1.In vitro effect of Fructus ligustri lucidi and its monomers on the migration of mouse melanoblasts

Objective To study the effects of Fructus ligustri lucidi and its monomers, tyrosol and oleanotic acid, on the migration of mouse melanoblast cell line (NCCmelb4M5). Methods Cultured NCCmelb4M5 cells were treated with Fructus ligustri lucidi (0.0625, 0.125, 0.25, 0.5, 2 mg/mL), tyrosol (0.02, 0.04, 0.08, 0.16, 0.8 mg/mL) and oleanolic acid (0.0625, 0.125, 0.25, 0.5, 2.5 mg/mL), respectively,for 48 hours followed by the detection of cell proliferation with MTT assay. The working concentration of the three drugs was determined according to the results of MTT assay. Scratch and transwell assays were performed to observe the effect of Fructus ligustri lucidi and its monomers at working concentration on the migration of NCCmelb4M5 cells. Results Based on the results of MTT assay, the working concentration of Fructus ligustri lucidi, tyrosol and oleanolic acid was determined at 0.125 mg/mL, 0.08 mg/mL and 0.0625 mg/mL respectively, and at these concentrations, these drugs exhibited a cytotoxity lower than that of absolute alcohol with no obvious stimulation of cell proliferation. Scratch and transwell assay revealed a promoting effect of both Fructus ligustri lucidi and tyrosol on melanoblast migration (P<0.05), while oleanolic acid had little effect on melanoblast migration. Conclusions The extract of Fructus ligustri lucidi has a significant stimulatory effect on the migration of mouse melanoblasts, and tyrosol may be an active component of Fructus ligustri lucidi associated with confirmative effect on migration of mouse melanoblasts.
2.Preliminary evaluation of hypersensitive C-reactive protein rapid quantitative chemiluminescent detection kit

Objective To evaluate the performance of the hypersensitive C-reactive protein(hs-CRP) rapid quantitative chemilu-minescent detection kit .Methods According to National Committee for Clinical Laboratory Standards (NCCLS) EP10-A2 docu-ment ,hs-CRP rapid quantitative chemiluminescent detection kit was employed to measure the CRP at low ,medium and high con-centration levels of quality control serum .Bias ,total imprecision and their slope rates ,intercepts ,carryover ,non-linearity and drift were calculated ,and its clinical acceptability was evaluated .Results Bias and total imprecision of hs-CRP rapid quantitative chemi-luminescent detection kit were within the allowable ranges ,the average values of slope rates ,intercepts ,carryover ,non-linearity and drift were 1 .005 7 ,0 .537 8 ,0 .789 6% ,0 .019 2 ,0 .036 0 ,respectively ,the differences showed no statistically significance ( P>0 .05) .Conclusion hs-CRP rapid quantitative chemiluminescent detection kit has good accuracy and precision ,stable perform-ance ,and consistent with the clinical testing requirements .
3.Evaluation of mycosis fungoides with reflectance confocal microscopy

Objective To investigate the reflectance confocal microscopy (RCM) features of mycosis fungoides (MF).Methods Four male and two female patients with MF,who were aged 7-73 years with a clinical course of 1-5 years,were collected from June 2011 to May 2012 in the department of dermatology.Of the six patients,two were in patch stage,three in plaque stage,and one in tumor stage.All the cases were confirmed by histopathological,immunohistochemical and relevant examinations.For each patient,three lesions were selected and investigated using RCM,and one of the three lesions was subjected to histopathological examination.Results RCM features of MF included hyperkeratosis,disarray of honeycomb of stratum spinosum,reduced refractivity of dermal papillary rings,roundish or round scattered hyperrefractive cells in the epidermis,infiltration of a small quantity of inflammatory cells in the papillary dermis.Occasionally,some round to oval hyporefractive cells aggregated to form vesicle-like structures.These RCM features correlated well with histopathologic findings.Conclusion Combined with clinical manifestations,RCM may serve as a noninvasive auxiliary tool for the diagnosis of MF.
4.Relationship between the clinical onset of cerebral infarction in anterior cycle and properties of carotid atheromatous plaque

