Objective By observing the curative effect of Yangxue Zhiyang Capsule (YZC) on guinea pigs and senile rats, therapeutic effect of YZC for senile pruritus and its mechanism was assessed. Methods Animal models with pruritus induced by exogenous histamine were used to observe the effect of YZC in relieving itching. Senile rat models of blood deficiency induced by acetophenylhydrazine (APH) were applied to measure the effect of YZC on red blood cells (RBC) counting, blood hemoglobin (Hb), plasma estrogen (E2), testosterone (T) and proesterone(PO), sexual organ weight and the amount of sebaceous gland and its secretory function. Results YZC could raise the itching threshold of guinea pigs with pruritus, elevate RBC amount and Hb content, increase E2,T and PO levels, elevate the weight of sexual organs and the number of sebaceous glands, and promote the secretory function of sebaceous glands in rats with blood deficiency. Conclusion YZC has good effect in relieving itching of guinea pigs with pruritus and nourishing blood of rats with blood deficiency by raising the levels of sex hormones, increasing the number of sebaceous glands and stimulating sebaceous gland secretion.
2.Diagnosis of Abdominal Organs Diseases by Contrast-Enhanced Ultrasonography

Objective To investigate the efficacy of contrast-enhanced ultrasound (CEUS) in diagnosing liver, gallbladder, spleen and renal diseases. Methods 27 patients with liver diseases, 30 patients with gallbladder diseases, 5 patients with renal diseases, 5 patients with renal trauma and 6 patients with spleen disease were examined by CEUS. Results There were 3 cases of primary hepatocellular carcinomas, 4 cases of metastatic hepatic carcinomas, 4 cases of liver abscess, 10 cases of hepatic hemangiomas and 3 cases of liver repture with active bleeding. The diagnostic coincidence rate of CEUS was 90%. 30 patients with gallbladder polyposis were diagnosed by CEUS, and the diagnostic coincidence rate was 100%. 5 cases with renal diaseses included 1 of renal abscess, 1 of renal column hypertrophy and 3 of renal hemangioma. In 5 cases with renal trauma, there were 3 cases with renal contusion and 2 cases with renal rupture and active bleeding. The coincidence rate was 61%. In 6 cases with spleen diseases, there were 1 case with splenic infraction, 1 case with splenic abscess and 4 cases with splenic rupture and hemorrhage. The coincidence rate was 100%. Conclusion CEUS has great value of clinical application in diagnosis of liver, gallbladder, spleen and renal diseases.
3.Experimental study on thrombus-targeted ultrasound contrast agent enhancing acute thrombus in normal canine femoral vein

Objective To evaluate the enhancement effect of the thrombus-targeted ultrasound contrast agent on acute thrombus in normal canine femoral vein.Methods Acute thrombi were created in both sides of the femoral vein in ten canine models.Targeted ultrasound contrast agent was given in a dose of(0.06) ml/kg in ten canines via intravenous bolus injection from forelimb of canine,the images of the acute thrombus were taken at 0,2,4,8,12,14 minutes after infusion of ultrasound contrast agent and stored in magneto optical disks.The images were assessed qualitatively by two independent observers and quantitatively using acoustic densitometry(AD) to determine the contrast enhancement effect of acute thrombus in normal canine femoral vein.Results After infusion of targeted ultrasound contrast agent,the thrombus was enhanced considerably and easy to be detected.AD analysis demonstrated that peak indencity(PI) and area under curve(AUC) increased with the changes of time,reaching the peak at 8th minutes after infusion of targeted ultrasound contrast agent,then decreased.PI and AUC increased at the 8th minute after infusion of targeted ultrasound contrast agent than before [((333.21)?(38.56))dB vs((168.18)?(28.18))dB,((884.40)?(94.62))dB vs((439.65)?(98.54))dB,respectively,P
4.Effects and mechanism of magnetic microsphere combination of cisplatin with micron ferric oxide particle on human hepatoma cell in vitro

