1.Retrospective analysis of a tuberculosis outbreak among junior high school students in Chongqing

Objective:
To analyze changes in tuberculosis infection among junior high school students before and after tuberculosis exposure, so as to provide a reference for improving school tuberculosis prevention and control measures and policy formulation.
Methods:
Retrospectively collect data on a tuberculosis outbreak that occurred in a grade of a junior high school in Chongqing in 2025, including tuberculosis screening records of students in this grade upon their enrollment in 2022 (1 156 students) and after two tuberculosis outbreaks in 2023 (206 students) and 2025 (171 students). The Wilcoxon signed rank test for paired design was used to compare the induration diameters of the subjects, and the Chi square test was adopted to analyze the rate of tuberculosis infection among students.
Results:
In the tuberculosis outbreak in 2023, the rate of tuberculosis infection among close contacts ( 11.84 %) and the rate of tuberculosis infection among freshrman at school enrollment (12.89%) showed no statistically significant difference ( χ 2=0.25, P >0.05). The rate of tuberculosis infection of close contacts in the 2025 tuberculosis outbreak (55.56%) was higher than that in the 2023 outbreak (11.84%) ( χ 2=30.42, P <0.01). Among the 106 students included in the cohort analysis, the median induration diameter was 3.50 (1.50, 7.50) mm in 2023 and 8.75 (4.25, 11.50) mm in 2025, with a statistically significant difference ( Z=-5.76, P <0.01). There was no statistically significant difference between the infection rate in 2022 (16.98%) and that in 2023 (10.38%) ( χ 2=1.96, P =0.16). The infection rate in 2025 (43.40%) was higher than those in 2022 and 2023 ( χ 2=17.55, 29.39, both P <0.017). The seroconversion rate of students in the same class in 2025 ( 58.00 %) was higher than that of students in different classes (16.07%), with a statistically significant difference ( χ 2=20.19, P <0.01). All 72 individuals with latent tuberculosis infections identified during the pandemic in 2023 and 2025 refused to undergo prophylactic treatment.
Conclusions
The lack of preventive treatment may be the underlying cause of the successive outbreaks during the epidemic. Early detection of infection sources and standardized outbreak management are crucial to controlling the spread of the epidemic.
2.Research progress on early screening methods for occupational noise-induced hearing loss

Occupational noise-induced hearing loss (NIHL) is an irreversible sensorineural hearing loss that severely endangers workers’ health, making early screening crucial. This article reviewed the research progress on early screening methods for occupational NIHL, introduced the testing mechanisms of three core screening methods—tympanometry, otoacoustic emissions, and extended high-frequency audiometry —and summarized their clinical application advantages and limitations. It is proposed that multimodal combined detection (e.g., the combination of tympanometry, otoacoustic emissions, and extended high-frequency audiometry) can significantly improve the accuracy and comprehensiveness of early screening. Meanwhile, future studies with prospective cohort design are encouraged to verify the long-term monitoring value of each method and to strengthen the joint development of screening technologies with cutting-edge approaches such as machine learning, in order to further improve screening efficiency and provide stronger protection for workers’ hearing health.
3.Phenotypic and genotypic analysis of five fetuses with Harlequin ichthyosis due to variants of ABCA12 gene.

OBJECTIVE:
To explore the clinical and genetic characteristics of five fetuses with Harlequin ichthyosis (HI).
METHODS:
Five fetuses with HI diagnosed at Guangdong Women and Children Hospital between 2017 and 2024 were selected as study subjects. Clinical and laboratory data were collected and reviewed. Whole exome sequencing (WES) was carried out, and candidate variants were verified by bioinformatic analysis. This study was approved by the Medical Ethics Committee of the hospital (Ethics No.: 202401024).
RESULTS:
The five fetuses had presented with ectropion, eclabium and contracture and flexion of fingers and toes. WES revealed that all had harbored compound heterozygous or homozygous variants of the ABCA12 gene. Among the eight types of variants, five were unreported previously.
CONCLUSION
The compound heterozygous or homozygous variants of the ABCA12 gene probably underlay the HI in the five fetuses. Clinicians should be vigilant about the possibility of HI in fetus with ectropion, eclabium, and contracture and flexion of fingers and toes.

