1.Change of Kv Channel Interacting Protein 1 and Gamma-Aminobutyric Acid-ergic Neurons in Hippocampus of Pentylenetetrazole Induced Epilepsy Rats

Objective To explore the role of Kv channel interacting protein 1(KChIP1)in the process of epileptic seizure and the relationship between KChIP1 and gamma-aminobutyric acid-(GABA)ergic neurons.Methods Normal female Sprague-Dawley rats were treated with pentylenetetrazole to make acute pentylenetetrazole models of epilepsy.Laser Scanning Confocal Microscope(LSCM)combined with double-labeled immunohistochemical technique was applied to observe the expression of the KChIP1 and the GABAergic neurons in the hippocampus of rats.Results The number of KChIP1-postive neurons in the hippocampus was significantly increased in the acute pentylenetetrazole model rats(P<0.05).There was no significant difference in the number of double-labled neurons(P>0.05),nor of the GABA-postive neurons between the model rats and the controls.The ratio of double-labeled neurons/total positive neurons was 63.9% in the hippocampus.Conclusion The KChIP1 might be involved in epileptogenesis of pentylenetetrazole induced seizure.The KChIP1 was associated with GABAergic neurons,whereas it may be functionally different from GABA.
2.Usage Analysis on Traditional Chinses Medicine Decoction Pieces and Granules in Zhuhai Hospital of Guangdong Provincial Hospital of Traditional Chinese Medicine During 2011-2014

Objective To provide references for reasonable procurement planning, TCM decoction pieces reserving and clinical medication guarantee by analyzing the usage of TCM decoction pieces and granules from 2011 to 2014 in Zhuhai Hospital of Guangdong Provincial Hospital of Traditional Chinese Medicine (hereinafter referred to as Zhuhai Hospital). Methods The sale data of TCM decoction pieces and granules during 2011-2014 in the database of Zhuhai Hospital were analyzed. Sales quantity, sales amount of the category and single pieces of TCM decoction pieces and granules ranking the first 10 were analyzed. Results The sales quantity and amount of TCM decoction pieces and granules were increasing from 2011 to 2014. The usage weight ratio and usage amount ratio between TCM decoction pieces and granules was stable. The top 10 categories and single pieces in sales quantity and amount were also stable. Conclusion Zhuhai Hospital has its own characteristics in TCM types, categories and dosage forms. TCM decoction pieces play an important part in clinical treatment in Zhuhai Hospital.
3.Resilience and related factors in rescue soldiers

Objective To research the resilience and related factors of the rescue soldiers. Methods Resilience Scale for Adults( RSA ), Self-report Symptom Checklist 90 (SCL-90), Eysenck Personality Questionnaire(EPQ) and Coping Style Questionnaire were applied in this survey to 1401 rescue soldiers. Results ①The soldiers' resilience score was (2.49±0.57) ,significantly lower than college students( t= 16.619, P=0. 000).The rescue soldiers with the year in army above 3 were significantly higher than that below 2 years in social competence( t=2.581, P=0.01 ) ,social resources( t=2. 158, P=0.031 ) ,structured style( t=3.254, P=0.001 ) and RSA scores(t=2.455, P=0.014). There exist significantly differences in various education level of the rescue soldiers in perception of self(t=3.732, P=0.024), planned of future ( t = 9.324, P = 0.000 ) , social competence ( t = 8. 838, P = 0.000 ), social resources ( t = 13. 660, P = 0. 000 ) and RSA scores (t=9.805, P=0.000) , and college ≥ senior high school ≥ junior high school. ②Compared to the lower group in the RSA, the higher group scored significantly higher in extraversion (t=16. 204, P=0.000) and positive coping( t = - 18. 171, P=0.000), while lower in SCL-90 (t=8.461, P=0.000) and neuroticism (t=8.833, P=0.000 ).③Resilience have the obvious positive correlation with the education ( r= 0. 116 ), extraversion ( r= 0. 463 ) and positive coping(r=0.500) ,and negative correlation with the neuroticism(r=-0.251 ) and SCL-90( r= -0.260). It can be assumed that positive coping,extraversion and neuroticism had good predictive ability(35.9%) to resilience. Conclusion The rescue soldiers have low resilience than college students, positive coping, extraversion, and neuroticism are important psychological factors for rescue soldiers.
4.In vitro induction and detection of human B cells producing HLA antibodies

