1.Therapeutic Observation of Acupuncture-moxibustion plusMEI Te-lan’s Manipulations for Lumbar Intervertebral Disc Herniation (Blood Stagnation Pattern)

Objective To observe the clinical efficacy of acupuncture-moxibustion plusMEI Te-lan’s manipulations in treating lumbar intervertebral disc herniation (LIDH) due to blood stagnation.Method Totally 248 LIDH patients due to blood stagnation were randomized into a treatment group of 123 cases and a control group of 125 cases. The treatment group was intervened by acupuncture-moxibustion plusMEI Te-lan’s manipulations, while the control group was by acupuncture-moxibustion plus tuina at lower back. The short-form McGill Pain Questionnaire (SF-MPQ) and Oswestry disability index (ODI) were observed before and after intervention, and the clinical efficacies were compared between the two groups.Result The total effective rate and recovery-markedly effective rate were respectively 80.5% and 52.0% in the treatment group, versus 72.8% and 20.0% in the control group. There was a significant difference in comparing the recovery-markedly effective rate between the two groups (P<0.05). In the treatment group, the Pain Rating Index (PRI) score, Visual Analogue Scale (VAS) score, Present Pain Intensity (PPI) score, and ODI score were significantly changed after 3-day, 6-day, and 20-day treatment compared to the scores before treatment (P<0.05). After 20-day treatment, the PRI, VAS, PPI, and ODI scores of the treatment group were significantly different from that of the control group (P<0.05).Conclusion Acupuncture-moxibustion plusMEI Te-lan’s manipulations is an effective method in treating LIDH due to blood stagnation.
2.Venous Catheter Infection in Malignancy Patients and Its Countermeasures

OBJECTIVE To make the patients using the central venous catheter safely when given chemotherapy and intravenous nutritional therapy,and to decrease the catheter infection by establishing effective infection-preventing systems. METHODS Many infection-preventing actions were carried out,including a three-level management,improving the workers′ training in catheter operation and maintenance,and health education. RESULTS Comparing with the matched group,the infection rate of the experimental group was much lower after the actions.The actions prevented the increase of infection effectively. CONCLUSIONS Building and carrying out the systems are proved to be important measures to decrease the catheter infection and ensure the quality of life among patients during the therapy in hospital.
3.Inhibitory Effect of Rupixiao Granule on Hyperplasia of Mammary Gland in Rabbits

Objective:To determine the inhibitory effect of Rupixiao ( RPX) granule on mammary gland hyperplasia ( HMG) in rabbits and explore the possible mechanism to provide reference for clinical medication. Methods:Rabbit model of mammary hyperpla-sia was established by estradiol benzoate and progesterone. Xiaoyao pills and tamoxifen were used as the positive control, and the RPX granule group was respectively at low, medium and high dose (0. 525,1. 05,2. 1 g·kg-1). Each group was with intragastric adminis-tration for 30 days. The levels of E2 , PROG, FSH, LH and PRL in serum and the expression of VEGF in the homogenate tissue were determined by an Elisa method. The breast tissue of rabbits in each group was withdrawn to observe the structure changes after Hema-toxylin-eosin staining ( HE) . Results:Ovarian preservation-benzoic acid, estradiol combined with progesterone could be used to estab-lish mammary gland hyperplasia in rabbits. RPX granule could significantly decrease the serum levels of E2 and PRL (P<0. 01)and increase that of PROG (P<0. 01 or P<0. 001). The results of pathological section showed that the breast structure in RPX granule high dose group could be restored to the level of the blank control group, which showed its therapeutic effect on HMG was better than that of tamoxifen and Xiaoyao pills. The expression of VEGF in each treatment group significantly decreased compared with that in the model group by the Elisa reagent (P<0. 001). Conclusion:RPX granule has good efficacy in HMG rabbits. The inhibitory effects of RPX granule on HMG in rabbits maybe related to the inhibition of VEGF in rabbit breast tissues, and the inhibition of RPX granule was similar to that of tamoxifen and Xiaoyao pills.
4.Puerarin extraction and identification of solution wine to protect liver function in mice

