1.Pathological Significance of CK5 Expression in Primary Cutaneous Amyloidosis

Objective To analyze the pathological significance of CK5 expression in primary cutaneous amyloidosis(PCA).Methods The expression of CK5 in superficial dermis of PCA group and the control group[lichen planus(LP),lupus erythematosus(LE)]were detected by CK5 monoclonal antibody.The infiltration densities of CD3-positive T lymphocytes and CD68-positive macrophages in superficial dermis of PCA and control group were measured respectively by immunohistochemical staining using anti-CD3 antibodies and anti-CD68 antibodies.The skin lesions of PCA and control groups were analyzed by immunofluorescence to detect whether CK5 was phagocytosed by macrophages in superficial dermis.Results Totally,39 cases of PCA all were CK5-positive.Some control cases were positive.The number of CD3-positive T lymphocytes and CD68-positive macrophages in 8 cases of PCA group was lower than that of control group.The result of immunofluorescence colocalization of monoclonal anti-CK5 antibodies and anti-CD68 antibodies in 5 cases of PCA lesions was negative;that in 2 cases of LE lesions were both positive,and that in 2 cases of LP lesions were both negative.Conclusion Amyloid protein may be derived from the basal keratinocytes after interface damage.The amyloid protein deposits may be related to the number decrease or the functional defect of macrophages.
2.Extraction Process Optimization of Fufang Jinsha Lidan Granule by Orthogonal Experiments

Objective:To optimize the extraction technology of Fufang Jinsha Lidan granule. Methods:According to the properties of traditional Chinese medicines in the formula, orthogonal experiments were used to optimize the extraction process of Fufang Jinsha Li-dan granule. The volume of water, boiling time and boiling frequency were used as the three influencing factors with three different lev-els in the orthogonal experiments. Moreover, the content of paeoniflorin and the yield of extract were chosen as the evaluation indices. The orthogonal experiments were carried out according to the L9 (34 ) orthogonal table. Results:The optimal extraction process of Fu-fang Jinsha Lidan granule was as follows:boiled twice, and one hour per time with 12-fold amount of water ( soaked for 30 min with 14-fold amount of water for the first extraction process) . Conclusion:The extraction process has such properties as high extraction rate, stability, simple operation, high yield of extract and controllable quality, which is worthy of wide application.
3.Effect of lncRNA-1700020I14Rikon the fibrosis in mouse mesangial cells in high glucose concentration

Objective To construct the lncRNA-1700020I14Rik plasmid and detect its effect on the fibrosis of mice mesangial cell (MMC) cultured with high glucose medium.Methods RT-qPCR was used to measure the expression of 1700020I14Rik in MMC cultured with low glucose medium or high glucose.Total RNA was extracted from MMC and cDNA was got by RT-PCR.The whole fragment of lncRNA-1700020I14Rik amplified by PCR was constructed into plasmid pcDNA3.1(+) through PCR.Lipidosome 3000 was used to transfect the plasmid into the MMC cultured with high glucose medium and RT-qPCR was used to measure the expression level of 1700020I14Rik.Western blot was used to analyze the expression of fibronectin, collagen Ⅳ and TGF-β1.Results 1700020I14Rik was significantly down-regulated in MMC cultured with high glucose and it was significantly up-expressed in the MMC after transfecting with pcDNA3.1(+)-1700020I14Rik.The expressions of fibronectin, collagen Ⅳ and TGF-β1 were down-regulated by 1700020I14Rik.Conclusions The plasmidpcDNA3.1(+)-1700020I14Rik is able to effectively express the lncRNA-1700020I14Rik.Over-expression of 1700020I14Rik may protect mesangial cells from fibrosis conduced in high glucose medium.
4.Analysis of symptoms and quality of life in children with allergic rhinitis.

OBJECTIVE:
To evaluate symptoms and quality of life in children with allergic rhinitis, and to evaluate the correlations among the skin prick tests results, symptoms and quality of life.
METHOD:
Visual analog scale (VAS) was used to assess the symptoms severity, rhinoconjunctivitis quality of life questionnaire (RQLQ) was used to assess quality of life, the skin prick tests results were recorded, and statistics analysis was carried out among them.
RESULT:
(1) There were significant differences among the five aspects of RQLQ (F = 32.03, P < 0.01), nasal symptoms aspect was the most affected aspect of quality of life, moreover, there were significant differences among all the items of RQLQ (F = 7.35, P < 0.01), the scores of nasal blockage, sneezing and rhinorrhea were the highest, there were no significant differences among them. (2) VAS was significantly correlated with RQLQ, moreover, the scores of unable to get to sleep and embarrassed by symptoms were significantly correlated with rhinorrhea VAS scores (r = 0.230, P < 0.05; r = 0.325, P < 0.01), the score of wake up during the night was significantly correlated with nasal itching and sneezing VAS scores (r = 0.385, P < 0.01; r = 0.231, P < 0.05), the score of can't concentrate was significantly correlated with the scores of unable to get to sleep and wake up during the night (r = 0.316, P < 0.01; r = 0.525, P < 0.01), the score of irritable was significantly correlated with the scores of unable to get to sleep (r = 0.243, P < 0.05). (3) The results of the skin prick tests were related with VAS and RQLQ.
CONCLUSION
Nasal symptoms severity may significantly correlated with the quality of life. Rhinorrhea, nasal itching and sneezing may be the main factors affecting the quality of sleep, while the quality of sleep may affect non-hay-fever symptoms and emotions, rhinorrhea may be the main factor affecting embarrassed by symptoms. The allergen level was related with the symptoms severity and quality of life.

