1.Randomized controlled study of topical administration of nonsteroidal anti-inflammatory drug for moderate or severe dry eye

BackgroundDry eye is a multi-factorial-induced tear film and ocular surface disorder.Immunoinflammation plays a key role in the pathogenesis of dry eye.As inhibitor of the cyclo-oxygenase pathway,nonsteroidal anti-inflammatory drugs play an anti-inflammatory and anti-hypersensitivity role,and it can be a potential treatment for dry eyes.ObjectiveThis study was to investigate the effectiveness of nonsteroidal anti-inflammatory drugs (0.1%topical pranoprofen) on moderate to severe dry eyes and its mechanism.MethodsThis was a small sample of randomized controlled clinical trial.Thirty right eyes of 30 patients with moderate to severe dry eyes were included in the study according to the diagnosis criteria and randomized into two groups.The patients of the trial group received topical administration of 0.1% pranoprofen plus 0.1% sodium hyaluronate,and those of the control group received the topical 0.1% sodium hyaluronate only.Ocular surface inflammation index scores (OSDI) and ocular surface fluorescine staining (OSS) scores were measured under the slit lamp,and tear film break-up time (BUT),Schirmer Ⅰ test values were evaluated.The expression of human leucocyte antigen-DR (HLA-DR) and CD11b in conjunctiva epithelial cells were detected by impression cytology and flow cytometry (FCM).All the indexes were compared between the two groups before and after treatment.Informed consent was obtained from all patients.ResultsThere were no significant differences in terms of age and gender and their baseline values between the trial group and control group (t=0.412,P=0.684;x2=0.240,P=0.624),and so were all the indexes (P>0.05).Compared with the control group,the OSDI,OSS scores and cells positive for HLA-DR were lowered but the BUT was delayed in the trial group on day 15 ( t=2.43,P=0.03;t=2.83,P=0.01;t=3.29,P=0.00;t=3.23,P=0.00 ).No significant differences were found in the Schirmer Ⅰ test value and CD11b expression between these two groups (t=0.17,P=0.87;t=0.28,P=0.79).The OSDI,OSS scores and BUT were significantly improved,and the number of cells positive for HLA-DR were reduced 15 days after administration of drugs in comparison with before treatment in the trial group ( t =12.30,10.70,6.10,7.92,P =0.00 ).However,there were no comparable alteration seen in these indexes before and after the usage of drugs in the control group ( P>0.05).Positive correlations were found in HLADR expression with OSDI and OSS ( r =0.601,P =0.018 ; r =0.586,P =0.022 ) and a negative correlation in HLADR expression with BUT (r=-0.697,P=0.004) on day 15 in the trial group.ConclusionsTopical usage of 0.1% pranoprofen is beneficial for remitting the ocular signs and symptoms in moderate to severe dry eyes.This study illustrates that topical usage of 0.1% pranoprofen can down-regulate the expression of inflammatory markers in conjunctival epithelial cells.

3.Self-made vancomycin quality-control serum applied in blood drug concentration monitoring

Objective To evaluate self -made vancomycin quality -control serum used in blood drug concentration monitoring .Methods To prepare 10,20,30 μg·mL-1 vancomycin quality -control serum. The preparation, repeatability, sample stability and clinical application were studied.Results The results of low, medium and high concentra-tion of vancomycin quality-control serum was 98.0%-101.5%.The content change of vancomycin was less than 2% while saved at -30 ℃within 6 months.Conclusion The method for preparation of self-made vancomycin quality-control serum is simple, cost-effective, accurate and reliable, which can be used as daily laboratory quality control saved at -30℃.
4.The research on applying ozone air disinfector at dynamic state to ultrasonic scaling treatments

Objective To explore the method of intervening measure applied at dynamic state during ultrasonic scaling treatments and may control the aerosols and spatters in dental department so that we may provide safe clinical environment for dental personnel and patients. Methods 40 patients received ultrasonic scaling treatments were divided into experiment and control group. The experimental group received measures by the ozone air disinfector at the beginning of ultrasonic scaling treatments (the ozone air disinfector worked 5 minutes) and the control group was not received the device treatment.. Air samples were collected at different appointed period. Results A Mann-Whitney test was conducted to determine significant difference between the ultrasonic scaling treatments with the ozone air disinfector and the ultrasonic scaling treatments without the device at different appointed time. The general bacterial population in the air were lower in experimental group than in control group (P < 0.01). Conclusions The ozone air disinfector applied at dynamic state during ultrasonic scaling treatments may control the quantities of colony-forming units of aerosols and spatters in dental clinic. After dynamic disinfection, the quantities of colony-forming units may conform to the National Disinfection Technology Criteria.
5.Fitting of moisture adsorption models for Astragali Radix aqueous extract

AIM To fit the moisture adsorption models for Astragali Radix aqueous extract.METHODS For the aqueous extract prepared by decocting,the moisture adsorption kinetics curve and adsorption/desorption isotherms were drawn by gravimetry,and then the data on moisture adsorption were fitted.The fitted models were optimized according to evaluation indices of root mean square error (RMSE),residual sum of squares (RSS) and coefficient of determination (R2).RESULTS The double exponential model for moisture adsorption kinetics curve and Peleg model for adsorption/desorption isotherms were considered as the two best models with regard to their fitting effects (R2 values were the closest to 1,RMSE and RSS values were the closest to 0).Among them,various adsorption/desorption isotherms all belonged to type Ⅱ ("S" isotherm),while the higher equilibrium moisture content of desorption isotherm than that of adsorption isotherm contributed to obvious type H3 hysteresis phenomenon.CONCLUSION Double exponential model and Peleg model can well fit the moisture adsorption kinetics curve and adsorption/desorption isotherms of Astragali Radix aqueous extract,respectively.
6.Expression of TET and 5-HmC in Trophoblast Villi of Women with Normal Pregnancy and with Early Pregnancy Loss

