This paper highlights research on the gene transduction with signal factors into germ cells .
2.Effects of Acupuncture at Tianzhu on Lumbar Disc Herniation of Blood Stasis Type

Objective To observe the effect of acupuncture at Tianzhu (BL10) on Blood Stasis Type lumbar disc herniation (LDH). Methods From October, 2013 to October, 2015, 108 LDH cases were randomized into observation group (n=53) and control group (n=55) in accordance with the order of visiting. The observation group received Tuina and acupuncture mainly at Tianzhu, while the control group only received Tuina, for four weeks. The clinical efficacy and improvement of Japanese Orthopaedic Association (JOA) scores were record-ed, and the Visual Analogue Scale (VAS) and serum levels of interleukin-6 (IL-6) were measured before and after treatment. Results The VAS score and IL-6 content decreased in both groups after treatment (t>4.220, P<0.001), and decreased more in the observation group than in the control group (t>3.745, P<0.001). The total effective rate was 94.34% in the observation group, more than 81.82% in the control group (χ2=3.997, P=0.046). The incidence of impromvement of JOA scores was 90.57%in the observation group, more than 76.36%in the control group (χ2=3.920, P=0.048). Conclusion Acupuncture at Tianzhu based on Tuina is more effective on Blood Stasis Type lumbar disc herniation, in term of pain and function, which may relate with decrease of the level of IL-6.
3.Effect of fluoxetine on the viability and lipopolysaccharide-induced tumor necrosis factor-α release of primary cultured rat astrocytes

ObjectiveTo investigate the effects of fluoxetine on viability and lipopolysaccharide (LPS)-induced TNF-α release of primary cultured rat astrocytes.MethodsThe cells were plated on 96-well tissue culture plates and treated with (5,10,20,40) μM fluoxetine for 24 hours,MTT method was used to measure the cell viability.The cells were plated on 48-well tissue culture plates,treated sequentially with (5,10,20,40) μM fluoxetine and 1 μg/ml LPS,and enzymatic linked immunosorbent assay(ELISA) was used to detect the TNF-α level of the cell supematant.ResultsCompared to viability of the control group( OD value:0.20 ± 0.017 ),fluoxetine at the dose of 20 μM,40 μM increased the viability of astrocytes ( OD value:0.23 ± 0.013,0.24 ± 0.012 ) (P <0.05,P<0.01 ).Treatment with LPS for 24 hours,the level of TNF-α was significantly increased ( 53.84 ±24.84) pg/ml compared to the control group( 8.00 ± 10.87)pg/ml (P < 0.01 ),fluoxetine at a dose of 10 μM can suppressed LPS-induced TNF-α release from astrocytes ( 28.85 ± 3.36 ) pg/ml (P < 0.05 ).ConclusionFluoxetine can increase astrocytes viability and suppress LPS-induced TNF-α release from astrocytes.

The paper is to report the establishment of three methods for determination of methyl salicylate-2-O-beta-D-galactopyranoside (1-4)-beta-D-glucopyranoside (MSG) by HPLC, UV or potentiometric titration. The results determined by the three methods turned out to be of no significant difference (P>0.05). The method was chosen according to MSG difference test demands.
5.Effects of novel path of lymph node dissection combined with peripheral vascular occlusion for the treatment of gastric cancer in stage ⅢC

Objective To investigate the clinical efficacy of a novel path of lymph node dissection combined with peripheral vascular occlusion for the treatment of gastric cancer in stage ⅢC.Methods The clinical data of 142 patients with gastric cancer in stage Ⅲ C who were admitted to the 175th Hospital of PLA from January 2006 to December 2009 were retrospectively analyzed.Seventy patients received novel path of lymph node dissection combined with peripheral vascular occlusion (combined group) and 72 patients received simple lymph node dissection via the novel path (simple group).The positive rate of mRNA expression of carcinoembryonic antigen (CEA) and the distal metastatic rate of tumor cells between the 2 groups were compared.Patients were followed up via outpatient examination,phone call and in-patient examination till January 2013.The measurement data and the enumeration data were analyzed using the t test and the chi-square test,respectively.Results The operation time,volume of blood loss and number of lymph nodes dissected were (184 ± 26) minutes,(282 ± 80) mL and 28 ± 5 in the combined group,(180 ± 28)minutes,(355 ± 85)mL and 27 ± 5 in the simple group,with no significant difference between the 2 groups (t =0.882,5.267,0.496,P > 0.05).The positive rates of mRNA expression of CEA of the combined group and the simple group were 10.0% (7/70) and 9.7% (7/72),with no significant difference between the 2 groups (x2 =0.003,P > 0.05).The positive rates of mRNA expression of CEA after lymph node dissection of the combined group and the simple group were 2.9% (2/70) and 16.7% (12/72),with significant difference between the 2 groups (x2 =8.240,P < 0.05).The positive rates of mRNA expression of CEA after total gastrectomy of the combined group and the simple group were 4.3% (3/70) and 4.2% (3/72),with no significant difference between the 2 groups (x2=0.001,P > 0.05).All patients were followed up at postoperative year 1,and 139 patients were followed up at postoperative year 3.The median time of follow-up was 33.5 month (range,12.0-42.0 month).The 1-year distal metastatic rate of the combined group and the simple group were 7.1% (5/70) and 8.3% (6/72),with no significant difference between the 2 groups (x2=0.070,P >0.05).The 3-year distal metastatic rate of the combined group and the simple group were 33.3% (23/69) and 51.4% (36/70),with significant difference between the 2 groups (x2=4.660,P <0.05).The distal metastatic rates of distal parenchymatous organs of the combined group and the simple group were 10.1% (7/69) and 44.3% (31/70),with significant difference between the 2 groups (x2=20.390,P < 0.05).Conclusions Novel path of lymph node dissection combined with peripheral vascular occlusion can block the lymph node and blood backflow,and thus effectively reduces the distal metastatic rate of gastric cancer in stage ⅢC after operation.

