1.The Promotion of Setting up Analysis Technique of Molecular Biology on the Experimental Skills in Laboratory Medicine

To adapt the developing requirement of clinical laboratory diagnostics,the Analysis Technique of Molecular Biology was set up for Grade 2003 in laboratory medicine in our college.The students showed great interests and enthusiasm in this subject,especially in laboratory course.The studentsof Grade 2003 made a better score than those of Grade 2002 in the final Test of Basic Experimental Skills,which included the scores of basic operation,experiment theory and technical skill.These results showed that setting up the Analysis Technique of Molecular Biology could improve student's ability in experimental skills.
2.Distribution of multiple myeloma literature in Chinese periodicals

Objective To analyze the distribution of multiple myeloma (MM) literature in Chinese periodicals and to find out the core periodicals about MM.Methods Periodical literature that involved with MM in the article titles from January 2008 to December 2014 on the Wanfang medical database was collected and searched.Bibliometric method was used to analyze journal date and journal distribution.Results 456categories of periodicals had published literature about MM.The number of articles about MM published in the top 10 journals was more than 30,accounting for 27.00 % of all published MM literature,which were highly effective core journals.The number of articles about MM published in the top 3 journals was more than 100,accounting for 15.59 % of all published MM literature.The number in Journal of Leukemia & Lymphoma had reached 187,accounting for 6.43 % of the total.Conclusions The core journals about MM provide the key approach to help MM medical personnel to select useful information efficiently.Furthermore,the core journals are not only the significant gist for authors who plan to contribute their articles to relevant MM periodicals,but also the necessary reference tools for medicine workers who engage in basic and clinical research and teaching.
3.Effect of inhibition of hPOT1 expression mediated by siRNA on Caspase-3 in HeLa cells

Aim To inhibit the expression of human protection of telomere gene hPOT1 in HeLa cells using vector based on RNA interference(RNAi) technique and to investigate its effects on the activation of Caspase-3 in HeLa cell.Methods Three recombinant plasmids(our group constructed before) containing different hPOT1 target sequences(pmU6-shRNA1,pmU6-shRNA2 and pmU6-shRNA3)were transfected into HeLa cells by liposomal.The expression inhibition of hPOT1 was detected by RT-PCR and EMSA.The expression and activation of Caspase-3 were inspected by RT-PCR,Western blot analysis and Caspase-3 kit.Results After 48 hour′s transfecting,hPOT1 mRNA and protein level in HeLa cells reduced and the expression of Caspase-3 mRNA had no change.Caspase-3 zymogen decreased but its activity increased.Conclusions hPOT1 expression in HeLa cells can be significantly inhibited by using siRNAvectored RNAi and the down-regulation of hPOT1 expression can activate Caspase-3.
4.Influence of the nitric oxide-donor of sodium nitroprusside on the expression of hippocampal neurons gene cpp32

BACKGROUND: Alzheimer disease is a senile degenerative disease of nervous system. Neuron apoptosis is regarded as one of possible reasons,and neuron culture in vitro is a common method to research the mechanism of apoptosis.OBJECTIVE: To observe the influences of nitric oxide-donor, sodium nitroprusside (SNP), on the expression of gene cpp32 in the cultured hippocarnpal neurons in vitro.DESIGN: A randomized controlled animal experiment. SETTING: Institute of Biochemistry and Molecular Biology in Guangdong Medical College.MATERIALS: The experiment was carried out in the Institute of Biochemistry and Molecular Biology, Guangdong Medical College, and newborn (< 24 hours) Sprague-Dawley rats were used.METHODS: The hippocampl neurons of rats were primarily cultured, and then treated with SNP of different terminal concentrations (0, 25, 50, 100,200, 400 μmol/L) for 24 hours, and the expressions of mRNA and protein were analyzed with RT-PCR and Western blot respectively. The hippocampl neurons of rats were treated with SNP of different terminal concentrations (0, 25, 50, 100, 200, 400 μmol/L) for 12 hours, and the activity of CPP32 enzyme was detected with CPP32 activity detected kit.MAIN OUTCOME MEASURES: The expressions cpp32 mRNA and CPP32 protein and activity of CPP32 were detected.RESULTS: The cpp32 mRNA expression was unchanged as the increasing dose of SNP, but the pro-CPP32 was activated and the activity of CPP32 was increased significantly at 50 μmol/L SNP which was 3.02 times of that in the control group, and reached to the maximal value at 100 μmol/L which was 3.47 times of that in the control group.CONCLUSION: SNP cannot increase the cpp32 mRNA expression, but can increase degradation of pro-CPP32 and activate CPP32.
5.Research in correlation between quality of life and acceptance of disability of colostomy patients