5.Construction and application of traditional chinese medicine evidence-based big data platform

Objective Build an evidence-based big data governance platform centered around intelligent Traditional Chi-nese Medicine(TCM),incorporating TCM characteristics,to promote the development of intelligent TCM healthcare,and en-hance the efficiency of medical research and practical applications..Methods We integrate hospital TCM diagnosis and treat-ment process data and other kinds of clinical data..And we employ artificial intelligence and big data technologies to effectively clean and standardize the data,among other data governance and data preparation tasks,in order to build a Chinese Medicine Clinical Data Center System.Results The platform has achieved the collection and governance of millions of real medical re-cords,providing strong support for health management and clinical research in TCM,particularly promoting research in TCM's syndrome differentiation and treatment,prescription research,and drug reactions.Conclusion By constructing a big data gov-ernance platform based on Traditional Chinese Medicine(TCM),we have achieved value-added effective management and utili-zation of TCM data,particularly enhancing the precision of TCM diagnosis and treatment and the speed of research output,thus driving the modernization process of TCM.
6.Risk factors of extrauterine growth retardation in very preterm infants with birth weight less than 1 500 g

Objective:To study the risk factors of extrauterine growth retardation (EUGR) during hospitalization in very preterm infants (VPIs) with birth weight (BW) <1 500 g.Methods:From Jan 2015 to Dec 2020, clinical data of VPIs admitted to neonatal department our hospital were retrospectively studied. The infants were assigned into EUGR group and non-EUGR group according to their weight at discharge. Multivariate logistic regression analysis was used to analyze the risk factors of EUGR in VPIs.Results:A total of 969 VPIs were enrolled, including 400 cases of EUGR (41.3%). Multivariate logistic regression analysis showed that Z-score of BW ( OR=0.057, 95% CI 0.037-0.088, P<0.001) was closely correlated with the occurrence of EUGR and growth velocity (GV) after regain BW ( OR=0.537, 95% CI 0.479-0.602, P<0.001) was a protective factor for EUGR. Maternal hypertension during pregnancy ( OR=1.895, 95% CI 1.059-3.394, P=0.031), asphyxia at birth ( OR=2.508, 95% CI 1.265-3.347, P=0.004) and moderate to severe bronchopulmonary dysplasia (BPD) ( OR=2.660, 95% CI 1.503-4.708, P=0.001) were risk factors for EUGR at discharge. Conclusions:EUGR is still common in VPIs. Increased GV after regain BW, prevention and treatment of moderate to severe BPD may reduce the incidence of EUGR at discharge in VPIs.
7.The status and development of acupuncture in northern Thailand.

To introduce the status and development of acupuncture in Thailand, and elaborate on the development of acupuncture education and acupuncture clinics in northern Thailand. It is pointed out that there are still a small number of Chinese medicine clinics in northern Thailand, a shortage of college acupuncturists, and there is no traditional Chinese medicine themes Confucius Institute and other issues. The author considered that through the cultivation of acupuncture skills and diagnosis and treatment ability, the development of Chinese medicine graduate education, the establishment of Confucius Institute of traditional Chinese medicine, to expand the influence of acupuncture in northern Thailand, hoping to provide references for the cooperation and exchanges between China and Thailand in acupuncture .

Dysfunction of the Hippo pathway enables cells to evade contact inhibition and provides advantages for cancerous overgrowth. However, for a significant portion of human cancer, how Hippo signaling is perturbed remains unknown. To answer this question, we performed a genome-wide screening for genes that affect the Hippo pathway in Drosophila and cross-referenced the hit genes with human cancer genome. In our screen, Prosap was identified as a novel regulator of the Hippo pathway that potently affects tissue growth. Interestingly, a mammalian homolog of Prosap, SHANK2, is the most frequently amplified gene on 11q13, a major tumor amplicon in human cancer. Gene amplification profile in this 11q13 amplicon clearly indicates selective pressure for SHANK2 amplification. More importantly, across the human cancer genome, SHANK2 is the most frequently amplified gene that is not located within the Myc amplicon. Further studies in multiple human cell lines confirmed that SHANK2 overexpression causes deregulation of Hippo signaling through competitive binding for a LATS1 activator, and as a potential oncogene, SHANK2 promotes cellular transformation and tumor formation in vivo. In cancer cell lines with deregulated Hippo pathway, depletion of SHANK2 restores Hippo signaling and ceases cellular proliferation. Taken together, these results suggest that SHANK2 is an evolutionarily conserved Hippo pathway regulator, commonly amplified in human cancer and potently promotes cancer. Our study for the first time illustrated oncogenic function of SHANK2, one of the most frequently amplified gene in human cancer. Furthermore, given that in normal adult tissues, SHANK2's expression is largely restricted to the nervous system, SHANK2 may represent an interesting target for anticancer therapy.