Objective To induce and detect human B cells producing HLA antibodies in vitro and by an ELISPOT assay. Methods Human peripheral blood mononuclear cells (PBMC) from 7 healthy vol-unteers were isolated by density gradient centrifugation, and cultured with the stimulation of PWM alone or PWM + SAC for 5 d. The lg levels in culture supernatants and the number of Ig producing B cells were de-termined by ELISA and ELISPOT assay, respectively. The optimal stimulation protocol for PBMC to produce optimal Ig in vitro was anticipated to be obtained. Using the optimal pre-cuhure and ELISPOT conditions, PBMC from 9 alloimmunized subjects were analyzed for the presence of B cells secreting HLA antibodies. Results There was a tendency towards more viable cells following the activation with PWM and SAC vs PWM alone in a 5-day culture (P =0. 052). The IgG levels in the supernatants were found to be comparable for the two culture conditions whereas the lgM levels were increased ( P = 0.03 ) in the presence of the com-bination of PWM and SAC. In 6 subjects a specific signal was obtained with our ELISPOT assay. The presence of HLA antibodies in the respective pre-cuhure supernatants supported the specificity. Conclusion The activation of PBMC with the combination of PWM and SAC, and the application of HLA-specifie ELIS-POT assay can succeed in the induction and detection of human B cells producing HLA antibodies.
5.Analysis on the emotional sate and the related influencing factors in children with nocturnal enuresis

Objective To investigate the emotional state and its influencing factors in children with nocturnal enuresis,so as to improve clinical attention and give appropriate intervention.Methods Totally 166 nocturnal enuresis patients aged 9 -15 years old were admitted to Beijing Children′s Hospital Affiliated to Capital Medical University from August 2011 to August 2014.Specifically,Screen for Child Anxiety Related Emotional Disorders (SCARED)and Depres-sive Self -Rating Scale for Childhood (DSRSC)were utilized to test the patients′emotional state.The comparison be-tween the general norm data of the healthy children and the psychological test of enuresis children were performed,the lat-ter emotional state and its influence factors were evaluated.Analysis of variance and t -test were performed.Frequency comparison was done by using χ2 test.Influence factors analysis was made by Logistic or multiple regression analysis. Results The incidence of children′s anxiety with enuresis was 39.2%(65 /166 cases).Anxiety overall scores of children with enuresis amounted to 21 .16 ±11 .88 which were significantly higher than the norm(14.40 ±9.41 )(P <0.05).The diagnosis rate of depression in the NE patients was 9.0%.No significant difference was found in the depression total scores(P >0.05).The emotional disorders mainly appeared in patients with primary enuresis,without family history,poor grade and poor relationships with family members and peers.The school performance was an important factor for enuresis patients with anxiety disorders(P <0.05).Conclusions Children with nocturnal enuresis are more likely to have anxiety disorders than normal children.Their emotional states is related to the enuresis type,family history,school performance, family and peer relationship.Therefore,attention should be paid to the emotional state of children with nocturnal enuresis. According to the adverse factors for nocturnal enuresis patients,effective treatment and appropriate psychological interven-tion should be given at the same time.
6.Hollow hydroxyapatite combined with human bone morphogenetic protein-2 microspheres for bone defect repair

