1.Detection of five periodontal pathogens in coronary atherosclerotic plaque

ObjectiveTo detect five periodontal pathogenic bacteria in coronary atherosclerotic plaques.MethodsAtherosclerotic plaque specimens were obtained from 101 patients who scheduled for coronary artery bypass graft surgery.The bacteria DNA was obtained from coronary atherosclerotic plaques using the chelex-100 method.The extracted DNA were examined by PCR.ResultsWithin the 101 pieces of atherosclerotic plaque samples Porphyromonas gingivalis( Pg,31% ),Tannerella forsythensis(Tf,42% ),Prevotella intermedia( Pi,26% ),Fusobacterium nucleatum( Fn,21% ),Actinobacillus actinomycetemcomitans( Aa,23% ).PCR products were sequenced and were compared with GenBank sequences,the homology was 99%-100%.ConclusionPeriodontitis might affect the development of atherosclerosis and there is a correlation between coronary heart disease and chronic periodontitis.
2.Effect of long-term wearing rigid gas permeable contact lenses on the stability of tear film

· AIM:To investigate the effect of long-term wearing rigid gas permeable (RGP) contact lenses on the stability of tear film,and to observe its safety.· METHODS:Totally 60 myopia patients (120 eyes) wearing RGP in our hospital from October 2014 to January 2016 and 30 low myopia patients (60 eyes) without a history of wearing RGP were selected.There were 26 patients (52 eyes) wearing RGP at daytime were included into daytime wearing group;34 patients (68 eyes) only wearing RGP at night were set as the night wearing group.And patients without wearing RGP were set as control group.Schirmer Ⅰ test (S Ⅰ t),tear film fern experiments,and break-up time (BUT) of tear film were performed before wearing RGP,wearing for 1 mo,wearing for 6mo,wearing for 1a,wearing for 2a,respectively.· RESULTS:After wearing RGP,the BUT and S Ⅰ t of daytime wearing group and night wearing group significantly decreased,and were significantly lower than those of the control group at the time points,the difference was statistically significant (P< 0.05).After wearing for 6mo,1a and 2a,the BUT and S Ⅰ t of daytime wearing group was significantly lower than that of the night wearing group,the statistic found significant difference (P<0.05).The three groups had no statistically significant differences in tear fern classification experiment before wearing RGP and wearing for 1 and 6mo (P>0.05).The number of the eyes ≥ Ⅲ in daytime wearing group and night wearing group was significantly higher than that of the control group after wearing for 1 and 2a,the difference was statistically significant (P< 0.05),but the difference between daytime wearing group and night wearing group found no significant difference (P>0.05).· CONCLUSION:Wearing RGP can result in reducing tear secretion and decreasing the stability of tear film.
3.Correlation analysis of the degree of demodex infection with corneal cell density and ocular surface function in BKC patients

AIM:To investigate the correlation between demodex infection with corneal cell density changes and ocular surface function in patients with blepharo kerato conjunctivitis(BKC).
METHODS: Ninety-four patients with BKC(BKC group)at Department of Ophthalmology of our hospital from July 2019 to July 2020 were selected as the research objects, in addition, 80 matched healthy volunteers were selected as control group. The BKC patients were divided into infected group(45 cases)and uninfected group(49 cases)according to whether they were infected with demodex. According to the number of demodex detected in eyelashes, there were 17 cases of suspicious infection, 18 cases of moderate infection and 10 cases of severe infection. All subjects were examined by laser confocal microscopy, and the cell density in the superficial stromal layer of the central cornea and peripheral cornea was calculated. The ocular surface function \〖Schirmer test, Ocular Surface Disease Index(OSDI)\〗, eyelid margin abnormality score, corneal fluorescence stain and tear film break-up time(TF-BUT)of patients with BKC were examined, and the correlation between demodex infection with corneal cell density and ocular surface function in patients with BKC was analyzed.
RESULTS: Compared with those in the control group, the cell density in the superficial stromal layer of the central cornea and peripheral cornea was lower in the BKC group(P<0.05), and the OSDI, eyelid margin abnormality score and corneal fluorescence stain score were higher(P<0.05); the cell density in the superficial stromal layer of the central cornea and peripheral cornea of patients in uninfected group, patients with suspicious demodex infection, moderate demodex infection and severe demodex infection decreased in turn(P<0.05), and the OSDI, eyelid margin abnormality score and corneal fluorescence stain score increased significantly in turn(P<0.05); the degree of demodex infection was negatively correlated with the cell density in the superficial stromal layer of the central cornea and peripheral cornea in patients with BKC(P<0.05), and was positively correlated with OSDI, eyelid margin abnormality score and corneal fluorescence stain score(P<0.05).
CONCLUSION: The severity of demodex infection has a significant negative correlation with the cell density in the superficial stromal layer of the central cornea and peripheral cornea in patients with BKC, has a significant positive correlation in patients with ocular surface dysfunction.

