1.Analysis on distribution and drug resistance of Escherichia coli infection

Objective To analyze the distribution of Escherichia(E .) coli in clinical infection and its drug resistance situation to provide the scientific evidence for the control of outside hospital infection and nosocomial infection and clinical rational drug use . Methods E .coli isolated situation among various types of clinical samples in our hospital during 5 years from January 2010 to De‐cember 2014 ,its department distribution and drug resistance were analyzed .Results The isolated 2 405 strains of E .coli were mainly originated from urine samples (1 049 strains ,43 .60% ) and sputum samples (562 strains ,3 .4% ) .In which the detection rate of extended‐spectrum beta‐lactamase(ESBLs)producing E .coli was 57 .92% ;the resistance rate of E .coli to the most antibacterial drugs including penicillins ,cephalosporins ,fluroquinolones ,aminoglycosides and sulfonamides was more than 50% ,the drugs with the resistance less than 10% were imipenem(0) ,meropenem(0) ,piperacillin/tazobactam(4 .6% ) ,cefoperazone/sulbactam(6 .4% ) and cefoxitin (7 .7% ) ,the resistance to partial third and fourth generation cephalosporins of ceftriaxone ,cefotaxime and cefepime showed the significantly increasing trend .Conclusion E .coli has relatively serious drug resistance situation in hospital‐acquired in‐fections ,clinic should strengthen the pathogen distribution monitoring and drug resistance detection for avoiding the generation of more drug resistant strains in order to reduce the bacterial drug resistance and the hospital infection rate .
2.Determination of Nitrite by Catalytic Kinetic Photometry with Surfactant Sensitizer

A catalytic kinetic photometric method for the determination of nitrite with surfactant sensitizer has been developed. The method is based on the catalysis of NO2- to the discoloring reaction of pyronine B oxidized by potassium bromate in the solution of dilute H3PO4. When cetylpyridine bromide was used as a sensitizer of the reaction, the sensitivity increased by 2.5 times approximately. There was linear relationshop of lg(Ao/A) with nitrite in the range 0.005 ~ 0.15 mg/L. The detection limit was 0.004 mg/L. The method was used in the determination of trace nitrite in water and vegetable samples with satisfactory results.
3.The fixation method of electrode of improved pediatric video electroencephalogram and its applica-tion effect

Objective To determine whether the fixation method of electrode of the improved pediatric video electroencephalogram (VEEG) is better than the traditional fixation method. Methods A total of 746 children patients who needed VEEG monitoring were divided into the experimental group and the control group with random digit table with 373 patients in each group. Improved fixation method and traditional fixation method of electrode were adopted during VEEG monitoring in the experimental group and the control group. The advantage and disadvantage of the two fixation method were compared through evaluation of skin injury of scalp and deltoideus triangularis, electrode loss and electrode breakage. Results The incidence of skin injury of scalp and deltoideus triangularis, electrode loss and electrode breakage in the experimental group were 7, 0, 29, and 12, which were less than 29, 6, 71, and 37 of the control group, the differences were of statistical significance (χ2=14.126, 20.371, 13.652, P<0.01 or 0.05). Conclusions Improved electrode fixation method during pediatric VEEG can avoid skin injury of scalp and deltoideus triangularis, reduce electrode loss and electrode breakage, besides, it also can reduce medical cost and discomfort of children. The modified electrode fixation method proved to be superior to the traditional method and it is worth popularization and application.
4.Inhibitory effect of metformin on TNF-? expression in hepatic tissue of OLETF rats

