1.Analysis of Phosphate in Rhizosphere Soil Solution Samples Using Capillary Electrophoresis

Analytical method of capillary zone electrophoresis with indirect UV detection was optimized and validated to determine the phosphate in rhizosphere soil solution samples.The effect of detection wavelength, electrolyte composition, separation voltage and temperature were investigated.Analyses were performed in a 56 cm uncoated fused-silica capillary(length to the detector window) with reversed electroosmotic flow, anodic detection in 205 nm, separation voltage-20 kV, and temperature 25 t.The electrolyte contained 32 mmol/L tris-hydroxymethyl-aminomethane, 4 mmol/L 1,2, 4-benzene-tricarboxylic trimellitic acid and 0.3 mmol/L tetradecyl-trimethylammonium bromide, pH 8.5.Under the conditions, the negative effect of the inorganic anion, such as Cl~-, SO_4~(2-) and NO_3~-, can be reduced on phosphate.The detection limit was 0.68 mg/L(S/N=3), and the recovery was 87.2%-99.4%.The method has been applied to the determi nation of the concentration of phosphate in the rhizoshere soil solution.
2.The detection and significance of cardiac troponin Ⅰ, myoglobin and creatine kinase MB in old patients with acute myocardial infarction

ObjectiveTo explore the changes and significances of cardiac troponin Ⅰ (cTnⅠ),myoglobin (Mb) and creatine kinase MB (CK-MB) in old patients with acute myocardial infarction(AMI).MethodThe levels of cTnⅠ, Mb and CK-MB in 89 cases with AMI patients (AMI group) and 100 healthy controls (control group) were detected and compared. ResultsThe levels of cTnⅠ, Mb and CK-MB in AMI group were ( 8.15 + 3.26) μ g/L, (478.45 ± 96.87 ) μ g/L and ( 128.17 ± 53.26 ) U/L, while were (0.03 ±0.02) μ g/L, (21.61 + 9.38 ) μ g/L and (9.53 ± 2.94) U/L in control group, the levels of cTnⅠ, Mb and CK-MB in AMI group were significantly higher than those in control group(P < 0.05 ). The sensitivity rate of cTnⅠ,Mb and CK-MB for detecting AMI was 95.5% (85/89), 97.8% (87/89) and 87.6% (78/89), and the specificity rate was 98.0%(98/100), 82.0%(82/100) and 94.0%(94/100). ConclusionThe levels of cTnⅠ, Mb and CK-MB are significantly increased in AMI patients, which cTnⅠ for detecting AMI has high sensitivity and specificity.
3.Influence of postural intervention to ambulation of patients after video-assisted thoracoscope surgery

Objective To study the influence of postural intervention on ambulation of patients after video-assisted thoracoscope surgery? Methods One hundred and forty eight patients having undergone chest cardiac surgery were randomly divided into the control group(n = 75)and the observation group(n = 74)? The former received routine nursing intervention and the latter postural intervention besides routine nursing? The two groups were compared in terms of the time for ambulation and the ability in ambulation? Result The ambulation in the observation group was earlier than the control group and the ability in ambulation was better compared to the control group(both P < 0?05)? Conclusion The postural intervention after thoracic surgery is effective in pushing patient’s ambulation and improve their ability?
4.Observation of ankle pump motor in the prevention of deep vein thrombosis after surgical operation

Objective To evaluate the influence of ankle pump motor on deep vein thrombosis after surgical operation.Methods 138 cases of patients after surgical operation who were assessed as high risk patients,were randomized into the study group (70 cases)and the control group (68 cases),routine nursing was adopted in the control group,while in the study group,ankle pump motor was adopted in addition.The incidence of deep vein thrombosis was compared between two groups.Results No deep vein thrombosis was found in the study group,while 1 case of thrombosis was detected in the control group,but there was no statistic difference.Conclusions Ankle pump motor was easy and convenient,and can prevent deep vein thrombosis by promoting the reflux of the lower limbs,improving the recovery of patients after operation,and increasing nursing quality as well.
5.Research progress of Piezo mechanosensitive ion channels in bone and joint tissues

Mechanosensitive ion channels are mechanoreceptors that convert mechanical stimulation on the cell surface into intracellular electrical signal or chemical signal. In recent years, Piezo protein as a mechanically activated cation channel in eukaryotes has gradually become a research hotspot. Piezo1 and Piezo2 proteins are expressed in different organs and tissues of mammals, and tissues such as bones and joints are directly related to mechanical conduction. The intracellular effects caused by the conduction of mechanical force play a very important role in the treatment and prevention of orthopedic diseases. In this paper, the researches on Piezo protein mechanically activated cation channels in bone tissue, cartilage tissue, bone marrow tissue, intervertebral disc tissue and orthopedic diseases were reviewed.
6.Diagnostic value of genexpert MTB/RIF in tuberculosis and population characteristics of retreated patients

