4. Interstitial cell of cajal regulates motility of gallbladder smooth muscle in guinea pigs
Academic Journal of Second Military Medical University 2010;31(7):721-724
Objective: To prove that the interstitial cells of cajal (ICCs) are the pacemaker of gallbladder smooth muscle in guinea pigs. Methods: The contraction of gallbladder was observed by isometric transducer and spontaneous electrophysiology was recorded in vivo and in vitro. ICCs were damaged with methylene and illumination. The ICCs were isolated by collagenase digestion and the morphology of single ICC was analyzed. C-kit expression was detected by laser confocal microscopy to identify the isolated ICCs. Spontaneous currents of ICCs were recorded with EPC-10 Patch Clamp. Results: Slow wave and action potential were recorded in gallbladder in vivo, with the frequency of slow wave being (58±3.2) bpm (n=16). Methylene blue and illumination treatment led to decrease or disappearance of slow wave and action potentials. Some cells isolated from gallbladder smooth muscle ([5±1.2]%, n=12) showed the morphological characteristics of ICCs under light microscope, with (21±4)% of the cells were positive for c-kit. When the membrane potential was at -60 mV, the frequency of spontaneous inward currents was (58.4±3.5) bpm (n=21) and the amplitude was -(72±3.5) pA. Conclusion: ICC-like cells are present in the gallbladder smooth muscle of the guinea pigs, and participate in the pacemaking of spontaneous current and action potential of gallbladder smooth muscle.
5. Mesenchymal stem cells: A delivery vehicle for cancer therapy
Academic Journal of Second Military Medical University 2011;32(10):1030-1034
Stem cells possess unlimited self-renewal properties and the ability to produce multiple differentiation progenitors. Mesenchymal stem cells (MSCs) can specifically migrate to multiple kinds of tumors and their metastases in a systemic manner(to all parts of the body). Therefore genetically modified MSCs can serve as vehicle of anticancer factors to inhibit growth of tumor. But many questions remain to be answered and the precise mechanisms remain unclear. This paper reviews the mechanism of MSC homing, MSCs as delivery vectors for cancer therapy and its advantages over traditional drug vehicle. MSCs as delivery vectors can be used for cellular therapy in future cancer treatment.

OBJECTIVE:To systematically review the efficacy and safety of pituitrin combined with phentolamine versus pitui-trin alone in the treatment of bronchiectasis hemoptysis(BH),and provide evidence-based reference for the clinical treatment. METHODS:Retrieved from PubMed,Cochrane Library,EMBase,CJFD,Wanfang and VIP database, the randomized controlled trails(RCT)about pituitrin combined with phentolamine(test group)versus pituitrin alone(control group)in the treatment of BH were collected, and afterdata extract and quality evaluation ,Meta-analysis was performed by using Rev Man 5.2.3 statistics soft-ware. RESULTS:A total of 43 RCTs were included, involving 3 094 patients. Results of Meta-analysis showed the significant effi-cacy [RR=1.50,95%CI(1.38,1.62),P<0.001] and effective rate [RR=1.25,95%CI(1.20,1.30),P<0.001] in test group were sig-nificantly higher than control group, inefficacy [RR=0.29,95%CI(0.23,0.36),P<0.001], hemoptysis stopped or remission time [MD=-2.00,95%CI(-2.43,-1.57),P<0.001],incidence of headache [RR=0.36,95%CI(0.22,0.59),P<0.001],incidence of chest tightness [RR=0.41,95%CI(0.26,0.63),P<0.001],incidence of abdominal pain [RR=0.26,95%CI(0.16,0.43),P<0.001] and incidence of elevated blood pressure [RR=0.28,95%CI(0.14,0.56),P<0.001] in test group were significantly lower than control group, there were significant differences in 2 groups. CONCLUSIONS:Pituitrin combined with phentolamine has better efficacy and safety than pituitrin alone in the treatment of BH. Due to the limit of methodological quality and sample size,it re-mains to be further verified with more rigorously designed and long-term follow-up of large-scale RCT.

Objective To investigate the dynamic changes of metallothionein(MTs)gene expression and explore the important significance of MTs during hepatocarcinogenesis. Methods One hundred and twenty-five SPF 5 -8-week old male C57BL/6J mice were randomly divided into control group and model group.Diethylnitrosamine (DEN) was given to the mice at a dose of 100 mg/kg, ip, and 50 mg/kg, ip, in the first and next week, respectively. The mice were given ethanol (53%, 5 mL/kg/day, 5 days/week) from the third week of experiment till 35 weeks. At 1, 3, 9, 13, 24 and 35 weeks of the experiment, liver samples were taken for histopathological examination of liver damages and incidence of HCC. The liver index and malondialdehyde (MDA) of liver homogenate were determined. All liver tissue samples were examined by histopathology using hematoxylin and eosin (HE), Masson and reticular fiber staining.Real-time RT-PCR was used to analyze the mRNA expression level of liver metallothionein-1 /2 (MT-1 /2) in different periods. Results Progressive liver damages in model group mice were identified in different periods.Hepatocytes abnormal tission and abnormal liver plate structure, architecture often characteristic of HCC could be seen in approximately 50% of mice at 35 weeks. In addition to these, a higher liver index also were seen at 35 weeks.Increased MDA levels in the mouse liver tissues were observed in each stage.Real-time RT-PCR analysis showed that significantly increased transcription of MT-1 and MT-2 at 1, 3 and 9 weeks, then gradually declined and even below the normal level. Conclusions MTs gene expression levels in mouse liver tissues are changed from significantly increased in the early stage of injury to decreased expression combined with distinct fibrosis. Our findings further demonstrate that the down-regulation of MTs level is closely correlated with hepatocarcinogenesis.
