Osteoblasts (OB) isolated from newborn SD rats were cultured in vitro.After treatment with different concentrations of 17?-estradiol (10~(-11)-10~(-6)mol/L),the mRNA expressions of osteoprotegerin (OPG) and osteoclast differentiation factor (ODF) in OB were measured by RT-PCR.17?-estradiol increased the expression of OPG in OB with the maximal effect at the concentration of 10~(-8) mol/L.No significant difference was observed in the expression of ODF in OB with different concentrations of 17?-estradiol.The therapeutic effect of estrogen on osteoporosis appears to be related to the enhanced OPG expression in OB at physiological concentration of estrogen.

Objective: To investigate the effect of sole and combined utilizing of carriers to prepare solid dispersion for improving dissolution rate of hydrophilic and lipophilic components in extracts of Salviae Miltiorrhizae Radix et Rhizoma (Danshen). Methods: Solid dispersions were prepared with PVP10 and F127 as sole carrier or their combinations by Spray Drying (SD) method. The prepared powders were characterized by various methods. Three hydrophilic (danshensu, rosmarinic acid, and salvianolic acid B) and two lipophilic components (cryptotanshinone and tanshinone IIA) of Danshen were determined as bioactive markers. The simulating factor (f2) was utilized to evaluate similarities of dissolution profiles of hydrophilic and lipophilic components, respectively. Results: SEM and PXRD results showed that solid dispersion prepared by SD method was amorphous spheroid particles without intensive characteristic peaks. The hydrophilic components were completely released from the bulk Danshen extract and from the prepared solid dispersions. However, no lipophilic components could be completely dissolved from solid dispersions prepared by sole carrier. When combined using carriers (e.g. PVP10-F127 = 5:4), both of the lipophilic and hydrophilic components could be rapidly and completely released from the solid dispersion. Combined use of carriers could increase the wetting angle without changing the specific surface area. Conclusion: The combined use of carriers significantly increases the in-vitro release of lipophilic components in Danshen extract, which: may be attributed to the increased wettability of powders. The simulating factor evaluation results suggest that the release profiles of either the lipophilic or the hydrophilic components in Danshen extract are complete and similar, which could meet the requirements of Chinese Pharmacopoeia.

Objective To understand the pathogenetic role and clinical significance of interleukin 6(IL－ 6 ),tumor necrosis factor(TNF) and soluble interleukin－ 2 receptor(sIL－ 2R) in patients with cerebral infarction(CI) .Methods The IL－ 6,TNF and sIL－ 2R levels were measured in 46 CI patients with ELISA. Results The serum IL－ 6,TNF and sIL－ 2R levels of CI group were much higher than those of controls group .The variation of IL－ 6,TNF and sIL－ 2R levels was closely related to the size of cerebral infarction. The serum IL－ 6,TNF and sIL－ 2R levels of recovery period were much lower than those of acute period in patients with CI .Conclusions The IL－ 6 ,TNF and sIL－ 2R take part in the pathologic process after CI .The measurement of IL－ 6, TNF and sIL－ 2R is useful in determining the size of cerebral infarction.

Objective To explore the clinical application of laparoscopic splenectomy in treatment of spleen disease at our hospital.Methods We reviewed laparoscopic splenectomy carried out at our hospital since 1995,patients were grouped by date.63 laparoscopic splenectomies were divided into six groups.Operation time,intraoperative blood loss,postoperative hospital stay,time to feeding,days of drainage,amount of drainage,postoperative complications and indications for surgery were compared.Results Patient's age averaged at 44.19 years,body mass index averaged at 23.75,3 patients were converted to open surgery.Mean operating time,blood loss,postoperative hospital stay,time to feeding,converting rate in the 53 cases which had the surgery after 2003 were much better than the 10 cases before 2003.Surgical indications for laparoscopic splenectomy were limited to hematopoietic disease related splemegaly before 2003,the indication range significantly expanded after 2003,during which laparoscopic splenectomy were mainly applied to treat spleen tumors.Conclusions There is a marked learning curve in laparoscopic splenectomy after 10 surgeries before 2003 we have achieved the level.With the suitable approach,apparatus and skillful technique,laparoscopic splenectomy is safe and feasible to treat tumors of the spleen.

