1.Clinical neuropsychological and functional neuroimaging features in patients with cerebellar infarction

Objective To investigate the neuropsychological and functional neuroimaging features in patients with cerebellar infarction (CI).Methods We analyzed 59 CI patients (27 left CI,32 right CI) and 26 healthy control subjects who received standard and experimental cognitive testing and neuroimaging study.We compared the cognitive manifestations between the groups with Student' s t test.Results Patients with CI(left/right) achieved significantly lower scores in auditory verbal learning test (AVLT) of memory test (12.27 ± 1.37 vs 9.33 ± 1.90/10.25 ±2.20,t =6.46,4.26,P <0.05),Associative Learning of Clinical Memory Scale (22.77 ± 3.07 vs 18.67 ± 1.98/16.84 ± 3.55,t =5.74,6.69,P < 0.05),symbol digit modalities test (SDMT) of visuospatial test(42.54 ±6.32 vs 20.85 ±9.57/34.84 ± 16.10,t =9.68,2.47,P < 0.05),and errors responses (RE) of Wisconsin card sorting test for executive function (16.77 ± 2.64vs 52.22 ± 16.29/54.47 ± 16.27,t =11.15,12.89,P < 0.05).Patients with left CI had significantly lower scores in correct responses percentage (RCP; 58.71 ± 10.93 vs 78.43 ± 5.26,t =-8.41,P < 0.05)and significantly higher scores in the trials to compete first category (RF; 23.59 ± 9.79 vs 14.12 ± 3.75,t =4.68,P < 0.05).Those finding suggests left CI would cause impairment on abstract conceptualization and concept formation; The patients with right CI had significantly lower scores in total memory quotient (86.69 ± 7.56 vs 112.02 ± 9.70,t =-11.17,P < 0.05),higher scores in perseverative responses (RP ;44.59 ± 17.50 vs 8.23 ± 3.46,t =11.47,P < 0.05) and nonperseverative responses errors percentage (nRPE; 44.00 ±20.67 vs 10.58 ± 2.35,t =9.07,P < 0.05).It means right CI would cause serious deficits on memory,cognitive shift and attention.The fibers between cerebellum and frontal,parietal lobe were reduced in CI patients,compared with healthy control.Conclusions These results suggest that cerebellum participated in the formation of part of cognitive function by connection with cerebrum.After CI,that the fibers contacted with the prefrontal and parietal reduced is the possible mechanisms for cognitive impairment.
2.Construction of siRNA expression vector targeting IGF-1R and its ability to induce cell apoptosls in hmnan lung cancer cells

Objective Insulin-like growth factor-1 receptor (IGF-1R),similar to insulin receptor,is one of the families of re- ceptor tyrosine kinases.,which has been found to be overexpressed in a variety of cancer.It is the main proliferation and survival sig- nal molecule in cancer cell and plays an important role in cancer growth and progress.Blocking signal transduction of IGF-1R by vari- ous strategies can suppress tumor growth and induce regression of established tumor.This study is to construct the siRNA expression vector targeting IGF-1R and to evaluate its ability to induce cell apoptosis in human lung cancer cells.Methods Two siRNA expres- sion vector,pENTR/U6-shRNA-1 and pENTR/U6-shRNA-2 targeting IGF-1R,were constructed using pENTR/U6 vector,and a vector targeting hieiferase gene,pENTR/U6-shRNA-Iuc,was constructed as control.After vectors were transfected into A549 for 48h, knockdown of IGF-1R mRNA and protein and Akt phosphorylation were accessed,and DNA ladder and flow cytometry were used for cell apoptosis.Results siRNA expression vectors targeting IGF-1R were successfully constructed,which was confirmed by PCR and DNA sequencing,pENTR/U6-shRNA-1 and pENTR/U6-shRNA-2 demonstrated the expression were (22.1?2.5) % and (80.1? 3.9) % in IGF-1R mRNA level,(15.2?3.1)% and (47.1?4.1)% in protein level,respectively,compared with pENTR/U6- shRNA-luc.Suppression of IGF-1R by pENTR/U6-shRNA-1 blunted Akt phosphorylation,increased cell apoptosis induced by 3% ethanol,and retained 77.5 % A549 cells in the G0/G1 phase.Conclusion siRNA expression vector targeting IGF-1R can effectively suppress the expression of IGF-1R expression in A549.This study suggests that DNA vector-based RNAi has the potential to be effec- tive and practical cancer gene therapy strategy.
3.Clinical significance of serum cytokines IL-1beta, sIL-2R, IL-6, TNF-alpha, and IFN-v in acute coronary syndrome.

