1.Clinical value of improved lateral lymph node dissection for stageⅢ lower rectal cancer

Background and purpose:The extent of lymph node dissection for the stageⅢ lower rectal cancer is still a subject of debate. Some Japanese researchers recommend improved lateral lymph node dissection for stageⅢ lower rectal cancers while American scholars claim that total mesorectal excision is sufficient. This study aimed to explore the clinical significance of improved lateral lymph node dissection for stageⅢ lower rectal cancer in patients treated with radical resection.Methods:Sixty-six patients with stageⅢ lower rectal cancer were enrolled. Among these patients, 31 had been treated with radical resection combined with improved lateral lymph node dissection, whereas the others received radical resection without improved lateral lymph node dissection.Results:In the group of improved lateral lymph node dissection, five patients had positive lateral node including four poorly differentiated adenocarcinoma and one mucinous cell carcinoma. Compared with the group without improved lateral lymph node dissection, the group of improved lateral lymph node dissection showed significant difference in sexual disturbance, dysuresia and operation duration (P<0.05), but not in the presence of anastomotic fistula and blood loss during operation (P>0.05). Furthermore, patients had lower rate of pelvis recurrence and better 5-year rate of survival for the group of improved lateral lymph node dissection (P<0.05).Conclusion:Radical resection with improved lateral lymph node dissection may decrease the pelvis recurrence rate and increase survival rate in patients with stageⅢ lower rectal carcinoma.

Objective:To image workstations in the necessity of desktop computer and data lines are connected and fixed in the desktop computer of defects, design a set of wireless transmission of picture archiving and communication system.Methods: Notebook computer with a wireless network using a wireless router, switch and set up a set of wireless image text transmission equipment.Results:The wireless transmission of honeycomb like structure covered with 802.11n standard data rates up to 600Mbps, frequency of 5Hz, multiple security protection by the hospital image filing requirements, which makes medical information resources to achieve full sharing. Conclusion: In 802.11n Wireless LAN PACS system architecture, the basic realization of the wireless transmission and real-time image display, image data for clinicians can within the hospital patients with real time view and its important role in the future development of hospital.

In order to investigate the role of ?-endorphin in the regulation of immune function,we ob-served in vitro and in vivo effects of ?-endorphinon on mitogen--induced spleen lymphocyte pro-liferation,interleukin—1 and interleukin—2 production,It was found that ?-endorphin at 10~(-8)-10~(-14)mol/L could enhance Con A induced proliferation and interleukin-2 production of spleniclymphocyte in vitro.B—endprphin couled also increase LPS induced interleukin—1 productionby peritoneal macropages.The augmentative effect of ?-endorphin on lymphocyte proliferationand interleukin—2 production was abrogate by naloxone.Intravenous injection of ?—endorphinalso increased the lymphoproliferation,interleukin—1 and interleukin—2 production.Our resultsindicated that ?-endorphin enhanced immune function of rats and this action of ?-endorphin maybemediated by opioid receptor.
6.Quantitative Determination of Octanoate Content in Albumin Product By Acid Base Titration

The quantitative determination of octanoate content in albumin product was performed by acid base titration directly after the water phase was mixed up with the ether phase using the mixture solvent solution. The three factor optimum seeking principle was used to seek the optimum concentrations and volumes of components in mixture solvent solution and the optimum conditions of extraction. The method had its recovery rate of 99. 8+0. 89%(n=6) and variation coeff-cient (CV)of 0. 89%(n= 16),indicating that the method was a precise and reliable one.
7.Primary study of the inhibitive effect of naphthalene acetic acid on K562 cell proliferation