4.Administration of Psoralea corylifolia L. (Buguzhi) during pregnancy causes mild liver injury in mouse mothers and weaned offspring

Background: Psoralea corylifolia L. (Buguzhi, BGZ), known for its efficacy in supporting pregnancy and preventing miscarriage, has been used in China for over 1000 years. Recently, BGZ has been identified as a potential cause of drug-induced liver injury. However, its safety during pregnancy remains unclear, which significantly hinders its routine clinical application. Objective: To investigate the effects of BGZ administration during pregnancy on the liver of mouse mothers and their weaned 21-day-old offspring. Methods: Mice were orally administered BGZ at doses of 2.5 and 10 g/kg during pregnancy, with BGZ withdrawal during the lactation period. Liver histopathology (hematoxylin-eosin staining), biochemical analysis, and evaluation of liver bile acid metabolism were performed after the lactation period. Results: BGZ administration at doses of 2.5 and 10 g/kg during pregnancy, followed by withdrawal during the lactation period, caused mild liver damage in both mothers and their 21-day-old offspring. Serum total bile acid (TBA) levels were elevated compared with those in the control group. Additionally, changes were observed in the levels and proportions of various bile acids (BAs) in the liver, suggesting mild effects on BA metabolism. Conclusion: BGZ administration during pregnancy caused mild liver damage and increased serum TBA levels in both mouse mothers and their 21-day-old offspring. This phenomenon may be associated with imbalanced BA metabolism in the liver. Based on the present study and the limited toxicological research on BGZ, pregnant women should avoid prolonged use of BGZ. If BGZ is administered during pregnancy, serum TBA levels should be monitored, and if elevated, BGZ should be discontinued.
5.Toxicological evaluation of aristolochic acid II following single and repeated oral administration over a 24-week period

Background: Aristolochic acid II (AAII), a major nephrotoxic and carcinogenic component of aristolochic acids (AAs), has been less studied compared with its well-characterized analog, aristolochic acid I (AAI). Although AAs are known to induce carcinogenesis via DNA adduct formation, the toxicity mechanisms, environmental prevalence, and long-term health impacts of AAII remain poorly understood. Objective: This study aimed to systematically evaluate AAII’s acute and chronic toxicity, carcinogenic mechanisms, and environmental exposure patterns using integrated murine models and phytochemical analyses to clarify its toxicological profile and associated health risks. Methods: C57BL/6J mice were used in the following experiments: (1) determination of AAII content in 3 commonly used Aristolochia medicinal materials via liquid chromatography-mass spectrometry/mass spectrometry; (2) acute toxicity testing with single doses of 10, 20, or 40 mg/kg; and (3) chronic exposure with 1 or 10 mg/kg administered every other day for 24 weeks, followed by 21 to 40 weeks of postexposure monitoring. Histopathological examination, whole-exome sequencing, biochemical assays, and micronucleus tests were performed to assess multi-organ damage, tumorigenesis, genomic mutation signatures, and direct clastogenicity. Phytochemical analyses were used to evaluate environmental distribution. Results: (1) A single 40 mg/kg dose of AAII induced dose-dependent renal tubular degeneration without hepatotoxicity; (2) the 10 mg/kg group showed significant mortality (20%), tumor incidence (33.3%, primarily forestomach and bladder transitional cell carcinomas), persistent renal interstitial fibrosis, and subclinical hepatic injury. Chronic exposure to 1 mg/kg still induced 13.3% mortality and 15.5% tumor incidence over a 64-week period; (3) whole-exome sequencing revealed a predominance of C>T mutations and pathway enrichment in chemical carcinogenesis and cytochrome P450-mediated metabolism, indicating reactive metabolite-driven mechanisms distinct from classical AA-DNA adducts; and (4) no histopathological changes were observed in nontarget organs (brain, heart, and testes), and micronucleus assays confirmed the absence of direct clastogenicity. Conclusion: This study highlights the delayed carcinogenic risks of low-dose chronic AAII exposure and emphasizes the need to update regulatory frameworks to ensure the safe use of aristolochiaceae-containing herbal products.
6.Progress on ketogenic diet in children with autism spectrum disorder

