1.The early stage of operative treatment for traumatic floating knee

Objective To discuss the opportunity of operation and selective in the early stage of treatment method. Methods To analyze the clinical data of 31 cases of patient traumatic floating knee since June 1999 to Au-gust 2006 at my hospital. The results of operative treatment and clinical condition were reviewed. Results In this group there were wound infection 5 cases,amputation 1 case,two stage plant the bone technique and inner fiction 2 cases. After following up patients 1 to 3 years(average 25 months). All of the patients were recovered. According to kerstin at working out standard the limbs function of floating knee damage was evaluated so as to obtain the good percentage of 87 % of the knee joint function. Conclusion A.s usually, the complication of multiple organs injury in floating knee damage was most likely to occur if there had been position priority disposed of important internal organs damage and shock strives in 24 hours properly. The fixed thighbone and shinbone fracture with the emergency dis-posal of important blood vessel damage can reduce the amputation rate.
2.Echocardiographic evaluation of epicardial fat thickness in coronary artery disease

Objective To evaluate epicardial fat thickness change in coronary artery disease patients by using echocardiography.Methods 36 patients with coronary artery disease convinced by coronary angiography and 36 volunteer underwent transthoracic echocardiographic examination.Besides conventional parameters of heart,epicardial fat thickness on right ventricle were measured.Results Subjects with coronary artery disease showed higher epicardial fat thickness than volunteer subjects with predominant peripheral fat distribution:(9.97?2.88)mm vs (4.34?1.98)mm(P=0.005) and (7.19?2.74)mm vs (3.43?1.64)mm(P=0.004) in men and women,respectively.Conclusion Transthoracic echocardiography provides a new convient method for evaluating epicardial fat thickness prediction in coronary artery disease.
3.Studies on Renal Histological Parameters in SD Rats and Beagles

[Objective] To investigate the renal histological parameters in SD rats and beagles and to supply evidence for renal toxicological and pathological diagnosis. [Methods] Forty SD adult rats and 20 adult beagles (half males and half females) , were respectively executed at 12 weeks and 12 months old. Then, kidney tissue was fixed in 10% neutral formalin, embedded in paraffin, made into slices at 5?m and stained by HE. The larger diameter and shorter diameter of glomerulus in the renal cortex and outer stripe of rats and beagles were determined by Spot Advanced image analysis software. The differences of the above parameters appearing in different species and genders were compared. [Results] The larger diameter was about 96 ?m, and the shorter diameter 76 ?m in the renal cortex of the beagles, and the larger was about 120?m and the shorter 94 ?m in the outer stripe of the beagles. The differences of the larger and shorter diameters were insignificant between the male and female beagles, but the larger and shorter diameters in the outer stripe were longer than those in the cortex. The larger diameter and shorter diameter were about 87 ?m and 64 ?m respectively in the renal cortex of SD rats, and the larger was about 88 ?m and the shorter 64 ?m in the outer stripe of SD rats. The differences of the larger and shorter diameters were insignificant between the male and female beagles, as well as in the outer stripe and the cortex. [Conclusion] The nephron in the outer stripe is larger than that in the cortex of beagles but no difference of the nephron exists in SD rats. The nephron in beagles is larger than that in SD rats, and there is no difference in the nephron between male and female beagles and SD rats.

Objective To discuss the early stage treatment of reduction and the curative effect in coxa dislo- cation with acetabular fracture. Methods According to the different type of coxa dislocation with aeetsbular frac- ture,22 patients were given the distinctive operation and drawn reduction. As to 22 cases combination with acetabu- lar fracture and basis of difference type of the fracture were selected the method of suitable operative entrance or in- ner fixation. In this gronp,22 patients were followed up about 6 month to 3 years(average 15.3 months) after opera- tion. According to the standard of America orthopedic surgery study were commented on curative effect. Results Excellent 15 cases,well 4 casea,pass 2 cases,bad 1 case. Total fine rate were 86% .Conclusion Each kinds type of the hip dislocation cases should needed emergency reduction. The operation treatments were selected first method to settlement the bone shatter displacement in acetabular fracture or unsuitable joint and unsteady joint.

