Objective To research the method for constructing an ECG acquisition platform on Zigbee based network with multi-node. Methods The platform is based on an SOC Zigbee chip CC2430. Furthermore,the hardware design and software ECG sampling process is introduced respectively. Results As the Zigbee can support dynamic routing on the platform,we can get ECG data in abundance,and meanwhile,users get more room and freedom. Conclusion It's a right nice scheme for colleting ECG data.

Objective To generate a comD gene knock-out mutant of Streptococcus pneumoniae,and determine the correlation of comD gene and the bacterial resistance against β-lactam antibiotics and understand the effect of closantel down-regulating comD, comE and comC mRNA levels. Methods A suicide plasmid pEVP3comD was constructed for comD gene knock-out and a comD gene knock-out mutant (comD-)was generated through homologous recombination and insertion inactivation. PCR and immunofluorescence method were used to identify the comD- mutant and real-time fluorescence quantitative PCR was applied to detect the changes of comD, comE and comC mRNA levels before and after closantel treatment in comD-mutant and wild-type strain. Double agar dilution method was performed to determine the sensitivity of comD- mutant and wild-type strain to penicillin G and cefotaxime. Results The comD gene in genome DNA of the generated comD- mutant was inactivated by sequencing and immunofluorescence detection. 50 μ mol/L or 100 μmol/L closantel had a function to down-regulate the comD, comE and comC mRNA levels ( P ＜ 0. 05) whereas 25 μmol/L closantel did not. Both the MIC values of penicillin G and cefotaxime inhibiting comD- mutant were 32 μg/ml which was significantly higher than that of wild-type strain (0.06 μg/ml and 1 μg/ml). Conclusion In this study a comD gene knock-out mutant of S. pneumoniae was successfully generated. There is a close correlation between comD gene and β-lactam antibiotics resistance of S. pneumoniae. Closantel has a function to inhibit the competence formation of S. pneumoniae through down-regulating the transcription levels of comD, comE and comC genes.

Objective To observe the influence of exercise intensity on cardiorespiratory function in the elderly with coronary heart disease using simplified 24-action Taijiquan and jogging. Methods A total of 115 sedentary elderly patients with heart disease were randomly divided into a control group ( no exercise), a low-intensity exercise group, a medium-intensity exercise group (whose exercise was simplified 24-action Taijiquan at 40-60% of their individual maximum oxygen consumption (VO2max) for 90 to 200 minutes per week) and a high-intensity exercise group ( whose exercise was jogging on a treadmill at ＞ 70% of their maximum oxygen consumption for 200 minutes per week). The regime was maintained for 6 months. All patients accepted a treadmill exercise test before and after the 6 months of exercise to evaluate the changes in VO2max, maximum heart rate ( HRmax), VO2max/HRmax and exercise time (t). Results ( 1 ) After 6 months of exercise, VO2max, HRmax, VO2/HRmax and t had increased sisnificantly in all three exercise groups. Work capacity had increased 18.6% , 33% and 34.4% and HRmax had increased 9.3% , 13.8%, 19.1%. There was no difference between before and after in the control group. (2) Compared with the control group the average VO2max, HRmax and t of all three exercise groups had increased significantly. VO2 max was 9.4% , 26% and 23. 1% better; HRmax was 6.7% , 8.8% and 12% better; t was 3.5,4 and 4.9 minutes longer. (3) Compared with the low-intensity exercise group, VO2max, HRmax and t in the medium- and high-intensity groups increased significantly more. VO2max increased 16.6% and 13.7% more; HRmax increased 2.1% and 3.2% more and average t was 0.5 and 1.4 minutes longer. (4) There was no significant difference between the medium- and high-intensity exercise groups in terms of VO2max. (5) The incidence of self-reported adverse responses to exercise such as dizziness, chest stuffiness and fatigue in the high-intensity exercise group increased significantly compared with the other 3 groups. Conclusions Exercise can gradually improve cardiorespiratory function in the sedentary elderly with heart disease. The effects of medium-intensity exercise are similar to those of high-intensity exercise, but medium intensity is safer.