Objective:To explore the reliability of Xpert MTB/RIF (Xpert) in the diagnosis of pulmonary tuberculosis and to understand the characteristics of retreated patients, so as to provide clues for the investigation of retreated patients.Methods:905 patients sputum with suspected tuberculosis from July 2019 to December 2019 were collected from the Tuberculosis Department of Changsha Central Hospital. Xpert was used for rapid screening of tuberculosis. MGIT960 and Roche culture method were used for rapid sputum culture and drug sensitivity, respectively. Rifampin (RIF) resistance was compared between Xpert and Roche culture. The gender, age, RIF resistance, occupation and diabetes mellitus of patients with positive Xpert were compared, and the risk factors of RIF resistance were analyzed by binary logistic regression.Results:The total detection rate of Xpert was 28.7%(260/905) in 905 suspected pulmonary tuberculosis patients; RIF resistance rate accounted for 20.8%(54/260) in 260 positive patients, and the main mutation types were probe-E and probe-D. There was no significant difference between traditional drug sensitivity and Xpert method in RIF resistance detection ( P>0.05); There was no significant difference between Xpert method and traditional drug sensitive cases in predicting isoniazid (INH) resistance ( P=0.293); there were significant differences in age ( t=-3.835, P<0.05), gender, RIF resistance rate and proportion of farmers between newly treated and retreated patients (χ 2=5.091, 16.862, 4.808, P<0.05); Binary logistic regression showed that retreatment was a risk factor for RIF resistance ( OR=3.739, P=0.004). Conclusions:Xpert has high clinical value in the diagnosis of pulmonary tuberculosis and RIF resistance; The retreated patients were older, and the proportion of male farmers was larger, with higher resistance rate to RIF.
7.A biomechanical model of knee osteoarthritis and stress characteristics on the cartilage surface

BACKGROUND:Knee osteoarthritis is a chronic disease characterized as cartilage degeneration and hyperostosis to impact the soft tissues around the joints, thereby resulting in joint pain, and it can be aggravated and lead to joint deformity after weight-bearing. OBJECTIVE:On the basis of imaging parameters of knee osteoarthritis patients and normal controls, a model was established. And then, knee squatting movement was simulated on the model and stress data from the cartilage surface of patients and normal controls were colected and compared, thereby to obtain the stress characteristics of the cartilage surface in knee osteoarthritis patients. METHODS: There were 30 knee osteoarthritis patients and 30 healthy volunteers in the study. CT and MRI data from these participants were colected and analyzed using Mimics software and simpleware software to establish a model. Based on this model, cartilage-related stress data were harvested from knee osteoarthritis patients and healthy volunteers and compared to summarize the stress characteristics of the articular cartilage in patients with knee osteoarthritis. RESULTS AND CONCLUSION:In knee osteoarthritis patients, the stress on the cartilage surface from standing to squatting was increased in a nonlinear manner. During the process of squatting, the mediolateral stress was higher in the knee osteoarthritis patients than healthy controls (P < 0.05); however, there was no difference in the proximodistal and anteroposterior cartilage surface stress between the two groups (P > 0.05). These findings indicate that the knee osteoarthritis musculoskeletal model and finite element model are established successfuly, and these models are closer to the real motion characteristics of the knee joints, based on which, quantitative biomechanical data of the knee can be provided for knee osteoarthritis treatment.
8.Effect of Neurotrophin 3-chitosan on Endogenous Neurogenesis and Motor Function after Motor Cortex Injury in Rats

