2.Investigation and Analysis on Dysmenorrhea and Constitution of Students in Adolescent Age

Objeclive To explore relationship between TCM constitution and primary dysmenorrhea of female students in adolescent age,and to observe clinical effect of treating dysmenorrhea of students whose constitution tends toward Qi-deficiency and Yang-deficiency with wanning meddian therapy.Methods Relationship between TCM constitution and dysmenorrhea was performed in 789 students by questionnaire.Results Students with dysmenorrhea accounted for 50.3%,in which students with mild constitution accounted for 70.2%,and students with constitution tending toward Qi-deficiency and Yang-deficiency accounted for 80%.Conclusion Dysmenorrhea is associated with constitution.Warming meridian therapy can adjust constitution tending toward Qi-deficiency and Yang-deficiency.
3.Analysis of HBV precore 1896 site mutation and its HBV genotype and other laboratory features

Objective:To analyze the relationship between hepatitis B virus(HBV)gene mutation at 1896 in precore region with genotype and replication of HBV and the liver function of patients.Methods:HBV precore 1896 site mutation,the genotype of HBV and serum content of HBV DNA were determined by PCR in 60 patients positive of HBV DNA.Chemiluminescence miacropaticle immunoassay(CMIA)was used for detection of serum HBeAg and HBeAb.Liver function parameters were ob- tained by routine biochemistry method.Results:The alanine aminotransferase(ALT)level in HBV with 1896 site mutation was significantly higher than that in the wildtype virus.Site mutation at 1896 had no correlation with HBeAg,HBV genotype and HBV DNA content.HBV DNA content in patient with genotype C was significantly higher than that with genotype B(P
4.Effects of Morroniside on Activation of Caspase-3 in Rats with Focal Cerebral Ischemia/Reperfusion

Objective To investigate the effects of morroniside on activation of caspase-3 in rats with focal cerebral ischemia/reperfusion. MethodsThe model was induced with occlusion of middle cerebral artery with suture embolus, ischemia for 30 min, and reperfusion for 72 h in rats. Vitamin E used for the positive control. The activity of caspase-3 was detected with spectrophotometry. ResultsCompared with sham group, the caspase-3 activity increased obviously in model rat. Compared with model group, Morroniside (30 mg/kg,90 mg/kg,270 mg/kg) decreased the activation of caspase-3 remarkably, which was dose-dependent (P<0.05). ConclusionMorroniside may reduce apoptosis by decreasing the activation of caspase-3 in rats.
5.Effects of Morroniside on IL-1β in Rat Cortex with Focal Cerebral Ischemia/Reperfusion

ObjectiveTo investigate the effects of morroniside on IL-1β in rat cortex with focal cerebral ischemia/reperfusion.MethodsThe animal model was induced by occlusion of middle cerebral artery with suture embolus, ischemia for 30 min, and reperfusion for 72 h in rats. The content of IL-1β was detected with enzyme linked immunosorbent assay(ELISA).ResultsCompared with sham group, the content of IL-1β increased obviously in model rat. Compared with model group, morroniside(30 mg/kg, 90 mg/kg, 270 mg/kg) and colchicine (0.06 mg/kg) decreased the content of IL-1β significantly (P<0.001).ConclusionMorroniside may protect the cortex from inflammatory factor IL-1β.

Cerebral ischemia-reperfusion results in damage on neuron, leading to genes and proteins related to apoptosis activation. At the same time, generous cytokines released after cerebral ischemia-reperfusion can induce the apoptosis of the neuron. Many current studies have showed that the major damage mechanisms on apoptosis of the neuron are mitochondrion impairment, calcium overload, increased levels of oxygen radicals and so on. The advance research on the mechanism contributes to explore new neuroprotective drugs, and further identify the target and therapeutic effect of drug treatment.

8.Lipid compounds from Echinacea purpurea.

OBJECTIVETo study the lipid constituents from Echinacea purpurea.
METHODThe compounds were isolated by chromatography method and the structures were identified on the basis of spectral analyses.
RESULTFive compounds were isolated and identified as, 1 beta, 6 alpha-dihydroxy-4(14)-eudesmene(1), (2E, 4E, 8Z, 10E)-N-isobutyl-2,4,8,10-dodecatetraenamide(2), (2E, 4E, 8Z, 10Z)-N-isobutyl-2,4,8,10-dodecatetraenamide(3), cerotic acid(4), hyxacosyl alcohol(5).
CONCLUSIONCompounds 1,4 and 5 were obtained from the plant for the first time.

9.The role of neuronal and glial activation in spinal cord in the mechanism of neuropathic pain following three different peripheral nerve injuries in rats

Objective To determine if there is any difference in neuronal and glial(astrocytic and microglial)activation in the spinal cord in three rat models of neuropathic pain.Methods Twenty-four SD rats weighing 150-200 g were randomly divided into 4 groups(n=6 each):Ⅰ control group;Ⅱ chronic constrictive injury group(CCI);Ⅲ spinal nerve ligation group(SNL)and Ⅳ spared nerve injury(SNI).No operation was performed in control group.In CCI group left sciatic nerve was exposed and loosely ligated with catgut.In SNL group the L_5 spinal nerve was exposed and ligated with silk suture and cut.In SNI group tibial nerve and common fibular nerve were ligated and cut.Pain threshold was measured using plantar tactile stimulator(Ugo,Basile Co. Italy)every other day from 3 days before until 15 days after operation.50% paw withdrawal threshold was measured using up-and-down sequential mechanical stimulation of different intensity(0.45,0.70,1.20,2.00, 3.63,5.50,8.50,15.10 g)applied to the plantar surface of the injured paw.On the 15~(th) day after operation after pain threshold was measured the animals were anesthetized with intraperitoneal 10% chloral hydrate 400 mg? kg~(-1).The L_(5,6) segment of the spinal cord was isolated.Neuronal,astrocytic and microglial activation was determined by immuno-histochemistry with antibodies of c-Fos(a proto-oncogene protein),GFAP(an astrocyte marker)and OX-42(a microglial marker).Results The 50% paw withdrawal threshold reached the lowest level on the 7~(th) day after operation.The lowest level was maintained until the 15~(th) day after operation in group CCI,SNL and SNI.The 50 % paw withdrawal threshold was(14.1+1.5)g in control group,(2.5+0.5)g in CCI group, (1.5?0.6)g in group SNL and(0.8?0.4)g in group SNI.The number of c-Fos positive neurons in laminae Ⅳ-Ⅵ of dorsal horn was significantly greater in group CCI,SNL and SNI than in control group,but there was no significant difference among the 3 peripheral nerve injury groups.The activation of astrocytes and microglias in laminae Ⅰ-Ⅳ of dorsal horn was significantly increased in group CCI,SNL and SNI than in control group but there was no significant difference among the 3 peripheral nerve injury groups.Condusion There is no significant difference in activation of neurons and astrocytes and microglias in the ipsilateral dorsal horn among the 3 pain models.
10.A study on polymorphism of salivary esterase in personal identification.

OBJECTIVE:
To research the value of polymorphism of salivary esterase(Set) in paternity and personal identification.
METHODS:
Phenotype and genotype of human salivary esterase were detected in 114 liquid saliva samples from the Chinese population by disc electrophoresis and fast blue RR staining assay.
RESULTS:
The frequency of Set type was F 22.81%, FS 50.88%, S2 6.31%. The estimated gene frequency of SetF was 0.4825 and SetS was 0.5175. The PE was 0.1875 and the DP was 0.6199.
CONCLUSION
Polymorphism of salivary esterase (Set) was practical in paternity and personal identification.