Objective To explore the effect of fetal thymus transplantation on enhancing the immunity of patients with late-stage malignant tumors. Methods The whole thymus of 24～32 weeks fetus was transplanted into the forearm of the patients with late-stage milignant tumors by microvascular surgery. The levels of serum IgA,IgG and IgM were measured before and at the third, 6th and 12th months after operation. No immunosuppressive regimen was performed in all the patients after operation. The follow-up period was 1 to 2 years. Results The clinical symptoms of all the patients improved after treatment, and no rejection reaction occurred. Objective To detect the expression of hypoxia inducible factor-lα (HIF-1α) in both mRNA and protein and reactive oxygen species (ROS) levels in human epithelial ovarian cancer,and investigate the abnormal state of oxygen metabolism and their interaction in human epithelial ovarian cancer develoment.Methods The expression of HIF-1α in mRNA and protein in epithelial ovarian cancer were detected by real time reverse transcription-polymerase chain reaction and immunohistochemical Elivision plus two-step method,and the level of ROS was detected by fluorescence microplate reader.Results The relative content of HIF-1α mRNA in the epithelial ovarian cancer group was 4.466±0.754,which was higher than that of the benign control group (6.073±1.053) (t =-7.701,P =0.000).The expression of HIF-1α mRNA was raised gradually in tissues of the benign ovarian tumor,early and advanced stage of epithelial ovarian cancer (t =5.301,P =0.000).The expression of HIF-1α mRNA in the epithelial ovarian cancer had positive correlation with grade and lymph node metastasis (t =3.724,-3.851,P =0.000).The positive expression rate of HIF-1 α protein in epithelial ovarian cancer was 63.49 ％ (40/63),which was higher than that of the benign group (U =433,P =0.005),which had positive correlation with lymph node metastasis and stage (x2 =4.289,12.360; P =0.038,0.000).The expression of ROS in the epithelial ovarian cancer was 110.239±18.414,which was higher than that of the benign group (58.366±20.360) (t =11.068,P =0.000),which had positive correlation with grade,lymph node metastasis and stage (t =-2.055,2.816,-4.138; P =0.044,0.007,0.000).There was a moderate positive correlation between the expression of HIF-1α mRNA and ROS in epithelial ovarian cancer (r =0.572,P =0.000),and that was similar in low differentiation group and advanced group,there was a relatively high positive correlation in lymph node metastasis group (r =0.703,P =0.001),there was a relatively low correlation between HIF-1α mRNA and ROS (r =0.376,P =0.002).Conclusion Hypoxia is the basic characteristic of human epithelial ovarian carcers.This study suggested that HIF-1α is associated with human epithelial overaian cancer prognosis.ROS and HIF-1α might be involved in the genesis of human epithelial ovarian cancer,and the regulation of ROS to HIF-1α may play the important roles in the invasion and metastasis process of human epithelial ovarian cancer.The expression of HIF-1α is regulated by ROS at the transcriptional and protein levels,probably mainly on protein level.

In the present experiment the high dose of LRH-A was injected into pregnant rats on Day 9 of pregnancy. Morphology and histochemistry of corpus luteum were studied. It was found that 24 hours after injection LRH-A could effectively cause the corpus luteum to degenarate. A great many large lipid droplets occured in the luteal cells, and the activity of 3?-HSD and of SDH of these cells decreased. At the same time the activity of AcP increased in a few of luteal cells. 48 hours after injection the effects outlined above became more distinct, and the activity of G-6-PD also began to decrease. The activity of 20 ?-HSD in both control and experimental groups was negative 24 hours after injection, while the activity of the AIP increased distinctly in endothelium of the capillary network of corpus luteum. These changes indicated that the high dose of LRH-A had damaged corpus luteum and reduced its synthetic and secretory function of progesterone which, we believe, plays a main role in terminating pregnancy.

[Objective] To determine the effects against human simplex virus type I(HSV-1) and adenovirus type Ⅲ(ADV-3) anti-viral in vitro and in vivo of eye drop prepared by mixed extract of Heartleaf houttuynia and Lonicera japonica.[Methods] By using different numbers of host cells with cytopathic effect(CPE) as the observation index,models of HSV-1 infected Vero cells and ADV-3 infected HeLa cells were established to determine the anti-viral effects in vitro of HLD with different dosages.Rabbit eye conjunctivitis model infected by HSV-1 was established to observe the anti-viral effects in vivo of HLD with different dosages,and the biopsy specimens of pathogenic eye conjunctive tissue were examined by routine pathological methods.In these experiments,a commercial acilovir eye drop(ACV) was used as the control.[Results] The minimum dosages of HLD to completely inhibit the CPEs of HSV-1 and ADV-3,the host cells were 32 mg/ml and 64 mg/ml respectively.After treatment with HLD,the inflammatory reactions of rabbit eye conjunctivitis caused by HSV-1 infection were obviously weak or disappeared.The curative rates of 1,3 and 6 g/ml HLD treating experimental rabbit eye conjunctivitis were 50.0%,83.3% and 100% respectively.Furthermore,the curative effect using 6 g/ml HLD has no significant difference compared to that using ACV drop.[Conclusion] HLD has a certain anti-viral effect in vitro and a stronger anti-viral effect in vivo,indicating the potential of HLD for developing a new drug to prevent or treat viral eye conjunctivitis.

