Objective:To isolate monocytes from human peripheral blood mononuclear cells( PBMC) ,induce macrophages, and identify the function of macrophages. Methods:Monocytes were isolated from PBMC using magnetic activated cell sorting( MACS) anti-CD14 microbead.Sorted CD14+and CD14-cells were checked by flow cytometer to evaluate the efficiency of sorting. The sorted CD14+cells were cultured in IMDM media with 10%human AB serum and 10 ng/ml M-CSF for 7 days to generate macrophages, which were identified by morphological features and phagocytosis function. Results:A high purity of monocytes was obtained by MACS anti-CD14 microbead. The percentage of CD14+cells was 10% and 85.8% before and after sorting, respectively. The macrophages were approximately 40-45 μm in maximum diameter and had the fried egg colony morphological features after 7 days culture. The lymphoma ( Raji) cells were efficiently engulfed by macrophages. Conclusion: The high purity of CD14+monocytes is isolated from PBMC and monocyte-derived macrophages efficiently engulfed lymphoma cells.

Aim To investigate the brain targeting of ginkgolide B prodrug (PGB ) and its mechanism. Methods The liquid chromatography tandem mass spectrometry (LC-MS /MS)method was applied to in-vestigate the pharmacokinetics of PGB in rat brain tis-sue after intravenous injection of PGB.Also the brain targeting was evaluated on the basis of the pharmacoki-netic parameter of PGB.The incomplete cerebral is-chemia model was induced in mouse, the effect of PGB on cerebral capillary permeability was observed by Ev-ans blue method.High performance liquid chromatog-raphy (HPLC)was used to determine the partition co-efficients (logP)of PGB in octanol-water system.PGB and P-glycoprotein (P-gp)was docked by using Mole-gro Virtual Docker (MVD)software to predict its bind-ing abilities with P-gp.The interaction of PGB with ATPase activity of human P-gp membrane was esti-mated by measuring inorganic phosphate liberation. Results The brain targeting of PGB was evaluated by treatment effective (TA ) and drug targeting index (DTI),the calculated value were 6.87 and 4.1 4 re-spectively.Preventive medication of PGB could signifi-cantly decrease cerebral capillary permeability (P <0.05).The lipo-hydropartition coefficient of PGB was higher than that of GB,their logP data were 1 .03 and 0.61 respectively.PGB displayed the stronger binding affinity with P-gp than GB according to the molecular docking calculations, their MolDock Score toward P-gp were -1 43.36 and -1 1 6.40KJ·mol -1 respectively. ATP-hydrolisis showed that PGB increased ATPase activity with a Km of approximately 237.75 μmol · L -1 ,however GB with a Km of approximately 841 .24μmol·L -1 .PGB might interact with P-gp with a high-er affinity and exhibit more effect than GB.Conclu-sion PGB is characterized by its brain targeting. Higher liposolubility of PGB results in good blood-brain permeability, which is advantageous to its brain targe-ting.Besides,PGB can effectively inhibit the efflux effect of P-gp to GB because of its increased P-gp AT-Pase activity.

OBJECTIVETo study on the separation from essential oil-in-water emulsion of Citri Reticulatae Pericarpium Viride by ultrafiltration and acetoacetate extraction methods respectively, and the comparison of the oil yields and chemical compositions.
METHODEssential oil-in-water emulsion of Citri Reticulatae Pericarpium Viride was separated by ultrafiltration and acetoacetate extraction methods respectively, and the chemical compositions were analyzed and compared by GC-MS.
RESULTUltrafiltration method could enrich essential oil more and its chemical compositions were more similar to the essential oil prepared by steam distillation method.
CONCLUSIONUltrafiltration method is a good medium to separate essential oil from essential oil-in-water emulsion of Citri Reticulatae Pericarpium Viride.

Objective To observe the effect of local injections of Pseudomonas Aeruginosa Preparation joint compres-sion bandage in lymph leakage after breast carcinoma operation. Methods A total of 60 patients of lymph leakage after breast carcinoma operation were randomly divided into the observation group and the control group , the pations of ob-servation group was treated with pseudomonas aeruginosa preparation joint compression bandage treatment ,while the pations of control group was treated with the puncture pumping local joint compression bandage treatment , the clinical efficacy of two groups were compared. Results The treatment group cure rate was 76.7%, effective rate was 96.7%, control group 20.0%, 66.7% respectively. The difference was statistically significant (P<0.05).After treatment, KPS score of the observation group was significantly better than the control group, drainage time, hospitalization time was shortened (P<0.05), the expected survival was not significantly different (P>0.05).There were no serious adverse reac-tionsin two groups. Conclusion Pseudomonas Aeruginosa Preparation joint compression bandage has definite clinical effect in lymph leakage after breast carcinoma operation, less adverse reaction.

Objective To analyze the clinical effect of a bifurcated ball frame designed for undergraduate research to assist hand-knee position, avoid physical exertion and increase comfort. Methods Randomized controlled trials consisted of 260 occipital (posterior) primiparous women born during labor, and were randomly divided into the observation group and the control group with 130 cases each. The observing group's mothers were on the yoga ball on the device. The angle and height of the frame and the ball plate were adjusted according to the maternal body type,so that the yoga ball at the chest and the cross could effectively support the maternal weight loss knee burden, and both sides of the small abdominal yoga ball could be effectively posted. After the adjustment was completed, the maternal relaxes and gently shakes the body in one direction so that the small yoga ball on the ventral side forms a continuous soothing push. The control group leaned forward on both knees (with protective gear) and supported themselves with both hands or both fists, swaying the pelvis or crawling forward.Comparing the two groups of vaginal delivery rate, maternal postural maintenance time, maternal dehvery experience, and effective correction rate of occipital position. Results The time of postoperative maintenance in the observation group was (49.51 ±10.62) minutes, and that of the control group was (18.31 ± 9.72) minutes. The difference between the two groups was statistically significant (t=6.69,P <0.05).The effective correction of abnormal fetal position rate, spontaneous delivery rate and comfort rate were 82.30% (107/130),77.69% (101/130),and 99.23% (129/130),respectively in the observation group, and 27.69% (36/130),33.85% (44/130),3.84% (5/130),respectively in the control group, the differences between the two groups were statistically significant (x2 =4.06,4.15,5.61,all P <0.05).Conclusions The undergraduate design of this sort of ball delivery group has a significant effect on the extraversion of the assisted position to the fetal position.