OBJECTIVESTo explore serum cytokines levels (including IL-1beta, sIL-2R, IL-6, TNF-alpha, and IFN-upsilon) and their significance in patients with acute coronary syndrome (ACS) and the subsequent follow-ups, with attempt to estimate the role of various serum inflammatory markers in the diagnosis and assessment of ACS.
METHODSThe study population include 40 patients with acute myocardial infarction (AMI), 40 patients with unstable angina pectoris (UAP), and 40 controls. Among the 80 patients, 60 patients attended a follow up 4 months later. Serum inflammatory markers including IL-1beta, slL-2R, IL-6, TNF-alpha, and IFN-upsilon were measured by enzyme linked immunosorbent assay.
RESULTSSerum IL-1beta, sIL-2R, IL-6, TNF-alpha were significantly higher in AMI group or UAP group compared to the control group and became significantly lower 4 months later in the follow-up patients. Serum levels of IFN-upsilon shows no significant difference between AMI group or UAP group and controls, also showing no significant change when measured in follow up patients. There was no correlation between serum creatine kinase-MB isoenzyme levels and serum inflammatory markers either in UAP or AMI group. Furthermore, when divided into two subgroups using Wagner's QRS scoring system in the AMI group, there is no difference of each serum inflammatory marker between < or = 6 scores group and > 6 scores group.
CONCLUSIONSerum levels of certain inflammatory markers may have some diagnostic value for ACS, and can be a useful marker reflecting disease stability.

4.Effect of Cynomorium songaricum polysaccharide on telomere length in blood and brain of D-galactose-induced senescence mice.

OBJECTIVETo study the effects of Cynomorium songaricum polysaccharide (CSP) on telomere length in blood and brain tissues of aged mice in order to provide some evidence for CSP's development and applying in the clinical uses.
METHODKunming mice were intraperitoneal injected D-galactose (500 mg x kg(-1) x d(-1)) to make the aging models, and different dosages of CSP (20, 40, 80 mg x kg(-1)) were given by gavage for 56 days. The average length of telomere was determined by real-time fluorescence quantitative PCR.
RESULTThe relative T/S ratio of the group high and middle dosages of CSP in blood were 1.64 +/- 0.36 and 1.33 +/0.28, respectively, and higher than that of the group of senescence 1.01 +/- 0.13 (P < 0.01). Values of the group of high, middle, and low dosages of CSP in brain tissues were 3.34 +/- 0.58, 2.30 +/- 0. 75 and 1.55 +/- 0.58, respectively, and significantly higher than that of the group of senescence 1.04 +/- 0.33 (P < 0.01).
CONCLUSIONCSP can exert the anti-aging effects by increase telomere length f senescence mice.

5.Chemical constituents from Polygonum posumbu

AIM To study the chemical constituents from Polygonum posumbu.METHODS The 95% ethanol extract from P.posumbu was isolated and purified by silica,Sephadex LH-20 and recrystallization,then the structures of obtained compounds were identified by physicochemical properties and spectral data.RESULTS Fourteen compounds were isolated and identified as n-hexacosanol (1),n-tetradecanoic acid (2),β-sitosterol (3),daucosterol (4),gallic acid (5),quercetin-3-O-(2″-O-galloyl)-β-D-glucopyranoside (6),quercetin-3-O-β-D-glucopyranoside (7),quercetin (8),o-phthalic acid bis-(2-ethyl decyl)-ester (9),inositol (10),sucrose (11),succinic acid (12),n-butyl-β-D-fructopyranoside (13),α-D-glucose (14).CONCLUSION Compounds 1,2,9,10 and 14 are isolated from genus Polygonum for the first time,compounds 1-14 are isolated from this plant for the first time
6.Mediating effect of coping efficacy on relationship between psychological stress and adjustment in college freshmen

