate (γ = -0.005 7x + 9.197 3, R2 = 0.998 3) and between feeding cycle and specific growth rate (γ = -0.468 2x + 10.574, R2 = 0.998 8).

3.Reasonable and safe application of Shuxuetong injection and intravenous medication's combined application in acute cerebral infarction's therapy.

Shuxuetong injection is a kind of compound injection which is made from traditional Chinese medicine Hirudo and Pheretime, which has a clear anticoagulant, fibrinolytic promoting, blood rheology improving, blood lipids regulating and cell protecting effect, and the injection has been widely used in clinical. Especially, the injection has often been combined with other Chinese and modern medicine in the treatment of cerebral infarction disease. However, there are still many non-standard and irrational aspects in clinical practice so as to make a more reasonable and safer use of Shuxuetong injection. In order to avoid the occurrence of adverse reactions to provide a reference for regulating the use of the injection,the paper systematically expounds the Shuxuetong injection's main clinical problems and the reasonable combination.

4.Relevance between quality craft and safe medication of Shuxuetong injection.

Shuxuetong (SXT) injection is the first animal original injection in traditional Chinese medicine, the main component are the leech and the earthworm. SXT has got five national invention patents, and is recognized as the most potent medicine expelling blood stasis agent, which indication is the only one be clearly approved by SFDA as the acute phase of cerebral infarction. Modern biological extraction technology is adopted to prepare SXT, the entire production process using only saline as a solvent. Patented product process is induced to maximize the retention of medicinal components and activity, as well as to remove invalid substances as variant protein,high molecular weight substances which causes allergic reactions. SXT have been isolated and identified class 7 of56 compounds, the molecular weight is from 100 to 1 700 Da, mainly including peptides, glycopeptides, endogenous small molecules.

5.Effect compound decoction on notoginsenosides in Panax notoginseng.

OBJECTIVETo explore the effect of compound decoction on notoginsenosides in Panax notoginseng.
METHODNotoginsenoside R1, Rg1, Re, Rb1 and pH were used as the parameters to investigate the changes on the content of notoginsenosides in different compound extractions by heating for two hours and their correlation with pH.
RESULTWhen the pH values of solution of P. notoginseng with Fructus ligustri, P. notoginseng with Eupolyphaga seu steleophaga, P. notoginseng with Pheretima asiatica, and Zhitangjiang Fang (free of Hirudo) were rept higher than 5.7, the reserved rate (RR) of notoginsenside were higher than 90%; When the pH values of decoetion of P. notoginseng with Salvia miltiorrhiza, P. notoginseng with Paeonia lactiflora, P. notoginseng with Platycodon grandiflorum, P. notoginseng with Arctium lappa were kept 4.5-5.5, their RR of notoginsenside were 60% - 85%; When the pH values of the decotction of P. notoginseng with Hirudo nipponica was decreased to 3.4, its RR of of notoginsenside was 38.4%; When the pH values of Zhitangjiang Fang extraction was regulated by 0.1% NaOH solution to pH 6. 3, and the RR of notoginsenside increased to 97%.
CONCLUSIONThe pH of other Chinese herbal medicines extraction with P. notoginseng compound is a critical effect on the stability and yields of notoginsensides.

6.Study in the mechanism of Shuxuetong injection on antithrombosis and thrombolysis.

OBJECTIVETo study the effect of Shuxuetong injection on cerebral tissue and brain microvascular endothelial cell(BMEC) in secreting tissue-type plasminogen activator (tPA).
METHODThe experiment in vivo was designed for three groups: sham-operation group, focal cerebral ischemia model group, Shuxuetong injection-treated group. The gene expression and the activity of tPA on brains were detected by RT-PCR and chromogenic substrate analysis respectively after treatment for five days; The experiment in vitro: The cultured BMEC were divided into experimental group and control group. The Shuxuetong injection were added into experimental group and the BMECs in the experimatal groups were treated for 24 h. The gene expression and the activity of tPA on cultured BMEC were detected by RT-PCR and chromogenic substrate analysis respectively.
RESULTThe gene expression and the content of tPA on both cerebral tissue and BMEC in the experimatal groups were significantly higher than that in the control groups.
CONCLUSIONShuxuetong injection could accelerate secretion of tPA, and therefore enhance fibrinolysis.

7.Bacterial Identification in the Medicinal Leech.

The use of medicinal leech(Hirudo medicinalis) has been increased during the past years and is a very common procedure for treatment of venous congestion in flaps and replanted part in plastic surgery. One of the main complication in using leeches is infection. We used the medicinal leeches in venous congestion after total ear reconstruction(8 cases) and replanted finger(1 case). A bacteriologic study of the leeches is undertaken before applying it to the patient and an antibiotic susceptibility test was also performed. In 6 patients. 6 pathogenic organisms were identified, and in 2 cases 2 different organisms were identified simultaneously.

Study on some pharmacological effects of earthworm in experimental methods, aiming at treating hypertension. Study results denoted that earthworm solution 2%, 4% all have vasodilator effect and inhibit the constriction of small intestine. In the study on dogs, earthworm solution at the dose 30g/Kg of body weight, 60 minutes after drinking, the tension began down and diminishing arterial tension continuously during 6 hours.
