3.Changes of SGOT and SGPT enzym activities in blood of experimental mice after long-time exposing to Bassa

100 white mice were divided in two groups, 50 mice for each one. The experimental group was exposed to Bassa 50EC with dose of 30 mg/kg body weight orally during 60 days. The other one was taken distilled water with a similar dose and time. The results showed that SGOT and SGPT enzyme activities in blood of experimental mice increased after exposing to Bassa.

5.Relationships with Alanine Aminotransferase Activity, and Anthropometric and Biochemical Measures in Obese Children.

PURPOSE: To investigate the physical and biochemical parameters related with elevated serum alanine aminotransferase (ALT) levels in obese children. METHODS: One hundred forty-two obese or overweight children who visited the out-patient clinics of Chungnam National University Hospital between January 2006 and August 2008 were enrolled. Physical measures and biochemical tests were performed in all patients. Liver sonography was performed in 43 patients. They were divided into the following 2 groups based on ALT levels: group I, normal ALT levels (n=65); and group II, elevated ALT levels (n=77). We compared the physical measures, biochemical results, and ultrasonographic findings of the livers in both groups. Other causes of elevated serum ALT levels were ruled out. RESULTS: The male-to-female ratios were 1.6:1 in group I and 7.6:1 in group II. Among physical parameters, the waist circumference-to-height ratio and hip circumference-to-height ratio were significantly higher in group II (p=0.001 and 0.046, respectively). Among biochemical parameters, aspartate aminotransferase (AST), gamma-glutamyltransferase (gamma-GT), and total cholesterol levels were significantly higher in group II (p<0.001, 0.001, and 0.001, respectively). The AST/ALT ratio was <1 (mean, 0.55) and statistically lower in group II. There was a positive correlation between the serum ALT level and gamma-GT (p<0.001, r=0.750), and a positive correlation between the serum ALT level and the waist circumference-to-height ratio in group II (p<0.001, r=0.401). CONCLUSION: The results suggest that the waist circumference-to-height ratio and gamma-GT may be associated with elevated alanine aminotransferase activity in obese children.

6.Aspartate and alanine aminotransferase in Fasciola hepatica.

The activity and distribution of aspartate aminotransferase (EC 2.6.1.1) and alanine aminotransferase (EC 2.6.1.2) in adult Fasciola hepatica have been studied. Fasciola hepatica was fractionated by differential centrifugation into nuclear, mitochondrial and cytosolic fractions. The activity of GOT and GPT was measured by the method of Reitman and Frankel. Isozyme patterns of those enzyme were also examined by DEAE-cellulose column chromatography. The results obtained were as follows: The activity of aspartate and alanine aminotransferase was about 0.55 unit and 0.92 unit per 1 g of Fasciola hepatica, respectively. The activity of those enzymes was relatively low compared with those in mammalian tissues. The distribution of aspartate aminotransferase in the subcellular organelles showed that 71 percent of the activity was in cytosolic, 24 percent in mitochondrial and 5 percent was in nuclear fraction. About 22 percent of the total alanine aminotransferase activity was found in the mitochondrial fraction, about 66 percent in the cytosolic fraction. Aspartate aminotransferase from cytosolic fraction was separated into two types of isozymes, whereas alanine aminotransferase from cytosolic fraction gave only one active peak on DEAE-cellulose column chromatography.

7.High Energy Electron Dosimetry by Alanine/ESR Spectroscopy.

Dosimetry based on electron spin resonance(ESR) analysis of radiation induced free radicals in amino acids is relevant to biological dosimetry applications. Alanine detectors are without walls and are tissue equivalent. Therefore, alanine ESR dosimetry looks promising for use in the therapy level. The dose range of the alanine/ESR dosimetry system can be extended down to l Gy. In a water phantom the absorbed dose of electrons generated by a medical linear accelerator of different initial energies (6~21 MeV) and therapeutic dose levels(1~60 Gy) was measured. Furthermore, depth dose measurements carried out with alanine dosimeters were compared with ionization chamber measurements. As the results, the measured absorbed doses for shallow depth of initial electron energies above 15 MeV were higher by 2~ 5% than those calculated by nominal energy CE factors. This seems to be caused by low energy scattered beams generated from the scattering foil and electron cones of beam projecting device in medical linear accelerator.

