1.Conventional Segmentation and Large Fractionated Three-dimensional Conformal Radiotherapy in the Treatment of Non Small Cell Lung Cancer with Superior Vena Cava Syndrome

Objective To explore the efficacy and safety of large and conventional fractionated three dimensional conformal radiation therapy for non small cell lung cancer with superior vena cava compression syndrome, and to provide clinical reference for improving the prognosis of patients with non small cell lung cancer. Methods The general data of 63 patients with non small cell lung cancer complicated with superior vena cava syndrome treated in Lu'an Hospital of Traditional Chinese Medicine from January 2010 to December 2012 were analyzed retrospectively. According to the different treatment methods, the patients were divided into observation group ( n =28) and control group ( n =35) .The control group was given conventional fractionated three-dimensional conformal radiation therapy, three dimensional conformal radiation therapy was given to the observation group. The clinical efficacy, toxicity, and 1, 2 years survival rate of the two groups were compared. Results The complete remission rate of observation group was 96.43%, which was significantly higher than 74.29%in the control group (P<0.05) . The complete remission in the observation group was 89.29%in two weeks, which was significantly higher than 62.86%in the control group (P<0.05) . The total effective rate of observation group was 85.71%, which was significantly higher than that 62.86%in the control group (P<0.05) . The clinical benefit rate of observation group was 96.43%, and there was no significant difference between the two groups (P>0.05) . There was no significant difference in the incidence of adverse reactions between the two groups (P>0.05) . There was no significant difference in 1, 2 year survival rate between the two groups (P>0.05) . Conclusion Three dimensional conformal radiation therapy for non small cell lung cancer patients with superior vena cava syndrome is superior to conventional treatment, and does not increase the toxic side effects, which is worthy of clinical attention.
2.Effect of Aitongxiao Granules Containing Serum on Proliferation and Apoptosis of SMMC-7721 and Bel-7404 Cells in Vitro

Objective To observe the anti-cancer effect in vitro of Aitongxiao Granules containing serum on SMMC-7721 and Bel-7404 liver cancer cells, thus revealing the anti-cancer mechanism of the treatment in adjusting proliferation and apoptosis. Methods Male Sprague-Dawley rats were administered by gastrogavage Aitongxiao decoction of 86.4, 43.2, 21.6 g/kg twice a day for 1 week. Using serum pharmacologic method, the proliferation of SMMC-7721 and Bel-7404 liver cancer cells were determined by MTT chromatometry after co-cultured with medicated serum containing different concentration of Aitongxiao decoction. The apoptosis of SMMC-7721 and Bel-7404 liver cancer cells were detected by flow cytometry. Results The research in vitro showed that Aitongxiao Granules containing serum low, medium and high dose groups had varying degrees of inhibition on hepatoma cells, and this inhibition showed a concentration dependence within a certain range. Aitongxiao Granules containing serum could induce apoptosis of human hepatoma SMMC-7721 cells and Bel-7404 cells. Conclusion Aitongxiao Granules showed effect of inhibiting proliferation and inducing apoptosis on SMMC-7721 and Bel-7404 cells in vitro.
3.Application value of fast blood glucose meter in emergency inspection of blood glucose

Objective To explore the clinical application value of fast blood glucose meter in the emergency test of blood glu‐cose .Methods 186 critical patients in or hospital from August 2013 to August 2014 were selected for conducting the clinical re‐search .The average blood sugar was detected in all the research subjects by using the fast blood glucose meter and the fully auto‐matic biochemical analyzer .Results The consistency rate of detection results by the two kinds of detection method reached 22 .04% ,the difference of data was relatively small .In addition ,the detection time for the fast blood glucose meter was significantly shorter than that of the fully automatic biochemical analyzer ,the difference showed the statistical significance (P< 0 .05) .Conclu‐sion The detection results of the fast blood glucose meter have no too large difference with those of fully automatic biochemical an‐alyzer and have the same guidance significance to the diagnosis and treatment .The fast blood glucose meter has the advantages of quickness and convenience ,short time‐consuming and can be used in emergency .
4.Extraction Process Optimization of Fufang Jinsha Lidan Granule by Orthogonal Experiments

Objective:To optimize the extraction technology of Fufang Jinsha Lidan granule. Methods:According to the properties of traditional Chinese medicines in the formula, orthogonal experiments were used to optimize the extraction process of Fufang Jinsha Li-dan granule. The volume of water, boiling time and boiling frequency were used as the three influencing factors with three different lev-els in the orthogonal experiments. Moreover, the content of paeoniflorin and the yield of extract were chosen as the evaluation indices. The orthogonal experiments were carried out according to the L9 (34 ) orthogonal table. Results:The optimal extraction process of Fu-fang Jinsha Lidan granule was as follows:boiled twice, and one hour per time with 12-fold amount of water ( soaked for 30 min with 14-fold amount of water for the first extraction process) . Conclusion:The extraction process has such properties as high extraction rate, stability, simple operation, high yield of extract and controllable quality, which is worthy of wide application.
5.Association of serum testosterone with lean body mass, body fat content, and bone mineral density in postmenopausal females

