1.Effects of seven sevoflurane and propofol anesthesia on heart rate and myocardial enzymes in elderly patients with coronary heart disease

Objective To investigate the effects of seven halothane and propofol anesthesia on heart rate and myocardial enzymes in elderly patients with coronary heart disease (CHD), and to analyze the safety of clinical application of the two kinds of anesthetic drugs.MethodsA total of 70 cases of elderly patients with coronary heart disease treatment January 2012 to December 2014 general anesthesia surgery as the research object, randomly divided into A group and B group were 35 patients in.A group were treated with seven sevoflurane anesthesia, group B patients were given propofol anesthesia, observe the change of the two groups of patients before and after anesthesia hemodynamic index and myocardial enzyme etc.index.ResultsA group of patients with HR 5min after induction and 10min were significantly lower than before the surgery, and the difference was statistically significant (P<0.05);HR stabilized after the induction of 15min and 20min, and compared with before surgery had no significant difference;patients in group B HR and 10min 5min after induction there was no obvious change compared with that before operation, no statistical significance;and after the induction of 15min and 20min HR increased, the difference was statistically significant (P<0.05).A group and B group in patients with systolic and diastolic blood pressure were lower than before anesthesia in varying degrees, and group B more significantly, the difference was statistically significant (P<0.05).A group and B group of patients after 6 h, cTnl and CK-MB in 12h and 24h than 15min before surgery has increased significantly, and A group cTnl and CK-MB were lower than B group, the difference was statistically significant (P<0.05).ConclusionThe clinical application of seven sevoflurane anesthesia for elderly patients with coronary heart disease during propofol is feasible, but seven halothane on blood pressure and heart rate of patients affected with propofol, improve the myocardial enzyme level, and better reduce the damage of perioperative myocardial, more suitable for anesthesia in elderly coronary heart disease.
2.Experimental study on IL-3 gene expression induced by TSPG

Objective:To explore the effect of TSPG (total saponins of panax ginseng) on gene expression of IL-3 which is one of the important hematopoietic growth factor at early stage of hematopoiesis.Methods:Techniques of RT-PCR and in situ hybridization were used.Results:A band corresponding to IL-3 was observed after RNA extraction from TSPG stimulated Jurkat cells were subjected to RT-PCR,while such band was not observed after RNA extraction from unstimulated Jurkat cells were subjected to RT-PCR.Comparing with the control group,both quantity and intensity of IL-3mRNA expression in fetus thymocytes and splenocytes induced by TSPG were obviously promoted.Conclusion:TSPG can induce human lymphocytes to express IL-3 mRNA,thus promote human hematopoiesis at early stage.
3.Study on the preparation of mouse embryonic stem cell feeder layer

Objective:To establish a stable and effective system of mouse embryonic fibroblasts(MEFs) feeder layer in order to separate and culture mouse embryonic stem cells(ESC) in vitro.Methods:the MEFs were isolated from the mouse embryos at 13.5~14.5dpc(days post-coitus) through primary tissue digestion and tissue piece culture.The optional planting density of cell was determined according to the cellular growth curve detected by MTT and the morphology in the culture dish under inverted microscope.The optional concentration of mitomycin C and the time treating with mitomycin on MEFs were determined by MTT assay.Murine blastcysts(3.5dpc) were cultured on the feeder layer treated by mitomycin C and ESC growth was observed.Result:The method of tissue digestion and tissue piece culture is an efficient way to obtain primary MEFs;1?105~2?105/ml cells is an optional planting density;MEFs proliferation could be effeciently repressed after being treated with mitomycin C 10?g/ml for 3.5 hours or 20?g/ml for 2.5 hours,and the quantity of MEFs could maintain at least for 7 days.ES clone could be obtained from the mouse blastcysts(3.5dpc) after being cultured on the feeder layer.Conclusion:MEFs feeder layer could effectively support mouse embryonic stem cells to grow.

5.EXPERIMENTAL STUDY FOR THE EFFECT OF TSPG ON THE EXPRESSION OF IL-3 IN HEMATOPOIETIC STROMAL CELLS

Objective Panax ginseng is a well-known Chinese traditional medicine functional in “invigorating qi”,and saponins are one of its main effective fractions.Our study investigates the effect of total saponins of panax ginseng(TSPG)on human hematopoiesis at early stage and its possible regulative mechanism to clarify the hematonic mechanism of Panax ginseng. Methods The techniques of culturing hematopoietic progenitor cells in vitro,bioassay of hematopoietic growth factor(HGF),immunocytochemistry and nucleic acid in situ hybridization were used to study the effect of TSPG on IL-3’s expression in hematopoietic stromal cell and its possible mechanism. Results TSPG directly added into culture system in vitro can markedly increase the colony forming yields of CFU-Mix;the different conditioned culture media prepared with TSPG can promote the proliferation and differentiation of CFU-Mix;the protein and mRNA expression of IL-3 in BMSC and cells of EcV304,THP induced by TSPG has been much intensified.Conclusion TSPG may regulate hematopoiesis at early stage by activating IL-3's gene expression in hematopoietic stromal cell,which is the most important element in hematopietic inductive microenvironment(HIM).
6.The progress of traditional Chinese medicine treatment of sjogren's syndrome with pulmonary fibrosis

