1.Effects of agmatine on neuronal nitric oxide synthase in hippocampus of morphine withdrawal rats

BACKGROUND: Agmatine can enhance the analgesic effect of morphine,and antagonize the tolerant and dependent effect of morphine.OBJECTIVE: To observe the effects of injecting agmatine on the neuronal nitric oxide synthase (nNOS) in hippocampus of morphine withdrawal rats.DESIGN: A randomized controlled experimental study.SETTING: Department of Anesthesiology, the General Hospital of Chinese PLA.MATERIALS: All the experiments were carried out in the Department of anesthesiology, the General Hospital of Chinese PLA between April and July 2004. Eighteen healthy SD rats were randomly divided into saline control group (n=6), morphine group (n=6) and agmatine-treated group (n=6).METHODS: The rats in the saline control group were treated with subcutaneous injection of physiological saline (10 mg/kg), those in the morphine group were treated with 5-day preconditioning, subcutaneous injection of morphine of 10, 20, 30, 40 and 50 mg/kg respectively, twice a day, and those in the agmatine-treated group were treated with subcutaneous injection of agmatine (10 mg/kg) at 30 minutes before morphine was given, but at 6 hours later, before morphine was given for the last time, the rats in the morphine group and agmatine treated group were also given intraperitoneal injection of naloxone (5 mg/kg) to induce morphine withdrawal symptoms.The number of times of the morphine withdrawal symptoms (including physical signs of trembling like a wet dog, chewing, irrigating, drooling, diarrhoea, etc.) were recorded within 1 hour, and the reduction of body mass was calculated according to the different value of body mass before and after the withdrawal symptoms induced by naloxone. The rats were killed under anesthesia after praxiological detection, and then hippocampus was taken out and made into frozen sections, and the nNOS was detected with immunohistochemical staining. The CMIAS systemwas applied for imaging analysis, and the average value of the integral absorbance (A) values in 5visual sights for each section was taken as the integral A value of positive neuron.MAIN OUTCOME MEASURES: ① The detected results of morphine withdrawal symptoms in each group; ② The changes of the nNOS expressions in hippocampus of rats in each group.RESULTS: All the 18 rats were involved in the analysis of results. ① The detected results of morphine withdrawal symptoms in each group: The withdrawal symptoms of trembling like a wet dog, chewing, irrigating, drooling,diarrhoea and reduction of body mass in the agmatine treated group were all obviously lower than those in the morphine group [(2.0±1.3), (5.0±1.1);(0.3±0.4), (1.8±0.7); (3.2±1.2), (6.8±3.1); (0.2±0.4), (1.2±0.9); (2.7±2.1),(6.7±2.1); (6.0±3.0), (12.8±2.7) times, P < 0.01], and close to those in the saline control group (P > 0.05). ② The changes of the nNOS expressions in hippocampus of rats in each group: The positive neurons of nNOS in hippocampus mainly distributed in CA1 region, the cytoplasm was stained buffy, and the round nuclei were stained pale purple by haematine. The immunofluorescent A value of positive neuron in the agmatine-treated group was significantly decreased as compared with that in the morphine group (24.32±8.31, 50.82±15.13, P< 0.01), and almost the same as that in the saline control group (24.32±8.31, 15.24±1.88, P > 0.05).CONCLUSION: Agmatine can inhibit the morphine withdrawal syndrome and decrease the expression of nNOS in hippocampus CA1 region of morphine-withdrawal rats. Hippocampal nitric oxide pathway takes part in the inhibitory effect of agmatine on morphine withdrawal syndrome.
2.Therapeutic effect of desloratadine citrate disodium tablets combined with narrow band ultraviolet B in the treatment of pityriasis rosea

Objective To observe the clinical efficacy of desloratadine citrate disodium tablets combined with narrow band ultraviolet B in the treatment of pityriasis rosea.Methods According to the digital table,88 pityriasis rosea patients were randomly divided into the two groups.45 cases of the study group were treated by desloratadine citrate disodium tablets combined with narrow ultraviolet B,43 cases of the control group were treated by desloratadine citrate disodium tablets alone.The both two groups applied Binghuangfule cream.The therapeutic effect and safety of the two groups were compared.Results The cure rate in study group was 71.11%,which of the control group was 51.16%,the difference was not statistically significant (x2 =3.69,P > 0.05).The effective rate of the treatment group was 91.11%,which of the control group was 69.77%,the difference between the two groups was statistically significant (x2 =6.43,P < 0.05).Conclusion The result reveals that desloratadine citrate disodium tablets combined with narrow band ultraviolet B is more effective and worthy to be used in clinical practice.