Objective To determine the relationship between serum testosterone level and lean body mass, body fat content, and bone mineral density (BMD) . Methods The study involved 185 healthy females in Changsha, aged 45 ~81. Fasting serum testosterone was measured by radioimmu-noassay. Hologic QDR 4500A fan beam X-ray bone densitometer was used to measure the BMD of anteroposterior lumber (AP, L_(1~4)) and total hip, to measure the bone mineral content, BMD, body fat content and muscle tissue weight of head, trunk, ribs, pelvis, spine, upper limbs, lower limbs and the total body. Body weight, lean body mass and body fat percentage were calculated. SPSS 11.0 software was used to conduct regression analysis. Results (1) Serum testosterone showed no correlation with lean body mass, body fat content, and body fat percentage. (2) Serum testosterone was positively related with the BMD of lumbar spine and hip, but showed no correlation with the BMD after adjustment of age and years since postmenopause. (3) Lean body mass showed significant positive correlation with the BMD of different sites. Total body fat content showed positive correlation with the BMD of total hip, while body fat percentage showed negative correlation with the BMD of the whole body. Conclusion Keeping lean body mass benefits postmenopausal women to maintain bone mineral content, and taking androgen should still be cautious.
6.Study on Sterility Test after Using Non-PVC Bivalve Soft-bag Injection in PIVAS

OBJECTIVE:To study sterility test after using Non-PVC bivalve soft-bag injection in PIVAS. METHODS:The test was divided into 3 groups according to the type of transfusion solution packaging and dispensing environment. Group 1 received Glucose solution using bivalve soft-bag,dispensed in PIVAS;group 2 received Glucose solution using bivalve soft-bag,dispensed in wards area;group 3 received Glucose solution using plastic bottle,dispensed in wards area. After puncturing 1,3,6,9 times (n=80),finished products placed in ward for 0,2,4,6 h(n=20),and then sterility test was conducted with membrane filtra-tion method stated in second part of Chinese Pharmacopoeia (2010 edition). Infusion contamination of 3 groups was analyzed at 9th puncture. RESULTS:The growth of bacteria was not found in group 1;the positive detection rate of group 2 and 3 were 2.5%and 3.8%(n=320). The total positive detection rates after puncturing 1,3,6,9 times were 0,0.4%,0.4%,7.5%(n=240);the positive detection rates of group 1 were all 0,those of group 2 were 0,1.25%,0,8.75%and those of group 3 were 0,0,1.25%, 13.75%(n=80). After 9 times of puncture,the positive detection rates of group 1 after placing 0,2,4,6 h were all 0,those of group 2 were 25%,5%,0,5%;those of group 3 were 5%,15%,5%,30%(n=20). CONCLUSIONS:The use of the Non-PVC bi-valve soft-bag injection in PIVAS can effectively prevent microbial contamination.
7.Changes of chemical components and anti-tumor activity of total ginsenosides from ginseng stems and leaves tranformed by submerged fermentation of ganoderma lucidum

Objective:To study the biotransformation rule of the chemical components of total ginsenosides from ginseng stems and leaves tranformed by submerged fermentation of ganoderma lucidum,and to lay the foundation for further development and research of the medicinal fungus of ginseng stems and leaves.Methods:The total ginsenosides from ginseng stems and leaves were fermented by ganoderma lucidum,the changes of chemical components before and after fermentation were measured by thin-layer chromatography;the levels of total ginsenosides from ginseng stems and leaves before and after fermentation were detected by ultraviolet spectrophotometry;the levels of ginsenoside Rb1,Rg3,Rd,CK,Re,Rg,and Rh1 before and after fermentation were measured by high performance liquid chromatography.The human hepatocellular carcinoma SMMC-7721 cells were divided into blank control group,low,medium and high doses of total ginsenosides from ginseng stems and leaves groups and medicinal fungus of ginseng stems and leaves groups.The dosages of total ginsenosides from ginseng stems and leaves and medicinal fungus of ginseng stems and leaves were 5,10 and 20 mg·L-1,respectively.The inhibitory rate(IR) of the cells was determined by MTT assay.Results:The results of thin-layer chromatography showed that the saponins before and after fermentation occured mutual transformation;the levels of total ginsenosides from ginseng stems and leaves before and after fermentation were 749.98 and 602.26 mg·g-1,respectively;the levels of panaxadiol saponins Rb1,Rg3,Rd and CK before fermentation were 24.54,2.21,87.22 and 0 mg·g-1,and the levels of panaxatriol saponins Re,Rg and Rh1 were 151.34,77.02,and 3.06 mg·g-1.After fermentation,the level of Rb1 was decreased by 61.45%,the levels of Rg3,Rd and CK were increased by 238.91%,34.43% and 268.00%.The levels of Re and Rg1 were decreased by 63.75% and 64.41%,and the level of Rh1 was increased by 408.88%;the IR of the SMMC-7721 cells in medium dose of medicinal fungus of ginseng stems and leaves group was higher than that in medium dose of total ginsenosides from ginseng stems and leaves group(P<0.05);the IR of the SMMC-7721 cells in high dose of medicinal fungus of ginseng stems and leaves group was higher than that in high dose of total ginsenosides from ginseng stems and leaves group(P<0.01).Conclusion:The total ginsenosides from ginseng stems and leaves transformed by the submerged fermentation of ganoderma lucidum can significantly change its chemical components,which have stronger anti-proliferation activity in the SMMC-7721 cells.
8.High efficient expression of human endostatin in E.coli and its antiangiogensis activity