Objective Pueraria extract puerarin,HPLC assay puerarin extract and compare different doses of correlation with the hang-over effect of puerarin evaluate different doses of puerarin liver hangover effect. Methods Extracted under optimal conditions obtained in the previous experiment puerarin spare,HPLC method for qualitative and quantitative detection of alcohol extract of kudzu root ( PRE) ,the male Kunming mice were randomly divided into control group,positive control group and puerarin group,each group of 10. Give mice fed pueraria extract,30 min after administration of liquor,drunk mice sobering observation time and the determination of mouse liver ADH,GOT,GPT con-tent in order to investigate the effect of puerarin on drunken mice. Results HPLC fraction was measured at 8 times the volume of 70% etha-nol,60 ℃ constant temperature water bath shaker at 30 min for optimal extraction conditions puerarin extraction. Compared with the positive control groups:low,medium and high doses of alcohol extract of pueraria can significantly shorten the time to sober up drunken mice,the dose of PRE could effectively inhibit the absorption of alcohol,reduce liver tissue ADH,GOT,GPT,the effects of high doses of PRE absorption of alcohol was small. Conclusion HPLC method capable of puerarin extract the qualitative and quantitative determination of puerarin on liver injury caused by acute alcoholism a protective regulatory role,and the hangover effect of puerarin dose showed a good positive correlation.
5.Genotyping of 67 norovirus strains

Objective To investigate the prevalent situations of norovirus infection and genotype distributions in 2014 in Suzhou area .Methods A total of 322 fecal specimens were collected from infants with suspected viral diarrhea at Children′s Hospital of Soochow University in 2014 .Norovirus genogroupⅠ and Ⅱ was detected by reverse transcription (RT )‐polymerase chain reaction (PCR) ,In an effort to identify norovirus genotypes , RNA dependent RNA polymerase region (region A ) and capsid region (region C) segment of some samples positive for norovirus was amplified by RT‐PCR .Comprehensive molecular characteristics of norovirus were obtained by sequence analysis of the same samples in different regions .Results Among 322 fecal specimens ,67 cases were positive for norovirus of G Ⅱ group ,and norovirus of GⅠ group was not found .The genetic fragments of region A was successfully detected in 42 strains .Among all 42 specimens ,there were 35 GⅡ .e strains ,3 GⅡ .7 strains ,2 GⅡ .17 strains and 2 GⅡ .12 strains .The genetic fragments of region C was successfully detected in 53 strains .Among these 53 specimens ,there were 44 GⅡ .4‐2012Sydney strains ,4 GⅡ .6 strains ,2 GⅡ .17 strains ,2 GⅡ .3 strains and 1 GⅡ .2 strain .Conclusions It′s indicated that G Ⅱ .4‐2012Sydney is the main genotype of norovirus causing viral diarrhea in Suzhou ,and other genotypes including the new GⅡ .17 variant ,GⅡ . 7/GⅡ .6 and GⅡ .12/GⅡ .3 recombinant strains also exist .
6.A new photodynamic therapy drug toward gastric cancer MGC803 cell.

OBJECTIVETo investigate the treatment efficiency of a new photodynamic therapeutic(PDT) drug synthesized by our laboratory toward MGC803 cells and related mechanisms.
METHODSBleaching method was used to evaluate the photostability of drug upon repetitive illumination. MTT assay was used to determine the ability of new drug killing MGC803 cells after PDT. Laser scanning confocal microscopy (LSCM) was applied to investigate the subcellular localization of drug in MGC803 cells (mitochondria and/or lysosomes). Hoechst staining and flow cytometry(Annexin V/PI double-staining) were performed to detect the death mode of MGC803 cells after PDT.
RESULTSThis new PDT drug had good stability to light irradiation after repetitive illumination. MTT assay showed no cytotoxicity towards MGC803 cells only by drug or only by irradiation(P>0.05), but intense lethal effect was observed with drug and light combination(P<0.05). The phototoxicity of medicine increased with the elevation of concentration, the LD50 was 1.74 μmol/L, and reaching plateau at the concentration of 3.12 μmol/L, even increasing the concentration. LSCM found that drug localized in lysosomes of MGC803 cells. Hoechst staining showed that the death mode of cells was mainly necrosis and Annexin V/PI double-staining proved this result further.
CONCLUSIONThis new PDT drug is an effective PDT sensitizer for MGC803 cells and the death mode of cells is mainly necrosis.

7.The role of cell pathway in apoptosis of fibroblasts from proliferative scar induced by steroid or IFN.

OBJECTIVEThis paper is to investigate the effects of steroid or IFN alpha-2b on apoptosis and cell pathway of fibroblasts from keloids, hypertrophic scars and normal skins and different responses of different fibroblasts.
METHODS6 samples from keloid, hypertrophic scar and normal skin were collected respectively and fibroblasts from different sources were cultured in vitro. After different fibroblasts were treated with dexamethasone (0.1 mg/ml) or IFN alpha-2b (1000 U/ml), Bax and Bcl-2 protein expressions were detected in situ by immunohistochemical staining; DNA ladders of different fibroblasts were observed by gel electrophoresis; and relative activated (phospho-) ERK1/2 and JNK pathways were detected by method of FACE ELISA.
RESULTSDexamethasone could induce apoptosis of fibroblasts from keloids, hypertrophic scars and normal skins through activating (phospho-) ERK1/2 and JNK pathways; IFN alpha-2b could not induce apoptosis of fibroblasts from different sources. IFN alpha-2b could inhibit (phospho-) ERK1/2 pathway and could not affect (phospho-) JNK pathways of fibroblasts from keloid and hypertrophic scar. IFN alpha-2b could affect neither (phospho-) ERK1/2 pathway nor (phospho-) JNK pathways of fibroblasts from normal skin.
CONCLUSIONSThe responses of different fibroblasts to steroid or IFN alpha-2b were different.