5.Clinical Study on Treatment of Mid-Late Stage Gastric Carcinoma by Compound Xiansu Capsule (仙酥胶囊) Combined with Chemotherapy

Objective: To assess the effect and mechanism of compound Xiansu capsule (仙酥胶囊, XSC) combined with chemotherapy in treating gastric carcinoma of mid-late stage. Methods: Sixty-one patients of the test group were treated by XSC combined with chemotherapy and 30 patients of the control group treated with chemotherapy alone. The effect of treatment and cell mediated immunity of patients were observed. Results: The effective rate of the test group and the control group was 32.8% and 13.3% respectively (P<0.05), the toxic reaction occurrence caused by chemotherapy was less in the former than that in the latter group (P<0.01). The CD8 level of patients in the test group decreased, and CD4/CD8 level was raised obviously, which suggested that XSC had immuno-regulating effect on T-cell. Conclusion: XSC could enhance the efficacy and reduce the toxic and side-effect of chemotherapy. To regulate the cell mediated immunity of patients is possibly its mechanism.
6.Three Dimensional Orbital Magnetic Resonance T2-Mapping in the Evaluation of Patients with Graves' Ophthalmopathy

The clinical application of orbital magnetic resonance (MR) T2-mapping imaging in detecting the disease activity of Graves' ophthalmopathy (GO),and the predictive values of therapy response to intravenous glucocorticoid (ivGC) were investigated.Approved by the local institutional review board (IRB),106 consecutive patients with GO were included in this prospective study.All subjects were divided into two groups according to the patients' clinical activity score (CAS):the CAS positive group (CAS ≥3) or the CAS negative group (CAS <3).T2 relaxation time of extraocular muscles (T2RT;ms) and the areas of four extra-ocular muscles (AEOMs;mm2) were measured by 3D T2-mapping MR sequence before and after methylprednisolone treatment,so as the CAS and some ophthalmic examinations including visual acuity,intra-ocular pressure,eyeball movement,diplopia and proptosis.In addition,24 healthy volunteers were recruited as the control group.The mean T2RT and AEOMs in CAS positive group were higher than those in CAS negative group.Both CAS positive and negative groups had significantly higher mean T2RT and AEOMs than the control group (P<0.01).There was a positive correlation between T2RT and AEOMs values in GO patients,both of them had a positive correlation with CAS and the ophthalmic examinations.It was concluded that to evaluate the activity of GO,CAS was mostly related to inflammation symptoms of ocular surface,more than that,T2RT and AEOMs were also related to abnormal findings of the ophthalmic examinations including high ocular pressure,impaired eyeball movement,diplopia and proptosis.T2RT and AEOMs can reflex the inflammation state of ocular muscles better.CAS combined with 3D T2-mapping MR imaging could improve the sensitivity of detection of active GO so as the prediction and evaluation of the response to methylprednisolone treatment.
7.Advances in the study of inhibitors of kinases and nuclear factors for treating allergic asthma.

Currently, about 300 million people worldwide are affected by asthma. Most of these sufferers inhale immunosuppressants (ie corticosteroids) and beta-adrenergic receptor agonists for their asthma treatment. However, about 5%-10% of patients of asthma have poor response to such treatment. Investigation of kinase signaling pathway and nuclear transcription factor as a target molecule in the treatment of allergic asthma has been the concern of scholars home and abroad. This paper reviewed inhibitors of kinase signaling pathway and nuclear transcription factors for the treatment of asthma.