BACKGROUND:Hydroxyapatite (HA) is a good scaffold material, and recombinant human bone morphogenetic protein-2 (rhBMP-2) possesses a strong osteogenic ability, therefore, by which preparing a novel composite material wil be helpful for bone repair. OBJECTIVE:To explore the effects of the hol ow HA/rhBMP-2 microspheres on the osteogenesis and biomechanics of rabbit bone defects. METHODS:Forty-eight male healthy adult New Zealand white rabbits were randomly divided into three groups (n=16 per group), including composite, single and control groups. Radical defect models were prepared, and the hol ow HA/rhBMP-2 and hol ow HA scaffolds were implanted into the composite and single groups, respectively. The control group received no treatment. At the 1st day of 4, 8, 12, and 16 weeks after implantation, the level of serum alkaline phosphatase was detected, and the bone healing was assessed through X-ray, three-dimensional CT, radionuclide bone scan and biomechanics testing, respectively. RESULTS AND CONCLUSION:The level of serum alkaline phosphatase, X-ray scale scores, osteogetic effect, region of interest volume, three-dimensional CT and biomechanical strength in the composite group were superior to those in the single group. In the meanwhile, the bone healing was unsatisfactory in the control group. Our findings indicate that the hol ow HA/rhBMP-2 artificial bone exhibits a good osteogenic ability and mechanical strength, contributing to bone healing.
7.Study of differentially expressed genes in laryngeal squamous cell carcinoma by cDNA microarray

Objective: To screen for the differentially expressed genes in laryngeal squamous cell carcinoma and normal laryngeal tissue using cDNA microarray. Methods: The PCR products of 4 096 genes were spotted on a chemical material coated glass plates in array. The DNAs were then fixed on the glass plate by a serial of treatments. The total RNAs were isolated from the tissues, and then were purified to mRNAs by Oligotex. Both the mRNAs from the laryngeal squamous cell carcinoma and normal tissue were reversely transcribed to cDNAs with the incorporations of fluorescent dUTP, for preparing the hybridization probes. The mixed probes were then hybridized to the cDNA microarray. After high stringent washing, the cDNA microarray was scanned for the fluorescent signals and showed the differences between 2 tissues. Results: Among the 4 096 target genes, there were 36(0.88%) genes whose expression levels differed between the carcinoma and normal tissues in all 4 cases. Bioinformatical analysis of those genes had been performed. Conclusion: DNA microarray technology is an effective technique in screening for differentially expressed genes between 2 different kinds of tissue. Further analysis of the obtained genes will help to understand the molecular mechanism of malignant carcinoma. [
8.Placental Foxp3 expression in patients with preeclampsia and correlation of Foxp3 gene lo-cus 924 (rs2232365) polymorphism with preeclampsia

Objective To detect changes of Foxp3 expression in the decidua in patients with preeclampsia and investigate the correlation of Foxp3-924 (rs2232365) polymorphisms with preeclampsia. Methods From October 2011 to December 2012, 252 normal pregnant women and 156 preeclampsia patients of Han nationality from the same geographic region were tested for Foxp3-924 genotypes by polymerase chain reaction with sequence-specific primer (PCR-SSP). Sixty-eight of the patients with preeclampsia (33 with mild and 35 with severe preeclampsia) and 30 of the normal pregnant women were also examined for Foxp3 expression in the decidua using immunohistochemical method. Results Foxp3 positive expression rates in the decidua was 51.52%in mild preeclampsia and 28.57%in severe preeclampsia cases, significantly lower than that in the control group (86.67%, P<0.05). In preeclampsia patients, the frequencies of Foxp3-924G/G, G/A, and A/A genotypes were 0.1346, 0.4615 and 0.4038, respectively, and the frequencies of Foxp3-924A and Foxp3-924 G were 0.6346 and 0.3654, respectively. The genotype frequencies of Foxp3-924G/G, G/A and A/A in the control group were 0.1508, 0.4087 and 0.4405, respectively, and the frequencies of Foxp3-924 A and Foxp3-924 G were 0.6448 and 0.3552, respectively. No significant differences were found in the gene frequencies of Foxp3-924G/A between preeclampsia patients and the control group (P>0.05). Conclusion The expression level of Foxp3 in the placental tissue of preeclampsia patients is significantly lower than that in normal pregnant women, suggesting that lowered Foxp3 expression decreases the immunosuppressive function and causes imbalance of immune tolerance between maternal-fetal to induce preeclampsia. Foxp3-924 polymorphisms is not significantly correlated with the occurrence of preeclampsia.