Objective To explore the effect of naphthalene acetic acid(NAA) on animal cells. Methods K562 cells were treated in vitro with different doses of NAA (0, 50 and 100?mol/L). The cell proliferation, cell cycle, cell division index and apoptosis was observed by means of MTT, cell enumeration, and flow cytometry. The sister chromatid exchange (SCE) and micronucleus were tested. Results The proliferation of K562 cell was inhibited by NAA compared with that of the controls; the cell cycle was arrested at G1, and the cell division index reduced; apoptosis cells increased lightly in NAA experiments; the frequency of SCE and micronuclear formation were increased by NAA treatment. Conclusion NAA has antiproliferative activities on K562 cell.
8.PARTIAL PURIFICATION OF LEUKOREGULIN AND LYMPHOTOXIN PRODUCED BY PHYTOHEMAGGLUTiNIN AND DITERPENE ESTER ACTIVATED HUMAN SPLEEN CELLS

In order to provide highly purified preparation of human ieukoregulin (LR) for the exploration of its anticancer mschatiism, crude preparations of LR and lymphotoxin(LT) derived from human spleen cell cultures induced with phytohem-agglutinin and diterp;ne ester l2-O-tetradecanoylphorbol-i3-acetate were purified by using four purification nmhois : (a) Con A-Sepharose 48 affinity chromatography, by which crude human LR or LT was purified to maximum specific activity of 85 333 U/mg protein or 13333 U/mg protein with a recovery of 28.1% or 31.9%, respectively 5 (b) DEAE-Sjphadex A-50 ion-exchange chromatography, by which partially purified LR or LT obtained after Con A-Sepharose 4B affinity chromatography was further purified 2.44-fold or 3.8o-fold with a recovery of 96% or 92%, respectively ; (c) DEAE-22-celluiose ion-exchange chromatography, by which crude human LR or LT was purified to maximum specific activity of 11 294 U/mg protein or 13 176 U/mg protein with a recovery of 87.5% or 89.6%.respectively ; (d) Blue-Sepharose CL-6B affinity chromatography, by which crude human LR or LT was purified to maximum specific activity of 51.89 U/mg protein or 2594 U/mg protein with a recovery of 53.75% or 5t3.G7%, respectively.Our data indicate that Con A-Sepharose 4B affinity chromatography, DEAE-Sephadex A-50 ion-exchange chromatography.DEAE-22-cellulose ion-exchange chromatography and Blue-Sepharose CL-6B affinity chromatography may be useful in the purification of LR or LT.

10.INTERRELATIONSHIPS OF HUMAN LEUKOREGULIN WITH LYMPHOTOXIN AND RELATED CYTOTOXINS

The cellular sources of leukoregulin (LR) and lymphotoxin (LT) from human peripheral blood leukocytes or spleen cells were examined The ratios of LT activity to LR activity in lymphokine preparations from different individuals were not constant but varied over 640-fold, suggesting that LR and LT activities were mediated by distinctly different entities.Purified LR was used to examine the relative susceptibilities of human or mice cells to LR.Most human tumor cells were very sensitive to LR while human normal cells and mice normal or tumor cells were less sensitive or resistant.Crude preparations of human interferon (Hu IFN)-? or Hu IFN-? were more cytotoxic to human tumor cells than partially purified natural Hu IFN-? or highly purified rHu IFN-?D, and crude or purified LR were more cytotoxic to human tumor cells than Hu IFN-? Hu IFN-?.Hu IFN-? activity but not cytotoxic activity could be removed from preparatons containing Hu IFN-?,LR and LT activities with the aid of monoclonal antibody to Hu IFN-?. Natural killer cytotoxic factor (NKCF) supernatants from the co-culture of human spleen cells and NK susceptible cell line K562 or Molt-4 were strongly cytotoxic to various tumor cell lines.The NKCF activity was completely adsorbed after incubation with Molt-4 cells and completely inactivated by incubating at pH2 for 24 h.The putative receptor for LR or LT was studied using adsorption assay.The present study reveals that human LR activity can be adsorbed from crude LR supernatants by incubation with native or formalin-treated K562 or SMMC-7721 cells at 37℃ or 4℃; LT activity can be adsorbed with L929 cells.Therefore, LR appears to be a cytotoxic/cytostatic factor that is distinct from LT, 1FN and NKCF.
