Objective To observe the protective effect of α-lipoic acid (LA) pretreatment on hypoxia/reoxyg-enation injury in vascular endothelial cells and preliminary explore its mechanism. Methods HUVEC were cultured intro and devided into control group,hypoxia/reoxygenation model group and LA group. Cell morphology was observed under inverted phase contrast microscope,cell viability was detected by MTT,total antioxidant capacity (TAOC),nitric oxide(NO),malondialdehyde (MDA),superoxide dismutase (SOD) activity or level were measured by various commercial kits respectively. Apoptosis of HUVEC in each group were detected by flow cytometry , cleaved caspase-3 level was measured by western blot. Results As compared to control group, the survival rate, TAOC, SOD, NO activity were significantly lower(P<0.05), and MDA level, apoptosis rate and apoptosis protein levels were significantly higher (P < 0.05) in model group. However,LA can increase the survival rate,TAOC, SOD,NO activity (P < 0.05),reduce MDA level and cell apoptosis (P < 0.05)in a dose dependent manner. Conclusion LA pretreatments provide protection against hypoxia/reoxygenation injury in HUVEC. LA exerts protective effects through inhibition of HUVEC apoptosis.

To investigate the therapeutic effects of recombinant humanized anti-HER2 antibody (Herceptin) on Her-2/neu-overexpressing metastatic human breast cancer, 7 patients with Her-2/neu-overexpressing metastatic human breast cancer were involved in this study. The results indicated that the therapeutic effective rate was 71. 4%. CR. PR, SD. PI) were seen in 2. 3, 1, 1 cases, respectively. No side reactions were observed. The results revealed that the therapeutic effect of Herceptin is intimately linked to Her-2/neu expression of breast cancer.

The aim of this paper is to report the study on gene silencing efficiency of siRNA targeted against mouse VEGFR2 (siVEGFR2) in vitro mediated by polyethyleneimine (PEI) and its anti-tumor effect in vivo. CY3-labeled siRNA was compounded into PEI and transfected into MS1 cells. Confocal microscopy was used to image the subcellular distribution of siRNA in MS1 cells. Semi-quantitative RT-PCR and Western blotting were used to evaluate VEGFR2 gene silencing induced by siVEGFR2/PEI complexes. A tumor-bearing nude mice model was established to compare the anti-tumor effect after delivered by local and systemic routes. siVEGFR2/PEI complex-transfected cells exhibited much fluorescence in cytoplasm with no evidence of nuclear accumulation. The expression levels of VEGFR2 mRNA and protein in PEI-transfected cells were significantly down-regulated compared with that in blank group, the silencing efficiency were 28.2% and 23.6% respectively. The tumor sizes in mice intratumorally injected with siVEGFR2/PEI complexes (189.429 +/- 17.562 mm3) were reduced definitely compared to that in mice injected with siVEGFR2/PEI complexes via vein route (315.507 +/- 20.491 mm3), or to saline groups (365.844 +/- 20.713 mm3). The study demonstrated that PEI could effectively transfect siRNA into cells and silence the VEGFR2 gene expression. Intratumoral delivery is more suitable for non-targeted modified PEI/siRNA complexes to inhibit the tumor growth in vivo. The present data lay a solid foundation to further study on the gene silencing mechanism for PEI-medicated RNAi and its anti-tumor efficiency in vivo.

OBJECTIVE:To establish an HPLC method for the determination of lipoic acid in lipoic acid sustained-release tablets.METHODS:HPLC was conducted on a Hypersil ODS with acetonitrile-0.05 mol?mL-1 potassium dihydrogen phosphate(45∶55,adjust pH to 2.0)as mobile phase at a flow rate of 1.0 mL?min-1.The injection volume was 10 ?L;the detection wavelength was set at 219 nm and the column temperature was 25 ℃.RESULTS:The linear range of lipoic acid was 9.99～399.70 ?g?mL-1(r=0.999)with an average recovery rate of 99.17%(RSD=0.40%);the intra-day and inter-day RSD were all less than 2.6%.CONCLUSION:The method is simple and accurate,and suitable for the determination of the content of lipoic acid in lipoic acid sustained-release tablets.

