2.Therapeutic observation of grain-sized moxibustion for chemotherapy-induced myelosuppression for non-small cell lungcancer

Objective: To observe the clinical efficacy of grain-sized moxibustion in treating chemotherapy-induced myelosuppression for non-small cell lung cancer (NSCLC) and its effect on quality of life (QOL). Methods: Eighty NSCLC patients admitted to the Inpatient Department of Zhejiang Cancer Hospital between September 2016 and March 2018 were recruited and divided into an observation group and a control group by random number method, with 40 cases in each group. The two groups both received chemotherapy with paclitaxel plus cisplatin (TP regimen). The control group received oral administration of leucogen tablets starting from the first day of chemotherapy, 20 mg each time, three times a day, for consecutive 14 d; the observation group was additionally given grain-sized moxibustion, once a day, five days per week at a two-day interval, until the fourteenth day. The myelosuppression severity was observed and compared between the two groups prior to chemotherapy, at the 3rd, 7th and 14th days of chemotherapy; the QOL in the two groups was evaluated before chemotherapy, at the 14th and 21st days of chemotherapy. Results: Regarding myelosuppression, the peripheral blood indicators increased significantly at the 3rd day of chemotherapy in both groups (P<0.05 or P<0.01); at the 7th and 14th days of chemotherapy, the peripheral blood indicators presented a decreasing tendency in the two groups, but the level in the observation group was still significantly higher than that before chemotherapy (P<0.01); at the 3rd, 7th and 14th days of chemotherapy, the peripheral blood indicators in the observation group were higher than those in the control group (P<0.05 or P<0.01); the occurrence rate of myelosuppression in the observation group was significantly lower than that in the control group (P<0.01). The QOL score in the observation group was markedly higher than that in the control group at the 14th and 21st days of chemotherapy (both P<0.05). Conclusion: Grain-sized moxibustion can effectively improve myelosuppression after chemotherapy for NSCLC, reducing its occurrence and enhancing the patient's QOL.

4.Role of calpain-fibronectin signaling inestrogen-induced migration in breast cancer cells

Aim To study the effect of estrogen on the migration of breast cancer cells and the possible underlying mechanisms.Methods Human breast cancer cell lines MCF-7(estrogen receptor, ER+) and MDA-MB-468(ER-) were employed as a model system.Cells were treated with E2 and pretreated with CANP inhibitor(calpeptin)where needed.Wound-healing assay was applied to evaluate cell migration, Western blot assay was performed to observe protein level, and fibronectin expression was silenced by specific siRNA transfection.Results ① Treatmentof MCF-7 and MDA-MB-468 cellswith E2(50 nmol·L-1) increased cell migration by(51.55±5.50) %(P<0.01)and (40.78±4.78)%(P<0.05), respectively;② E2 significantly up-regulated the expression of FN protein in MCF-7 and MDA-MB-468 breast cancer cells, which was 2.11 times(P<0.05)and 1.86 times(P<0.01), respectively;③ Pretreatment with calpeptin(10 μmol·L-1) decreased E2-induced cell migration by (49.55±6.44)%(P<0.05) in MCF-7 and(36.85±4.40)% (P<0.01)in MDA-MB-468 cells;④ Calpeptin pretreatment inhibited E2-induced fibronectin up-regulation by(80.12±4.55)% and(78.84±5.70) %(P<0.01), respectively;⑤ Knockdown of FN with siRNA suppressed cell E2-induced migration by(40.65±5.80)%(P<0.01)in MCF-7 and(40.88±6.02)%(P<0.05)in MDA-MB-468 cells.Conclusion E2 stimulates the migration of breast cancer cells with or without ER expression and a calpain-FN signaling pathway may be involved in the E2 action.
5.The role of neuronal and glial activation in spinal cord in the mechanism of neuropathic pain following three different peripheral nerve injuries in rats

