1.Effect of cAMP on myocardial apoptosis induced by ischemia/ reperfusion injury in rat

ve To investigate the effect of intravenous cAMP on the myocardial infarct size, the ultrastructure of myocardium and myocardial cell apoptosis and the possible mechanism of myocardial protection affected by cAMP against ischemia /reperfusion (I/R) injury. Methods Forty SD rats of either sex weighing 250-280g were anesthetized with abdominal sodium pentobarbital 4.5mg/100g, tracheotomized and mechanically ventilated (VT = 2ml/100g, RR = 60bpm) . Myocardial I/R was produced by tying and untying of left anterior descending coronary artery. Ischemia lasted 30 min and reperfusion 2h. Rats were randomly divided into 3 groups: control group (n = 8) in which left anterior descending coronary artery was exposed and a piece of silk thread was placed around the artery but not tied; I/R group (n = 16) in which normal saline 1ml was injected into sublingual vein before I/R; cAMP group (n = 16) received intravenous cAMP 1mg/kg 5min before I/R. The animals were then sacrificed and heart was harvested for determination of myocardial infarct size (by TTC) and ultrastructure examination (electron microscope) . Apoptosis was identified by TUNEL and apoptosis index (AI) was obtained. The expression of Fas, Bcl-2 protein was measured by immunohistochemical technique. Results The infarct size was smaller in cAMP group than that in I/R group . Myocardial apoptosis and necrosis were quite obvious in I/R group whereas in cAMP group the ultrastructure of myocardium was fairly normal. The AI in I/R group was significantly higher than that in cAMP group (P 0.05 ) . Conclusions cAMP can protect myocardium from I/R injury by modulating the expression of Fas and Bcl-2 protein and inhibit apoptosis following myocardial I/R.
2.Mechanism of Pengjiayin Preparation in the Treatment of Ectopic Endometrium in SD Rats

Objective To explore the mechanism of Pengjiayin preparation in the treatment of ectopic endometrium in SD rats. Methods To study the mechanism of Wudan Pill and Pengjiayin preparation in the treatment of ectopic endometrium in immunological way. SD rat models of endometriosis were established, and the expression of Survivin and Caspase-3 protein associated with apoptosis was detected in ectopic and eutopic endometrium in rats with the immunohistochemical method, simultaneously apoptotic cells were detected by TUNEL. Results Compared with the control group, the expression of the caspase-3 was increased in the low dosage group, the midium dosage group and the high dosage group of the Pengjiayin preparation, and the increase was also observed with the Wudan Pill group (P
3.Effects of Microiontophoretically Applied Some Different Transmitter Receptor Antagonists on the Evoked Response of “Pain” Units of Hypothalamic Paraventricular Nucleus

Effects of microiontophoretically applied transmitter receptor antagonists on the evoked response of “pain” units of PVN in rats by sciatic stimulation were observed. The results showed: (l)The evoked response of 7 out of 38 PVN “pain” units could be blocked by atropine (7/38); 5 out of 27 by hexamethonium; 6 out of 31 by phentolamine (6/31); and 4 out of 25 by propranolol (4/25). Seventeen out of 52 were blocked by cyproheptadine (17/52), but 3 out of 52 were augmented by cyproheptadine (3/52). (2) The evoked response of the same “pain” unit could be blocked by two antagonists: the evoked response of 2 out of 27 PVN “pain” units could be blocked by atropine and cyprohepadine; 3 out of 20 by hexamethonium and cyproheptadine; 2 out of 25 by prepamolol and cyproheptadine. These results suggest that the noxious somatic input to PVN involves 5-HT, cholinergic and adrenergic transmitter receptor mechanisms and that the convergence of various transmitter systems on PVN “pain” unit is indicated.
4.Traditional Chinese medicine health management guidance to the community of puerperal women’s breast-feeding

Objective By combining the traditional Chinese medicine(TCM) health care service and the current community puerperal women's health management, to explore the feasibility of TCM health management in the grass-roots community on puerperal women’s breast-feeding. Methods 440 patients meeting the inclusion criteria of puerperal women according to the neighborhood of residence were recruited into a non TCM intervention group of 225 people, and a TCM intervention group of 215 people. The non TCM intervention group was given health guidance on puerperal women. On such basis, the TCM intervention group was further cooperated with appropriate technical guidance of TCM. Puerperal women breastfeeding before and after the intervention was studied. Results After the intervention, the milk shortage of puerperal women breastfeeding rate of TCM intervention group was higher than the non TCM intervention group, 76.9% (60/78) vs. 56.9% (41/72), and mixed feeding rate was lower 21.8% (17/78) vs. 40.3%(29/72), the difference was statistically significant (χ2=5.916, 5.178; P < 0.05). The rate of breast feeding and mixed feeding rate were also statistically different between the TCM intervention group and the non TCM intervention group 78.1%(168/215) and 17.2% (37/215) vs. 62.7% (141/225) and 33.3% (75/225), (χ2=11.860, 14.226; P<0.01). Conclusion TCM health management guidance on puerperal women in the community can effectively improve the hypogalactia puerperal women's breastfeeding rate.
5.Influence of blood glucose level at hospitalization on no-reflow and prognosis in non-diobetic patients with myocardial infarction after emergency coronary intervention

