2.Complication and its Prevention during the Treatment of Femoral Shaft Fracture with Interlocking Intramedullary Nail

Objective: In order to popularize the use of interlocking nail, the authors reviewed 81 femoral shaft fractures in 80 patients who were treated with interlocking nail from July 1988 to October 1997. Material and Method: There are 60 males and 20 females. Forty-one fractures were of left femur and 40 of right. The average age was 33 years(range 15-79 years) . Results: The results showed that 60 cases of fractures that followed up over half a year had bony union. The complications included one bone union failure due to incorrect indication. two fractures during the operation, misinserting the proximal screw in 2, and misinserting the distal screw in 3, lengthening of the extremity in 2, valgus knee in 2, deep infection in 1, delayed infection in 1, nail broken in 3, proximal screw broken in 1, distal screw broken in 1, distal screw loosening in 2. Conclusion: 1) The indication must be correctly chosen. 2) Well-trained operative technique for interlocking nail is necessary to reduce the compli cation. 3 )The patient with old or comminuted fracture fixed with G - K nail should properly delay the time to start weight bearing.
3.Pharmacokinetic Study on Hyperoside in Beagle's Dogs

Objective To develop and validate a simple,rapid,sensitive,and reproducible HPLC method for determination of hyperoside in plasma of dogs and for the subsequent pharmacokinetic (PK) study.Methods An accurate and reproducible HPLC-UV method was developed and validated for the determination of hyperoside in plasma of dogs,using kaempferol as internal standard.The plasma samples of dogs following ig administration of hyperoside were analyzed for the detection of quercetin after enzymatic hydrolysis treatment with combined β-glucuronidase and sulphatase.The analytes were separated on a Diamonsil C18 column (250 mm × 4.6 mm,5 μm).The mobile phase consisted of methanol-buffer solution (0.1mol/L NH4Ac + 0.3 mmol/L EDTA-Na2)-acetic acid (60:40:1) and was delivered at a flow rate of 1mL/min.The UV detector was set at 370 nm and the column temperature was maintained at 35 ℃.The sample injection volume was 20 μL.Data were collected and analyzed using the ANASTAR software.PK parameters were calculated with DAS software (2.0).Results Linear relationships were validated over the range of 0.01-1μg/mL for hyperoside (r =0.9997).The intra-and inter-day precision values for all samples were within 10.0%,and the accuracies of intra-and inter-day assays were within the range of 92.4%-102.4%.The validated method was successfully used to determine the hyperoside concentration in plasma of dogs for up to 12 h,after a single ig administration (25 mg/kg).The mean PK parameters for male and female dogs were as follows:Cmax (0.18 ± 0.05) and (0.16 ± 0.05) μg/mL,AUC0-∞ (0.79 ± 0.34) and (0.86 ± 0.27) μg/(mL·h),t1/2(ka) (0.89 ± 0.41) and (0.88 ± 0.28) h,respectively.Statistical analysis on the PK of hyperoside in male and female groups showed that sex had no significant impact on the PK of hyperoside (P > 0.05).Conclusion The method is able and sufficient to be used in drug PK studies of hyperoside.
4.Rhein retards the progression of type 2 diabetic nephropathy in rats

Objective To assess the effect of rhein on diabetic nephropathy in rats with type 2 diabetes mellitus. Methods Diabetes was induced by high-lipids and high-sucrose diets (20% sucrose, 10% lard, 2.5% cholesterol, 1% cholic acid and 66.5 % regular chow, w/w) for 1 month and then intraperitoneal injection with 25 mg/kg streptozoticin(STZ) in female inbred Wistar rats. Rhein was continuously given 100 mg?kg-1?day-1 for 6 months 1 week after STZ injection in rhein-prevented group, and 1 month after STZ injection in rhein-treated group. Blood and urinary biochemistry and renal morphology were evaluated at divers times. Results Rhein ameliorated hypertriglyceridemia and hypercholesterolemia, but had few effect on the level of plasma glucose in both rhein-prevented group and rhein-treated group. Excretion of urinary protein reduced markedly and stable-state plasma glucose (SSPG) level decreased in both groups as compared with diabetic group. Examination of kidney sections from both rhein-prevented group and rhein-treated group showed that the structural lesions of glomeruli including mesangial expansion, diffuse glomerulosclersis and thickening of glomerular basement membrane, as well as artery injury such as arteriolar hyalinosis, atherosclerosis were markedly improved as compared with diabetic group. Morphometry of glomeruli from both rhein-prevented group and rhein-treated group showed that glomerular grow and mesangial hypertrophy occurred in diabetic group were ameliorated. The accumulation of fibronectin examined by immunostaining in glomeruli was reduced in both groups. Conclusion Therapeutic intervention with rhein can halt the progression of diabetic nephropathy and prevent the development of glomerulosclerosis and vascular injuries in this animal model of type 2 diabetes mellitus.