Objective To investigate the TNF-? expression in hepatic tissue of Otsuka Long-Evans Tokushima Fatty(OLETF) rats(an insulin resistance model) with abnormalities of glucose-lipid metabolism and effect of metformin on TNF-? expression.Methods OLETF rats were randomly assigned to two groups(OLETF and OLETF/M).The OLETF/M group was treated with metformin [100 mg/(kg?d)],the OLETF group and age-matched LETO(nondiabetic Long-Evans Tokushima Otsuka rats) group were fed with placebo.Twenty-two weeks after treatment,we analyzed hepatic triglyceride level and measured expression of TNF-? by western-blot(protein) and quantitative RT-PCR(mRNA).Results Twenty-two weeks after treatment,the level of fasting glucose,insulin,triglyceride,free fatty acids(FFA) and hepatic triglyceride increased significantly in OLETF group as compared with LETO group.Western-blot method showed upregulated expression of TNF-? protein in OLETF group(1.57-fold vs LETO group,P
5.The Effect of Salvia Miltiorrhiza on Gentamicin Ototoxicity-Induced Activity of Nitric Oxide Synthase in Cochlea of Guinea Pig

Objective: Our purpose was to explore the protective role of Salvia Miltiorrhiza (SM) on gentamicin (GM) ototoxicity. Methods: We used NADPH-diaphorase in histochemical staining and image quantitative analysis technique, combined with auditory brainstem response (ABR) measurement to investigate the change of nitric oxide synthase (NOS) activity in cochlea of guinea pig after the injection of GM and SM. Results: Gebtamicin and SM significantly reduced NOS activity in cochlear and the threshold of ABR compared with GM alone (P<0.01); and the threshold of ABR was in high correlation with NOS activity (rGM= -0.8236 ~ -0.8662; rSM+GM= -0.8628 ~ -0.9172, P<0.01). Conclusion: Salvia Miltiorrhiza can reduce NOS activity in cochlea, alleviate GM ototoxicity, and ameliorate hearing function.
6.Localization and expression of three nitric oxide synthase isoforms in the cochlea of guinea pigs and the effects in the hearing process of inner ear

BACKGROUND: Nitric oxide (NO) is synthesized by catalysis of nitric oxide synthase (NOS). Three distinct isoforms of NOS have been detected in different structures of the guinea pig cochlea. However, there are still rather controversies about the definite localization and expression of NOS isoforms in guinea pig cochlea.OBJECTIVE: To investigate localization and expression of three NOS isoforms in the cochlea of guinea pigs, and to explore the effect of NO on auditory physiology and pathophysiology of inner ear.DESIGN: An observed and controlled study.SETTING: Department of Physiology, Jinzhou Medical College; Department of Orthopedics, the Second Affiliated Hospital of Jinzhou; Department of Otorhinolaryngology, Jinzhou Central Hospital.MATERIALS: The study was performed in Hearing Research Laboratory of China Medical University from May to November 2000. Ten healthy male albino guinea pigs, with body mass of 250-300 g, with sensitive Preyer's reflexes and normal drum membrane and external auditory canal,were selected.METHODS: After anesthesia, guinea pigs were decapitated and the temporal bones were removed immediately. The round and oval windows were opened, perfused with 40 g/L paraformaldehyde, and the specimens were immersed in the same fixative solution for 2 hours. Decalcification of the cochleae was performed in 100 g/L EDTA solution for 1 week. Then the specimens were placed in 250 g/L sucrose solution overnight and embedded in OCT. Cryostat (20 μm) sections were prepared for immunohistochemistry.MAIN OUTCOME MEASURES: Localization and expression of three NOS isoforms in the cochlea of guinea pigs.RESULTS: All data of ten guinea pigs was entered the final analysis without any loss. [1] Neuronal-type nNOS and endothelial-type eNOS immunoreactivity were localized in inner and outer hair cells of the organ of Corti, as well as in spiral ganglion cells. In addition, staining for nNOS and eNOS were also seen in the marginal cells of stria vascularis, spiral ligament cells, and in nerve fibers. [2] Inducible-type iNOS immunoreactivity was also detected in above each structure of the guinea pig cochlea under physiological conditions.CONCLUSION:NO may play an important role in neurotransmission,blood flow regulation, and cytotoxicity.
7.Therapeutic effect of indole-3-carbinol on pig serum-induced hepatic fibrosis in rats.