Objective To determine if there is any difference in neuronal and glial(astrocytic and microglial)activation in the spinal cord in three rat models of neuropathic pain.Methods Twenty-four SD rats weighing 150-200 g were randomly divided into 4 groups(n=6 each):Ⅰ control group;Ⅱ chronic constrictive injury group(CCI);Ⅲ spinal nerve ligation group(SNL)and Ⅳ spared nerve injury(SNI).No operation was performed in control group.In CCI group left sciatic nerve was exposed and loosely ligated with catgut.In SNL group the L_5 spinal nerve was exposed and ligated with silk suture and cut.In SNI group tibial nerve and common fibular nerve were ligated and cut.Pain threshold was measured using plantar tactile stimulator(Ugo,Basile Co. Italy)every other day from 3 days before until 15 days after operation.50% paw withdrawal threshold was measured using up-and-down sequential mechanical stimulation of different intensity(0.45,0.70,1.20,2.00, 3.63,5.50,8.50,15.10 g)applied to the plantar surface of the injured paw.On the 15~(th) day after operation after pain threshold was measured the animals were anesthetized with intraperitoneal 10% chloral hydrate 400 mg? kg~(-1).The L_(5,6) segment of the spinal cord was isolated.Neuronal,astrocytic and microglial activation was determined by immuno-histochemistry with antibodies of c-Fos(a proto-oncogene protein),GFAP(an astrocyte marker)and OX-42(a microglial marker).Results The 50% paw withdrawal threshold reached the lowest level on the 7~(th) day after operation.The lowest level was maintained until the 15~(th) day after operation in group CCI,SNL and SNI.The 50 % paw withdrawal threshold was(14.1+1.5)g in control group,(2.5+0.5)g in CCI group, (1.5?0.6)g in group SNL and(0.8?0.4)g in group SNI.The number of c-Fos positive neurons in laminae Ⅳ-Ⅵ of dorsal horn was significantly greater in group CCI,SNL and SNI than in control group,but there was no significant difference among the 3 peripheral nerve injury groups.The activation of astrocytes and microglias in laminae Ⅰ-Ⅳ of dorsal horn was significantly increased in group CCI,SNL and SNI than in control group but there was no significant difference among the 3 peripheral nerve injury groups.Condusion There is no significant difference in activation of neurons and astrocytes and microglias in the ipsilateral dorsal horn among the 3 pain models.
6.Molecular cloning of tomato LeHsp110/ClpB gene and its effect on the thermotolerance in plant.

The heat shock protein ClpB is a member of the Clp family and functions as molecular chaperones. ClpB is related to the acquired thermotolerance in organisms. A cDNA of 3144 bp was screened out of a tomato cDNA library. The polypeptide deduced from the longest ORF contains 980 amino acid residues, and was classified into HSP100/ClpB family based on the result of molecular phylogenesis analysis. Thus it was named as LeHSP110/ClpB according to its calculated molecular weight. LeHSP110/ClpB was characteristic of heat-inducibility but no constitutive expression, and was demonstrated to locate in chloroplastic stroma. An antisense cDNA fragment of LeHsp110/ClpB under the control of CaMV 35S promoter was introduced into tomato by Agrobacterium tumefactions-mediated method. At high temperature, the mRNA levels of LeHsp110/ClpB in antisense transgenic plants were lower than those in control plants. The PS II of transgenic plants is more sensitive to high temperature than that of control plants according to data of Fv/Fm. These results clearly showed that HSP110/ClpB plays an important role in thermotolerance of high plants.

7.Development of virulent heat-evil-induced thrombosis animal model.

OBJECTIVETo develop a virulent heat-evil-induced thrombosis animal model, and provide a rational animal model for pathogeny and pathogenesis research of thrombosis-related diseases, anti-thrombosis activity screening and pre-clinical studies of CAHT formula.
METHODSD rats were pretreated with carrageenin (Ca) intraperitoneal injection, followed by intravenous injection of endotoxin (LPS from E. coli O111:B4) 50 microg x kg(-1) 16 h later. Thrombosis in rat tails were observed during 12-24 h after injection of LPS. The inflammatory mechanism of this model were investigated by analyzing serum level of TNF-alpha, IL-6, TXB2 and 6-keto-PGF 1alpha, CD11b/CD18 expression of white blood cells (WBC) and P-selectin expression of vessel walls.
RESULTIn LPS/Ca model group, thrombosis can be clearly observed in the distal part of rat tails after 12-24 h of LPS/Ca treatment. High level of TNF-alpha and IL-6 can be measured in serum. The expression of CD11b/CD18 in WBC and P-selectin in vessel endothelium significantly increased and the number of WBC in peripheral blood markedly decreased shortly after LPS/Ca treatment. The adherence of white blood cells to vessel endothelium which can be seen by microscope mainly contributed to the decrease of WBC. The results indicated that there was obvious inflammation after treatment with LPS/Ca, suggesting that inflammation was the key mechanism for this model.
CONCLUSIONThis model was developed through treatment of LPS in combination with Ca, of which LPS is considered to be an exotic virulent heat-evil in TCM, while the inflammatory molecules produced in this model, such as TNF-alpha, IL-6, CD11b/CD18 and P-selectin belong to internal virulent heat-evils, so this animal model consists of pathogeny and pathogenesis of virulent heat-evils. virulent heat-evil.