BACKGROUND: Is the inhibition of the hematopoietic stem progenitor cell (HSPC) proliferation and differentiation after human cytomegalovirus (HCMV) infection associated with abnormal expression of infected cell proliferated gene?OBJECTIVE: To observe the HOXB6-mRNA expression in the process of proliferation and differentiation of HCMV-infected HSPC into colony-forming unit granulocyte-macrophage (CFU-GM) and colony-forming unit erythroid (CFU-E).DESIGN: A controlled observation.SETTING: Laboratory for Molecular Biology, Affiliated Hospital of Luzhou Medical College, Lanzhou, Gansu Province, China.MATERIALS: All cord blood (CB) specimens were provided by the Obstetrics Department of Affiliated Hospital of Luzhon Medical College. They were collected from the umbilical vein of normal term neonates delivered spontaneously. All neonate mothers were healthy and HBS-Ag-negative. HCMV-IgM antibody revealed by routine ELUSA and HCMV-DNA checked by PCR were undetectable. Written informed consent for the laboratory measurements was obtained from each neonate mother, and the protocol was approved by the hospital's Ethics Committee. HCMV-AD169 strains were obtained from the Institute of Virology, Chinese Academy of Preventive Medicine. All-trans retinoic acid (ATRA, lot No. 20010126) was provided by Chongqing Huapont Pharm. Co., Ltd., China.METHODS: This study was performed at the Laboratory of Molecular Biology (state-level), Affiliated Hospital of Luzhou Medical College of Luzhou Medical College from April 2006 to April 2007. Cord blood mononuclear cells were separated for HSPC culture. According to different interventions, the study consisted of 4 groups. Control group: no HCMV virus solution was added and equal volume of culture medium was added instead. HCMV group: 105 PFU/mL HCMV-AD169 virus solution was added to the culture system. ATRA group: ATRA was added into the cultivation system at the final concentration of 60 μ mol/L. HCMV+ATRA group: ATRA was added into the HCMV group, and its final concentration was also 60 μ mol/L.MAIN OUTCOME MEASURES: In each group, cells were harvested on days 3,7 and 12. HOXB6 mRNA expression levels in CFU-GM and CFU-E were detected by real-time fluorescent-based quantification PCR.RESULTS: In the control group, both CFU-E and CFU-GM expressed HOXB6-mRNA. The HOXB6 mRNA expression was increased as a function of time. The HOXB6-mRNA expressed by CFU-E reached its peak level on day 12, while that expressed by CFU-GM reached its peak level on day 7. Compared to control group, the expression levels of CFU-E and CFU-GM HOXB6-mRNA genes in normal cord blood were significantly lower in the HCMV group (P<0.05)and significantly higher in the ATRA group (P<0.05) at each time point after HCMV infection. Furthermore, the expression levels were significantly higher in the ATRA+HCMV group than in the HCMV group at each time point(P<0.05-0.01).CONCLUSION: HOXB6-mRNA expression is stable and lasting in the proliferation and differentiation of HSPC into CFU-GM and CFU-E. HCMV could down regulate HOXB6 gene expression, and ATRA could up regulate HOXB6 gene expression.