[Objective] To probe into the psychological health status of middle-aged intellectuals and its influencing factors. [Methods]A cluster sampling of 1 474 middle-aged intellectuals was measured by the SCL-90 table and was analyzed by the Reliability Analysis, Principal Component Analysis and Analysis of Variance. [ Results ] The measurement table is highly reliable (Cronbach a = 0.949) and the Principle Component Analysis has main factors (A -1.97) and cumulative contribution rate is 80% . [Conclusion]Marital status, income and gender are the main influencing factors of psychological healthy status in middle-aged intellectuals.

ObjectiveTo generate the prokaryotic expression systems of vwA1 and vwA2 genes of Leptospira interrogans serogroup Icterohaemorrhagiae serovar Lai strain Lai,and to determine the correlation among the target recombinant expressed products rVwA1 and rVwA2 and platelet-associated hemorrhage.MethodsThe vwA1 and vwA2 genes of L.interrogans strain Lai were amplified using high fertility PCRs and then the amplification products were sequenced.The prokaryotic expression systems of vwA1 and vwA2 genes were generated by using routine genetic techniques.The expression and dissolubility of rVwA1 and rVwA2 proteins were examined by SDS-PAGE plus Bio-Rad Gel Image Analyzer.Ni-NTA affinity chromatography was used to extract the expressed rVwA1 and rVwA2.Real-time fluorescence quantitative RT-PCRs were performed to determine the changes of vwA1-mRNA and vwA2-mRNA levels in the leptospires of L.interrogans strain Lai before and after infection of human umbilical vein endothelial cell line (HUVEC).The ability of leptospiral rVwA1 binding to human platelets was detected by flow cytometry.A hydrolytic test plus SDS-PAGE were applied to examine the cleavage of leptospiral rVwA2 by a human von Willebrand factor lysase (ADAMTS13).ResultsThe nucleotide and putative amino acid sequences of the cloned leptospiral vwA1 and vwA2 genes were 100％ identities compared to the reported corresponding genes.The generated prokaryotic expression systems of vwA1 and vwA2 genes could express soluble rVwA1 and rVwA2,respectively.When L.interrogans strain Lai infected HUVEC for 8 h,both the vwA1-mRNA and vwA2-mRNA levels were significantly up-regulated (P＜0.05).The result of flow cytometry showed that the leptospiral rVwA1 was able to combine with human platelets with a 60.8％ binding rate. Human recombinant vWF-A2 ( rhvWF - A 2 ),but not the leptospiral rVwA 2,could be hydrolyzed by human ADAMTS 13.Conclusion The vwA1 and vwA2 genes may play roles during infection of L.interrogans species,and the function of vwA1 gene product is referring to the hemorrhage in leptospirosis.

Objective To determine the role of flagellar motor protein MotA in the pathogenesisassociated chemotaxis and colonization of Campylobacter jejuni. Methods The motA gene as well as Kan~r gene and plus-motA gene segments for motA gene knock-out were amplified by PCR and the target amplification fragments were sequenced after cloning. A suicide plasmid(pBlueskrit-Ⅱ-SK~(motA-kan)) and a motA gene knock-out mutant (motA~-) were constructed based on homologious recombination. By using semisolid plate migration test, hard agar plus (HAP)-based chemotactic test towards sodium deoxycholate (SDC) in vitro, and jejunal colonization test in BALB/c-ByJ mice were performed to determine the differences of flagellar motility, chemotaxis towards SDC and colonization in murine jejunum between motA~- mutant and wild-type strain. Results The nucleotide and amino acid sequences of the cloned motA gene were 100% identical to the reported corresponding sequences. The results of PCR, sequencing and continuous passage culture in antibiotics-contained medium demonstrated that both suicide plasmid and motA~- mutant were successfully generated. The diameters of clonies on semisolid plate and 0.2 mol/L SDC-induced chemotactic tings in HAP as well as the bacterial numbers adhering to the surface of murine jejunal mucosa and in jejunal content of motA~- mutant were significantly less than those of wild-type strain(P<0.05). Conclusion A motA gene knock-out mutant of C. jejuni was successfully constructed in this study, motA is an essential gene for flagellax motility, pathogenesis-associated chemotaxis and colonization of C. jejuni.
