1.Application of reflectance confocal microscopy combined with dermoscopy in assessing melanin and blood ;vessels in chloasma lesions

Objective To investigate the morphology and quantity of melanin and blood vessels in chloasma lesions by using reflectance confocal microscopy(RCM)and dermoscopy, and to explore the relationship of melanin and blood vessels with lesion color. Methods RCM was used to evaluate the quantity and distribution of melanin in lesions of 135 patients with chloasma, and to observe the morphology of melanin and dendritic melanocytes in 54 of the 135 patients, and dermoscopy was performed to assess the quantity and morphology of blood vessels in lesions of the 54 patients. Statistical analysis was carried out by Spearman correlation analysis to assess the relationship of melanin and blood vessels with lesion color. Results Increased quantity of melanin was observed in both the epidermal layer and dermo?epidermal junction in all the 135 patients, and in only the superficial dermis in 36(27%)patients. No patients showed increased quantity of melanin in only the dermis without epidermal involvement. Among the 54 patients, there was a positive correlation between the quantity score and morphology score of melanin(r=0.73, P<0.001), between the quantity score and morphology score of blood vessels(r=0.87, P<0.001), as well as between the total melanin score and total blood vessel score(r=0.554, P<0.001). Lesion color was positively correlated with the quantity and morphology of melanin(r = 0.51, 0.39, respectively, both P < 0.05)and blood vessels(r = 0.46, 0.44, respectively, both P<0.05). Conclusions RCM and dermoscopy can be used to evaluate melanin and blood vessels in chloasma lesions. There is a positive correlation between the quantity and morphology of melanin and between those of blood vessels in chloasma lesions, and lesion color is positively correlated with the quantity and morphology of melanin and blood vessels.
2.Preliminary study of the accuracy of hepatic volume measurement by 3D + medical imaging visualization processing system

Objective To investigate the accuracy of liver volume measurement in patients with primary hepatocellular carcinoma by using the 3D + medical imaging visualization processing system. Methods Thirty patients who were clinically diagnosed as primary hepatocellular carcinoma and underwent surgical resection before preoperative assessment were selected from January 2016 to November 2016. Preoperative thin-slice CT images were transferred into the 3D + medical imaging visualization processing system for hepatic volume measurement, the predictive resection volume were measured and statistically analyzed with that of intraoperative specimen according to water measurement method. Results All patients had complete resection of tumor, the average volume of predictive resection calculated by the 3D software was (408.84 ± 177.07) ml, the average volume of intraoperative specimen measured by the water test was (399.95 ± 176.15) ml, the difference was not statistically significant (t=1.502, P=0.144), and the average percentage error was (6.21 ± 4.73) %. Pearson correlation analysis showed that there was significant correlation between them (r=0.983, P<0.05). Conclusion 3D+medical image visualization processing system is a simple, intuitive and accurate method for liver volume measurement, which can provide the basis for the accurate measurement of liver volume and the implementation of complex hepatectomy, and guide the precise hepatectomy.
4.DAZL and male infertility: an update.

DAZL, a member of the DAZ family, plays a key role in human spermatogenesis. It regulates the promoter via genetic modification, especially DNA methylation, promoting the transcription of DAZL. Besides, the relation of DAZL SNPs with male infertility has been a hot spot of research for many years. It is deduced that local and ethnic factors may be associated with the failure of spermatogenesis. This article presents an overview on the results of recent studies, changes in the DNA methylation of DAZL in spermatogenesis, and the association of DAZL SNPs with male infertility, aiming to provide a new theoretical basis and clinical strategy for the treatment of male infertility.

5.Interaction of E3 ligase HUWE1 and eukaryotic translation initiation factor eIF4E.

To explore the regulation of eIF4E, we screened the protein interacting with eIF4E from human cDNA library by using yeast two-hybrid system. Several clones interacting with eIF4E were identified. One of them was homologous with HUWE1 (HECT, UBA and WWE domain containing 1, also named as ARF-BP1, HECTH9 or HUWE1). Cell co-immunoprecipitation showed that eIF4E could bind to HUWE1 in mammalian cells. We also found that HUWE1 bearing the HECT domain is necessary for its association with eIF4E.