8.Postoperative Serum Glutamic Oxalacetic Transaminase and Serum Glutamic Pyruvic Transaminase Changes in Biliary Patients.

The SGO-T and SGP-T values were studied among 19 non-biliary surgical patients for control and 21 biliary tract surgical patients from Feb. to August 1969 in Chung Nam Medical Centor. The results obtained are as follows: 1. The SGO-T and SGP-T evels in the non-biliary surgical patients was in the normal range during pre-operative and postoperative days. 2. In patients with biliary disease SGO-T and SGP-T level were high beyond normal range on the pre-operative day. On the 1st postoperative day, these values were more elevated about 20 % and 11 %, respectively, but decreased gradually on the second and third postoperative days. 3 In biliary patients, the values were lower than pre-operative level on the 3rd postoperative day. On the other hand, values had not dropped to pre-operative level on the 3rd postoperative day in non-biliary patients.

9.Voriconazole Therapeutic Drug Monitoring is Necessary for Children with Invasive Fungal Infection.

PURPOSE: To determine the clinical significance of voriconazole therapeutic drug monitoring (TDM) in the pediatric population. METHODS: Twenty-eight patients with invasive fungal infections administered with voriconazole from July 2010 to June 2012 were investigated retrospectively. Fourteen received TDM, and 143 trough concentrations were analyzed. All 28 patients were assessed for adverse events and treatment response six weeks into treatment, and at the end. RESULTS: Out of 143 samples, 53.1% were within therapeutic range (1.0-5.5 mg/L). Patients administered with the same loading (6 mg/kg/dose) and maintenance (4 mg/kg/dose) dosages prior to initial TDM showed highly variable drug levels. Adverse events occurred in 9 of 14 patients (64.3%) in both the TDM and non-TDM group. In the TDM group, voriconazole-related encephalopathy (n=2, 14.3%) and aspartate aminotransferase (AST) or alanine aminotransferase (ALT) elevation (n=8, 57.1%) occurred with serum levels in the toxic range (>5.5 mg/L), whereas blurred-vision (n=2, 14.3%) occurred within the therapeutic range (1.18 mg/L and 3.9 mg/L). The frequency of voriconazole discontinuation due to adverse events was lower in the TDM group (0.0% vs. 18.2%, P=0.481). Overall, 57.2% of the patients in the TDM group versus 14.3% in the non-TDM group showed clinical response after 6 weeks (P=0.055), whereas 21.4% in the TDM group versus 14.3% in the non-TDM group showed response at final outcome (P=0.664). In the TDM group, >67.0% of the serum levels were within therapeutic range for the first 6 weeks; however 45.5% were within therapeutic range for the entire duration. CONCLUSION: Routine TDM is recommended for optimizing the therapeutic effects of voriconazole.

10.Retrospective Study on Hepatic Enzyme Changes in Patients Treated with Risperidone and Olanzapine.

OBJECTIVES: Hetapic dynfunction caused by the use of atypical antipsychotics in patients with psychiatric disorders has not been frequently reported compared to the use of typical antipsychotics. This retrospective study was aimed at assessing the differences of hepatic dysfunctions between patients treated with risperidone and olanzapine through reviewing computerized-medical records. METHODS: Charts of 667 risperidone- and olanzapin treated patients hospitalized at the department of psychiatray, Kangnam St. Mary's Hostpital, between 1998 and 2002, were reviewed. The frequencies of hepatic enzyme elevation in association with clinical variables such as time course of the use of antipsychotics were analyzed. RESULTS: Significant elevation in alanine aminotransferase (ALT) to upper reference range and 2 fold range were seen in olanzapine monotherapy patients, but not in risperidone monotherapy group. There was no difference between olanzapine and risperidone monotherapy groups in time course of hepatic dysfunction. There was no difference in hepatic enzyme elevation between resperidon and olanzapine combination therapy groups. CONCLUSION: The results suggest potential difference in degree of antipsychotic induced hepatic dysfunction between risperidone and olanzapine treatment. Well-desigend prospective study would be nessassary to extend our putative results.