Objective:To explore the influence of different blood glucose level at hospitalization on no-reflow and prognosis in patients with acute myocardial infarction (AMI),but without diabetes after emergency percutaneous coronary intervention (PCI).Methods:A total of 678 non-diabetic AMI patients undergoing PCI were selected.Ac-cording to blood glucose level at hospitalization,patients were divided into normal group (n=312),blood glucose level higher group (higher group ,n=201)and blood glucose level extremely high group (extremely high group,n=165).Clinical characteristics,no-reflow after PCI and incidence of cardiovascular events within 30d after PCI were compared among three groups.Results:Compared with normal group and higher group,there were significant rise in age,percentages of >1 location of myocardial infarction and Killip's ≥ class II,white blood cell count instant af-ter hospitalization,peak value of creatine kinase (CK)and CK-isoenzyme (CK-MB)in extremely high group,P <0.01 all;Compared with normal group,the age,peak value of CK and CK-MB significantly rose in higher group (P<0.01 all).Along with blood glucose rose,incidence rate of no-reflow significantly increased (17.0% vs.38.8%vs.49.7%),left ventricular ejection fraction (LVEF)on 30d after PCI gradually reduced [(68.6± 5.7)% vs. (62.2±6.4)% vs.(55.6±6.9)%]in above-mentioned 3 groups in turn,there was significant difference between any two groups,P <0.05 or <0.01.Conclusion:Stress hyperglycemia possesses great influence on no-reflow and prognosis in non-diabetic AMI patients after PCI.
6.Antagonistic effect of scutellarin on toxicity of selenium in human liver cells L-02

Object To investigate the antagonistic effect of scutellarin on the toxicity of selenium in human liver cells L-02. Methods MTT method was used to observe the effects of scutellarin and selenium on cell growth. The morphological changes of cells were observed by fluorescent microscope. The MDA level, thiol content, and total antioxidative capacity of cells were also measured. Results The optimum condition for scutellarin to promote cell growth was 0.5 mmol/L in 48 h culture period. Comparing with the only selenium treated group, 0.5 mmol/L scutellarin significantly antagonized the inhibition of cell growth caused by 1 and 5 ?mol/L selenium, suppressed the selenium-induced cell damage and the increase of MDA content, as well as increased the thiol content and cell antioxidation capacity. Conclusion Scutellarin markedly antagonizes the toxicity of selenium in human liver cells L-02 by inhibiting lipid peroxidation and promoting cellular antioxidation capacity.
7.Content Determination of 4 Flavonoids Constituents in Yinqiao Capsules by HPLC

OBJECTIVE:To establish a method for the determination of 4 flavonoids constituents in Yinqiao capsules. METH-ODS:HPLC method was adopted. The Hypersil ODS C18 column was used with the mobile phase A of methanol-water-acetic acid (10∶88∶2,V/V/V)and B of methanol-water-acetic acid(88∶10∶2,V/V/V)in gradient elution at the flow rate of 1.0 ml/min;the de-tection wavelength was 327 nm,the column temperature was 25 ℃,and the volume was 10 μl. RESULTS:There was a good linear relationship between the amount of quercetin and peak area in the range of 0.050 9-1.018 0 μg(r=0.999 8),kaempferide in the range of 0.050 2-1.004 0 μg(r=0.999 5),isorhamnetin in the range of 0.051 0-1.020 0 μg(r=0.999 4)and rutin in the range of 0.050 4-1.007 0 μg(r=0.999 8). RSDs of precision,stability and repeatability tests were <2%. The average recoveries were 100.09%(RSD=0.93%,n=9),99.83%(RSD=0.75%,n=9),100.51%(RSD=1.17%,n=9) and 101.19%(RSD=1.08%,n=9). CONCLUSIONS:The method is amount specific,stable and reproducible and can be used for the quality control of Yinqiao capsules.
8.Analysis of monitoring results of iodine deficiency disorders in Zhongwei City of Ningxia Hui Autonomous Region after marketization reform of salt industry