Objective:To explore the mediating effect of coping efficacy on the relationship between psychological stress and adjustment.Methods:This study was a longitudinal study.Totally 267 college freshmen (137 males and 130 females,aged 19 to 21) were selected to fill in the China College Student Psychological Stress Scale (CCSPSS),China College Student Adjustment Scale (CCSAS) and Coping Efficacy Questionnaire (CEQ),each measurement interval two months,three times totally.Results:The CCSPSS scores were negatively correlated with scores of CCSAS and CEQ cognitive dimension (r =-0.29--0.08,Ps < 0.05),and positively correlated with measurement period (represented by a continuous and dummy variable,r =0.08,P < 0.05).The CCSAS scores were positively correlated with scores of CEQ cognitive dimension (r =0.25,P < 0.01),and negatively correlated with measurement period (r =-0.13,P < 0.01).The CEQ cognitive dimension scores was negatively correlated with measurement period (r =-0.14,P < 0.01).The mediating effect analysis showed that coping efficacy played a partly mediating role between adjustment and psychological stress,measurement time,and the mediating effect were accounted for the total effect 45.7%,33.6%.Conclusion:It suggests that psychological stress and measurement time may not only directly affect the adjustment,but also indirectly affect adjustment through mediating effect of coping efficacy.
7.Multidrug-resistant Clinical Enterobacter cloacae Isolates: Analysis of Their ESBLs Gene Types and Molecular Epidemiology

OBJECTIVE To understand the distribution and epidemiology of ESBLs in ceftazidime or cefotaxime-(resistant) clinical Enterobacter cloacae isolates.METHODS Twenty seven ceftazidime or cefotaxime-resistant(nonrepetitive) E.cloacae were collected from 27 patients hospitalized at the Huashan Hospital,Shanghai.PCR and(sequencing) were performed to understand the distribution of ESBLs in E.cloacae;rep-PCR was(performed) to(understand) the epidemiology of ESBLs in E.cloacae.RESULTS CTX-M-3 like(ESBLs) were the most prevalent in our study(48%);this was the first report of VEB-1-like ESBLs in the member of(Enterobacteriaceae) in China,and the first report of the ESBLs VEB-1-like and CTX-M-3-like in an isolate simultaneously;the majority of(ESBLs) producers exhibited the same rep-PCR pattern,but harbored different ESBLs gene.(CONCLUSIONS) In our study,ESBLs have become prevalent in clinical E.cloacae isolates,and become an important factor of E.cloacae isolates resistant to extended-spectrum beta(-lactams).
8.InvestigatIion on the work stress and psychological status of nursing staff in a general hospital

Objective To study the work stress and psychological status of nursing staff as well as their relationship,so as to provide scientific references for alleviating work stress and improving mental health.Methods 532 nurses insome general hospital were chosen by target sampling,and self-designed questionnaire,work stress scale and SCL-90 were used to investigate their basic information,work stress and psychological status.Results Work stress analysis showed that,among 5 dimensions of nursing staff' s work stress,the score of nursing,nursing profession and work was the highest,respectively (2.73 ± 0.62),(2.61 ± 0.61).The score of somatization,obsessive-compulsivesymptoms and interpersonal sensitivity in SCL-90 was respectively (1.80 ± 0.50),(2.07 ± 0.55),(1.82 ± 0.52),higher than the norm which is (1.37 ± 0.48),(1.62 ±0.58),(1.65 ± 0.51),and the differences were statistically significant (t =17.37,15.43,6.49,respectively;P < 0.01) ; the score of depression,paranoid,psychosis,anxiety,hostility,terror in SCL-90 was respectively higher than the norm (P < 0.01).There was no statistically significantdifference of mental health scores between nurses of different genders (P > 0.05).Score of somatization was relatively higher in nursing staff who were over 40 years old,officially employed or nurses-in-charge and above (P < 0.05).Score of interpersonal relationship,anxiety,hostile and horror was relatively higher in nursing staff who were under 40,non-officially employed or below nurses-in-charge (P < 0.05).Logistic regression analysis showed that,professional title,nursing for patients,management and interpersonal relationship,nursing profession and work were the four main influencing factors for nursing staff' s mental health,and the order of importance was professional > nursing profession and work > nursing for patients > management and interpersonal relationship.Conclusions Nursing staff' s work stress is high and their psychological status is not good enough in general hospital.Scientific measures should be taken by hospital manager to alleviate nursing staff' s work stress and improve their mental health.
9.Study on diet, physical activities and body mass index in Chinese population in 2002.