Increasing evidence suggests that epigenetic dysfunction may influence the stability of normal pregnancy.The ten-eleven translocation (TET) family and 5-hydroxymethylcytosine (5-hmC) were found to be linked with epigenetic reprogramming.The present study aimed to examine the expression of the TET family and 5-hmC in the villi of human embryos and compared their expression between normal pregnancy and early pregnancy loss (EPL).Embryonic villi were collected from normal pregnant women (control) experiencing medical abortion and from EPL patients at gestation ages of 6,7 and 8 weeks.The mRNAs of TET family were analysed using quantitative polymerase chain reaction (qPCR),and TET proteins using Western blotting and immunohistochemical analysis.The MethylFlashTM Kit was used to quantify the absolute amount of 5-methylcytosine (5-mC) and 5-hmC.Our results showed that the expression of the TETs and 5-hmC in the normal villus decreased with increasing gestational age.Immunohistochemistry revealed that the TET proteins were expressed in the cytoplasm of trophoblasts and their expression was the highest in the 6-week tissue samples,which was consistent with the qPCR and Western blot results.The expression of TET1,TET2,and TET3 was lower in the villi in EPL group than in normal pregnancy group (P<0.05 for all).It was concluded that the TET family and 5-hmC are critical in epigenetic reprogramming of human embryo.The findings also suggest that a deficiency of TETs in the villus might be associated with human EPL.
7.Lung microRNA profile in chronic cyanotic piglets with decreased pulmonary blood flow.

BACKGROUNDCyanotic congenital heart defects with decreased pulmonary blood flow due to lung ischemia, hypoxia, and others lead to infant morbidity and mortality more than acyanotic heart disease does. Despite the great effort of medical research, their genetic link and underlying microRNAs molecular mechanisms remain obscure. In this study, we aimed to investigate microRNAs regulation during cyanotic defects in lung of immature piglets.
METHODSCyanotic piglet model was induced by main pulmonary artery-left atrium shunt with distal pulmonary artery banding. Four weeks later, hemodynamic parameters confirmed the development of cyanotic defects and pulmonary lobe RNA was extracted from all animals. We studied the repertoire of porcine lung microRNAs by Solexa deep sequencing technology and quantified highly expressed microRNAs by microarray hybridization. Furthermore, we quantitated selected microRNAs from cyanotic and control piglets by quantitative RT-PCR.
RESULTSAfter surgical procedure 4 weeks later, the cyanotic model produced lower arterial oxygen tension, arterial oxygen saturation, and higher arterial carbon dioxide tension, hematocrit and hemoglobin concentration than controls (all P < 0.05). In 1273 miRNAs expressed in the immature piglets lungs, 2 most abundant microRNAs (miR-370 and miR-320) demonstrated significant difference between cyanotic and control group (all P < 0.05).
CONCLUSIONOur results extended lung microRNA profile in immature piglets and suggested that miR-370 and miR-320 are significantly up-regulated in cyanotic lung tissues.

9.Effects of acupuncture on rehabilitation of nervous functions in the stroke patient of different OCSP types.

OBJECTIVETo probe into effects of acupuncture on rehabilitation of nervous functions in the stroke patient of different OCSP types.
METHODSThe stroke patients were randomly divided into 4 subtypes (LACI, TACI, PACI, POCI) based on the OCSP clinical classification. The patients of every subtype were randomly sub-divided into an observation group and a control group. The patients in the 2 groups were treated with routine western medicine, and acupuncture at Baihui (GV 20), Sanyinjiao (SP 6), Taixi (KI 3), Xuehai (SP 10), etc. were added with reinforcing and reducing manipulation in the observation group and without manipulation of the needle in the control group. They were assessed with daily living ability scale (BI) at the time of enrollment, the end of 1, 3, 6, 12 months after the stroke, respectively.
RESULTSThere was no significant difference between the two groups for BI in LACI (P>0.05); there were significant differences 3, 6 and 12 months after the stroke between the two groups in PACI, POCI (P<0.05); in the TACI, there was a significant difference between the two groups for BI 3 and 6 months after stroke (P<0.05), with no significant difference 12 months after stroke (P>0.05).
CONCLUSIONAcupuncture has different effects on rehabilitation of daily living function in the stroke patient of different OCSP types.

10.Therapeutic effect of dense-packing autologous hair grafting technique for restoration of seborrheic alopecia

Objective To investigate the therapeutic effect of dense-packing auto hair grafting technique on the restoration of seborrheic alopecia. Methods With local anesthesia, a scalp strip was harvested from the back of the head. Under operating microscope (Various graft was created from the scalp strip, including micro-grafts with 1-2 hairs, mini-grafts with 3-4 hairs and sliver graft with 5-6 hairs. In the alopecia recipient area, micro slots were made with a small triangle-edged needle for the micro-grafts,mini slits were made with mini blade for the mini-grafts and foramen ovale were made with a slot punch. The grafts were then implanted into these holes. Results 32 cases of seborrheice alopecia were treated with the above mentioned technique from March 2007 to July 2009. Postoperative following up for 12-24 month showed that the grafted hairs were growing well with average 90 % survival of the hair. 81 % of the patients obtained satisfactory results with only one stage operation. Six patients needed the second operation to improve the appearance. All of the patients were satisfied with the appearance. Conclusions The dense-packing hair grafting technique with various grafts not only saves time of operation, but also obtains dense grafted hair and well appearance. The results are satisfactory to most patients with only one stage operation.