Objective To observe the effects of neurotrophin 3 (NT3)-chitosan on motor function, and proliferation and differentiation of the neural stem cells (NSCs) in the injury area and subventricular zone (SVZ) in rats with motor cortex injury. Methods Sixty-five Wistar rats were divided into control group (n=7), injury group (n=29) and NT3-chitosan group (n=29). The motor cortex was aspirated and re-moved as cerebral injury model. NT3-chitosan was immediately implanted into the injured area after operation, and the control group re-ceived no intervention. Pellet reaching test was performed to detect the recovery of the forelimb function, HE staining was used to observe the lesion cavity size, and immunofluorescence staining was used to observe the proliferation and differentiation of NSCs 3 days, 7 days, 14 days, 1 month, 2 months and 3 months after operation. Results The grasp success rate was higher (F>6.00, P≤0.05), and the lesion cavity size was significantly smaller (F>629.5, P<0.001) in the NT3-chitosan group than in the injury group. In the NSCs differentiation experi-ment, the number of BrdU cells at all the time points was significantly higher in the NT3-chitosan group than in the injury group (F>171.43, P<0.001). In the NSCs proliferation experiment, the number of BrdU positive cells was still significantly higher in the NT3-chitosan group than in the control group and in the injury group (F>155.06, P<0.001), the number of Dcx positive cells was significantly higher in the NT3-chitosan group than in the injury group (F=62.367, P<0.001), and the number of BrdU/Dcx positive cells was significantly higher in the NT3-chitosan group than in the control group (F=33.527, P<0.001). Conclusion NT3-chitosan could activate NSCs in the SVZ, and pro-mote endogenous neurogenesis and forelimb function recovery in rats after motor cortex injury.
9.Clinical study of compound glutamine for adjuvant therapy after radical surgery in gastric cancer

Objective To evaluate the clinical effect of compound glutamine in the adjuvant treatment after radical surgery of gastric cancer.Methods A total of 63 cases with gastric cancer after radical resection were randomly divided into treatment group and control group, according to random number table.32 patients in treatment group received simultaneous three-dimensional conformal radiotherapy with capecitabine monotherapy regimen, while oral administration of compound glutamine enteric-coated capsules.31 cases in control group only received chemoradiotherapy.Results In treatment group, the quality of life score for the excellent and good accounted for 40.6% and 34.4%, significantly higher than 16.1% and 12.9% in control group respectively (x2 =4.63, P =0.03;x2 =4.00, P =0.04).The incidence of nausea and vomiting, abdominal pain and diarrhea in treatment group respectively were 31.3% and 37.5% , significantly less than 61.3% and 64.5% in control group with statistical significance (x2 =5.72, P =0.02;x2 =4.60, P =0.03).The incidence of gastrointestinal reactions in treatment group were mainly Ⅰ and Ⅱ degrees, and Ⅲ-Ⅳ degree of gastrointestinal reactions were not appear.While the antidiarrheal drugs (21.9% vs.48.4%) and analgesics drugs (15.6% vs.38.7%) in the treatment group were significantly reduced compared with the control group (x2 =4.87, P =0.03;x2 =4.26, P =0.04).Conclusion By administration of compound glutamine in the course of adjuvant chemotherapy after radical surgery of gastric cancer can improve the quality of life, reduce chemotherapy-induced gastrointestinal side effects, improve treatment compliance, and successfully complete the treatment.
10.Mechanism Study on Protective Effect of Extracts from Rhizoma Anemones Raddeanae on Hepatic Fibrosis Induced by Porcine Serum in Rats

This study was aimed to observe the protective effect of extract from Rhizoma A nemones Raddeanae (RAR) on hepatic fibrosis induced by porcine serum in rats. A total of 68 SD rats were randomly divided into 5 groups, which were the normal group, model group, RAR group, extraction of RAR (EXRAR) group, Fu-Zheng Hua-Y u(FZHY) group. Each rat was intraperitoneally injected with 0.5~0.6 ml of porcine serum twice a week for 15 suc-cessive weeks to establish liver fibrosis model. Intragastric administration was given after the model was successfully established. The FZHY group was given FZHY capsule (0.525 g·kg-1). The RAR group was given RAR decoction (0.7 g·kg-1). The EXRAR group was given EXRAR (0.071 g·kg-1). The model group and normal group were given e-qual amount of physiological saline. The medication was given once a day. And the treatment course was 8 weeks. At the end of the 23th week, rats were sacrificed. Contents of SOD and MDA in blood serum were assayed. The protein expressions of α-SMA and TGF-β1 in liver tissues were detected by SABC. The results showed that compared with the model group, content of MDA decreased in the EXRAR group, RAR group and FZHY group (P<0.05), and content of SOD increased obviously (P<0.05). In the model group, expression of α-SMA and TGF-β1 increased, with dark brown dyeing and diffusion area. Expression area and strength of the FZHY group, RAR group, and EXRAR group were ob-viously weak with tasteless interval dyeing and no formation of typical pseudolobule in comparison with the model group. The color rendering index showed that compared with the model group, the protein expression of α-SMA and TGF-β1 decreased obviously in liver tissues of the FZHY group, EXRAR group, and RAR group (P< 0.05). It was concluded that RAR and its extract had a good antifibrosis effect. And the EXRAR had basically the same antifibrosis effect as RAR. It was assumed that the possible mechanism was related with the inhibiting of hepatic stellate cell (HSC) activation and the expression of TGF-β1 as well as the resisting of lipid peroxidation.