Autism spectrum disorder(ASD)is a neurodevelopmental disorder that originates in early childhood. The precise pathogenesis of ASD remains unclear,and current interventions show limited effectiveness in improving patient outcomes. Various treatment strategies are being explored.The ketogenic diet(KD),as a high-fat,low-carbohydrate,moderate-protein diet,gains attention as a potential therapeutic option for ASD. This article reviews the current application of the KD in the treatment of ASD,with a detailed exploration of its efficacy in alleviating ASD symptoms,clinical uncertainties,and potential mechanisms of action. These mechanisms include its effects on gut microbiota,neural excitation-inhibition balance,inflammation,oxidative stress,and so on. The review aims to provide reference for further scientific research and practical clinical applications.
7.Research the effect of 4℃ refrigerated stored apheresis platelets based on platelet metabolomics

[Objective] To investigate the differences in metabolomics between apheresis platelets stored at 4℃ and at 22℃ with agitation, aiming to provide a theoretical basis for the cold storage of apheresis platelets. [Methods] Samples were collected at four time points (d1, d5, d10, d15) for platelets stored at 4℃ (experimental group) and two time points (d1, d5) for platelets stored at 22℃ with agitation (control group). Liquid chromatography-tandem mass spectrometry (LC-MS/MS) technology was used to detect changes in platelet metabolome levels under different storage conditions. Platelet functional activity was assessed by thromboelastography (TEG) for maximum amplitude (MA) values and flow cytometry for CD62P activation rates. [Results] Metabolites in the glycolytic pathway, key metabolites in the tricarboxylic acid cycle (citrate, α-ketoglutarate), metabolites in the purine metabolism pathway (adenine, inosine monophosphate, guanine, etc.) and amino acid metabolites significantly decreased by d5 in the control group, whereas they remained stable in the experimental group. The content of fatty acid metabolites, such as prostaglandin G2, 13(S)-HOTrE, and linoleic acid, significantly increased in the control group. Statistically significant differences in MA values were observed between the two groups at d1 and d5 (P<0.05). However, in the experimental group, as the storage time extended, the MA values at d10 and d15 showed no significant difference compared to the control group at d5 (P>0.05). The CD62P activation rate between the two groups was statistically significant (P<0.05). Additionally, the CD62P activation rate of platelets in the 22℃ group increased rapidly from d1, while it rose gradually in the 4 ℃ group. [Conclusion] Platelets stored at 4 ℃ exhibit more stable metabolic activity and slower functional deterioration, which is beneficial for extending the effective storage period of platelets.
8.Characterization of lower limb coordination and cortical activation during walking in adolescents with spastic cerebral palsy

