Objective To investigate the effect of aloperine(ALO)on interleukin-1β(IL-1β)-induced chondrocyte injury and its mechanism. Methods Chondrocytes were randomly divided into control(Con)group,IL-1 β group,IL-1β + ALO-L(25 mg/L)group,IL-1β + ALO-M(50 mg/L)group and IL-1 β + ALO-H(100 mg/L)group;Con group,IL-1βgroup,IL-1β + miR-NC group and IL-1β + miR-16-5p group;Con group,IL-1β group,IL-1β + si-NC group and IL-1β + siSOX5 group. Cells in IL-1β group were treated with 10 ng/mL IL-1β,while no treatment was given in Con group. The transcription levels of miR-16-5p and SOX5 mRNA in chondrocytes were detected by qRT-PCR;The contents of IL-6,TNF-αand IL-1β were detected by ELISA;The expression levels of Bcl-2,Bax and SOX5 protein were detected by Western blot and the apoptosis was detected by flow cytometry. Results Compared with IL-1 β group,the contents of IL-6,TNF-α and IL-1βin IL-1β + ALO-L group,IL-1β + ALO-M group and IL-1β + ALO-H group decreased significantly(t = 5. 002~20. 653,each P < 0. 001),the apoptosis rate decreased significantly(t = 5. 473~17. 371,each P < 0. 001),the expression level of Bcl-2 protein increased significantly(t = 7. 800~16. 100,each P < 0. 001),and the expression level of Bax protein decreased significantly(t = 4. 993~14. 311,each P < 0. 001);The mRNA transcription level of miR-16-5p gene increased significantly(t = 6. 688~16. 545,each P < 0. 001),while the mRNA transcription level and protein expression level of SOX5 gene decreased significantly(t = 4. 609~15. 393,each P < 0. 001). Compared with the IL-1β + miR-NC group,the mRNA transcription level of miR-16-5p in the IL-1β + miR-16-5p group increased significantly(t = 17. 106,P < 0. 001),the contents of IL-6,TNF-α and IL-1 β decreased significantly(t = 15. 030~20. 013,each P < 0. 001),the apoptosis rate decreased significantly(t = 12. 273,P < 0. 001),the expression level of Bcl-2 protein increased significantly(t = 15. 652,P < 0. 001),and the expression level of Bax protein decreased significantly(t = 12. 999,P < 0. 001). Compared with IL-1β +si-NC group,the expression level of SOX5(t = 13. 444,P < 0. 001),IL-6,TNF-α and IL-1β in IL-1β + si-SOX5 group decreased significantly(t = 14. 087~17. 103,each P < 0. 001),the apoptosis rate decreased significantly(t = 11. 991,P < 0. 001),the expression level of Bcl-2 protein increased significantly(t = 13. 864,P < 0. 001),and the expression level of Bax protein decreased significantly(t = 11. 818,P < 0. 001). Conclusion Alo inhibited the apoptosis of chondrocytes induced by IL-1β,thus reducing the injury of chondrocytes,of which the mechanism might be regulating the expression of miR-16-5p and SOX5 and the secretion of inflammatory factors in chondrocytes.
3.Clinical analysis of ketogenic diet for 25 children with infantile spasms
Chinese Journal of Applied Clinical Pediatrics 2015;30(7):532-534
Objective To assess the efficacy, compliance and safety of ketogenic diet (KD) for children with infantile spasms, thus to provide an effective basis for the treatment of infantile spasms. Methods Twenty-five patients with infantile spasms were treated with KD.The gas chromatography-mass spectrometry and high performance liquid chromatography-mass spectrometry (HPLC-MS/MS) were used, except for metabolic diseases,to detect the blood routine, urine, stool, blood Iipids, liver function,as well as electrocardiogram (ECG),electroencephalograph (EEG),ultrasound of the urinary system and other related checks of all patients. The KD antiepileptic drug therapy was given after all children were admitted to hospital, and KD started directly in all patients without fasting. The efficacy compliance and safety were followed up in this study. Results KD maintenance therapy for 1 month,3 mouths,6 months were given to 25 cases (100%,25/25 cases),to 22 cases (88%,22/25 cases),and to 10 cases (40%,10/25 cases),respectively.Among them 7 patients (28%,7/25 cases) were in complete control of seizures within 1 month.Up to Class Ⅱ,Ⅲ efficacy level accounted for 32% (8/25 cases),12% (3/25 cases),respectively.Thirteen cases of children had varying digestive symptoms,10 cases of them developed asymptomatic hypoglycemia, and 9 cases of children had sleep trouble during KD treatment, but after symptomatic treatment, children's adverse reactions almost subsided.Only 4 cases of children discontinued treatment due to adverse reactions. All children's seizures were fully controlled. Conclusions The effect of KD is sure in the treatment of infantile spasms.Although KD has some adverse reactions, but it can be tolerated;serious body organ dysfunction treated by KD + antiepileptic drugs combination therapy for 1 to 6 months is not found yet,so the therapy is worthy of promotion clinically.
