ObjectiveTo observe the effect of pioglitazone on the metabolism of glucose and lipid in patients with Impaired Glucose Regulation(IGR).Methods60 cases of IGR received conventional education on diabetes prevention, while the patients in intervention group(30 cases) accepted pioglitazone 15 mg once a day for 12 weeks.Blood pressure(BP),body weight, waist circumference, hip circumference were measured to calculate body mass index(BMI) and waist-hip ratio(WHR).The Fasting blood glucose(FBG),2 h postprandial blood glucose, glcosylated hemoglobin(GHb,HbA1C),fasting serum insulin(FINS),postprandial serum insulin(2hINS),total cholesterol(TC),triglyceride(TG),high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-C),hepatic function, renal function were determined before and 12 weeks after intervention.ResultsAfter intervention,BP,BMI,FBG,2hBG,FINS,2hINS,HbA1C,TC,TG,LDL-C had been decreased significantly in intervention group(P<0.05 or P<0.01).There were significant differences between intervention group and control group(P<0.05 or P<0.01) in all indexes except body weight,WHR,hepatic function, renal function.ConclusionPioglitazone can obviously improve the metabolism of glucose and lipid in patients with IGR.

Object To investigate the in vivo antioxidant effect of the extract from Rosmarinus officinalis L. and its active substances. Methods The contents of MDA, the activites of SOD and GSH Px in serum, heart, liver, brain and skeleton muscle were determined in oxidative stress mouse model caused by exercise. Results It was found that in serum, liver, heart and skeleton muscle except the brain, the contents of MDA were decreased and the activities of SOD and GSH Px were increased by 250 mg/kg and 500 mg/kg of total phenolic diterpenes (TPD) extract taken. Conclusion The results showed that R. officinalis has prominent antioxidant effect in exercise mice and the active constituents may be phenolic diterpenes.

To clone cDNA of human leptin gene and obtain leptin protein for future study on leptin binding proteins. The cDNA of human leptin with 6 x his-tag was cloned by over-hang extension PCR protocol using human genomic DNA as template, and subcloned into in vitro expression vector pIVEX2.3MCS, and the fusion protein was expressed in vitro by Rapid Translation System (RTS) (RTS500 cycle primer Kit and RTS500 ProteoMaster of Roche company). The apparent molecular weight(19.46 kD) and the immuno-specificity of the fusion protein were confirmed by SDS-PAGE and Western blot, and the expressed fusion protein stayed mainly in the supernatant of the reaction mixture in soluble form. This work provides us solid basis for further study on new leptin-associated proteins.

0bjective To evaluate the heart function of patients with refractory nephmtic syndrome during a combination therapy of methylprednisolone(MP)and cyclophosphamide(CTX).Methods From January 2004 to December 2007.30 patients with refractory nephrotic syndrome.who were in hospital and received a combination therapy of MP and CTX.were recorded for their heart rate and blood pressure before and during treatment,as well as observed for the complications.ResulIs Comparing with those before treatment.patients' heart rates were much quickly and their blood pressure were remarkablv higher during the combination treatment. And there was a statistic siglnificance(P<0.05).When patients received MP treatment.complications.such as cardiopalmus and dizziness, proned to emerging in the third cycle. When they received CTX treatment, complications, just like sicchasia, vomit and cardiopalmus.appeared in the first cycle. In addition. Myelosuppression and lesion of liver function often occurred during the sequence therapy. Conclusions When patients received a combination therapy of MP and CTX,we should monitor their heart funcion SO as to promptly make propotional measure and prevent complications according to variation of heart rate and blood pressure.

China Journal of Chinese Materia Medica 2008;33(21):2439-2442
By studying the literatures on the adverse reaction of Yuxingcao injection, the clinical features of ADR were summed up, and the causes of ADR were analyzed through raw material, technology, chemical composition, compatibility and the clinical usage. The causes of ADR induced by Yuxingcao injection are complicated, maybe both due to the medicine itself and the incorrect clinical usage. Multi-measures including manufacture, techniques, clinical usage and some other cases should be taken to prevent the occurrence of adverse reaction of Yuxingcao injection.

AIMTo study bioequivalences of bambuterol and its metabolites terbutaline in 20 healthy male volunteers.
METHODSA single oral dose of domestic bambuterol capsule or imported bambuterol tablet was given according to a randomized 2-way cross-over design. The plasma bambuterol and terbutaline concentrations were determined by high performance capillary zone electrophoresis (HPCZE).
RESULTSThe pharmacokinetic parameters of the capsule and tablet of bambuterol: AUC0-1 were (72 +/- 18) and (72 +/- 13) microgram.h.L-1, Cmax were (8.1 +/- 1.8) and (9.2 +/- 2.3) microgram.L-1, Tmax were (3.6 +/- 1.3) and (3.7 +/- 1.0) h, respectively; terbutaline: AUC0-t were (129 +/- 32) and (130 +/- 34) microgram.h.L-1, Cmax were (7.8 +/- 2.2) and (8.5 +/- 2.9) microgram.L-1, Tmax were (5.4 +/- 0.8) and (5.6 +/- 1.1) h, respectively. The bioavaiability of the capsule was (100 +/- 16)% (bambuterol) and (101 +/- 13)% (terbutaline).
CONCLUSIONThe results demonstrated that the two preparations of bambuterol and terbutaline were bioequivalent by analysis of variance, with two-one sided test at 90% confidential level.

Neuroscience Bulletin 2008;24(3):160-165
OBJECTIVETo re-confirm and characterize the biophysical and pharmacological properties of endogenously expressed human acid-sensing ion channel 1a (hASIC1a) current in HEK293 cells with a modified perfusion methods.
METHODSWith cell floating method, which is separating the cultured cell from coverslip and putting the cell in front of perfusion tubing, whole cell patch clamp technique was used to record hASIC1a currents evoked by low pH external solution.
RESULTSUsing cell floating method, the amplitude of hASIC1a currents activated by pH 5.0 in HEK293 cells is twice as large as that by the conventional method where the cells remain attached to coverslip. The time to reach peak at two different recording conditions is (21+/-5) ms and (270+/-25) ms, respectively. Inactivation time constants are (496+/-23) ms and (2284+/-120) ms, respectively. The cell floating method significantly increases the amiloride potency of block on hASIC1a [IC50 is (3.4+/-1.1) micromol/L and (2.4+/- 0.9) micromol/L, respectively]. Both recording methods have similar pH activation EC50 (6.6+/-0.6, 6.6+/-0.7, respectively).
CONCLUSIONASICs channel activation requires fast exchange of extracellular solution with the different pH values. With cell floating method, the presence of hASIC1a current was re-confirmed and the biophysical and pharmacological properties of hASIC1a channel in HEK293 cells were precisely characterized. This method could be used to study all ASICs and other ligand-gated channels that require fast extracellular solution exchange.
