Objective To analyze the DCE-MRI features and ADC value in triple-negative breast cancer and HER-2 overexpres-sion breast cancer,and to improve the j udgment of both characteristic in MRI.Methods To analyse the DCE-MRI imaging of TNBC and HER-2 overexpression breast cancer,measured the ADC value and compared the statistics.Results Compared with the shape, margin,the way of enhancement and ADC value of the TNBC and HER-2 overexpression breast cancer,the distinction has statisti-cally significant(P<0.05);there were no statistically significant difference among age,mass or nomass,categories of lesions and TIC,(P>0.05).Conclusion DCE-MRI features of the shape,margin,the way of enhancement and ADC values were useful for the diagnosis of TNBC and HER-2 overexpression breast cancer.

AIM: To isolate and identify triterpenes from inflorescence of Cremanthodium potaninii C.Winkl,collected from Qinghai province.METHODS: The triterpenes were isolated by column chromatography and their structures were elucidated through NMR.RESULTS: Seven triterpenes were isolated and identified as ?-amyrin(I),Taraxast-20(30)-ene-3?,16?-diol-3-O-palmitate(II),olean-12-en-3?,16?-diol-3-O-palmitate(III),Olean-12-en-3?,11?,16?-triol-3-palmitate(IV),lup-3?,16?-diol-3-O-palmitate(V),lup-3?,16?,28-triol-3-O-palmitate(VI),16?-hydroxypseudo tarsxasterol-3-O-palmitate(VII).CONCLUSION: All these compounds are obtained from Cremanthodium potaninii C.Winkl for the first time.

Collagen is a kind of natural biomedical material and collagen based three-dimensional porous scaffolds have been widely used in skin tissue engineering. However, these scaffolds do not meet the requirements for artificial skin substitutes in terms of their poor mechanical properties, short supply, and rejection in the bodies. All of these factors limit their further application in skin tissue engineering. A variety of methods have been chosen to meliorate the situation, such as cross linking and blending other substance for improving mechanical properties. The highly biomimetic scaffolds either in structure or in function can be prepared through culturing cells and loading growth factors. To avoid the drawbacks of unsafety attributing to animals, investigators have fixed their eyes on the recombinant collagen. This paper reviews the the progress of research and application of collagen-based 3-dimensional porous scaffolds in skin tissue engineering.

Objective To investigate the effects of ursolic acid (UA) on cisplatin (CDDP)-induced expres_sion of transient receptor potential vanilloid 1 (TRPV1) in mouse cochlea .Methods Sixty BALB/c mice were ran_domly divided into 4 groups (15 mice in each group) and received introperitoneal injection once daily for 5 days:Control group (normol saline) ,UA group (80 mg/kg/day) ,CDDP group (4 .5 mg/kg/day) ,and CDPP (4 .5 mg/kg/day) plus UA group (80 mg/kg/day) .The expression of TRPV1 in mouse cochlea was determined by immuno_histochemistry ,microscope image analysis and western blot ,and auditory thresholds were evaluated by auditory brainstem response (ABR) measurement .ResuIts The expression of cochlea TRPV1 and ABR threshold shift was significantly increased in the mice treated with CDDP (P< 0 .05) ,as compared with control mice .These effects were prevented by UA treatment (all P<0 .05) .Furthermore ,a linear relationship analysis revealed that the ex_pression of cochlea TRPV1 was significantly correlated with ABR threshold shift(|r|>0 .7 , P<0 .05) .ConcIusion UA effectively attenuated CDDP -induced ototoxicity and improved auditory function through inhibition of TR_PV1 .

Objective:To establish an HPLC method for the determination of levofloxacin in levofloxacin hydrochloride ophthalmic in situ gel. Methods:The chromatographic column was Agilent Zorbax C18(150 mm ×4.6 mm,5μm). The mobile phase consisted of hexane sulfonic acid sodium solution-methanol(72∶28). The flow rate was 1. 0 ml·min-1. The detection wavelength was 293 nm. The chromatographic column temperature was 40℃. The injection volume was 10 μl. Results:The calibration curve was linear within the range of 0. 040 3-0. 403 0 mg·ml-1(r=1. 000 0) for levofloxacin. The average recovery was 99. 8% (RSD=1. 08%,n=9). Conclusion:The method is accurate, simple and rapid, and suitable for the content determination of levofloxacin in levofloxacin hydro-chloride ophthalmic in situ gel.

Objective To analyze the impact of patients ages on propofol dosage in painless gastroscopy.Methods A retrospective analysis was conducted on the clinical data of 158 painless gastroscopy patients from January 2017 to June 2020.They were divided into the young group(18～44 years old,n = 57),the middle-aged group(45～59 years old,n = 51),and the elderly group(≥60 years old,n = 50)based on their age.The anesthesia status and safety of each group were compared.Results The results showed that the dosage of propofol,average total dosage of propofol,MAP,HR,RR,SpO2 levels in the young group were higher than those in the middle-aged and elderly groups when consciousness disappeared,and the middle-aged group was higher than the elderly group;The onset time,anesthesia recovery time,orientation recovery time,and departure time in the young group of patients were shorter than those in the middle-aged and elderly groups,and the middle-aged group was shorter than the elderly group(P<0.05).The incidence of airway obstruction,hypoxemia,mandible support,mask ventilation,adverse cardiovascular events,sedation related adverse events,and incidence of obstructed endoscopy in the elderly group were higher than those in the young and middle-aged groups.The incidence of smooth endoscopy was lower than that in the young and middle-aged groups(P<0.05).Conclusion Aging may increase the dosage of propofol in patients underwent painless gastroscopy under anesthesia,prolong the onset time,anesthesia recovery time,orientation recovery time,and departure time,increase stress reactions and adverse reactions,and strengthen monitoring for elderly patients in clinical practice.