Objective:To investigate the activation characteristics of the cerebral cortex and the coordination characteris-tics of the lower limb joints in adolescents with spastic cerebral palsy during walking,and to provide a theo-retical basis for the rehabilitation training of their gait coordination.Method:A total of eight adolescents aged 12-18 years with spastic cerebral palsy were included as the spas-tic cerebral palsy group and eight normal adolescents aged 12-18 years were included as the control group.Both groups walked on a treadmill at a fixed speed of 0.45 m/s.A Vicon infrared high-speed motion capture system and a functional near-infrared spectroscopy system were applied to collect the kinematic data and hemo-dynamic signals in the sensory-motor cortical region of the two groups while walking on the treadmill.The mean absolute value of the continuous relative phase(MACRP)and the mean of the standard deviation of the relative phase(DP)were calculated to evaluate the coupling coordination characteristics of the lower limb links.The relative concentration change of oxygenated hemoglobin(HbO2)in the cerebral cortex was calculat-ed to evaluate the activation characteristics of the cerebral cortex.Result:①Comparison results of lower limb coordination ability:During walking,the MACRP value of cou-pling coordination of hip-knee joints within the left lower limb of the spastic cerebral palsy group was signifi-cantly smaller than that of the control group(P=0.043,Cohen's d=1.113).The DP value of coupled variability within the limb of the spastic cerebral palsy group was significantly larger than that of the control group for both the hip-knee(P=0.003,Cohen's d=1.755)and knee-ankle(P=0.009,Cohen's d=1.503)joints.②Compari-son results of brain activation levels:Channels 11(P=0.025,Cohen's d=1.258)and 12(P=0.02,Cohen's d=2.310)in the supplementary motor area,channels 6(P=0.043,Cohen's d=1.112),14(P=0.006,Cohen's d=1.633),and 15(P=0.002,Cohen's d=1.886)in the precentral gyrus,channels 6(P=0.043,Cohen's d=1.112),7(P=0.002,Cohen's d=1.837),15(P=0.002,Cohen's d=1.886),and 17(P=0.002,Cohen's d=1.933)in the postcentral gyrus,channels 9(P=0.016,Cohen's d=1.363),10(P=0.045,Cohen's d=1.101)and 18(P=0.001,Cohen's d=2.221)in the superior parietal lobule,the activation levels of the above chan-nels of the spastic cerebral palsy group were significantly greater than those of the control group.Conclusion:During walking,adolescents with spastic cerebral palsy had hyperactivation of the somatosensory-motor cortical area of the brain and an unstable pattern of joint coordination within the lower limbs,with a high degree of variability.
9.Stat1 inhibits Foxp3 expression and Treg production

Objective:To explore the influence of Stat1 on Foxp3 expression and production of Treg.Methods:C57BL/6 mice were used and separated into normal control group and Stat1 specific inhibitor Fludarabine(Flud)treatment group.Ratio of CD4+Foxp3+Treg and expression of Foxp3 in spleen,lymph nodes and peripheral blood of mice in each group were detected by flow cy-tometry.Human Stat1 overexpression plasmid was constructed and transfected into human breast cancer MCF-7 cells,and expression changes of Foxp3 was detected by RT-qPCR and Western blot.Results:Compared with mice in normal control group,proportion of Treg and expression of Foxp3 in lymph nodes and peripheral blood of mice in Flud treatment group were increased,while Stat1 overex-pression resulted in decreased Foxp3 mRNA and protein expression in MCF-7 cells.Conclusion:Stat1 inhibits expression of Foxp3 and production of Tregs.
10.Balance ability and brain activation characteristics in adolescents with cerebral palsy under the external perturbation conditions

Objective:To explore the differences in balance ability and brain activation level between the cerebral palsy(CP)adolescents and the typically developing(TD)adolescents.Method:Eight adolescents with CP or TD were recruited in this study.The center of pressure(COP)was measured using an AMTI three-dimensional force platform to assess the standing balance under perturbation or non-perturbation condition.Brain activation level was also measured using the fNIRS device to collect the oxy-genated hemoglobin concentrations(ΔHbO2)in the supplementary motor area(SMA),precental gyrus(PreCG),postcentral gyrus(PoCG)and superior parietal lobule(SPL).Result:①Balance ability indicators:During the perturbation condition,DeltaAP,RMS,RMSAP,SPAP in the CP group were significantly greater than those in the control group.In the CP group,DeltaAP,DeltaML,RMS,RMSAP,SPAP during the perturbation condition were significantly greater than those during the non-pertur-bation condition.②Brain activation level indicators:During the perturbation conditions,the ΔHbO2 of SMA in the CP group were significantly greater than those in the control group.In the CP group,the ΔHbO2 of SMA,PreCG,PoCG during the perturbation condition were significantly greater than those during the non-per-turbation condition.Conclusion:Compared to the TD adolescents,the CP adolescents had poorer balance in the anterior-posterior direction and higher level of SMA activation in the external perturbation condition.Under the external perturba-tion condition,the balance ability decreased in the anterior-posterior direction,while the activation level of SMA,PreCG and PoCG increased in the adolescents with CP.
