1.Effects of different dose of edible liquor on cardiac structure and function in rats

Objective To investigate the effects of different dose of edible liquor on cardiac structure and function in rats ,and explore its underlying mechanisms .Methods 54 male SD rats were randomly divided into saline group ,alcohol group ,liquor group , each group were further divided into low-,middle-and high-dose groups .After 12 weeks of feeding ,left ventricular pressure was de-termined ,and left ventricular pressure amplitude and indexes of HW /BW were calculated .Myocardial fibrosis was observed by Masson staining ,AngⅡ content of myocardial tissue were detected by ELISA ,and ACE and ACE2 mRNA expressions were detec-ted by RT-PCR ,respectively .Results Left ventricular pressure amplitude and HW/BW index were increased by both liquor and al-cohol .The myocardial cell turbidity ,degeneration ,hyperplasia of fibrous tissue change were gradually worsened ,and the AngⅡ con-tents and the ACEmRNA expressions of the myocardial tissue were also increased by both liquor and alcohol .The ACE2 mRNA ex-pression of the myocardial tissue were higher by low-dose liquor and alcohol ,while the ACE2 mRNA expression of the myocardial tissue were lower by middle-and high-dose liquor and alcohol .Conclusion Long-term alcohol consumption can damage the balance of cardiac ACE and ACE2 ,causing the increase of ACE ,AngⅡ and the decrease of ACE2 ,resulting in myocardial fibrosis ,whose degree is related to the amount of alcohol consumption and drinking history .

Replacement therapy with plasma derived or recombinant F Ⅷ (rFⅧ) concentrates is only effective way to treat hemophilia A.Currently,the first,second and third generation rFⅧ are commercially available.Of the three products,the third generation rFⅧ products are best,because of no additional human or animal plasma proteins at any period.The use of rFⅧ concentrates has greatly improved the safety of replacement therapy in hemophilia A and reduced the risk of blood-borne pathogen transmission.In this article we review the current knowledge about the three rF Ⅷ concentrates,analyzing their main characteristic and their clinical hemostatic efficacy and safety.
3.Characteristics and clinical security of coronary artery drug-eluting stent

Technology and technique of coronary artery stent have been developed to a certain degree, restenosis rate of metal stent reduces to 15%, but coronary artery stent is still difficult for long-term effect of interventional therapy on coronary atherosclerotic heart disease. Drug-eluting stent is newly discovered during recent years, while it is prospect for preventing restenosis postoperatively. Although clinical efficacy of drug-eluting stent is encouraged, indication is still simple. In addition, some events related to drug-eluting stent remain poorly unknown. This study was designed to compare security between drug-eluting stent and metal bared stent, analyze major factors correlated with security of drug-eluting stent, and investigate the development of drug-eluting stent.
4.Preliminary study on change of immunologic factors within cervical microenvironment infected with HPV 16, 58 and integrated state of virus in cervical cells

Objective:To investigate the relationship between changes of the concentration of IL -12,IL-4,IFN-γwithin cervical local immune microenvironment and cervix infected with HPV 16,58,to understand the prevalence of physical status of HPV 16/58 DNA into the host genome in cervical lesions of different grades and to detect the relation between the integrated state of virus and the progress of cervical lesions.Methods: Detecting 89 cases of cervicovaginal lavage fluid and 140 cytological specimens of the cervix which from patients having had HPV-DNA genotyping assays and coming from the Affiliated Tumor Hospital of Guangxi Medical University from 2012.8 to 2013.5.89 cases of cervicovaginal lavage fluid were divided into 58 cases of HPV16 (+) and 31 cases of HPV58(+).Cytokines IL-12, IL-4, IFN-γin the cervical microenvironment were detected by ELISA assay.140 cases of cervical exfoliated cells were divided into 106 cases of HPV16(+) and 34 cases of HPV58(+).Using qRT-PCR to quantify the copy number of E2 and E6 genes and analyze the physical status of HPV 16/58 DNA according to E2/E6.Results:①The level of IL-4 in HPV16(+) was significantly higher than that in HPV58(+)(P<0.05).The levels of IL-12 and IFN-γwere lower than that in HPV58(+)(P<0.05).②Significant differences of DNA state were found between the groups of HPV 16(+) and HPV58(+)(P<0.05)and the rate of HPV16 DNA integration was significantly larger than that of HPV 58 DNA.Conclusion:The infection of HPV16 and HPV58 not only can lead to reduction of IL-12,IFN-γand increasing of IL-4 in cervical local immune microenvironment but also lead to viral integration in cervical cells,resulting in cervical lesions and cervical cancer through the internal and external changes in cervical cells .HPV16 is more likely to cause cervical cancer tissue than 58.