6.Experimental Study of Yougui Pills on Related Factors of SREBP Pathway in Kidney Yang Deficiency and Hyperlipidemia Rats

Objective To observe the effects of Yougui Pills (YP) intervention on SREBP pathway related factors of sterol regulatory element binding protein-1c ( SREBP-1c) , cholesteryl ester transfer protein ( CETP) and fatty acid synthase ( FAS) protein and gene expression in kidney yang deficiency hyperlipidemia rat model. Methods Sixty SD rats were divided into normal control group, model group (in the dosage of 2.43 g/kg), and YP group. Rat model of kidney yang deficiency hyperlipidemia was induced with intramuscular injection of hydrocortisone. Normal control group and model group were given normal saline, and YP group was given YP suspension intragastrically. After treatment, the hepatic pathomorphology of rats in the three groups was examined, and the contents of serum lipids were examined. Hepatic SREBP-1c, CETP and FAS protein expression was detected by Western-blotting method, and their mRNA expression was detected by real-time polymerase chain reaction ( RT-PCR) . Results Compared with the normal control group, serum triglyceride ( TG ) , total cholesterol ( TC ) , low-density lipoprotein cholesterol ( LDL-C ) contents were significantly increased (P<0.01), high-density lipoprotein cholesterol (HDL-C) was significantly decreased (P<0.01), and hepatic pathomorphological changes of hyperlipoidemia were obvious in the model group. Compared with the model group, TG, TC, LDL-C contents were significantly decreased ( P<0.01) , and HDL-C was significantly increased (P<0.01) in YP group. Model group had higher SREBP-1c, CETP and FAS mRNA and protein expression than the normal control group ( P<0.05) , while YP group had lower SREBP-1c, CETP and FAS mRNA and protein expression than the model group ( P<0.05) . Conclusion YP can decrease the blood lipid levels probably by down-regulating the expression levels of SREBP-1c, CETP and FAS gene and protein related to the the SREBP pathway in rats of kidney yang deficiency hyperlipidemia induced by intramuscular injection of hydrocortisone.
7.Morphometric Analysis of Liver Tissue in SD Rats and Beagle Dogs

Objective To investigate the parameters of liver tissue in SD rats and beagle dogs and to supply evidence for toxicologic pathological evaluation.Methods Forty SD rats and twenty beagle dogs,male in half,were used for the observation.SD rats were sacrificed at the age of 12 weeks and the beagles at the age of 12 months.After complete necropsy examination,all livers were fixed in 10 % neutral buffered formalin solution,embedded in paraffin,sliced at a thickness of 5 ? m and stained with hematoxylin and eosin(HE).The distance of central vein and portal area,and that of central veins in the neighbored lobules were measured by means of an image analyzer Spot Advanced.The difference of the above distance between beagles and rats,and that between male and female were compared.Results The distance of central veins in the neighbored lobules was about 883 ? m in the beagles and about 496 ? m in SD rats,and the distance between lobule center and portal area was about 511 ? m in beagle dogs and about 410 ? m in SD rats.There was no difference between the male and the female.Conclusion The distance of central veins in the neighbored lobules in beagle dogs is longer than that in SD rats,so does the distance between lobule center and portal area.The sex of the animal has no influnce on the size of the lobules.
8.Expression and clinical significance of Foxp3 in 84 cases of human endometrial carcinoma