目的探讨《国际功能、残疾和健康分类》(ICF)临床检查表(中文版)与美国脊柱损伤协会(ASIA)损伤分级和日常生活活动(ADL)在脊髓损伤患者评定中的相关性。方法采用临床测评方法,运用ICF临床检查表、ADL评定和ASIA损伤分级评定法,对50例脊髓损伤患者进行综合评定。结果受试者ICF得分与ADL总分和ASIA分级分之间呈明显负相关(P<0.05—0.01)。结论ICF临床检查表指标较ADL和ASIA更全面。

Objective To estabhsh an ischemic model of intratemporal facial nerve (IFN) via the mastoid process approach.Methods From February,2015 to December,2015,45 SD rats were divided randomly into an operation group (n=35) and a shame group (n=10) in random,the right side facial nerve was used for the operation and the left side served as the control in both groups.Establish the IFN ischemia model by interrupting the petrosal artery through the mastoid process approach.Facial nerve function were evaluated at the 12h and everyday postoperatively for 28 days.The degree of IFN swelling were studied by taking paraffin sections of the decalcified temporal bone containing the IFN instantly and at the 1st,3rd,7th,14th and 21st days postoperatively.Then calculated the ratio of the cross-sectional area of the IFN and the facial canal (FN/FC).The data of behavior assessment and FC/FN were analyzed using ANOVA.Twenty-eight days after the insult,took continuous sections of brainstem containing facial nucleus,then counted the number of the facial neurons.At last,analysed the results of both operation and control sides in each group by using the student-t test.Results Facial nerve paralysis developed at 12 hour after surgery,then continued deteriorated till the 7th day.By the 28th day postoperatively,all rats in surgery group recovered and data showed no significance statistically when compared with the shame group (P＜0.05).From the value of FN/FC in different groups,the nerve were found swelling in the facial canal was increasing from the 1st postoperatively and reach the peak value at the 7th day after surgery.By the 21st day,the FN/FC come to steady but remain significant statistically when compared with the contralateral side(P＜0.05).In the operation group,facial neurons of injury side exhibited significantly loss [(41.5±3.8)％] when compared with the shame group [(98.1±4.0)％](P＜0.05).Conclusion Rats with petrosal artery interrupted exhibited significant deficits.This approach involved less tissue injury,studies on the mechanisms and therapy could become more reliable using this approach.

To evaluate the biomechanical stability of femoral shaft fracture fixation using different locking plates combined with different screw layout, for two different fracture settings, we build six groups different length locking plate combined with different screw number and different screw layout, fix with the fracture models respectively, and use the biomechanical finite element method to analysis the models. Then we attain the axial displacement and equivalent stress distribution of the internal fixation system under the action of axial load. The research shows that filling relatively softer material in femoral fracture can guarantee the stability of the internal fixation system, the long plate combined with less screw layout is obviously better than the short plate combined with the all screw layout, the same length plate combined with the selective screw layout is more effective than combined with the all screw layout. And plate combined with screws fixed in four threaded hole of distal fracture make the screw system stress disperse, and avoiding screw fixed in proximal fracture can alleviate the stress concentration of screws.

Objective:To explore the mediating effects of resilience and depression on the relationship between social support and self-neglect.Methods:From July to October 2020, a cross-sectional survey was conducted among 549 empty-nest community-dwelling elderly in the community using the social support rating scale, Connor and Davidson resilience scale, geriatric depression scale and elderly self-neglect scale. SPSS 22.0 and AMOS 24.0 were used for data analysis, including descriptive analysis, correlation analysis and structural equation modeling.Results:The scores of social support, resilience and self-neglect of the empty-nest elderly were (38.63±7.47), (64.30±14.57) and (3.72±2.67) respectively, and 31.70% (174/549) of the subjects had depressive symptoms. The score of self-neglect was negatively correlated with the score of social support ( r=-0.597, P<0.01) and resilience ( r=-0.557, P<0.01), and positively correlated with depression score ( r=0.675, P<0.01). The score of social support was positively correlated with resilience score ( r=0.531, P<0.01) and negatively correlated with depression ( r=-0.597, P<0.01). Social support could affect self-neglect directly ( β=-0.485, P<0.05), and it could also influence self-neglect indirectly through the partial mediating effect of resilience ( β=-0.451, P<0.05). The mediating effect of resilience and depression accounted for 12.18% and 36.00% of the total effects respectively. Conclusion:Social support could influence self-neglect directly or indirectly through resilience and depression. The empty-nesters should be encouraged to participate more in social activities so as to improve their resilience, reduce the occurrence of self-neglect.