It is including the etiology and pathogenesis, specific prescription and medication, clinical proved recipe, single herb and experimental research to summarize the traditional Chinese medicine treatment of sjogren's syndrome with pulmonary interstitial fibrosis. After reviewing Literatures, the Chinese medicine to the sjogren's syndrome with pulmonary fibrosis has a vast developing prospect and deserves further study.
7.Research progress of anti-VEGF for the therapy in wet age - related macular degeneration

Wet age-related macular degeneration is the leading cause of the severe, irreversible vision loss in individuals over the age of 65 years. Anti-VEGF therapy has been shown to play a key role in the pathogenesis of wAMD, which is the front-line therapy admittedly. lt has no clear steady curative effect for some patients even if they accepted repeating Anti-VEGF therapy. For the purpose of visual acuity improved more, a steady flow of new therapy has emerged, such as function towards the same or different targets of antiangiogenesis, consolidating the effect by combination therapy, improving or simplifying the mode of administration, etc. This paper gives a brief review of the progress of anti-VEGF for the therapy in wet age-related macular degeneration.
8.Formation of cell sheet on acellular porcine corneal stroma with human corneal endothelial cells cocultured by mouse embryonic stem cell conditioned medium

Background Corneal transplantation faces a great challenge because of the shortage of corneal donors and difficulty of human corneal endothelial cells (HCECs) regeneration in vitro.So the study on tissue engineering cornea is still a main topic.Previous research showed that mouse embryonic stem cell conditioned medium (ESC-CM) improved the proliferative capacity of HCECs in vitro,and acellular porcine corneal stroma (APCS) was a good saffold material.However,whether HECEs cultured by mouse ESC-CM can form cell sheet in vitro were rarely studied.Objective This study was to investigate the potential that HCECs cultured by mouse ESC-CM form a monolayer cell sheet.Methods The supernatant of ESC-CM was collected after mouse ES-E14 cells were cultured,and the cultured medium was centrifuged and mixed with 75% human corneal endothelium medium (CEM)at a proportion of 1 ∶ 3 to prepare the 25% ESC-CM system.Primary cultures of HCECs were established from explants of corneal limbal with Descemet's membrane,and the cells were identified by using reverse-transcription PCR to determine the expressions of collagen Ⅷ (Col Ⅷ) mRNA and neuron-specific enolase (NSE) mRNA in the cells.APCS was prepared by decellularization with phospholipase A2 and bicarbonate solution,and the second generation of HCECs were inoculated on the sterilized APCS at a 800/mm2 density.The morphology of the cells was observed by hematoxylin-eosin staining under the phase-contrast microscope.The expressions of zona occludens protein-1 (ZO-1)and Na+-K+-ATPase in the cell sheet were detected by immunofluorescence staining.Results The second generation of HCECs cultured with 25% ESC-CM in vitro showed the hexagon in shape with positive expressions for Col Ⅷ mRNA and NSE mRNA.Decellularization APCS was transparent,and no corneal cells were seen,the structures of corneal collagenous fibres were regular.HCECs attached closely to APCS and formed monolayer sheet 7 days after culture on the APCS with the cell density of (2 694±143)/mm2.ZO-1 and Na+-K+-ATPase were positively expressed on the HCECs sheet.Conclusions Twenty-five percent ESC-CM promotes the proliferation and maintains the normal morphology of HCECs.APCS provide a good scoffold and microenvironment for the formation of HCEC sheet.The HCEC sheet Possesses the pump function of HCECs and is a good corneal donor for transplantation.
9.Training doctors specifically for the suburban and rural areas

The cultivation of medical and health personnel in the rural areas is currently confronted with severe challenges: very few graduating students go and work in the poverty-stricken or mountainous areas and there is a shortage of rural medical and health personnel. An effective mechanism for solving the problem in a short time has been the establishment of medical and health personnel training centers that recruit students specifically from the mountainous and poverty-stricken areas and assign them back there upon graduation. It has promoted the development of unique curriculums designed specifically for the training of personnel specializing in clinical medicine from rural grassroots. The establishment of long-term mechanisms that ensure the support of the development of the rural medical and health cause by urban medical and health human resources strongly guarantees the development of rural in-service education and non-academic training.
10.Therapeutic role on monoclonal antibody of alkaline phosphatase to vasogenic cerebral edema in rats

Objective To observe the therapeutic role on monoclonal antibody of alkaline phosphatase(AAP) to vasogenic cerebral edema in rats(VCE).Methods 60 Wistar rats were randomly divided into 3 groups:normal group,VCE group and AAP group.VCE animal model was made by injecting AAP.Brain water content of gray and white matter were measured by Moistrue Analyzer respectively.The permeability of the blood brain barrier (BBB) was determined by Evan's blue(EB) extravasation.Results The brain water content of gray and white matter in AAP group was reduced markedly,so the permeability of BBB was reduced(P0.05). Conclusion AAP can treat VCE well. Morbidity mechanism of VCE is related to the activity of alkaline phosphatase.