8.Early transplantation of double rectus abdominis musculo-cutaneous flaps from upper abdomen for the repair of patients with electrical injury in the wrist.

OBJECTIVETo observe the effect of the early transplantation of double rectus abdominis musculo-cutaneous flaps on the repair of electrical injury of the wrist.
METHODSThe study involved six patients suffering from circumferential deep electrical burn with only small amount of normal skin left on the dorsal side. The wounds were covered with double rectus abdominis musculo-cutaneous flaps raised from the upper abdomen with pedicles in both proximal and distal ends at an early postburn stage. The postoperative recovery of wrist function and wound repair were evaluated.
RESULTSThe wrist wounds in all the 6 patients were primarily healed, with perfect function and appearance.
CONCLUSIONEarly application of double rectus abdominis musculocutaneous flaps on the electrically injured wrists can promote the wound healing processes and plays important roles in the preservation of wrist function.

9.Analysis of non-bacterial respiratory pathogen infection in children with asthmatic diseases.

OBJECTIVETo investigate the association of non-bacterial respiratory pathogens with asthmatic diseases in children, and the clinical significance of total serum IgE levels and peripheral eosinophil count in infection with non-bacterial respiratory pathogens.
METHODSIndirect immunofluorescence assay was used to detect IgM antibodies against nine types of non-bacterial respiratory pathogens in the sera of 490 children with asthmatic diseases between September 2010 and September 2011. Pathogens were analyzed and total serum IgE levels and peripheral eosinophil count were measured in IgM-positive cases.
RESULTSOf the 490 children with asthmatic diseases, 47.6% (233 cases) were positive with IgM antibodies against non-bacterial respiratory pathogens, the most common being Mycoplasma pneumoniae (MP) (25.3%), followed by adenovirus (ADV) (8.9%) and influenza B virus (Flu B) (8.8%). Thirty-six cases suffered from co-infection of two or more non-bacterial pathogens, mainly comprising MP and other pathogens (94%). There were significant differences in the total detection rate of IgM antibodies among all age groups (0-30 days: 50.0%; 1-6 months: 67.3%; 0.5-1 year: 33.1%; 1-3 years: 57.3%; 3-8.9 years: 61.7%). The positive rate of IgM antibodies against respiratory pathogens was highest in children with bronchial asthma, followed by children with asthmatic bronchitis, and it was lowest in children with bronchiolitis. IgM-positive children had significantly decreased blood eosinophils and significantly increased total serum IgE levels.
CONCLUSIONSThe main non-bacterial respiratory pathogens include MP, ADV and Flu B in children with asthmatic diseases, and co-infection of MP and other non-bacterial pathogens is common. Infants aged 1 to 6 months have a higher infection rate than other age groups. Monitoring the changes in total serum IgE levels and peripheral eosinophil count has great significance for the clinical diagnosis and treatment of asthmatic diseases in children.

10.Posterior malleolar fracture: technique and clinical experience of the posterolateral approach.

OBJECTIVETo introduce the postero- lateral surgical approach to the posterior malleolar fracture and report its clinical outcomes in 32 cases.
METHODSThis study consisted of 32 cases, 22 males and 10 females with the mean age of 48 years (range, 21-63 years), suffering from posterior malleolar fracture. All cases were treated with the posterolateral surgical approach to the ankle. The average follow-up period was 28 months (range, 24-35 months). The clinical outcomes of these cases were evaluated on the basis of the Olerud-Molander Ankle (OMA) score and plain radiographs.
RESULTSAll cases showed radiological evidence of bony union at follow-up. The average OMA score was 82 points; 21 cases had excellent scores (90-100 points), 9 good (61-90 points), and 2 fair (31-60 points). The excellent-to-good rate was 93.8%. Although most cases did not show any wound dehiscence or necrosis, one patient had a superficial infection which healed after using antibiotic dressing and one had sural cutaneous nerve injury that underwent spontaneous remission without any treatment after three months. In addition, one presented with mild symptoms of peroneal tendonitis that disappeared after plate removal.
CONCLUSIONThe posterolateral approach offers an effective technique for fracture reduction and fixation of large posterior malleolar fragments.