6.Purification of Indoor Air Formaldehyde by Plants:a Review

The pollution hazard of formaldehyde in low level from indoor air is worldwide.Many researches have been focused on the ornamental plant purification for indoor air formaldehyde in low concentration.Although a numbers of literatures have documented that gaseous formaldehyde can be removed by many plant species,a long term purification efficiency of the plants for formaldehyde is uncertain.The research in the future works should be focused on phytotoxieity of formaldehyde,a long term purification efficiency of plants,soil contribution to formaldehyde removal under plant-soil system,effects of complex pollutants on the purification of plants for formaldehyde,and volatile organic compounds from the plants which can remove formaldehyde.
7.Effect Evaluation on Association Rules of Symptoms in TCM Treatment of Viral Pneumonia

This study was aimed to explore an effect evaluation method of traditional Chinese medicine (TCM) in the treatment of viral pneumonia. Association rules were used to analyze the therapeutic effect of heat, stagnation, and phlegm related syndromes from 297 respiratory syncytial virus (RSV) pneumonia children before and after treatment. The results showed that through the comparison of the frequent itemset of heat, stagnation, and phlegm related syn-dromes, in part of the time, the support degree of the experimental group was obviously lower than that in the con-trol group (P < 0.05). The frequent itemset of the experimental group disappeared 1~3 d earlier than the control group, which indicated the effect in the experimental group was better than the control group. It was concluded that association rules can be used for evaluating therapeutic effect of TCM in treatment of viral pneumonia. The effect evaluation method based on association rules of symptoms is more in line with the syndrome differentiation of TCM.
8.Expression, purification and immunogenicity analysis of recombinant MPB64 using baculovirus expression system

Objective:To express mycobacterium tuberculosis protein MPB 64 in baculovirus insect cell expression system ,and identify its immunogenicity.Methods:The target gene MPB64 was connected to pFastBac vector ,then the pFastBac-MPB64 plasmid which was harvested would transformed to DH10Bac competent,and the target gene was transposition into Bacmid by Tn7 transposase fragment,therefore Bacmid-MPB64 Shuttle vector was obtained.The shuttle vector was packaged by liposomes and transfected Sf 9 cells to harvest P1-generation virus ,then high titers of P4 generation virus was harvested by repeat transfected Sf 9 cells three times.The target protein MPB64 was purified from the supernatant by Q Sepharose FF and Ni affine chromatography ,which were used to immunize BALB/c mice.Antibody changes in serum would be detected ,and the proliferation of immunized mice spleen cells would be detected by MTT,detected the IFN-γsecretion by MPB64 stimulated spleen cells by ELISA method.Results: MPB64 successfully expressed in insect cells.The purity of target protein was over 90% and yield up to 35 mg/L after purification.Purified protein can effectively stimulate BALB/c mice to produce antibodies , increase the content of IFN-γmedium in mice spleen cells ,and significantly promoting proliferation in spleen cells between 0.2-100 μg/ml.Conclusion: MPB64 which has immunogenicity was successfully expressed in baculovirus insect cell expression system ,that open a new avenue for tuberculosis vaccine production.
9.Action mechanisms of prolactin and its receptors on penile erection and ejaculation.

Prolactin is a polypeptide hormone which mainly acts on the reproductive system and plays an important role in penile erection and ejaculation. Prolactin receptors have a variety of short forms apart from the classic long form, which are widely expressed in male reproductive glands. High levels of prolactin can induce erectile dysfunction and results in secondary male infertility, which are mainly associated with the inhibition of dopaminergic activity, reduction of the testosterone level, and contraction of the cavernous smooth muscle. Moreover, low levels of prolactin can result in ejaculatory dysfunction. This article updates the views on the expressions of prolactin receptors in the male reproductive system, the effects of prolactin on penile erection and ejaculation, and its action mechanisms.
