1.Application status of odds ratio for medical animal experiments

Aim To study the application status of odds ratio for medical animal experiments.Methods Odds ratio and seven kinds of animal models were used as retrieval strategy to search medical animal experiment related papers in several Chinese and English databases.Papers relating to each kind of animal model and using odds ratio in abstract and text were counted. Data from different databases were compared. Calculation of odds ratio was exemplified and the significance of different odds ratio values was illustrated in this paper.Results Few medical animal experiments cited odds ratio as statistics.Conclusions The importance of odds ratio has not been fully recognized in Chinese references.
2.Clinical and hematological features of malignant lymphoma com-bined with venous thromboembolism

Objective: To investigate clinical and hematological features of malignant lymphoma (ML) combined with venous thromboembolism (VTE). Methods:The clinical data of 65 patients with ML combined with VTE were collected between October 2010 and April 2014 and then retrospectively reviewed. Coagulation function and hemorheology were analyzed. Results:The proportion of males and females among patients with ML combined with VTE was 2.61:1. ML combined with VTE was mainly observed in patients with end-stage cancer. The highest incidence (81.54%) of ML combined with VTE was found in StageⅢB-Ⅳtumor. Among 65 patients with VTE, 43 (66.15%) were found after ML was diagnosed, 55 (84.62%) were deep vein thrombosis (DVT), 7 (10.77%) were pulmonary embolism (PE), and 3 (4.62%) were complicated with both DVT and PE. Common DVT locations were the upper limb and the neck vein of 37 cases (67.27%). Clinical manifestations of ML combined with DVT are limb pain, swelling, and increased skin temperature. Unexplained difficulty in breathing, chest pain, and syncope are also found in ML combined with PE. The total effective rate of 55 cases with DVT was 49.09%, and the rate of those cases with PE was 14.29%. The levels of platelet aggregation, D-dimer, high cut blood viscosity, low cut blood viscosity, plasma viscosity, hematocrit, assembly index of erythrocyte, and rigidity index of erythrocyte were significantly higher in patients with ML combined with VTE than in patients with ML alone;by contrast, APTT, erythrocyte sedimentation rate, deformability index of erythrocyte, and average velocity of blood flow were reduced in patients with ML combined with VTE than in patients with ML alone. Conclusion:DVT, which exhibits high incidence in males, is the most common type observed in ML combined with VTE. DVT is commonly manifested in the upper limb and the neck vein;furthermore, DVT is mainly observed in patients with end-stage cancer. Hematological indexes indicated changes in the sensitivity state of thromboembolism.
3.Effect of stellate ganglion block combined with morphine sustained-release tablets for controlling pain of advanced lung cancer

Objective To investigate the effect of stellate ganglion block combined with morphine sustainedrelease tablets for controlling pain of advanced lung cancer.Methods 90 cases with advanced lung cancer were randomly divided into the observation group and control group with 45 cases in each group.The control group was givenmorphine sustained-release tablets,the observation group received combination therapy with stellate ganglion block and morphine sustained-release tablets.The analgesic effects,quality of life scores and adverse reactions of two groups were compared.Results The effective rate of observation group was 93.33 % (42/45),which was significantly higher than that of control group 77.78% (35/45),the difference was statistically significant (x2 =4.54,P < 0.05).The morphine consumption of observation group was significantly less than that of control group(t =9.11,P < 0.05),duration of analgesia was significantly longer than that of control group (t =6.72,P < 0.05).Quality of life score in the observation group was significantly higher than that in the control group,the difference was statistically significant(Z =3.61,P < 0.05).The incidence rate of adverse reactions was 17.78% (8/45),which was significantly lower than that of the control group 46.67% (21/45),the difference was statistically significant (x2 =4.54,P < 0.05).Conclusion Stellate ganglion block combined with morphine sustained-release tablets for controlling pain of advanced lung cancer can significantly improve analgesia,reduce morphine consumption and adverse reactions,is worthy of clinical application.
4.Construction of recombinant adenovirus vector carrying human miR-133a and its expression in vitro

