7.Infectious bovine rhinotracheitis viral gG expression and gG-ELISA development.

Taking the genome DNA of Infectious Bovine Rhinotracheitis Virus (IBRV) as the template, the gG gene was amplified with PCR and cloned into the T cloning vector pMD18-T. After being identified by restriction digestion and DNA sequencing, the insert was subcloned into the expression vector pGEX-KG. Sodium docecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and Western blot assay showed that this gene was expressed as both soluble form and inclusion body by the transformed E. coli BL21 strain (DE3). The fusion protein was purified and used as the coating antigen to develop the indirect Enzyme-Linked Immunosorbent Assay (ELISA). Comparison between this gG-ELISA and commercial IBRV gB-ELISA Kit (IDEXX) was made in the detection of 380 cow serum samples. The results demonstrated an agreement of 92%. By using this novel gG-ELISA, 1248 cow serum samples were tested and the average positive rate of IBRV antibodies for imported cows is 21.7%, while the positive rate ranged greatly from 0.0%-41.5% for Hubei local Chinese Black and White Dairy Cows.

8.Longitudinal gracilis musculocutaneous flaps with a crossing boundary blood supply from the obturator artery.

OBJECTIVEThe traditional gracilis musculocutaneous flap is supplied by a branch of deep femoral artery, which enters the muscle in between the upper and middle third of it. So the flap barely reaches the pelvis and perineum region for reconstruction. By exploring the blood supply pattern we tried to rotate the flap Upon at the higher point starting at the obturator foramen in order to let it cover a bigger area.
METHODSanatomical reviewing of the blood supply of the gracilis branches of obturator, medial femoral circumflex and deep femoral arteries. Based on this a new type of longitudinal gracilis musculocutaneous flap supported only by the obturator artery was designed to reach the pelvis, female genitalia, pubic symphysis, inguinal area easily.
RESULTSThe new kind of flap has been applied to 9 patients for deformity repairing and tissue replacement in the pelvic and perineal area. All the flaps survived and achieved satisfactory result with 3 months to 3 years' follow up.
CONCLUSIONSLongitudinal gracilis musculocutaneous flaps supplied by the obturator artery can be used as regular musculocutaneous flap clinically.

9.Relationship between the efficacy of inhaled corticosteroids and the leukotriene expression in children with asthma.

OBJECTIVETo evaluate the relationship between leukotriene expression in blood polymorphonuclear leukocytes (PMNL) and the efficacy of inhaled corticosteroids (ICS) in children with asthma.
METHODSThirty-two children with asthma (5-12 years) and ten healthy children (control group) were enrolled. The asthmatic children were subdivided into ICS well-controlled and ICS poorly-controlled groups based on their clinical symptoms and lung function. The level of leukotriene C4 synthase (LTC4S) mRNA in PMNL was detected by fluorescence quantitative polymerase chain reaction. The level of LTC4S mRNA was expressed by the value of qCt, and the value of qCt was diversely correlated with the level of LTC4S mRNA expression. The concentration of urinary leukotriene E4 (LTE4) was measured using ELISA.
RESULTSThe expression of LTC4S mRNA in PMNL was significantly higher in children with asthma (qCt: 1.12+/-0.27) than that in the control group (qCt: 1.42+/-0.12; P< 0.05). The expression of LTC4S mRNA in PMNL in the ICS poorly-controlled group (qCt: 1.03+/-0.17) was significantly higher than that in the ICS well-controlled group (qCt: 1.24+/-0.33; P< 0.05) and the control group(1.42+/-0.12; P< 0.01). There was no significant difference in the level of urinary LTE4 among the the ICS poorly-controlled, the ICS well-controlled and the control groups.
CONCLUSIONSLTC4S mRNA expression in PMNL in asthmatic children increases, and the LTC4S mRNA expression in the ICS poorly-controlled group is higher than that in the ICS well-controlled group. This suggests that an increased leukotriene expression might be associated with poorly-controlled asthma.

10.Study on diet, physical activities and body mass index in Chinese population in 2002.

OBJECTIVETo describe the prevalence of behavior on diet, physical activities and body mass index (BMI) in different populations related to factors as education, occupation and geographical distribution.
METHODSIndicators including frequency of taking different foods, intake of cooking oil/fast foods, intensities of physical activities at work, proportion of taking physical exercises during the day, sedentary life style and BMI were calculated based on results from 17 questions of behavior risk factors surveillance (BRFS) questionnaire by weight on age structures from 2000 census.
RESULTSSeventy percentage of the people took vegetable and 40 percent took fruits 5-7 days per week, and over 50 percent of them ate pork/beef/mutton but few of them ate beans and eggs. 25 percent of the people ate chicken/duck/fish/and shrimps 5-7 times per week but another 40 percent ate them only less then once per week. 70% of the people almost never drank milk or milk-products. 15 percent of them consumed sweet and greasy foods 3-7 days per week and 30 percent of them ate smoked food 3-7 days per week in the past 30 days. The proportions of food intake were different under different geographical regions, education levels and occupations. 11.7% of the sample population cooked mainly with animal oil, and 33% of the students had ever been to McDonald's. 11.7%, 20.5%, 44.7% and 23.0% of the subjects engaged in sedentary, light, moderate or heavy physical activities respectively. 18.04% of the subjects took part in physical exercises with different proportions by gender, occupation, education and geographical settings. 8.3% people were slim which was defined as having BMI lower than 18.5 kg/m(2). 68.0% of the people had a BMI as 18.5-23.9 kg/m(2). 23.7% of the subjects were being overweight which was defined as having BMI greater than 24 kg/m(2), among which 8.5% people with a BMI of 24-24.9 kg/m(2).
CONCLUSIONThe prevalence of overweight and obesity will increase in the next 20-30 years, with the habits of taking more foods with high fat and energy but with less physical activities and keeping the idea as "fattier makes happier". Priorities should be given to changing the diet habit, avoiding over-intake of high fat and high energy plus increasing physical activities through publicity of knowledge on health, policy enforcement and development of supportive environment.
