1.Anti-inflammatory effect of astragalosides and its mechanisms of action

AimTo study the anti-inflammatory effect of astragalosides(AST) and its mechanisms of action. Methods The exudate volume, neutrophil count and protein content in exudate were measuredinthe carrageenan-induced air pounch model in rats. The content of PGE2 was assayed by radioimmunoassay,the activity of PLA2 by microacid titration assay, IL-8 by ELISA, and NO by nitrate reductase assay. The production of O2 in neutrophil was determined by cytochrome C assay. Results AST(40, 80 mg· kg -1) could markedly reduce the exudate volume, neutrophil count, protein content, the content of IL-8, and the production of O2.AST lowered PLA2 activity of neutrophil and accellular component in exudate, and it also decreased the contents of PGE2 and NO in exudate. Conclusion AST has an obvious anti-inflammatory effect on carrageenan-induced acute inflammation.Its mechanisms may be related to the inhibition of vascular permeability and leukocyte migration, as well as to the suppression of PLA2 activity and the reduction of IL-8, PGE2, NO and O2 production.

3.Sterilizing effect and residual bacteria of four different methods for sterilizing simple breathing vesicles.

OBJECTIVETo study the effects of 4 different methods for disinfection of simple breathing vesicles and microbial residue.
METHODSThe disinfection tests were divided into 4 groups: G1 group (43 cases) with 500 mg/L chlorine dioxide spray, G2 group (28 cases) with alcohol spray, G3 group (47 cases) with 50 mg/L trichloroisocyanuric acid (TCCA) immersion, and G4 group (46 cases) with 50 mg/L chlorine dioxide solution immersion. After 30 min of disinfection, each group was examined by bacterial culture and colony count. The residual bacteria were identified and typed.
RESULTSThe 4 methods showed significant differences in bacterial colony count (P<0.001). The rate of bacterial residue was 0% in G1 group, 53.6% in G2 group, 27.7% in G3 group, and 21.7% in G4 group, showing significant differences between the 4 groups (P<0.001). The residual bacteria included antibiotic-resistant common opportunistic pathogen such as Pseudomonas aeruginosa, Acinetobacter baumannii and Staphylococcus haemolytic.
CONCLUSIONSDisinfection with 500 mg/L chlorine dioxide spray is the best for simple breathing vesicles. Prolonged immersion in TCCA may lead to the growth of drug-resistant pathogens in the breathing vesicles.

4.Expression of E3 ubiquitin ligase UHRF2 in intrahepatic cholangiocarcinoma and its clinical significance

Objective:To explore the expression and clinical significance of ubiquitin-like with PHID and ring finger domains 2 (UHRF2) in intrahepatic cholangiocarcinoma (ICC).Methods:The expression of UHRF2 in 138 ICC was determined by immunohistochemistry,and the relationships between UHRF2 expression with prognosis and clinicopathological features of ICC were analyzed:Results:UHRF2 was highly expressed in ICC compared with paratumorous tissues.Level of UHRF2 was associated with high AFP level (P=0.010),poor tunor differentiation (P=0.026),lymph node metastasis (P=0.001),and advanced TNM stage (P=0.019).Hevated UHRF2 expression was suggested to be associated with poor prognosis and high recurrence rate in ICC patients (P< 0.001),and multivariable analysis indicated that UHRF2 was of theprognostic indicators of ICC (P<0.05).Conclusions:High level of UHRF2 is related to the malignant phenotype of ICC,and can be a prognostic and recurrent indicator of postoperative patients.