8.A study on apoptosis of fermented fnonotus obliquus on hepatocellular carcinoma HepG2 cells

Objective A variety of secondary metabolites can be obtained after the fermentation of medicinal fungi.The con-tent of these metabolites is much higher than that of cultivated medicinal fungi. We aimed to preliminarily study on the apoptosis of fer-mented Inonotus obliquus(FIO) on hepatocellular carcinoma HepG-2 cells. Methods The experiment was divided into 3 groups:the control group(The same volume of drug-free medium),the UIO group(200 mg/L) and the FIO group(200 mg/L). The contents of polysaccharides,terpenoids and polyphenols in unfermented Inonotus Obliquus (UIO) and FIO were determined by sulfuric acid phenol method,Folin-Ciocalteu method and colorimetric method. Effects of Fermented Inonotus obliquus on apoptosis of HepG-2 Cells were measured by MTT Method and Flow Cytometry. Results The contents of polysaccharides,terpenoids and polyphenols were sig-nificantly increased in FIO group [(10.140±0.849)mg/mL,(1.774±0.001)mg/mL, (9.979±0.022)mg/mL] compared with thatin UIO group [(7.161±0.305)mg/mL,(1.358±0.004)mg/mL,(6.314±0.237)mg/mL](P<0.01). Both UIO and FIO group induced apoptosis of HepG-2 cells according to Annexin V/PI double staining. Compared with the control group(4.3%),the apoptosis rate in-creased in UIO and FIO group (23.14%,27.37%) (P<0.05). In addition, the apoptosis rate was higher in FIO group than that in UIO group (P<0.05). Compared with the control group, the ratio of G0/G1phase cells was significantly increased and the ratio of S phase and G2/M phase was significantly decreased in UIO and FIO group (P<0.01). G0/G1phase cell ratio differences were statistically signif-icant compared with the UIO group (P<0.05) Conclusion FIO can better induce the apoptosis of HepG-2 cells than UIO.
9.Silica induced α-SMA expression in HBE cell line by targeting the PI3K/Akt pathway.

OBJECTIVETo explore the role of the phosphatidylinositol 3 kinase/protein kinase B (PI3K/Akt) pathway in silica-induced α-SMA (α smooth muscle actin) expression in HEB (human bronchial epithelial) cell.
METHODSThe cultured HBE cells were divided into 5 groups: control, silica, PI3K inhibitor (Ly294002), both PI3K inhibitor (Ly294002) and silica at the same time and the inhibitor 24 h ahead of silica. The final concentrations of PI3K inhibitor and silica were 10 µmol/L and 100 µg/ml, respectively. Western blots were used to detect protein expressions of Akt, p-Akt, TGF-β and α-SMA. The location and expression of α-SMA were measured by immunofluorescence assay.
RESULTSHBE cell line exposed to silica can induce Akt phosphorylation, in which expressions of p-Akt were up regulated 1 times at 48 and the highest at 72 h. The expressions of TGFβ increased remarkably at 12 h and the peak at 48 h after silica exposure, while the expressions of α-SMA increased at 24 h and the highest at 72 h. However, the PI3K inhibitor (Ly294002) significantly down regulated α-SMA expression. When the cell line exposed to the PI3K inhibitor ahead of silica 24 h, the expressions of p-Akt and α-SMA were more remarkably down regulated which were decreased 1.5 times and 7.6 times respectively compare to silica exposure group. But no significant changes were found for TGFβ expressions. The immunofluorescence assay showed that silica can induce α-SMA expression, which located in cytoplasma, and PI3K inhibitor can decrease the expression.
CONCLUSIONSSilica induced α-SMA expression in HBE cell line is by targeting the PI3K/Akt pathway and PI3K inhibitor can repress α-SMA expression.

10.Discovery and identification of WU polyomavirus in children from Zhejing region.

WU polyomavirus, which was firstly discovered in 2007, is a new human polyomavirus belonging to Polyomaviridae and containing circular double-stranded genomic DNA. In this study, the 278 clinical sputum specimens from children under 5 years old were collected from Wenzhou Medical College affiliated Wenling First Hospital, Zhejiang Province. Based on identification assay of WU polyomavirus previously reported, a WU polyomavirus was identified from clinical samples successfully, the positive rate was 0.4%. The sequences of PCR products were identical to that of VP2 gene and large T antigen gene derived from WU polyomavirus reported. The above results strongly suggested that the WU polyomavirus isolated was firstly found in Chinese children with acute lower respiratory tract infections. This study provides a firm basis for further research of WU polyomavirus.