Objective To explore the relationship between quality of life and acceptance of disability of colostomy patients.Methods Using convenience sampling method to investigate 111 colostomy patients.General information questionnaire,QLQ-C30 scale,QLQ-CR38 scale and Acceptance of Disability Scale were used to investigate patients' general condition,quality of life and acceptance of disability.Data was analyzed by SPSS 17.0.Results The general health condition of colostomy patients was better than reference value and the score of ADS was at an average level.There was a relationship between general health condition,functioning dimensions,symptom dimensions and acceptance of disability.Conclusions There was a close relationship between quality of life and acceptance of disability.The acceptance of disability should be improved to help patients to obtain better quality of life.

The‘eighteen clashes’and‘nineteen fears’have been tought of traditional prohibited combination in long time,but many examples show ginseng and trogopterus dung could be safely used after compatibility,so many scholars fouce on the essential of compatibility.While people can not reach an accordant consideration for compatibility of Ginseng with Trogopterus Dung,therefore we have a overview based on documents for about 30 years,which will provide evidences for researching systematically on the nature of compatibility of Ginseng with Trogopterus Dung.
7.Experimental investigation on tumorigenicity of Vero cell in the process of passage

Objective To produce an experimental information for the safety assessment of Vero cells during subculture. Methods Passage and freeze on Vero cells, and Vero cells in different passages in vitro and in vivo tumorigenicity were tested. The protein expression of different Vero cell passages was analyzed. Results Vero cells passaged to p270 and 14 cell banks were developed and stored for future evaluation. In vitro and in vivo tumorigenicity Lest results of Vero cells in different passages were negative. Conclusion Although the tumorigenicity test results in vitro and in vivo process were negative, the protein expression of more than p200 Vero cells were changed, which produced the experimental reference for the safety evaluation of the process during the Vero cell passage.
8.Magnetic Beads-Based Biological Mass Spectrometry Technology for Evaluating Therapeutic Efficacy of Acute Leukemia

A serum method for evaluating therapeutic efficacy of acute leukemia has been built by magnetic beads-based biological mass spectrometric technology. After analysis of quality control sample, the reproduci-)bility) of within-run assay and between-run assay was obtained and consistent with the previous reports. This result) manifested that the method used for serum peptides analysis was stable and reliable. Serum samples from 30 patients with primary acute leukemia and those with complete remission after treatment were extracted by metal-chelated magnetic beads and detected by MALDI-TOF-MS. Peaks at the m/z range 1000-10000 were analyzed by FlexAnalysis 2.4 software and Biosun_MS software. Approximately 27 differential peaks that were of statistical significance(p<0.05) were obtained. Ten peaks were up-regulated) and 17 peaks were down-regulated) after complete remission, respectively. The diagnostic model based on these differential peaks achieved) 100% sensitivity and 90% specificity. The results indicated that the model based on magnetic beads extraction and MALDI-TOF-MS detection acquires high sensitivity and specificity. Further sequence identification of differential) peaks is useful for evaluation of therapeutic efficacy and mechanism research on acute leukemia.)
9.An analysis of hyperinsulinemia in Bartter syndrome

