Chinese Journal of Emergency Medicine 2009;18(4):367-371
Objective To study the changes of apoptosis in brain after traumatic injury(TBI)treated with exogenous vascular endothelial growth factor(VEGF)gene therapy in oder to find out the role of exogenous VEGF gene in protectiog brain tissue.Method The injurea cerebral cortex from the rat models of brain with traumatic injury was injected with adenovirus(adenovirus,Ad)as the carrier of VEGF-165 gene(Ad-VEGF-165 Gene).RT-PCR and Western blot were used to detect the expression of VEGF mRNA and VEGF protein in the brain 6 h,24 h,3 d,7 d and 14 d after injury, and apoptosis in the injured location of brain was also detected by TUNEL successively after Ad-VECF-165 gene therapy. Results With exogenous Ad-VEGF-165 applied to the locally injured brain tissue after injury,VEGF mRNA and VEGF protein showed consistent expression and their expressions were significantly higher than those in trauma group and vehicle control group.Compared with the trauma group.the apoptosis in the gene therapy group 24 h,3 d and 7 d after injury presented a.significant reduction, and had close relationship with VEGF.(Control group:4.17±0.73;TBI group,24 h:47.18±6.01,3 d:79.44±11.23;TBI+VEGF group.24 h:28.72±5.31,3 d:54.18±7.66;P<0.05).Conclusions The exogenous VEGF gene therapy administered to have protective effects on the local brain tissue in rats with traumatic injury in a certain time.
3.In vivo animal models of cerebral vasospasm following subarachnoid hemorrhage
International Journal of Cerebrovascular Diseases 2010;18(6):477-480
Delayed cerebral vasospasm is the leading cause of death in patients with subarachnoid hemorrhage. A good in vivo animal model of cerebral vasospasm after subarachnoid hemorrhage helps to understand the occurrence, development of this cerebral vasospasm, and the complicated and various pathophysiological mechanisms. It will provide important information for clinical interventions
4.Experimental study on hemodynamics of traumatic brain injury using dynamic CT perfusion image
Journal of Third Military Medical University 2003;0(13):-
Objective To investigate the changes of region cerebral homodynamic and cerebral vascular autoregulation in traumatic brain injury(TBI) by dynamic CT perfusion(CTP) imaging in rat. Methods Forty-five SD rats underwent CTP,including 10 control rats and 35 TBI rats,on 1,6,24 h,and day 3,7,14,21 after TBI.The region of interest(ROI) was located near traumatic area within 2 mm and mirror area of relative brain hemisphere.False color pictures and dynamic rCBF,rCBV,MTT and PS were obtained in different time.Pathological examination was also done. Results The rCBF,rCBV on 1 h after TBI was lower than that of control group, and significantly higher on 6-24 h after TBI.They reached the peak on day 3.PS had a similar change compared with rCBF,rCBV,but a longer duration.MTT showed to moderate changes except an isolated tide on 3 days after TBI.The rCBF on mirror area of relative brain hemisphere was almost stable except 3 days after TBI,which was lower than that of control group.Meanwhile, the rCBV on mirror area of relative brain hemisphere was normal. Conclusion CTP is an advanced technique to investigate the changes of region cerebral homodynamic and cerebral vascular autoregulation in TBI.The early hypoperfusion and next hyperperfusion around traumatic areas after TBI suggest that the dysfunction of cerebral vascular autoregulation should be committed to the change of homodynamic in TBI.
5.Optimization of supercritical fluid extraction of paeoniflorin by response surface methodology
Chinese Traditional Patent Medicine 1992;0(08):-
AIM: To develop a method for optimizing of supercritical fluid extraction (SFE) of paeoniflorin. METHODS: On the basis of one-factor test and the Box-Behnken central-composite design response surface with 3 factors and 3 levels was adopted. A second order quadratic equation for SFE of paeoniflorin was built. Response surface and contour were graphed with the extraction rate of paeoniflorin as the response value. Based on the analysis of the response surface plots and their corresponding contour plots, the effects of extraction pressure, extraction temperature, and methanol concentration were explored. RESULTS: By using the new method discussed above, the optimum extraction condition was obtained as follows: 18. 9 MPa extraction pressure, extraction cosolvent temperature 59. 9 ℃,68. 5% methanol was chosen as cosolvent with the flow rate of 0. 4 mL/min,and extraction time 60 min. CONCLUSION: Under the optimized condition, extraction rate of paeoniflorin on theoretical value is 7. 85 mg/g.
6.Exploration on Management Technological Specification of Medical Equipment
Chinese Medical Equipment Journal 1993;0(05):-
Objective The diagnosis and treatment of modern hospital are greatly influenced by medical devices. With the widespread introduction of medical equipment, the scientific management how to carry on the instrument and appliance by the administrative department of hospital becomes an unavoidable problem. Methods Based on the definition of medical equipment, basic attributes and management skills, some key questions which the administrator should notice and solve in the scientific management are introduced, and gives out the principle and method. Results It is pointed out that scientific management can correctly guide the investment of equipment, and gain great benefit with a minimum input. Conclusion Investment according to the developmental strategy is proposed, which provides strong theoretical basis for leadership and administrative departments.
7.Foundational research of olfactory ensheathing cells transplantation in treating central nervous system disease
Chinese Journal of Tissue Engineering Research 2007;0(36):-
Central nervous system(CNS) disease induces functional deficits in persons.Simultaneously, there appears axonal destruction and the formation of scar tissue,as well as cavity and physical gaps in the lesions.Olfactory ensheathing cells(OECs),acting as prime candidates for autologous transplantation, secrete neurotrophic factors for neuroprotection, angiogenesis and outgrowth of both intact and lesioned axons to different degrees, change the response status of endogenous glia after lesion,as well as remyelinate axons after a range of demyelinating insults.OECs transplantation has become an ongoing clinical therapeutic approach for human CNS disease in recent years. Thus,OECs transplantation can be used for cell-mediated repair following a variety of CNS lesions. Here, the paper reviews the current status of foundational research of OECs transplantation in treating CNS disease underlying the wide variation of results obtained in the field.

10.New progress of myelodysplastic syndromes in the 55th ASH annual meeting
Journal of Leukemia & Lymphoma 2014;23(1):20-21,29
New progress of myelodysplastic syndomes (MDS) in the 55th ASH annual meeting on 5-azacytidine and decitabine was reviewed.Except in older females, the survival time for a diagnosis of MDS in the United States does not seem to have substantially improved since the introduction of 5-azacytidine into the market in 2004.However,clinical and mechanism data suggest that judicious,mechanism-based application and optimization of these agents will have increasingly meaningful impact. Moreover, these drugs are exemplars of an inspiring alternative to the traditional cytotoxic paradigm and should pave the way for additional relatively nontoxic but broadly effective agents.