6.Correlation Study of CYP2C19 Genotypes on Peri-procedural Myocardial Infarction in Patients With Percutaneous Coronary Intervention

Objective: To analyze the risk factors for peri-procedural myocardial infarction (PMI) in patients with percutaneous coronary intervention (PCI) and to study the impact of CYP2C19 genotypes on PMI. Methods: A total of 283 coronary artery disease (CAD) patients received elective PCI were enrolled, the patients were with normal level of cTnI prior PCI, without adverse reaction post PCI and cTnI levels were re-examined at 24 hours after PCI. The patients were divided into 2 groups: PMI group,n=83 and Non-PMI group,n=200. CYP2C19 genotypes were detected by DNA microarray chip method and based on its result, the patients were classiifed into 3 genotypes: Extensive metabolism type, Intermediate metabolism type and Poor metabolism type. The patients with ADP inhibition rate ≤ 20% was deifned as heightened platelet reactivity (HPR) which was measured by thrombelastograph. Baseline clinical condition was compared between 2 groups and the impact of CYP2C19 genotypes with other risk factors for PMI occurrence were studied. Results: The occurrence rate of PMI was 29.3% (83/283); the age, gender, BMI, smoking, blood levels of TG, LDL-C and uric acid, history of hypertension and diabetes were similar between 2 groups,P>0.05. Compared with Non-PMI group, PMI group had more implanted stents (2.1 ± 0.9) vs (1.5 ± 0.6) and longer total length of stents 56 (36-71) mm vs 33 (23-51) mm, bothP<0.05; lower ADP inhibition rate (36.0 ± 29.8) % vs (43.9 ± 26.4) %,P=0.02 and higher HPR occurrence rate 39.8% vs 22.0%,P=0.01. The ratios of CYP2C19 genotypes of Extensive metabolism, Intermediate metabolism and Poor metabolism were 37.5%, 45.2% and 17.3% respectively, while PMI patients in each genotype were 21.7%, 29.7% and 44.9% respectively. Logistic regression analysis indicated that HPR (OR=2.18, 95% CI 1.19-4.01,P=0.02), total length of stents (OR=1.03, 95% CI 1.00-1.06,P=0.007) and CYP2C19 Poor metabolism genotype (OR=2.8, 95% CI 1.24-6.28,P=0.01) were the risk factors for PMI occurrence. Conclusion: HPR, total length of stents and CYP2C19 poor metabolism genotype were the risk factors of PMI occurrence, while poor metabolism genotype might be the most important endogenous reason for PMI occurrence.

Acute respiratory distress syndrome (ARDS) caused by various reasons is a common severe neonatal clinical disease.The cure rate of the ARDS in newborns increases with the constantly progress of the neonatal intensive care and diagnosis medical technology.However,the motality of ARDS in newborn infants is still very high,about 30%-40%.There is no special therapy with neonatal ARDS and comprehensive treatment is adopted mainly according to its pathophysiologic change and clinical manifestation.The review will summarize the advances in the treatment of neonatal ARDS.

Objective To study the effect of angelica polysaccharides (APS) on recoverable ability from cryopreservation damage of umbilical cord blood (UCB) hematopoietic cells and APS with hematopoietic growth factors (HGFs) on in vitro expansion of UCB mononuclear cells (MNC) after thawing. 9. Metabolic imaging reveals a unique preference of symmetric cell division and homing of leukemia-initiating cells in an endosteal niche

The metabolic properties of leukemia-initiating cells (LICs) in distinct bone marrow niches and their relationships to cellfate determinations remain largely unknown. Using a metabolic imaging system with a highly responsive genetically encoded metabolic sensor, SoNar, we reveal that SoNar-high cells are more glycolytic, enriched for higher LIC frequency, and develop leukemia much faster than SoNar-low counterparts in an MLL-AF9-induced murine acute myeloid leukemia model. SoNar-high cells mainly home to and locate in the hypoxic endosteal niche and maintain their activities through efficient symmetric division. SoNar can indicate the dynamics of metabolic changes of LICs in the endosteal niche. SoNar-high human leukemia cells or primary samples have enhanced clonogenic capacities in vitro or leukemogenesis in vivo. PDK2 fine-tunes glycolysis, homing, and symmetric division of LICs. These findings provide a unique angle for the study of metabolisms in stem cells, and may lead to development of novel strategies for cancer treatment.