Objective To analys hyperinsulinemia in Bartter syndrome. Methods Twenty-three cases of Bartter syndrome [age (27 ±9) years;fasting serum potassium(2. 8 ±0. 5)mmol/L], 20 patients of aldosterone-producing adenoma [APA, age (45 ± 11 ) years, fasting serum potassium ( 3.0 ± 0. 4 ) mmol/L], 20 patients of idiopathic hyperaldosteronism [IHA, age (51 ± 11 ) years, fasting serum potassium (3.4 ±0. 2)mmol/L] were diagnosed in Peking Union Medical College Hospital from September 2003 to May 2008. All patients underwent 3-hours oral glucose tolerance test(3hOGTT), postural stimulation test and calculated HOMA-insulin resistance ( HOMA-IR ) and HOMA-insulin sensitivity ( HOMA-IS ) by Homeostasis model.Results The insulin area under curve-(229.0±162.4)mIU·L-1·h] was singnificantly higher than APA group [(227.7±158.6)mIU·-1·h].But HOMA-IR in Bartter group were similar to APA group( 1.96 ± 1.14 vs 1.41 ± 0. 91 ), and HOMA-IR in APA group was lower than IHA group ( 1.96 ± 1.14 vs 2.40 ± 1.60, P < 0. 05 ). There was no deference in HOMA-IS among three groups,but APA group had lower level. In all three groups, the peak of insulin secretion was delayed. Conclusion Bartter syndrome patients commonly present with hyperinsulinemia.
10.The effects of morphine on serum cortisol and catecholamine in premature infants receiving mechanical ventilation

Objective To study the effects of morphine on serum concentrations of cortisol (COR), adrenaline (ADR), noradrenaline (NAD) and dopamine (DOP) in premature infants requiring mechanical ventilation (MV).Method From January to November, 2017, premature infants (gestational age: 28~36 weeks and body weight :1 000~2 499 g) admitted to neonatal department of our hospital and received mechanical ventilation were prospectively studied.The participants were randomly assigned into morphine group and control group.Modified neonatal pain assessment scale ( NIPS ) and Ramsay scores were calculated at the beginning of MV (before the administration of morphine ) and at different time points after MV (2 h, 12 h, 24 h and 48 h after the administration of morphine ).Serum concentrations of COR , ADR, NAD and DOP were also measured at the beginning of MV and 24 h after MV/morphine.The scores and concentrations were compared between the two groups.Result A total of 60 infants were included, with 30 infants in each group.The modified NIPS and Ramsay scores and serum concentrations at the beginning of MV were similar between the two groups ( P>0.05).The modified NIPS and Ramsay scores at 2 h, 12 h, 24 h and 48 h after the initiation of MV were significantly lower in the morphine group comparing with the control group[2 h: (0.8 ±0.5) vs.(2.9 ±0.7), (0.9 ±0.6) vs.(2.3 ±0.6); 12 h: (1.1 ±0.6) vs.(2.8 ±0.8), (1.3 ±0.7) vs.(2.3 ±0.8); 24 h: (1.4 ±0.8) vs.(3.0 ±0.7), (1.6 ±0.6) vs. (2.4 ±0.8); 48 h: (1.3 ±0.7) vs.(2.7 ±0.9), (1.7 ±0.7) vs.(2.5 ±0.8)].Furthermore, serum concentrations of COR, ADR, NAD and DOP at 24 h after MV were also significantly lower in the morphine group [COR: (165.4 ±25.0) nmol/L vs.( 319.8 ±71.2) nmol/L, ADR: ( 30.3 ±4.5) ng/L vs. (45.4 ±4.3) ng/L,NAD: (138.0 ±16.5) ng/L vs.(230.5 ±26.1) ng/L, DOP: (43.6 ±7.2) ng/L vs.(86.3 ±10.0) ng/L, P<0.05].Conclusion Morphine is associated with lower serum concentration of catecholamine and lower NIPS scores during MV in premature infants.