Objective To estimate the present salt iodine content and iodine nutrition need of high risk population of iodine deficiency disorder in Meixian County. Methods Each primary school was selected from urban and rural areas(Xiyang Town, 20 kilometers away from Meixian County), the goiter rate of 8 to 10 year-old students was examined and urinary iodine and household salt iodine was sampled. Twenty to 40 year-old women of childbearing age nearby schools around the urban and villages around Xiyang Town were selected to collect their urine and salt samples. At urban hospitals and rural health centers, 0 to 2 year-old infant urine samples were collected, Thyroid gland was palpated and urinary iodine was determined by iodine in urine by As3+-Ce4+catalytic spectrophotometry, salt iodine was determined by direct titration. Results The goiter rates of 8 to 10 year-old students were 1.5 % (3/200), 1.0% (1/100) for the urban area and 2.0% (2/100) for rural area. Median of urinary iodine in 8 to 10 year-old students, infants, women of childbearing age averaged at 237.1 μg/L and 280.1, 234.7,187.6 μg/L respectively, with each being 287.4,245.0,205.5 μg/L in urban area and 278.9,228.5,176.4 μg/L in rural area. Women of childbearing age had a higher percentage of urinary iodine < 50.0 μg/L than students,students had a higher percentage than infants, each being 7.5%(15/200), 4.5%(9/200), 4.0%(4/100). The ration of urinary iodine > 300.0 μg/L was more in infants than in students, that in students was more than that in women of childbearing age, each being 33.0% (33/100), 30.0% (60/200),22.5% (45/200). The median of salt iodine was 27.2 mg/kg. The coverage of iodized salt was 100.0%(400/400). Ninty-seven percent(194/200) and 96.0% (192/200) of qualified iodized salt were consumed in urban area and in rural area. Conclusions The amount of iodine added to salt meets the requirement in the 3 kinds population risk of iodine deficiency disorder. But a higher iodine status has been found out in students and infants. It is reasonable to decrease the present salt iodine content.

Objective To investigate the meaning of expression of apoptosis related molecules NFKBp65 and p53 and GPX1-mRNA in patients with Keshan disease(KSD).Methods Sixteen chronic Keshan Disease patients were enrolled in KSD group according to electrocardiogram,chest X ray film and clinical examinations on 15,September in 2009,and 23 healthy people were included in control group from physical examination taken in The Second Affiliated Hospital of Xi'an Jiaotong University.Fresh blood(5 ml)was collected from antecubital vein of all subjects in the fasting state.Total mRNA and protein of blood sample were isolated using Trizol.GPX Assay Kit was used to detect GPX enzyme activity,and GPX1-mRNA expression was determined by SYBR Real-Time PCR.Meanwhile,expression of apoptosis related molecules NFKBp65 and p53 were determined by Western blot.Results GPX enzyme activity decreased significantly in KSD group[(108.61±14.10)U]compared with control group[(122.78±11.89)U,t=2.874,P<0.05],GPX1-mRNA level of KSD group(0.553±0.299)notably KSD group(0.802±0.057)compared with control group[(1.065±0.355),t=6.829,P<0.01].p53 increased in KSD group(1.604±0.191)compared with control group[(1.137±0.186),t=3.033,P<0.05].Conclusiom Decreased GPX1-mRNA expression may result in lower GPX enzyme activity of patients with KSD.Thus oxidative damage increases and cadioeyte apoptosis is activated by activating apoptosis signal pathway.

Objective To evaluate the neuroprotective effect and safety of neonatal hypoxic-ischemic encephalopathy(HIE)treated with recombinant human erythropoietin(rhEPO).Methods Fifty-three neonates with HIE were randomly divided into rhEPO treated group(n=29) with the dosage of 300 U/(kg?time),three times a week for 2 weeks and control group(n=24)without rhEPO.All supportive measures were same between 2 groups.Neurological scoring was evaluated at d3,d5 and d7 Neonatal behavioral neurological assessment(NBNA) was evaluated at d7,d14 and d28.The neurodevelopment quote was evaluated at age of 3 and 6 months.Blood pressure,liver and renal function,blood electrolytes and blood hemoglobin,platelet and reticular red blood cell count were monitored before and after treatment in all infants.Results The neurological scoring between two groups had no difference at d3.The significant difference was found at d7(P0.05).Conclusions Teraphy with rhEPO on neonatal HIE infants can promote neurological recovery,and there is no serious side effect with rhEPO treatment.