This study is to investigate the therapeutic effect and mechanism of indole-3-carbinol (I3C) on pig serum-induced liver fibrosis of rats. The liver fibrotic model of rats was induced by pig serum. After models were successfully established, rats in the treatment groups were administered with I3C through intraperitoneal injection or curcumin by intragastric administration, daily for 17 days. Hepatic hydroxyproline (Hyp) content was measured. The liver histology and immunohistochemistry with a-smooth muscle actin (alpha-SMA) were assayed. Hepatic stellate cells line, HSC-T6 was incubated with different concentrations of I3C (25, 50, and 100 micromol x L(-1)) for 24 h. The effect of I3C on cell apoptosis was identified by FITC-Annexin V/PI double labeled assay. And the mRNA expressions of Bax and Bcl-2 were measured by real time RT-PCR. The results showed that hepatic content of Hyp decreased by I3C treatment, as compared with the fibrotic model control. Histopathological changes, such as steatosis, necrosis, deposition of collagenous fiber reduced remarkably and the expression of alpha-SMA was significantly down-regulated in the I3C-treated groups (P < 0.01). Apoptosis analysis showed that I3C significantly increased HSC-T6 apoptosis rate and the expressional ratio of Bax to Bcl-2. The results indicated that I3C could effectively cure pig serum-induced liver fibrosis in vivo by inducing HSC apoptosis and promoting ECM degradation.

Objective To study the influence of nitric oxide synthase (NOS) inhibitor in a rat model of Parkinson’s disease. Methods Hemi-Parkinsonism rat model was established by stereotaxic 6-hydroxydopamine (6-OHDA) lesions in striatum in which nitric oxide synthase (NOS) was inhibited by L-Nitro-Arginine (L-NNA) and apomorphin-induced rotational behavior was measured. The immunohistochemical staining method was used to observe the change of striatal nNOS-positive neurons and nigral tyrosine hydroxylase(TH)-positive neurons. Results L-NNA dramatically protected 6-OHDA-injected rats against indices of severe injury to the nigrostriatal dopaminergic pathway, including decreases in numbers of TH-positive nigral neurons and rotational behavior. The nNOS-positive neurons showed no changes in numbers. Conclusions These results indicate that NO might mediate, in part, 6-OHDA-induced neurotoxicity and nNOS-positive neurons might resist the 6-OHDA neurotoxicity. NOS inhibitor may play a role in the protection of 6-OHDA neurons.
10.The value of autofluorescence bronchoscopy in assessment of tumor extent and guide of therapeutic strategy in central lung cancer

Objective To determine the sensitivity of autofluorescence bronchoscopy (AFB) in the assessment of tumor size and therapeutic strategy.Methods Patients with imaging suspected of malignancy were examined with both white light bronchoscopy (WLB) and AFB.The area of tumor infiltration,imaging information and pathological results were analyzed.Results A total of 212 patients were enrolled,including 180 male and 32 female.In 24 patients (13.2%),greater tumor volume was revealed by AFB than by WLB alone.In these patients,the median diameter of tumor was > 1 cm wider on AFB examination than on WLB.Therapeutic strategy was changed in 18 patients (9.9%) after receiving AFB,including 15 patients with expanded scope of removal and 3 patients with avoidance of surgery.In the univariate analysis,the pathological type of squamous cell carcinoma and tumor invasion in two or more segments of bronchus were independent predictive factors.Diagnostic sensitivity of AFB group was 85.7%,specificity 73.3%,positive predictive value 95.1%,false predictive value 45.8%.Diagnostic sensitivity of WLB group was 72.5%,specificity 60.0%,positive predictive value 91.7%,false predictive value 26.5%.Conclusion Our study suggests that compared with WLB alone,autofluorescence bronchoscopy plus WLB significantly improves the diagnostic value and treatment outcome of central lung cancer.