4.Morphological analysis of autophagy.
Acta Pharmaceutica Sinica 2016;51(1):39-44
Autophagy is an important homeostatic cellular recycling mechanism responsible for degrading injured or dysfunctional subcellular organelles and proteins in all living cells. The process of autophagy can be divided into three relatively independent steps: the initiation of phagophore, the formation of autophagosome and the maturation/degradation stage. Different morphological characteristics and molecular marker changes can be observed at these stages. Morphological approaches are useful to produce novel knowledge that would not be achieved through other experimental methods. Here we summarize the morphological methods in monitoring autophagy, the principles in data interpretation and the cautions that should be considered in the study of autophagy.

Results The scores by modified Barthel index (MBI) and functional comprehensive assessment (FCA) before intervention were significantly higher than those before intervention (P<0.01). The scores by MBI 1 month after intervention were significantly higher than those three months after intervention (P<0.01), indicating the longer the rehabilitation training, the better the effect. Conclusion The community rehabilitation care to patients with cerebral infarction sequelae can improve the quality of rehabilitation training and finally their quality of life.
7.The expression and significance of CDX2 and Ki-67 in colorectal tumor
Journal of Chinese Physician 2012;14(2):191-194
ObjectiveTo investigate the expression and significance of CDX2,Ki-67 in adenoma and colorectal adenocarcinoma, and the correlation between CDX2 and Ki-67.MethodsThe expression of CDX2 and Ki-67 was evaluated in the colorectal adenocarcinoma, adenoma and normal colorectal mucosa as control subjects by immunohistochemistry.ResultsThe magnitude of CDX2 were negatively correlated with the level of dysplasia in adenoma(rs=-0.38,P =0.02),and negatively correlated with differentiation, lymph node metastasis, diversion and Dukes stage of cancer in colorectal adenocarcinoma(rs=-0.49,rs=-0.37,rs =- 0.38,rs =-0.37,P =0.01).The expression and relativity of Ki-67 in the samples were contrary to the CDX2.The levels of CDX2 and Ki-67 expression had significant difference between the adenoma and adenocarcinoma, and showed negatively correlation in this two type tissues (rs=-0.69,P =0.00; rs =-0.40,P =0.00).ConclusionsCDX2 may be used as an effective marker to evaluate malignancy degree and prognosis of large bowel neoplasm patient. The levels of Ki-67 expression in colorectal adenocarcinoma are correlated with malignancy degree and clinical stage. The expression of CDX2 is negatively correlated with Ki-67.Combined detection of CDX2 and Ki-67 may be helpful for the judgment of colorectal tumor biological characters.
8.Effect of melatonin on breast cancer and its mechanisms
Chinese Pharmacological Bulletin 1987;0(02):-
Melatonin is a bioactive substance primarily secreted by the pineal gland.Increasing evidences indicate that melatonin is effective in reducing breast cancer development.Melatonin exerts its anticarcinogenic actions through a variety of mechanisms.Melatonin suppresses estrogen receptor gene, modulates several estrogen dependent signal transductions, inhibits cell proliferation and impairs the metastatic capacity and so on.It has been suggested that enhanced endogenous melatonin secretion or melatonin treatment is beneficial for breast cancer patients. This review describes the mechanisms of melatonin on breast cancer and its possible application.
9.Rehabilitation of Unilateral Spatial Neglect(review)
Chinese Journal of Rehabilitation Theory and Practice 2009;15(6):541-544
Unilateral spatial neglect is recognized as a significant disabling deficit, clinically defined as a failure to report, respond or orient to novel stimuli presented on the side opposite to a brain lesion. It is a predictor of poor functional outcome following right hemisphere lesion. Many different rehabilitation techniques or treatments have been put forward to alleviate, reduce or remediate spatial neglect. In the following review, we described, explained these Methods in terms of their underlying or motivatng mechanisms and evaluated in terms of the current evidence base for their effectiveness for clinical rehabilitation.