Objective To investigate the relationship between the clinical onset of cerebral infarction in anterior cycle and properties of carotid atheromatous plaque.Methods A retrospective analysis was carried out on clinical data of 624 cases of cerebral infarction in anterior cycle from December 2010 to December 2013 in our hospital.The carotid artery plaque Crouse score,index,and intima-media thickness (IMT) were used to evaluate carotid atherosclerosis and plaque lesions.The National Institute of Health stroke scale (NIHSS) questionnaire was used to evaluate neural function defect.Results Compared to the non-lesion side,the lesion side carotid artery plaque Crouse score was decreased and IMT was increased (P < 0.05) ; the lesion carotid artery plaque Crouse was lower,the higher NIHSS scores (r =-0.546,P =0.004) ; IMT score was higher,higher NIHSS score; carotid artery atheromatous plaque Crouse was negatively related to NIHSS score; IMT score was positively related to NIHSS score (r =0.672,P =0.003).Conclusions The clinical onset of cerebral infarction in anterior cycle was correlated with the carotid sclerosis degree.The Crouse score and IMT of the lesion have certain significance in predicting prognosis of cerebral infarction in anterior cycle.
5.Expression of NF-E2 related factor in vitiligo lesions

Objective To investigate the level of NF-E2 related factor 2 (Nrf 2) in vitiligo lesions.Methods Tissue samples were obtained by press suction blisters at lesional and donor sites of 12 patients with vitiligo who were managed with epidermal transplantation. Four lesional samples from the patients were subjected to primary culture and the level of Nrf 2 was detected by AEC immunohistochemistry after 48hours of culture. Western blotting was utilized to further detect the level of cytoplasmic and nuclear Nrf 2 in tissue samples from the other 8 patients with vitiligo. Results Immunohistochemistry revealed that Nrf 2 was predominantly expressed in cytoplasm, rather than nuclei, of keratinocytes in vitiligo lesions compared with the normal skin of patients. The level of nuclear Nrf 2 was significantly lower in lesions than that in normal skin (0.10 ± 0.03 vs 0.26 ± 0.03, P < 0.01) of the patients. In contrast, there was no significant dif- ference in the level of cytoplasmic Nrf2 between lesional and normal skin (0.61 ± 0.03 vs 0.60 ~ 0.02, P >0.05) of patients. Conclusion These results reveal an abnormality of nuclear translocation of Nrf 2 in vitili-go lesions.
6.Clinical Analysis of243Patients with Vitiligo and the Association with HLA-DQB1

Objective To analyze the clinical features and two HLA-DQB1alleles in their associa-tions with vitiligo.Methods Clinical data of243patients with vitiligo were collected and the HLA-DQB1al-leles were determined by a PCR-SSP assay in both patients and control groups.Results The mean age of the first onset was23.1years for the males,and23.7years for the females.The localized type was the major form in the initial stage.The segmental type mainly occurred in childhood.Age of the first onset was younger in pa-tients with familial history than those in patients without familial history.HLA-DQB1*0201allele was associ-ated with non-segmental type only.Conclusions①The pathogenesis might be different between non-segmen-tal and segmental vitiligo.②Polygenic inheritance may explain vitiligo with familial history.There might be a difference of genetic background between vitiligo with or without familial history.③HLA-DQB1*0201allele may be the susceptible gene or have close linkage with susceptible gene in vitiligo vulgaris.
7.Studies on expression, purification, crystal growth and optimization of putative transcription factor LytR from Streptococcus pneumoniae.

The aim of the present study was to obtain the crystal of transcription factor LytR of streptococcus pneumoniae for X-ray crystal structure and function analysis. The LytR gene of D39 strains from Streptococcus pneumoniae (S. pn) was cloned into the prokaryotic expression vector pET32a(+), then overexpression was obtained in the E. coli BL21 (DE3) through transformation of the recombinant plasmid that had been verified by colony PCR and sequencing. Soluble fusion protein with His-tag highly expressed by the induction of 0.5 mmol/L IPTG and was purified by a three step procedure, the purity of the purified LytR recombinant protein was over 90%. Preliminary screening of crystallization conditions was performed using the hanging-drop vapour-diffusing method with Hampton Crystal screen and PEG screen kits. The protein crystals X-ray diffraction data were collected from a single crystal and more stick crystals whose X-ray diffraction reached 4.0 A were obtained. These works laid the foundation for further research on the 3D structure of putative transcription factor LytR and its biological aspects.
