1.Antibiotic resistance analysis in hospitalized elderly patients with lower respiratory tract infection caused by Pseudomonas Aeruginosa

Objective To explore the characterization of antibiotic resistance in hospitalized elderly patients with lower respiratory tract infection caused by Pseudomonas Aeruginosa ,and provide scientific basis for clinical ra-tional use of antimicrobial drugs and prevent the emergence of pan-drug resistant bacteria .Methods The drug sensi-tive tests of 347 hospitalized elderly patients with lower respiratory tract infection caused by Pseudomonas Aeruginosa in Shandong provincial hospital affinited to Shandong university from May 2010 to June 2012 were analyzed retrospec-tively.Results The antimicrobial resistance of the 347 cases was rather serious .The resistant rates of ampicillin and cefazolin were both 100.0%.Followed by ampicillin/sulbactam,tobramycin,sulfamethoxazole/trimethoprim,the re-sistant rates were 95.1%,86.8%and 83.6%respectively.The resistant rate of imipenem has reached 19.6%,and the detection rate of pan-drug bacteria was 4.9%.Conclusion The antibiotic resistance in hospitalized elderly pa-tients with lower respiratory tract infection caused by Pseudomonas Aeruginosa has become more and more serious .In order to prevent the emergence and prevalence of pan-drug resistant bacteria , antibacterial agents should be chosen reasonably according to the results of drug sensitive tests .

4.Determination of Cinnamaldehyde in Zegui Longshuang Capsules by HPLC

Objective To establish a method for determining cinnamaldehyde in Zegui Longshuang Capsules. Methods A C18 column was used with Acetonitrile-Water (30∶70) as the mobile phase,and the detection wavelength was at 290 nm. Results The calibration curve of cinnamaldehyde was linear from 0.010 ?g to 0.507 ?g,and r=0.999 9. The average recovery was 98.55 %with RSD being 2.06 %. Conclusion This method is proved to be accurate and reliable,and can be used to control the quality of Zegui Longshuang Capsules.
5.Animal model of arterial vulnerable plaque for imaging study using balloon-injury method

Objective To establish animal model of arterial vulnerable plaques for molecular imaging study.Methods Fifteen New Zealand white rabbits were randomly divided into high lipid diet+balloon-injury group (A),high lipid diet group (B)and regular diet group (C).Ultrasound (US)and magnetic resonance (MR)imaging were used to dynamically observe the formation of plaque in abdominal aorta. Results were compared with blood lipid level and pathological indicators.Results At 4 weeks,several plaques could be seen in group A.The plaque number increased rapidly and reached to 22 at 12 weeks,which was in parallel with the change of blood lipid. Only a few plaques were observed in group B,while no vulnerable plaque was revealed in group C.All the plaques were judged to be soft plaques on US and MR images,which was consistent with the macrophages gathering and smooth muscle cell proliferating in plaques.Conclusion High lipid diet+balloon-injury is an ideal method to build animal model for molecular imaging of atherosclerotic plaque.
6.Investigate repairing effect of Lugua polypeptide on patients’ sensory and motor function with spinal cord injury

Objective To study repairing effect of Lugua polypeptide on patients with spinal cord injury and the related mechanism. Methods From August 2012 to August 2013,79 cases patients with thoracolumbar spinal cord injury as the research objects,and they were divided into two groups randomly.The control group (n=38)received conventional treatment,and the observation group (n=41)received intravenous drip of Lugua polypeptide on the basis of conventional treatment.The course is three months.The American spinal injury association(ASIA)sensory score,ASIA motor score and Bathel index (BI )were recorded in two groups before and after treatment.Analyse the repairing effect on patients with spinal cord injury. Results ASIA sensory score,ASIA motor score and Bathel index in two groups after treatment improved ,and the observation group improved significantly compared with control group(P<0.05).Conclusion Lugua polypeptide can improve sensory and motor function with spinal cord injury ,it has clinical value.
7.Correlation analysis of renal function and serum cystatin C in patients with kidney damage secondary to cirrhosis