OBJECTIVETo describe the prevalence of behavior on diet, physical activities and body mass index (BMI) in different populations related to factors as education, occupation and geographical distribution.
METHODSIndicators including frequency of taking different foods, intake of cooking oil/fast foods, intensities of physical activities at work, proportion of taking physical exercises during the day, sedentary life style and BMI were calculated based on results from 17 questions of behavior risk factors surveillance (BRFS) questionnaire by weight on age structures from 2000 census.
RESULTSSeventy percentage of the people took vegetable and 40 percent took fruits 5-7 days per week, and over 50 percent of them ate pork/beef/mutton but few of them ate beans and eggs. 25 percent of the people ate chicken/duck/fish/and shrimps 5-7 times per week but another 40 percent ate them only less then once per week. 70% of the people almost never drank milk or milk-products. 15 percent of them consumed sweet and greasy foods 3-7 days per week and 30 percent of them ate smoked food 3-7 days per week in the past 30 days. The proportions of food intake were different under different geographical regions, education levels and occupations. 11.7% of the sample population cooked mainly with animal oil, and 33% of the students had ever been to McDonald's. 11.7%, 20.5%, 44.7% and 23.0% of the subjects engaged in sedentary, light, moderate or heavy physical activities respectively. 18.04% of the subjects took part in physical exercises with different proportions by gender, occupation, education and geographical settings. 8.3% people were slim which was defined as having BMI lower than 18.5 kg/m(2). 68.0% of the people had a BMI as 18.5-23.9 kg/m(2). 23.7% of the subjects were being overweight which was defined as having BMI greater than 24 kg/m(2), among which 8.5% people with a BMI of 24-24.9 kg/m(2).
CONCLUSIONThe prevalence of overweight and obesity will increase in the next 20-30 years, with the habits of taking more foods with high fat and energy but with less physical activities and keeping the idea as "fattier makes happier". Priorities should be given to changing the diet habit, avoiding over-intake of high fat and high energy plus increasing physical activities through publicity of knowledge on health, policy enforcement and development of supportive environment.

10.Different levels of nitric oxide in seminal plasma of fertile and abnormospermic men.

OBJECTIVESTo understand the relation between nitric oxide (NO) level in seminal plasma and male fertility.
METHODSThe levels of NO in seminal plasma of 174 fertile males and 217 abnormospermia patients were measured by using the reductase of nitric acid, Greiss reagent and spectrophotometry.
RESULTS1. NO was found in all 174 samples (20-49 years) of fertile males which was (27.78 +/- 5.81) mumol/L. The NO level in seminal plasma in fertile males was became higher after age 40, and there was no significant difference between 20-29-year-old [(26.25 +/- 5.52) mumol/L] and 30-39-year-old[(28.11 +/- 5.87) mumol/L]. But the group of 40-49-year-old[(30.17 +/- 6.14) mumol/L] had a higher level of NO in seminal plasma than 20-29-year-old (P < 0.05). 2. The seminal plasma samples from nine types of abnormospermia were measured, which all had a higher level of NO than fertile males. In abnormospermia, the level of NO in seminal plasma of the patients with single abnormality increased slightly, two abnormality obviously increased, and the highest level of NO in seminal plasma appeared in three abnormality.
CONCLUSIONSThis results confirmed that proper level of NO in seminal plasma may regulate the spermatogenesis.