8.Expression of Bcl-w protein in human small intestinal adenocarcinoma and effect of Bcl-w siRNA on apoptosis in intestinal adenocarcinoma HuTu-80 cells.

OBJECTIVETo investigate the expression of apoptosis-related protein Bcl-w in adenocarcinoma of the small intestine, and the apoptotic effect of Bcl-w siRNA on small intestinal adenocarcinoma cells HuTu-80.
METHODSForty-two tissue samples were examined in our study, including 7 cases from human small intestinal adenocarcinoma, and 35 cases from normal small intestine served as control. The expression of Bcl-w was detected by immunohistochemistry (IHC). Western blot analysis was performed to confirm whether Bcl-w siRNA could effectively down-regulate Bcl-w protein after HuTu-80 cells were transfected with Bcl-w siRNA. The cells were treated with chemotherapeutic agent 5-Fu to observe whether Bcl-w protein-silecing affects the pro-apoptotic effect of 5-Fu. Flow cytometry analysis was used for assessment of apoptotic rate of HuTu-80 cells cultured with Bcl-w siRNA alone, with 5-Fu alone, and with combination of Bcl-w siRNA and 5-Fu, using untreated HuTu-80 cells as control.
RESULTSThe positive rate of Bcl-w expression was significantly higher in small intestinal adenocarcinoma than that in normal tissue (85.7% vs. 25.7%, P=0.005). Compared with the control group, Bcl-w siRNA transfection effectively down-regulated the expression of Bcl-w protein (P<0.05). The apoptosis in HuTu-80 cells was not increased significantly in Bcl-w-/-cells compared with that of control group (12.4±2.2)% vs. (8.6±1.7)% (P>0.05). However, compared with the 5-Fu group, the apoptosis in HuTu-80 cells was effectively enhanced after combination treatment with Bcl-w siRNA and 5-Fu (45.7±2.1)% vs. (71.6±3.2)% (P<0.05).
CONCLUSIONSBcl-w protein plays a significant role in the carcinogenesis of human small intestinal adenocarcinoma. Down-regulation of Bcl-w protein in small intestine adenocarcinoma HuTu-80 cells leads them susceptible to 5-Fu.

9.Post-term pregnancy and associated social factors among pregnant women in Jiaxing city, China, 1993 to 2000

Objective To understand the status of post-term pregnancy and associated social factors among pregnant women in Jiaxing city, from 1993 to 2000. Methods The study population consisted of 16 033 women who had delivered singleton live births in Jiaxing city, Zhejiang province, from 1993 to 2000. Chi-square test was employed to test the difference of proportions between respective groups. Multivariate logistic regression was conducted to examine the factors associated with post-term pregnancy. Results The incidence rates of post-term pregnancy were 8.7%, 10.6%, 9.4%, 7.4%, 4.3%, 2.6%, 3.3% and 4.0% from 1993 to 2000, respectively. Women who received premarital health care serviees had lower (5.6%) post-term incidence than those who did not (7.8%). Women who received county or above level health care services had lower (4.3%) post-term incidence than women who receive township level service (8.5%). Women who had primary school education had higher (11.5%) post-term incidence than women who received junior higher school education (6.8%), senior high school education (3.8%), or college education (2.9%), (P< 0.001). Farmers had higher (9.5%) post-term incidence than workers in the rural areas (8.9%), in urban areas (4.3%) or women with other professions (4.2%). Women whose husbands were rural workers had higher (9.8%) post-term incidence than farmers (9.4%), urban workers (5.0%)or with other professions (3.6%, P<0.001). Data from multivariate logistic regression showed that the risk of post-term pregnancy was associated with the length and the level of using health care services together with schooling and occupation. Conclusion The incidence of post-term pregnancy was decreasing in Jiaxing city during the period from 1993 to 2000 and the risk was associated with the length and level of health care service as well as the level of education and occupation of those women.
10.A study on polymorphism of salivary esterase in personal identification.

OBJECTIVE:
To research the value of polymorphism of salivary esterase(Set) in paternity and personal identification.
METHODS:
Phenotype and genotype of human salivary esterase were detected in 114 liquid saliva samples from the Chinese population by disc electrophoresis and fast blue RR staining assay.
RESULTS:
The frequency of Set type was F 22.81%, FS 50.88%, S2 6.31%. The estimated gene frequency of SetF was 0.4825 and SetS was 0.5175. The PE was 0.1875 and the DP was 0.6199.
CONCLUSION
Polymorphism of salivary esterase (Set) was practical in paternity and personal identification.
