1.Interaction of E3 ligase HUWE1 and eukaryotic translation initiation factor eIF4E.

To explore the regulation of eIF4E, we screened the protein interacting with eIF4E from human cDNA library by using yeast two-hybrid system. Several clones interacting with eIF4E were identified. One of them was homologous with HUWE1 (HECT, UBA and WWE domain containing 1, also named as ARF-BP1, HECTH9 or HUWE1). Cell co-immunoprecipitation showed that eIF4E could bind to HUWE1 in mammalian cells. We also found that HUWE1 bearing the HECT domain is necessary for its association with eIF4E.

2.Survey and related factors analysis of anxiety status among caregivers of stroke patients

Objective To investigate anxiety status among caregivers of stroke patients and analyze it's related factors.Methods 98 caregivers of stroke patients were surveyed by the general questionnaire and SelfRating Anxiety Scale (SAS).Results The anxiety scores of caregivers of stroke patients were significant higher than that in Chinese norm.Stroke patients with physical dysfunction,cognitive impairment and caregiver gender,educational level,daily care of the time,family income were significantly associated with anxiety of the caregivers.Conclusions Support information from various channels should be established and more emotional and spiritual assistances should be provided to the caregivers.
4.Analysis of relative factors of tumble among senile cerebrovascular diseases patients

Objective To study the related factors of tumble among senile cerebrovascular diseases patients,so as to effectively prevent it happening.Methods A total of 102 senile cerebrovascular diseases patients with the history of tumble within 12 months were investigated with self-designed questionnaire.Results The incidence of tumble among male patients was 42.16%,while female patients was6.86% ;living alone patients was 37.25%,while cohabit patients was 16.67% ;balance ability abnormal patients was 38.24%,while that normal patients was 22.55%.There was significantly difference between them(x2 =4.95,8.45,5.13 ;P <0.05 or P < 0.01).The tumble incidence among patients with different ages,medicine or not,nocturnal enuresis condition,eyesight,intelligence and different chronic diseases was significantly difference(P <0.05 or P <0.01).Conclusions The incidence of tumble among senile cerebrovascular diseases patients are related to many factors,so it is important to assessment their health condition and establish effective preventive measures to reduce the ratio of tumble.
5.Relationship between intracellular calcium and reactive oxygen species in sodium fluoride-induced injury in human neuroblastoma SH-SY5Y cells

Objective To explore the relationship between intracellular calcium levels ([Ca2+]1) and reactive oxygen species (ROS) in sodium fluoride (NaF)-induced injury in human neuroblastoma SH-SY5Y cells. Methods The levels of [Ca2+]1 and ROS were measured in different exposed times(0,3,6,12,18,24 h) respectively after SH-SY5Y cells were exposed to 40 mg/L NaF in vitro, and the optimal expose time was selected. Furthermore, the changes of [Ca2+]1, ROS and LDH levels in 40 mg/L NaF-treated groups incubated with 38.23 mg/L BAPTA-AM or 380.40 mg/L ethylene glycol-bis-(beta-aminoethyl ether)-N, N, N', N'-tetraacetic acid (EGTA) or 16.32 mg/L N-acetyl-L-cysteine(NAC) also observed at the optimal expose time(12 h), respectively. Results At 3,6,12,18 and 24 h, [Ca2+]1 level(5620.0±226.3,4775.5±85.6,3312.3±87.5, 3047.0±75.0,2717.0±66.5) was significantly increased, and so was the ROS level(4449.53±324.61,7463.07±117.43,20 227.33±178.04,8817.56±200.13, 7975.61±92.90) except at 3 h, compared with 0 h(2115.0±24.0,4098.01±21.22, all P<0.05). The levels of [Ca2+]1 and ROS reached the peak at 3 h and 12 h, respectively. [Ca2+]1 and LDH levels in NaF-treated group [3279.5±94.0, (1057.50±64.35)U/L], NaF+NAC treated group[ 3583.0±350.7, (561.02±85.50)U/L], NaF+EGTA treated groups[3701.5±157.7, (1074.50±86.97)U/L], and BAPTA-AM treated group[2766.5±38.9, (521.43±40.80)U/L] had increased, compared with the control[2022.5±118.1, (186.97±8.73)U/L], the difference being statistically significant (P<0.05). ROS levels in NaF-treated group (19 003.04±332.34), and NaF+EGTA treated group(19 170.12±95.46) was higher than that in the controls(4060.98±145.66), the difference being statistically significant (P<0.05). NaF and NAC had antagonistic effect on ROS and LDH levels (F=976.11,43.54,P<0.05). And NaF and BAPTA-AM had antagonistic effect on [Ca2+]1, ROS and LDH levels (F=15.65,1515.53,115.00, P<0.05). Conclusions NaF-related calcium is released from the site of intracellular calcium storage, which induces ROS production, both of them caused cytotoxicity and the increase of LDH level in human neuroblastoma SH-SY5Y cells.