Objective To investigate the effects and mechanism of magnetic microsphere combination of cisplatin with nano or micron ferric oxide particle on human hepatoma cell(HHC). Methods A human hepatoma cell line BEL-7402 was used in this study, 0.04?g/ml~8?g/ml of cisplatin(CDDP), cisplatin nano-ferric oxide magnetic microsphere(nCDDPmm) and micro-ferric oxide magnetic microsphere(mCDDPmm) were administered to the culture solution for culturing the BEL-7402 cell as the experimental groups,and in the control groups were administered with cisplatin 0?g/ml and magnetic microsphere without cisplatin respectively. The optic density of viable cell, cytotoxity index, growth curve of cell, cell cycle, proliferation index and apoptosis were assayed by MTT method, cell count and flow cytometry respectively. 5.Effects of PBDE-47 Single Exposed and Combined with PCB153 on Neurodevelopment in Rats

Objective To study the effects of 2,2',4,4'-tetrabromodiphenyl ethers(PBDE-47)single exposed and combined with 2,2',4,4',5,5'-hexachlorobiphenyl(PCB153)on learning and memory ability and the uhrastrncture of hippocampal CA1 region in rats.Methods Neonatal SD rats(CL grade)were randomly divided into groups,9 male and 9 female in each,then were exposed to single PBDE-47 at doses of 1,5,10 mg/kg and combined with PCB153 at dose of 5 mg/kg,through gavage for one time. Soya oil was used as the solvent control.The learning and memory ability and ultrastructure of neurons in hippocampal CA1 region were examined respectively 2 months after treatment.Results In the low dose groups(1 mg/kg PBDE-47 and 1 mg/kg PBDE-47+ 5 mg/kg PCB153 treated group),the ultrastructure of neurons in hippocampal CA1 region were as normal as that in the control group.As the dose increasing,in 5 mg/kg PBDE-47 group,the endoplasmic reticulum appeared swelling,expansion and degranulation.In 5 mg/kg PBDE-47 plus 5 mg/kg PCB153 group,the mitochondria appeared swelling,the ridge ruptured,the periplast puffed,and then the cytoplasm condensed.In the high dose groups(10 mg/kg PBDE-47 group and 10 mg/kg PBDE-47 plus 5 mg/kg PCB153 group),the neurons showed acutely denatured,the periplast puffed,the cell organelle dissolved and the mitochondria vacuolizated.The interaction between PBDE-47 and PCB153 on the latency period and the general pathway was presented in place navigation test(P

Objective To explore the correlation between hyperuricaemia and blood pressure, and blood lipid and glucose. Methods By using simple cluster sampling, 2 branch units from PetroChina Changqing Oilfield Company were selected, and all the 720 subjects with hyperuricaemia (HUA) were assigned to the HUA group; another 620 participants with normal uric acid (UA) level into the normal group. The correlation between blood uric acid and blood pressure,and blood lipid and glucose was assessed by Logistic regression. Results The odds ratio (OR) of those who had 1,2 or 3 abnormal status of hypertension,hyperlipidemia and impaired fasting glucose in the HUA group were much higher than the normal group (OR values were 4. 036,2. 562, and 4. 174, respectively). Logistic regression showed that male, systolic blood pressure ( SBP), GLU, total cholesterol ( TC), triglyceride ( TG), low-density lipoprotein cholesterol (LDL-C) were risk factors of H UA ( OR values were 7. 736,2. 309,1.721,2. 761 and 1. 411,respectively) ,while high-density lipoprotein cholesterol (HDL-C, OR = 0. 211 ) was a protective factor of HUA. Conclusions Gender,blood pressure and blood lipid may have correlation with blood UA. Multiple risk factors should be considered to improve the effectiveness of health education and health promotion.
7.Clinical study on relationship between sluggishness of lung-defensive qi and levels of vasoactive intestinal polypeptide and thromboxane B2