8.Cytological study on rat corneal new blood vessel after alkali burn

Background Microvessels are composed of two interacting cell types: endothelial cells and pericytes. Over the past decades, studies of corneal angiogenesis have concentrated mainly on endothelial cells, while interest in pericytes has lagged behind. Objective The present study aimed to investigate the recruitment of vascular endothelial cells and pericytes in rat corneas after alkali burn. Methods Corneal alkali burn models were established in the right eyes of 36 adult SPF SD rats by putting 4 mm medicators containing a 1% 1 mol/L NaOH solution at the central corneas for 30 seconds, and 3 matched normal rats were used as controls. Corneas were excised 1,2,3,5,7 and 10 days after surgery. Frozen sections that parallel with the corneoscleral limbus were constructed. Double immunofluorescence staining was used to observe the dynamic expression of CD31 and α-smooth muscle actin (α-SMA) in corneal tissue for the evaluation of the number of endothelial cells and pericytes. The pericyte coverage index (PCI) was calculated to quantify the recruitment of pericytes to neovascular sites. The use of experimental animals followed the Statement of Association for Research in Vision and Ophthalmology. Results CD31 was expressed in the superficial stromal layer of the cornea on the 1 st day, showing the presence of red fluoresence. The positive cell number for CD31 was gradually increased with the passage of time and proceeded into the deep stromal layer from days 2 through 5 but decreased after that. However,α-SMA was positively expressed on the 2nd day in the cornea after alkali burn with the presence of green fluorescence. The positive cell number for α-SMA was less than those of CD31 throughout the experimental period. The PCI was 0, 16.07%, 11.95%, 43.84%, 73.97% and 86. 21% , respectively, 1,2,3,5,7 and 10 days after surgery. Conclusion Pericytes recruitment to corneal new vessels may play a key role in the stabilization and maturation of angiogeneis.
9.Study on the correlation between monocular mild myopic juvenile myopia degree and dominant eye

?AlM: To study the correlation between monocular mild myopic juvenile myopia degree and dominant eye.
? METHODS: Totally 158 patients with juvenile monocularly mild myopia in our hospital from December 2012 to December 2013 were retrospectively analyzed, and cylindrical mirror astigmatism was used for spherical equivalent conversion. On the basis of the myopic degree, they were divided into three groups, 30 cases in group A (-0. 25~-0. 75D), 92 cases in B group (-1. 0~-2. 0D), 36 cases in group C (-2. 25 ~ -3. 0D). The card hole method was selected to measure dominant eye for subjectsat nearly 33cm and 5m. After glasses correction of ametropia, the far and near dominant eyes were received measurement again.
?RESULTS:The monocular mild myopia of dominant eye was compared with the non-dominant eye adjustment function, and there was no significant difference ( P>0. 05 ) . The dominant eye, non - dominant eye mean diopter and other correlations of eyes in the three groups were compared, and there was no significant difference (P>0. 05). The dominant eye of three groups at 5m was compared, and there was significant difference (P<0. 05). The dominant eye of three groups at 33cm was compared, and there was significant difference (P<0. 05). Dominant eye at different visual distance was compared, and there was significant difference (P<0. 05). Monocular uncorrected eye was compared with dominant eye mild afte myopia glassesr, and there was significant difference (P< 0. 05).
?CONCLUSlON: The formation of myopic anisometropia is related to the degree of clearly seeing objects, although glasses correction can improve the clarity and visual, but affect the choice of the dominant eye, because the myopic anisometropia appears most early in the dominant eye, therefore optician correction stage, which should be taken into consideration, and avoid the severity of the adolescent myopia.
10.Angiogenesis induced by micropocket assay on rat cornea

AIM: To explore the skills and characteristics of corneal neovascular model in rat induced by micropocket assay. ·METHODS: Nine eyes of nine Sprague-Dawley rats were studied. Pellets made of vascular endothelial growth factor (VEGF), poly-2-hydroxylethylmethacrylate and sucralfate were implanted into the corneal stroma nocloser than 1mm from the limbus. Biomicroscopic features of corneal neovascular were observed on 1,3, 5, 7th day after the implantation. ·RESULTS: On day 1 after operation, the limbal vessels were dilated, with no angiogenesis appeared. On day 3, angiogenesis began to invade peri-cornea with a brush shape, the area of CNV was (2.23±0.59) mm2. On day 5, new vessels reached the lower margin of pellet densely, the area of CNVwas (6.81±1.35)mm2. On day 7, new vessels continued to elongate, parts of them extended as loops toward the pellet, the area of CNV was (8.92± 1.79)mm2. Neither hyphema or other complications occurred.·CONCLUSION: Corneal neovascular induced by micropocket assay in rat grows steadily, with no complication, and is suitable for quantitative researches.