Background and Objectives: This study was to investigate the role of microRNA-29a-3p (miR-29a-3p) in human bone marrow mesenchymal stem cells (hBMSCs), and its relationship with steroid-associated osteonecrosis. Methods: and Results: The online tool GEO2R was used to screen out the differentially expressed genes (DEGs) in GSE123568 dataset. Quantitative real time-polymerase chain reaction (qRT-PCR) was performed to detect the expression of miR-29a-3p, forkhead box O3 (FOXO3), alkaline phosphatase (ALP), bone gamma-carboxyglutamate protein (OCN) and RUNX family transcription factor 2 (Runx2) in the hBMSCs isolated from the patients with steroid-associated osteonecrosis. CCK-8 assay was executed to measure cell viability; western blot assay was utilized to detect FOXO3, ALP, Runx2, OCN and β-catenin expression. Cell apoptosis and cell cycle were detected by flow cytometry. Immunofluorescence assay was used to detect the sub-cellular localization of β-catenin. Bioinformatics analysis and luciferase reporter gene assay were performed to confirm whether miR-29a-3p can combine with FOXO3 3’UTR. MiR-29a-3p was markedly up-regulated in the hBMSCs of patients with steroid-associated osteonecrosis, while FOXO3 mRNA was significantly down-regulated. Transfection of miR-29a-3p mimics significantly inhibited the hBMSCs’ proliferation, osteogenic differentiation markers’ expressions, including ALP, Runx2, OCN, and repressed the ALP activity, as well as promoted cell apoptosis and cell-cycle arrest. FOXO3 was identified as a target gene of miR-29a-3p, and miR-29a-3p can inhibit the expression of FOXO3 and β-catenin, and inhibition of miR-29a-3p promoted translocation of β-catenin to the nucleus. Conclusions MiR-29a-3p can modulate FOXO3 expression and Wnt/β-catenin signaling to inhibit viability and osteogenic differentiation of hBMSCs, thereby promoting the development of steroid-associated osteonecrosis.

Objective:To analyze the characteristics and clinical significance of ascites, peripheral blood lymph nodes, and cell subpopulations in patients with decompensated liver cirrhosis caused by alcoholic liver disease.Methods:Sixty decompensated liver cirrhosis patients with alcoholic liver disease admitted to the Affiliated Hospital of Jining Medical Unversity from July 2020 to July 2022 were selected as the observation group, and 40 healthy volunteers who underwent physical examinations in our hospital were randomly selected as the control group. The observation group was divided into A, B, and C grades based on the severity of the condition, according to the Child-Pugh grading of liver function. The differences in clinical data, peripheral blood lymphocyte subpopulation indicators, and ascites lymphocyte subpopulation indicators (CD3 + , CD4 + , CD8 + , CD20 + ) under different disease grades were analyzed, and the differences in peripheral blood lymphocyte subpopulation indicators between the two groups of people were compared. Resultsl:The levels of CD3 + , CD4 + , CD8 + , CD4 + /CD8 + , and CD20 + in the observation group were significantly lower than those in the control group (all P<0.05). Compared with Child-Pugh A grade patients, the expression of CD3 + , CD4 + , CD8 + , CD4 + /CD8 + , and CD20 + in peripheral blood of Child-Pugh B and C grade patients decreased significantly (all P<0.05); Compared with Child-Pugh B grade patients, the expression of CD3 + , CD4 + , CD8 + , CD4 + /CD8 + , and CD20 + in peripheral blood of Child-Pugh C grade patients decreased significantly (all P<0.05). Compared with Child-Pugh A grade patients, the expression of CD3 + , CD4 + , CD8 + , CD4 + /CD8 + , and CD20 + in ascites of Child-Pugh B and C grade patients decreased significantly (all P<0.05); Compared with Child-Pugh B grade patients, the expression of CD3 + , CD4 + , CD8 + , CD4 + /CD8 + , and CD20 + in ascites of Child-Pugh C grade patients decreased significantly (all P<0.05). The peripheral blood and ascites lymphocyte subsets were negatively correlated with the Child-Pugh grading of decompensated liver cirrhosis in alcoholic liver disease (all P<0.05). Conclusions:Patients with decompensated liver cirrhosis caused by alcoholic liver disease are accompanied by varying degrees of immune dysfunction. Detection of lymphocyte subsets in the patient′s peripheral blood and ascites has significant clinical significance for the diagnosis and prognosis of this disease.