Objective:To understand the surveillance of iodine deficiency disorders (IDD) in Zhongwei City of Ningxia Hui Autonomous Region after marketization reform of salt industry (2017).Methods:In 2018, Shapotou District, Zhongning County and Haiyuan County of Zhongwei City were divided into five sampling areas according to the location of east, west, south, north and middle. In each area, one township was selected, and three salt sales outlets were selected from each township; at the same time, three supermarkets were selected from the cities and towns of each county (district), and different kinds of salt samples in all sales outlets and supermarkets were tested semi-quantitatively. From 2016 to 2020, Shapotou District, Zhongning County and Haiyuan County of Zhongwei City were divided into five sampling areas according to the location of east, west, south, north and middle. One township was selected from each area, and one primary school was selected from each township, 40 non-boarding students aged 8 - 10 years old (age balanced, half males and half females) were selected from each primary school; at the same time, 20 pregnant women were selected from each township, and salt samples were collected from houses of children and pregnant women to test salt iodine content. From 2018 to 2020, according to the "Ningxia Iodine Deficiency Disorders Surveillance Program (2016 Edition)", random urine samples of children and pregnant women were collected to detect urinary iodine content. In 2018 and 2019, the thyroid volume of children was measured by B-mode ultrasound, and the goiter rate was calculated.Results:In 2018, 13 kinds of salt were supplied in the market of Zhongwei City, a total of 130 salt samples were tested, and 11 samples of non-iodized salt were detected, with a rate of 8.46%. From 2016 to 2020, the consumption rates of qualified iodized salt in Zhongwei City were 90.33% (813/900), 67.89% (611/900), 78.67% (708/900), 91.56% (825/901) and 94.44% (850/900), respectively. From 2018 to 2020, the medians urinary iodine of children aged 8 - 10 years old were 189.33, 195.64, and 222.10 μg/L, and the medians urinary iodine of pregnant women were 158.21, 158.01, and 171.84 μg/L, respectively. In 2018 and 2019, the goiter rates of children aged 8 - 10 years old were 1.75% (7/400) and 0.67% (4/600), respectively.Conclusion:After marketization reform of salt industry, the consumption rates of qualified iodized salt in Zhongwei City decrease in 2017 and 2018, however, children and pregnant women have sufficient iodine nutrition, and the goiter rate of children is controlled at a low level.
9.MRI follow-up and clinical analysis for postradiosurgical acoustic neuromas

Objective To assess the effect of the gamma knife radiosurgery (GKS) for acoustic neuromas, and to analyze the MRI features of the tumors after GKS. Methods GKS was performed in 309 patients with acoustic neuromas from December 1994 to December 2002 at our center. The Leksell Gamma Knife models B and C were used. 249 cases with 256 tumors were followed up with MRI, which included 107 males and 142 females. The median age of the patients was 48.7?14.2 years (10-77 years). GKS was applied as primary treatment for 189 cases (75.9%), and was an adjunctive management approach for 60 cases (24.1%). The median clinical follow-up period and radiological follow-up period was (26.4?21.3) months (6-101 months) and (22.4?19.0) months (6-93 months), respectively. Results Of the 256 tumors, 79 tumors (30.8%) regressed, 165 tumors (64.5%) remained stable, 12 tumors (4.7%) enlarged, and the overall tumor control rate was 95.3%. Among 30 tumors of NF-Ⅱ , 8 tumors (26.6%) regressed, 19 tumors (63.4) remained stable, and 3 tumors (10.0%) enlarged, and tumor control rate was 90%. The rate of hearing deterioration was 22.7%, the rate of permanent facial paralysis was 0%, the rate of transient facial paralysis was 5.9%, and the incidence of transient trigeminal neuropathies was 19.9%. On the image of follow-up MRI, 79 tumors (30.0%) showed loss of contrast enhancement (LOE). LOE was the imaging property of neuromas on follow-up MRI, which symbolized a chronic decrease in the vascularity of neuromas. The histological findings of LOE demonstrated radiation-induced tumor necrosis. The transient swelling of tumors could not be confounded with the growth of tumors. Conclusion GKS is confirmed to be an effective treatment for acoustic neuromas and plays an important role in the control of NF―Ⅱ, and GKS can prevent cranial nerve from injury to a great extent.
10.RP-HPLC Analysis and Pharmacokinetics Study of Serum Carthamin in Mice

OBJECTIVE:To establish an RP-HPLC analysis of Carthamin in mice and to study its pharmacokinetics.METHODS:The serum concentration of Carthamin was determined by RP-HPLC.The blood concentration-time curve was established and the main pharmacokinetic parameters were computed.RESULTS:The linear range of Carthamin was 0.558~55.8 ?g?L-1(r=0.999 2),with the lowest limit of detection at 0.005 ?g?L-1Carthamin in vivo assumed two-compartment model and rapid absorption.CONCLUSION:The proposed method is simple,sensitive and reproducible,and it met the standard for pharmacokinetic study.