OBJECTIVE: To explore the nature of pathology of sluggishness of lung-defensive qi and to offer objective experimental indexes for weifen syndrome (defensive phase syndrome). METHODS: According to the completely random design, the plasma levels of vasoactive intestinal peptide (VIP) and thromboxane B2 (TX2) of 19 patients with weifen syndrome and 13 patients with qifen syndrome (qi phase syndrome) were detected by radioimmunoassay. The plasma levels of VIP and TX2 at different stages of weifen syndrome and qifen syndrome were observed. RESULTS: The plasma levels of VIP in weifen syndrome and in the late stage of weifen syndrome increased greatly at different stages as compared to qifen syndrome and the blank group (P < 0.01), while the plasma level of TX2 of weifen syndrome was higher only at the late stage than the blank group and qifen syndrome (P < 0.01). As for the levels of VIP and TX2 in weifen syndrome with different internal organs infected, there was no significant difference (P > 0.05). CONCLUSION: VIP may be an index reflecting the pathology of weifen syndrome, and it is one of the material foundations of sluggishness of lung-defensive qi, but it has nothing to do with the infected internal organs. The level of TX2 increases only after the fever of patients with weifen syndrome subsided, so it can not be the basis for diagnosis of the early stage of weifen syndrome. It doesn't increase in qifen syndrome either, the mechanism remains to be further studied.
8.Effects of hydrogen-rich saline on inflammatory responses during transient cerebral ischemia-reperfusion in rats

Objective To evaluate the effect of hydrogen-rich saline on inflammatory responses during transient cerebral ischemia-reperfusion (I/R) in rats.Methods Forty-five male Sprague-Dawley rats,aged 5-6 yr,weighing 180-250 g,were randomly divided into 3 groups (n =15 each) using a random number table:sham operation group (group S),group I/R and hydrogen-rich saline group (group H).The rats were anesthetized with intraperitoneal 10% chloral hydrate 0.3 ml/100 g.Transient cerebral ischemia was induced by bilateral common carotid artery occlusion with hypotension for 15 min,followed by reperfusion.Five rats were randomly chosen from each group,and Morris water maze was used to assess the cognitive function starting from 5 days before establishment of the model.Place navigation test lasted for 5 consecutive days.The escape latency,swimming speed and swimming distance were recorded.Spatial probe test was carried out on 1 and 3 days after establishment of the model.The time of staying at the target platform quadrant,frequency of crossing the original platform and swimming speed were recorded.The rats were sacrificed after the end of spatial probe test on 3 days after the model was established,and hippocampi were removed to examine the morphology of neurons in hippocampal CA1 region with light microscope.Five rats randomly chosen from each group were sacrificed on 1 day after the model was established,and hippocampi were removed to detect the contents of tumor necrosis factor-alpha (TNF-α) and interleukin-1 beta (IL-1β).The rest 5 rats in each group were sacrificed,and hippocampi were removed for determination of nuclear factor kappa B (NF-κB) activity (by immuno-histochemistry).Results In place navigation test before the model was established,the escape latency and swimming distance were gradually shortened with the prolonging training time,and no significant change was found in the swimming speed with the prolonging training time in the three groups.Compared with group S,the time of staying at the target platform quadrant was significantly shortened,and the frequency of crossing the original platform was reduced on 1 and 3 days after establishment of the model,and the contents of TNF-α and IL-1β and NF-κB activity were increased on 1 day after establishment of the model in group I/R.Compared with group I/R,the time of staying at the target platform quadrant was significantly prolonged,and the frequency of crossing the original platform was increased on 1 and 3 days after establishment of the model,and the contents of TNF-α and IL-1β and NF-κB activity were decreased on 1 day after establishment of the model in group H.There was no significant change in the swimming speed during spatial probe test on 1 and 3 days after establishment of the model.Conclusion The mechanism by which hydrogen-rich saline reduces transient cerebral I/R injury may be related to inhibition of inflammatory responses in rats.

Objective: Diuresis and removing urinary calculus action of Jieshitong tablet were studied. Methods: Metabolic cage method of rats was adopted to be engaged in diuresis experiment; glycol and ammonium chloride had been given orally for 30 days to form concretion model so that a removing urinary calculus trial could be done. Results: High and low doses of Jeshitong tablet both had significant diuresis effect, total urinary output in 3h of drug group increased about 50% than that of control group. Concretio formative rates of Jeshitong tablet group significantly decresed; the stronger removing urinary calculus action, the higher Jieshitong concentraction was. Conclusion: Jieshitong tablet had diuresis and removing urinary calculus action.