AIMTo investigate the impact of 8-(N,N-diethylamino)-n-octyl-3,4,5-trimethoxybenzoate (TMB-8) on the change of cerebral blood flow(CBF) induced by 5-HT or KCl in rats.
METHODSThe CBF in rats was measured by a Laser-Doppler flowmeter. The cranial window field was superfused with artificial cerebral spinal fluid containing TMB-8, 5-HT or KCl.
RESULTS12.5, 25 and 50 mumol.L-1 TMB-8 showed no significant effect on rest CBF in rats. 12.5, 25 and 50 mumol.L-1 TMB-8 apparently inhibited the decline of CBF evoked by 1 or 2 mumol.L-1 5-HT. When persistent reduction of CBF were evoked by 1 mumol.L-1 5-HT, TMB-8 markedly increased the CBF in a concentration-dependent manner. The reduction of CBF induced by 20 or 40 mmol.L-1 KCl was also suppressed by 12.5, 25 and 50 mumol.L-1 TMB-8. While persistent reduction of CBF was evoked by 20 mmol.L-1 KCl. TMB-8 markedly increased the CBF in a concentration-dependent manner.
CONCLUSIONThese results indicate that TMB-8 is effective in preventing and treating the reduction of CBF induced by 5-HT or KCl, and improved the supply of blood in rat brain during ischemia.

7.Mutation analysis of p63 gene in the first Chinese family with ADULT syndrome.

BACKGROUNDADULT syndrome (acro-dermato-ungual-lacrimal-tooth syndrome) is a rare ectodermal dysplasia disorder known as autosomal dominant inheritance. Recent studies have linked p63 gene mutation to the development of this disease. However, the genetic characteristics of ADULT syndrome were still not well understood.
METHODSMutation analysis of p63 gene in the first Chinese ADULT syndrome family was performed using direct DNA sequencing.
RESULTSThe sequence analysis of exon 8 of p63 gene disclosed a heterozygous G>A substitution at nucleotide 893 (R298Q) in the proband. In addition, a single nucleotide polymorphism (SNP) rs16864880 in the downstream flanking region (DFR) of p63 exon 8 was also identified in this family. The proband and the paternal side including her father exhibited the C/G genotype at this position. The C/G variant frequency in the paternal was significantly higher as compared with the maternal (6/10 vs 0/6, P = 0.034).
CONCLUSIONSADULT syndrome may be caused by the p63 gene mutation, and it might have closer genetic association with the paternal side in this family.

8.Review on Pharmaceutical Pricing Reform of Urban Public Hospitals

Objective To describe and evaluate the strategies,status quo,effect and problems of pharmaceutical pricing reform in 4 batches of urban public hospitals in China.Methods Chinese database and website of Commission of Health and Family Planning were searched.Bibliometrics was done.Results A total of 33 studies were included by screening and reading the abstracts.Among them,4 were masters' theses,and 29 were research articles and reviews.Generally quantitative researches with strong study designs accounted for only a small proportion.Most of the local reforms achieved basic targets,such as cancelling medicine addition,increasing medical service price,improving hospital internal operating efficiency and controlling medical price,however problems still existed including incomplete adjustment for medical services prices and unmet needs of differentiated compensation plans.Conclusion The prices of medical services should make further adjustment.Efforts should still be made in terms of establishing dynamic compensation mechanism for urban public hospitals,coordinating the policy decision-making process for more integrated measurements including reforms on payment models and logistic process.
9.Oral lavage in coma patients with tracheotomy

Objective To compare oral lavage and traditional oral nursing method in coma patients with tracheotomy regarding their clinical effects.Methods 600 coma patients with tracheotomy suffering from pulmonary infection and/or oral complications were randomly divided into control group ( n =300 ) and observation group (n =300).The observation group was administered with gauze balls and oral lavage; the control group was nursed with the traditional method.The clinical effects of both groups with respect to oral stench,swollen bleeding gums,oral ulcers,pulmonary infections were compared.Results The improvement rate of oral stench,swollen bleeding gums,oral ulcers and pulmonary infections in the observation group surpassed that in the control group (P < 0.05).Conclusions Gauze balls and oral lavage in coma patients with tracheotomy is superior to the traditional method with respect with clinical effects,therefore it has clinical significance to some extent.
10.Establishment of an mouse model of iron-overload and its impact on bone marrow hematopoiesis.

OBJECTIVETo establish a mouse model of iron overload by intraperitoneal injection of iron dextran and investigate the impact of iron overload on bone marrow hematopoiesis.
METHODSA total of 40 C57BL/6 mice were divided into control group, low-dose iron group (12.5 mg/ml), middle-dose iron group (25 mg/ml), and high-dose iron group (50 mg/ml). The control group received normal saline (0.2 ml), and the rest were injected with intraperitoneal iron dextran every three days for six weeks. Iron overload was confirmed by observing the bone marrow, hepatic, and splenic iron deposits and the bone marrow labile iron pool. In addition, peripheral blood and bone marrow mononuclear cells were counted and the hematopoietic function was assessed.
RESULTSIron deposits in bone marrow, liver, and spleen were markedly increased in the mouse models. Bone marrow iron was deposited mostly within the matrix with no significant difference in expression of labile iron pool.Compared with control group, the ability of hematopoietic colony-forming in three interventional groups were decreased significantly (P<0.05). Bone marrow mononuclear cells counts showed no significant difference. The amounts of peripheral blood cells (white blood cells, red blood cells, platelets, and hemoglobin) in different iron groups showed no significant difference among these groups;although the platelets were decreased slightly in low-dose iron group [(780.7±39.60)×10(9)/L], middle dose iron group [(676.2±21.43)×10(9)/L], and high-dose iron group [(587.3±19.67)×10(9)/L] when compared with the control group [(926.0±28.23)×10(9)/L], there was no significant difference(P>0.05).
CONCLUSIONSThe iron-overloaded mouse model was successfully established by intraperitoneal administration of iron dextran. Iron overload can damage the hepatic, splenic, and bone marrow hematopoietic function, although no significant difference was observed in peripheral blood count.
