1.Utllizition of ELISA in determination of monoclonal antibody affinity constant in ascites

Antibody affinity influences the ant igen-antibody reaction in vivo and in vitro. It is important to routinely determine the antibody affinity in order to elucidate the impacts of this antibody characteristics on the immunoassay and the pathogenetic mechanisms of human diseases. In this paper, a simple method (modified Friguet' s) was reported to directly measure the experimental affinity data of monoclonal antibody in ascites by means of ELISA and to accurately determine the corresponding affinity constants of monoclonal antibody by amendment of underestimation of affinity constants due to the intrinsic properties of Friguet' s, and shows convenience and reliability.
2.Development of Incubators Temperature Network Supervisory System

Objective To develop a secure network system to monitor the temperatures in baby incubators.Methods By utilizing patented TAV temperature monitoring system,the running status of temperature-controlled equipments was recorded in digital way,and then it was analyzed and managed dynamically;and real time alarms could be given through networks.Results This system is a kind of security devices independent of baby incubators and can monitor temperatures in baby incubators well.Conclusion This system will not affect original functions of baby incubators,instead it can well monitor the temperatures in both imported and domestic incubators,so that it can improve the safety of facilities being used while decrease medical accidents.
3.Effect of telmisartan and fosinopril on the expression of angiotensin-con-vertion enzyme 2 in cardiomyocyte after myocardial infarction

Objective:To investigate the effect of telmisartan and fosinopril on the expression of angiotensin-convertion enzyms 2 (ACE2) in cardiomyocyte after myocardial infarction.Methods:Animal model of acute myocardial infarction were established by ligating the left anterior descending coronary artery of rats. Vehicle, telmisartan,fosinopril,or both drugs combined were giv- en to rats by intubation feeding,from 2 weeks before coronary artery ligation to 2 weeks there after. ACE2 protein expression of myocardium were semi-quantified by SP immunohistochemistry. ACE2mRNA were measured by Reverse Transcriptase Poly- merase Chain Reaction (RT-RCR). Results:Compare with sham operated group,ACE2 protein increased significantly in infarc- tion group,but not the ACE2 mRNA. telmisartan and fosinopril therapy caused a significant increase in ACE2 protein compare to infarction group,but only telmisartan caused a significant increase in ACE2 mRNA though all therapied groups showed higher ACE2 mRNA expression. Conclusion:Both telmisartan and fosinopril induced increases in cardiac ACE2 exprssion, whereas the combination of these two drugs do not associated with higher ACE2 expression.

5.Estrogen, anti-estrogen drugs, and thyroid cancer

Several researches have suggested that estrogen contributes to the initiation and development of thyroid cancer by binding to estrogen receptors (estrogen receptor α,estrogen receptor β,and G-protein-coupled receptor 30),activating gene or non-genomic pathways and regulating proliferation of thyroid cells.Studies on antiestrogen drugs based on inhibiting thyroid cell proliferation may provide a new target in treating thyroid cancer.

7.Coaxial double catheter technique in intracranial posterior circulation stenting

BACKGROUND: The posterior circulation artery is characterized by small caliber, long traveling, many curves and branches, so a stronger resistance will be encountered when a stent is delivered.OBJECTIVE: To investigate the effects of coaxial double catheter technique on intracranial posterior circulation stenting.DESIGN, TIME AND SETTING: A retrospective case analysis was performed at the Department of Neurology, Affiliated People's Hospital of Yunyang Medical College between December 2004 and February 2008.PARTICIPANTS: A total of 21 patients, 11 males and 10 females, aged 50-76 years old, were se/ected from the Department of Neurology, Affiliated People's Hospital of Yunyang Medical College.METHODS: All patients were confirmed as posterior circulation intracranial stenosis (stenosis > 70%) by digital subtraction angiography (DSA), magnetic resonance angiography (MRA), or CT angiography (CTA). They underwent cerebrovascular stent implantation, during which, single catheter technique was first applied, if failed, a coaxial double catheter technique was followed. A conventional group (single catheter technique successfully applied) and a double catheter group (coaxial double catheter technique successfully employed) were set. A written informed consent was obtained from each patient.MAIN OUTCOME MEASURES: Therapeutic results and biocompatibility of implanted stents.RESULTS: The single catheter technique was applied in 21 patients and 6 of them (28.57%) had successful stent deployment, The remaining 15 patients were managed by a coaxial double catheter technique, and the stent was successfully released in these patients. There was only 1 patient presenting with subarachnoid hemorrhage, and the success rate of stent deployment was 93.33%. Chi-square tests results showed that the coaxial double catheter technique produced significantly higher success rate of stent implantation than the single catheter technique (P < 0.05). Except this, no other adverse reactions were found. All stents covered the stenosis lesions completely, and fitted well with the blood vessel wall without rejection, showing the good biocompatibility of selected stents with the host.CONCLUSION: The coaxial double catheter technique can increase the longitudinal supporting strength and enhance the success rate of the intracranial posterior circulation stenting. Meanwhile, the risks of this procedure are low. Therefore, the technique deserves to be popularized.
8.Application and characteristics of self-assembling peptide nanofiber hydrogel in three-dimensional cell culture systems

BACKGROUND:Self-assembling peptide scaffolds have attracted much more attention among three dimensional biomaterials due to its biocompatibility, biodegradation, and tailor-made properties.
OBJECTIVE:To review the structure and design of functionalized self-assembling RADA peptide, and the recent advances in the use of RADA self-assembling peptide for three-dimensional cel culture in cel therapy applications.
METHODS:Total 224 literatures related to self-assembling peptide and tissue engineering from PubMed, Web of Science and CNKI databases were screened out for this review. The keywords were“self-assembly peptide, tissue engineering”in English and Chinese, respectively. Final y 48 of 224 articles about the design, fictionalization, and three-dimensional cel culture of peptide scaffolds were included in this review.
RESULTS AND CONCLUSION:Self-assembling peptide could undergo spontaneous assembly into wel-ordered interwoven nanofibers in water and rapidly form hydrogel, which physical y mimics the architecture of natural extracel ular matrix to ensure a real three-dimensional microenvironment for cel s. In terms of bio-function, this material can be tailor-made with various bioactive short peptide motifs to promote cel adhesion, proliferation, and differentiation.
9.Clinical features and aCGH analysis of one child with Phelan-McDermid syndrome

Objective To analyze the clinical features and results of array-comparative genomic hybridization (array CGH, aCGH) in a Chinese girl with Phelan-McDermid syndrome. Methods The clinical symptoms of a child with Phelan-Mc-Dermid syndrome were retrospectively analyzed. Routine G-banding was performed to analyze the karyotype, and the aCGH was used to analyze subchromosomal abnormalities. Results The routine karyotype analysis showed a normal female karyotype without abnomalities in chromosome number and structure. aCGH analysis finely mapped the deletion of Chr22q13.2-qter. Phel-an-McDermid syndrome was diagnosed for this case. Conclusions Phelan-McDermid syndrome can be diagnosed by the typi-cal and detailed clinical features in combination with the laboratory examinations of subchromosomal abnormalities. aCGH is one of the most valuable methods to analyze subchromosomal abnormalities and to diagnose Phelan-McDermid syndrome.