AIM: To examine the expression of human endostatin in E.coli , produce its fusion protein antibody and observe its biological activity. METHODS: Endostatin gene was amplified by polymerase chain reaction,recombined with plasmid vector pGEX-2T and induced expression with IPTG.The protein activity was tested by endothelial cell proliferation inhibitory assay.Inclusion body crudely purified was used to generate polyclonal antibody to detect its expression at mouse's liver and kidney etc. RESULTS: The protein expressed was 20 kD after digestion by thrombin,it appeared the anti-angiogenesis activity and Western blotting indicated the expression of endostatin in liver and kidney of mouse. CONCLUSION: The successful expression of human endostatin and the preparation of polycolonal antibody indicated its potential application in anti-angiogenesis therapy and diagnosis tumors.
9.Magnetic Beads-Based Biological Mass Spectrometry Technology for Evaluating Therapeutic Efficacy of Acute Leukemia

A serum method for evaluating therapeutic efficacy of acute leukemia has been built by magnetic beads-based biological mass spectrometric technology. After analysis of quality control sample, the reproduci-)bility) of within-run assay and between-run assay was obtained and consistent with the previous reports. This result) manifested that the method used for serum peptides analysis was stable and reliable. Serum samples from 30 patients with primary acute leukemia and those with complete remission after treatment were extracted by metal-chelated magnetic beads and detected by MALDI-TOF-MS. Peaks at the m/z range 1000-10000 were analyzed by FlexAnalysis 2.4 software and Biosun_MS software. Approximately 27 differential peaks that were of statistical significance(p<0.05) were obtained. Ten peaks were up-regulated) and 17 peaks were down-regulated) after complete remission, respectively. The diagnostic model based on these differential peaks achieved) 100% sensitivity and 90% specificity. The results indicated that the model based on magnetic beads extraction and MALDI-TOF-MS detection acquires high sensitivity and specificity. Further sequence identification of differential) peaks is useful for evaluation of therapeutic efficacy and mechanism research on acute leukemia.)
10.Research on the effects of sucking lollipops and pacifier on gastrointestinal function in infants after abdominal surgery

Objective To investigate the effects of early sucking a lollipop and pacifier to improve the recovery of gastrointestinal function in infant after gastrointestinal operation. Methods The infants after gastrointestinal operation were divided into four groups according to the random number table method, namely group Ⅰ, Ⅱ, Ⅲ and Ⅳ. Four groups were given different interventions in addition to routine nursing care after anesthesia awake, group Ⅰsucked a lollipop,while groupⅡsucking pacifier. Intervention time of 2 groups were 20-30 minites. GroupⅢsucked the pacifier 20-30 minites first, then sucked a lollipop 20-30 minites. Each group was given one intervention every 4 hours until infant eated. Group Ⅳ was given no interventions. Bowel sounds recovery time, first bowel movement time for the first time were compared. SPSS 17.0 software was used for statistical analysis. Results A total of 82 infants completed the clinical observation. Bowel sounds recovery time of group Ⅰ,Ⅱ,Ⅲ were (28.54±9.93), (34.10±9.43), (28.50±9.52) h, while first bowel movement time were (50.02±11.60), (57.65±15.28), (49.97±15.46) h, which were earlier than group Ⅳ whose bowel sounds recovery time and first bowel movement time were (39.55 ±11.22) h, (65.43 ±14.78) h. The bowel movement time and first bowel movement time of group Ⅲ were earlier than that of groupⅡ (P<0.05), but groupⅠandⅢhad no obvious difference (P>0.05). Conclusions Infants early sucking a lollipop and pacifier after gastrointestinal operation help gastrointestinal function recovery, but the lollipop is superior to the pacifier, lollipops and no interaction between pacifier.