Objective To investigate the correlation between serum cystatin C(Cys-C)and renal function in patients with renal damage secondary to cirrhosis.Methods 129 patients with liver cirrhosis were selected,according to the measured glomerular filtration rate (GFR) as the gold standard for evaluation of renal function,patients were divided into group A(cirrhosis with normal renal function),group B(cirrhosis following renal insufficiency),group C (cirrhosis secondary to renal failure).The concentration of Cys-C was compared among three groups,and the serum creatinine(Scr),serum proteins ball micro β2 (β2-MG),creatinine clearance (Ccr) were detected.Results Cys-C,Scr,β2-MG,Ccr and other indicators of A,B,C three groups increased or decreased with the severity of impaired renal function(t =6.65,6.85,6.58,4.54,all P < 0.05).The abnormal detection rates of Cys-C,Scr,Ccr,β2-MG in A group were 31.1%,11.1%,8.9%,26,7%,those in B group were 83.7%,30.2 %,34.9%,46.5%,those in C group were 100.0%,85.4%,87.8%,95.1%.Conclusion Serum Cys-C concentration is a sensitive marker to evaluate glomerular filtration,it has accurate and reliable clinical significance in the early diagnosis of liver cirrhosis secondary to kidney damage.
8.Ultrasound-guided percutaneous injection of lauromacrogol for the treatment of hepatic hemangiomas

Objective To investigate the effect of ultrasound- guided percutaneous injection of lauromacrogol in treating hepatic hemangiomas. Methods During the period from June 2010 to Jan. 2013 at authors’ hospital, a total of 32 patients with hepatic hemangiomas were treated with ultrasound-guided percutaneous injection of lauromacrogol. The largest lesion was 6.0 cm × 5.6 cm in size, and the smallest lesion was 3.0 cm × 2.6 cm in size. Before and after the treatment ultrasonography, hepatic functions and measurement of tumor size were performed, and the results were evaluated. Results A total of 54 times of puncturing were carried out for the 32 patients. Three times of puncturing were made in 2 cases , two times of puncturing in 18 cases, and single time of puncturing in 12 cases. Complete cure was achieved in 25 cases (78.1%) and effective result was obtained in 7 cases, with an effective rate of 100%. After the treatment, all patients complained of different degrees of pain at hepatic region, which were relieved spontaneously. Two patients developed fever (37.5 - 38.5℃), and after physical cooling the body temperature fell to normal within two days. One patient developed mild nausea and vomiting, which disappeared after symptomatic treatment. No severe complications occurred. Conclusion Ultrasound- guided percutaneous injection of lauromacrogol is a simple, safe and effective treatment for hepatic hemangiomas. Besides, this technique has no serious complications.
9.Establishment of a risk assessment model for laboratory biosafety of Crypto-sporidium

Objective To evaluate the risk assessment model of Cryptosporidium laboratory,so as to provide the basis for laboratory personnel engaging in the operation of Cryptosporidium. Methods Firstly,the risk factors of Cryptosporidium infec?tion in laboratory were determined by the literature and Delphi,and then the weights of risk factors were determined by fuzzy an?alytic hierarchy process. A risk assessment model for laboratory biosafety of Cryptosporidium was established. Results Com?pared to the indexes,based on the risk assessment model,stool sample processing was the two steps in the laboratory with high risk of infection and high risk factors,with the combination weights of risk possibility and hazard rating were 0.111 and 0.107, respectively. Conclusion The risk assessment model established is feasible. It can be used to make some suggestions for the re?lated laboratory staff.
10.Determination of Paclitaxel and Docetaxel in Plasma by Crossing Internal Standard Method and its Clinical Application

Objective:To establish an HPLC method for the determination of paclitaxel and docetaxel in plasma to provide refer-ence for the individualized treatment regimen and the evaluation of curative effect and adverse reactions. Methods:Paclitaxel and do-cetaxel were used as the internal standard for each other. The samples were precipitated by acetonitrile and separated on a DikMA Dia-monsil C18 column with a mixture of acetonitrile-water (55: 45) as the mobile phase. The flow rate was 1. 2 ml·min-1 . The column temperature was set at 25℃. Paclitaxel and docetaxel were detected by UV-detection (λ= 227 nm). Results: A linearity was ob-tained within the range of 0. 078-10. 0 mg·L-1 for paclitaxel and docetaxel. The limit of quantitation was 0. 039 mg·L-1 . The aver-age recovery of paclitaxel and docetaxel was 99. 85% and 100. 35%, respectively. The inter- and intra-day RSD were both less than 5% and the RSD for freeze-thaw stability was below 10%. The plasma concentration of paclitaxel in clinical samples was within the range of 0. 18-6. 16 mg·L-1 and obvious individual difference was shown. Conclusion:Therapeutic drug monitoring is very important due to the obvious differences in plasma concentration of paclitaxel and docetaxel. The established method is sensitive, accurate, con-venient and rapid in r the therapeutic drug monitoring, and is useful for the adverse drug reactions monitoring and pharmacokinetic study.