AIM:To construct recombinant adenovirus vector carrying human miR-133a and study its expression in human mesenchymal stem cells(hMSCs).METHODS:The PCR product containing miR-133a was amplified from human genomic DNA and inserted into the adenoviral shuttle vector pAdTrack-CMV.Then the recombinant shuttle plasmid linearized by pmeⅠwas cotransformed into competent E.coli.BJ5183 with the adenoviral backbone plasmid pAdEasy-1 to generate the recombinant adenovirus vector rAd-mir-133a.rAd-mir-133a was then packaged and amplified in human embryonic kidney 293(HEK293) cells.The purified rAd-miR-133a was used to infect the hMSCs and the expression of miR-133a was detected by non-quantitative RT-PCR and real-time PCR.RESULTS:The recombinant adenovirus shuttle vector pAdTrack-CMV-miR-133a was constructed and verified by restriction endonuclease analysis and DNA sequence analysis.rAd-miR-133a was successfully packaged and amplified in HEK293 cells.The transcriptions of primary miR-133a and mature miR-133a were over-expressed in the hMSCs infected with rAd-miR-133a.CONCLUSION:The recombinant adenovirus vector carrying human miR-133a is successfully constructed,which lay a foundation for miR-133a function study.
5.Two-dimensional speckle tracking imaging in predicting myocardial injury of Kawasaki disease patients with intravenous immunoglobulin resistant during acute phase

Objective To analyze the characteristics of left ventricular systolic function in Kawasaki disease (KD) patients with intravenous immunoglobulin (IVIG) resistant during acute phase by two-dimensional speckle tracking imaging (2D STI).Methods IVIG resistant patients (n=40) as well as age and gender matched IVIG responder patients (n=40) were selected from KD patients in acute phase.Patients in IVIG resistant group were further divided into coronary artery dilation (CAD) subgroup and no coronary artery dilation (NCAD) subgroup.Then conventional echocardiography,2D STI and laboratory indexes were acquired and compared between IVIG resistant group and IVIG responder group,as well as between CAD and NCAD subgroup.ROC curve analysis was used to determine threshold values of 2D STI measurements associated with IVIG resistance.Results Compared with IVIG responder group,coronary artery dilation,left ventricular mass and left ventricular mass index increased,systolic global longitudinal strain (GLS) and systolic global circumferential strain (GCS) decreased,albumin,erythrocyte sedimentation rate,C-reactive protein and platelet increased in IVIG resistant group (all P<0.05).Taking absolute GLS 16.8% as a threshold,the area under curve (AUC) was 0.769 (P=0.021),sensitivity,specificity in diagnosis of IVIG resistant was 79.27%,68.36%.Taking absolute GCS 15.9% as a threshold,AUC was 0.749 (P=0.038),sensitivity,specificity in diagnosis of IVIG resistant was 71.43%,57.28%.Conclusion IVIG resistant KD patients present significantly greater systolic dysfunction compared with responders in patients with KD,which may be the results of myocardium infection other than coronary artery lesions.2D STI may predict myocardial injury in IVIG resistant KD patients.
6.The construction of lentivirus-mediated RNAi vector containing hTERT.

OBJECTIVETo construct a recombinant lentivirus RNA interference (RNAi) vector carrying hTERT gene, and to obtain the titer of the lentiviral stock for investigating the expression in the eukaryotic cells and the effect on the hTERT gene silencing in the eukaryotic cells.
METHODSTwo complimentary oligos of small interference RNA (siRNA) with hairpin structures targeting the hTERT gene and a negative control were synthesized, then ligated with pLVTHM vector and sequenced. The recombinant vectors were then transfected with viral packaging mix into T293 cells, viral supernatant was harvested to determine the titer. U87 cells infected by virus were harvested and the expression of hTERT, telomerase activity and apoptosis were detected by reverse transcription-PCR(RT-PCR), TRAP assay and flow cytometry separately.
RESULTSSequencing data showed that the constructed plasmids contained the correct sequences of hTERT siRNA transcript templates. A vector producing cell line T293 was established, and the titer for transfection was obtained. RT-PCR and TRAP flow cytometry analyses demonstrated that hTERT shRNA expression construct could suppress the expression of hTERT and telomerase activity and induce apoptosis.
CONCLUSIONA lentivirus RNAi vector targeting hTERT gene was successfully constructed, which decreased the expression of hTERT and telomerase activity effectively and induced apoptosis. It has set up a research platform for the gene therapy of tumors which take hTERT as the target.