5.Advances in study on factors related to avascular necrosis of the femoral head after paediatric femoral neck fractures

Femoral neck fractures in pediatric fractures account for less than 1% which is very rare, and its mechanism is commonly caused by high-energy trauma. If children with femoral neck fracture cannot receive timely and effective treatment, they are at high risk of avascular necrosis of the femoral head (AVN), coxa vara, bone nonunion, premature physeal closure, leg length discrepancy and other complications. Surgical treatment is currently preferred over conservative treatment, which has a higher complication rate. Among them, AVN is one of the most common and the most difficult complications to manage. So far, no effective treatment measures and reliable predictors have been reported, and the related factors affecting the occurrence of AVN have also been controversial. Once femoral head necrosis occurs in children, the prognosis is not ideal due to the lack of appropriate treatment methods. Therefore, this paper reviews the research progress on the related factors of AVN after femoral neck fracture in children based on the literature reports in the past decade.
6.Heart function changes following transplantation of autologous bone marrow mononuclear cells in a canine model of heart failure induced by rapid ventricular pacing: Pathological image analysis of collagen fiber

BACKGROUND: Stem cell regeneration can repair injured myocardium. However, bone marrow mononuclear cells (BM-MNCs) transplantation for non-ischemic heart failure remains poorly understood.OBJECTIVE: To investigate effect of transplantation of autologous BM-MNCs on cardiac function in canine model of heart failure by rapid ventricular pacing. METHODS: Implantation and model control groups were subjected to model establishment of heart failure by rapid pacing of apex of right ventricle, and respectively injected with CM-DiI-labeled BM-MNCs and normal saline into myocardium. After 4 weeks, all dogs were sacrificed, and specimens of myocardium were collected from the apex, anterior wall and interventricular septum. All specimens were labeled by FITC. Myocardial fibrosis conditions of implanted cells were observed, collagen volume fraction was determined, and hemodynamic indexes were measured. RESULTS AND CONCLUSION: BM-MNCs labeled by CM-DiI and FITC were observed in the transplantation group showing yellow fluorescence, while in the control group FITC-labeled green fluorescence was seen. HE and Masson staining showed that inflammatory cell infiltration in interstitial matrix, displaying interstitial fibrosis and myocardial fibrosis in model control group, but no obvious inflammatory cell infiltration or myocardial fibrosis was observed in the transplantation group, indicating a success model establishment of heart failure by rapid ventricular pacing. Compared with model control group, the collagen volume fraction decreased significantly (P < 0.05), ejection fracture remarkably increased (P < 0.05), but left ventricular end-diastolic and end-systolic diameter remained unchanged in the transplantation group (P > 0.05). Autologous BM-MNCs in canine model of heart failure show myocardium-like cells differentiation, and improve heart function, which possibly associate with the ability of inhibiting the myocardial fibrosis.
8.MR Diagnosis of Stage Ⅰ Tumor Thrombus in Renal Cell Carcinoma

Objective To evaluate the accuracy of MR in the diagnosis of tumor thrombus in renal cell carcinoma.Methods 22patients with renal cell carcinoma proved pathologically were recruited in this study.All the patients underwent MR examination beforesurgery.The results of the MR were correlated with that of the operation.Results 8 renal veins could be found tumor thrombus during the surgery,and the other 14 renal veins and 22 inferior vena cava were absent of tumor thrombus.MR could found all of the 8 tumor thrombithat detected by the surgeons.In the other 14 cases,13 of them could be excluded of tumor thrombus by MR.Only in 1 patient,the MRresult was equivocal.CT was performed in 9 patients,the diagnosis was accurate in 7 cases,and was inaccurate in 2.Ultrasound wasperformed in 16 patients,the diagnosis was accurate in 13 cases,and was inaccurate in 3.Digital subtraction angiography was performed in 4 cases,the diagnosis was correct in all of them.Conclusion MR is perfect in the diagnosis of tumor thrombus in renal cell carcinoma oncondition that the renal vein is clearly defined by MR.It can be considered as the method of choice in the evaluation of tumor thrombus inrenal cell carcinoma.

10.ENHANCED PROTECTION OF FOLIC ACID AGAINST TERATOGENESIS BY COMBINED SUPPLEMENTATION WITH VITAMIN C, B_6 AND TAURINE

Objective:To study the effect of dietary supplementation of folic acid(FA),VC,VB 6 and taurine on teratogenesis of rats induced by cyclophosphamide. Methods:The teratogenesis was induced by cyclophosphamide. Dietary supplementation of FA, VC, VB 6 and taurine was used to depress teratogenesis in pregnant rats. Results:We found that cyclophosphamide administration resulted in decreased weight and length of the fetuses, increased rate of neural tube defects, and retarded ossification. Supplementation of the diet with FA alone or FA+VC+VB 6+taurine resulted in statistically greater fetal weight and length, decreased rate of neural tube defects, and increased ossification grade of supraoccipital bone. Conclusion:The mixture of FA+VC+VB 6+taurine is superior to FA alone in increasing fetal weight and length, reducing the incidence of neural tube defects, and ossification abnormalities of the supraoccipital bone.