Objective:To investigate the effect of Laurencia terpenoid extract (LET) on tumor inhibition,immune modulation and apoptosis of Sarcoma 180 cell. Method:The LD50 of LET was estimated by Horn assay. The models of S180-bearing mice were established and divided into 4 groups which were given LET 0 (control group), 25 (low-dose group), 50 (mid-dose group), 100 (high-dose group) mg/kg bw respectively for 10 d. Tumor inhibition rates were detected in treatment groups and control group respectively. To weigh exactly the thymus and spleen to calculate their indices. The proliferation effect of LET on spleen lymphocyte was evaluated by MTT assay. The apoptosis of tumor cell was assayed by flow cytometry. Results:The LD50 was more than 3 160 mg/kg bw. ObjectiveTo explore the childhood trauma experience and its relation to the eye emotion recognition ability in patients with schizophrenia.MethodsSeventy-one schizophrenic patients and fifty-two healthy subjects were assessed with the Childhood Trauma Questionnaire (CTQ) and the Eye Basic Emotion Discrimination Task (EBEDT).ResultsThe CTQ total score in the patients group was significantly higher than those in the control group (47.86 ± 13.26 vs 34.27 ± 7.27,P < 0.01 ).The subscores in emotional abuse,physical abuse,sexual abuse,emotional neglect and physical neglect of CTQ in the patients group were also significantly higher than those in the control group.(all P< 0.01 ).Compared with controls,the patients had significantly lower total accuracy( (86.74 ± 6.16vs73.42 ± 11.86) %,P < 0.01 ) and longer total reaction time ( (2776.75 ± 703.42 vs 4321.40 ± 1956.83 )ms,P < 0.01 )of EBEDT.The significant difference were also found on the accuracy and reaction time of the EBEDT six basic emotion between the two groups ( all P < 0.01 ).Correlation analysis indicated that CTQ total score were negatively correlated with the EBEDT total accuracy ( r=-0.390,P<0.01 ),and its sub-accuracy of happiness,surprise,sadness,and anger ( respectively r =- 0.419,r =- 0.451,r =- 0.391,r =- 0.344,P < 0.01 ) in the patients group.The correlation between CTQ total score and EBEDT total accuracy was also found in the control group.ConclusionChildhood trauma is related to eye basic emotion recognition in schizophrenia patients,suggesting that the trauma during childhood can affect the social recognition ability.
10.Effect of fluoxetine and pyrrolidine dithiocarbamate on the lipopolysaccharide-induced interleukin 6 release in cultured rat astrocytes

Objective To investigate the effects of fluoxetine and pyrrolidine dithidarbamate (PDTC) on the lipopolysaccharide(LPS)-induced interleukine-6(IL-6) release in cultured rat astrocytes.Methods The purified astrocytes were cultured in 48-well tissue culture plate and classified into control group,LPS group,fiuoxetine group and PDTC group.Control group and LPS group were cultured as usual,and fluoxetine group and PDTC group were cultured with fluoxetine or PDTC at different concentrations for 48 hours,and then LPS group,Fluoxetine group and PDTC group were incubated with 1 ug/ml LPS for 24 hours.Finally,the levels of IL-6 in the cell supernatant were detected by enzymatic linked immunosorbent assay (ELISA) method.Results The level of IL-6 in LPS group ((1975.46 ± 171.54) pg/ml) was significantly higher than that in control group((10633 ± 782.15)pg/ml) (P < 0.01).The levels of IL-6 were (6198.6 ± 379.4) pg/ml,(4973.6 ± 132.5) pg/ml and (4747.9 ±473.9) pg/ml respectively when the concentrations of fluoxetine were 10 μM,20 μM and 40 μM,and (4821.6 ±180.8) pg/ml,(4735.7 ±620.0)pg/ml and (3525.9 ± 240.0)pg/ml respectively when the concentrations of PDTC were 100 μM,150 μM and 200 μM.There was significant difference in the levels of IL-6 between LPS group and fluoxetine group (P < 0.05),as well as between LPS group and fluoxetine group (P < 0.05).Conclusion LPS can induce IL-6 release from astrocytes,while fluoxetine or PDTC at some concentrations can suppress LPS-induced IL-6 release.