Purpose To investigate the expression and clinical significance of Foxp3 ( cell surface marker of regulatroy T) mRNA and its protein in endometrial carcinomas and normal endometrial tissues. Methods Real-time fluorescence quantitative PCR and immuno-histochemical SP methods were used to detect the expressions of mRNA and protein in tumor tissue of 84 cases of endometrial carcino-mas and 40 cases of normal endometrial tissue, then to analyze the relationship between Foxp3 gene and clinical pathological character-istics of endometrial carcinoma specimens, such as differentiation, FIGO stage. Results Foxp3 mRNA and it′s protein expression of endometrial carcinomas were significantly higher than that of normal endometrial tissues. There were significantly relationships between Foxp3 mRNA expression and FIGO stage of endometrial cancer, Foxp3 mRNA expressions of III+IV stage was higher than that ofⅠ+Ⅱ stage endometrial carcinoma (P<0. 05). But the relationship between Foxp3 expression and differentiation degree reached differ-ent conclusions in the two detection methods. By immunohistochemistry the expression of Foxp3 protein was correlation with histological differentiation grade (rs =0. 72, P <0. 01). In poorly differentiated endometrial carcinoma Foxp3 + cell number was significantly higher than that in well differentiated endometrial carcinoma. By detection of real-time fluorescence quantitative PCR method, Foxp3 mRNA expression was not correlated with tumor grade (rs =0. 01, P=0. 35). Conclusion Foxp3 in endometrial carcinomas are high expressions. Immunohistochemical method has more clinical value than real-time fluorescence quantitative PCR test results. Foxp3 may be involved in the regulation of the development of endometrial cancers.
9.Inhibitory effects of LDL-ACM complex on subcutaneous implanted tumors in nude mice

AIM: In order to evaluate the applicable value of LDL as a targeted vehicle for chemotherapeutic agents, we investigated and compared the inhibitory effects of LDL-ACM complex and free ACM on nude mice's subcutaneous implanted tumors derived from gastric cancer cell lines, SGC-7901 and NKM-45. METHODS: LDL-ACM complex was prepared and the tumor model of nude mice was established by subcutaneous implantation of SGC-7901 and NKM-45. Then, the groups of nude mice developed subcutaneous implanted tumors were received either LDL-ACM complex or free ACM. Subsequently, the tumor size, weight and leukemia cell counts were measured and the rates of tumor-inhibition and the survival were compared among the groups. RESULTS: The inhibitory effects of LDL-ACM complex on the tumors, especially on SGC-7901 implanted tumors were much more obvious than that of free ACM. It was also indicated that the action of LDL-ACM complex was mediated by LDL receptor. CONCLUSION: These results showed that LDL-ACM complex had significant inhibitory effects on the implanted tumors and the effect might be mediated by LDL receptor.
10.Association of clinical features with mitochondrial DNA 3243 A to G mutation heteroplasmy levels in patients with maternally inherited diabetes and deafness

Objective To summarize the clinical phenotype profiles and mitochondrial DNA mutation in maternally inherited diabetes and deafness ( MIDD ) , and to improve the diagnosis and treatment of this disease in clinical practice. Methods Sixteen patients with MIDD in six families from Peking Union Medical College from 2007 to Dec 2014 were confirmed as carrying the mitochondrial ( mt) DNA 3243 A to G mutation. Sanger sequencing was used to detect the mt DNA 3243 A to G mutation. The peak height G/A ratio was used to determine mutation heteroplasmy levels. Results The patients with early onset of diabetes (35. 0 ± 14. 6 years), deafness, normal or lower body mass index ( BMI) , and maternal hereditary tendency suggested the diagnosis of MIDD. The peak height G/A ratio was significantly different according to the onset age of MIDD [≤25 years (61. 6 ± 20. 17)%;25-45 years (16.59±8.64)%;>45 years(6.37±0.59)%;P<0.01]. The peak height G/A ratio was negatively correlated with the onset age of MIDD(r=-0. 785,P=0. 001). Conclusion Early onset of diabetes with deafness, normal/lower BMI, and maternal hereditary tendency strongly suggests the diagnosis of MIDD. The peak height G/A ratio might provide a simple prediction regarding the onset age and severity of MIDD.
