2.Mechanism of Study on Gastric Mucosal Lesion Restoration in Rats Treated by Electroacupuncture

Objective Through comparing the similarities and differences between respairing effect on rat’s gastric mucosal lesion by electroacupure (EA) at Meridian of Foot Yangming and Meridian of Foot Shaoyang, to explore the specificity of the correlation between Meridian of Foot and Stomach and the mechanism of the healing effect by EA at meridian of Foot Yangming for curing the gastric lesion. Methods Forty rats were randomly divided into 4 groups:blank group, model group, stomach tunnel group, gallbladder tunnel group. All rats (except normal group) were made model by water restraint stress (WRS). After corresponding experimental process, epidermal growth factor (EGF) and transforming growth factor alpha (TGF-?) of plasma, gastric mucous tissue concentration was detected with radioimmunoassay, and the expression of EGFR mRNA of above tissue was detected by the reverse transcriptase-polymease chain reaction (RT-PCR) method. Results The gastric mucosal lesion index of Foot Yangming group decreased significantly in comparison with that of other groups, while EGF, TGF-? of gastric mucous tissue concentration level of Foot Yangming group was obviously higher than that of model group and gallbladder tunnel group. EA oould up-regulate the expression gastric mucosal EGFRmRNA, and there was a more significant difference in the group by EA at Meridian of Foot Yangming than that in the model group and gallbladder tunnel group. Conclusion The curative effect of EA at points of Meridian of Foot Yangming on the gastric mucosa may be realized by the synthesis, secretion and release of the related EGF family factor, such as EGF, TGF-? and so on. Such special regulation of electroacupuncture on gastric mucosal tissue was related to the gene expression of EGFRmRNA, and that may be further mechanism of the curative effect of EA at points of Meridean of Foot-Yangming on gastric mucosal lesion. There existed corresponding correlation between Meridian of Foot Yangming and Stomach.
3.Changes in proliferation and apoptosis of macrophages in the development of pulmonary fibrosis of rats

AIM: To observe the changes in proliferation and apoptosis of macrophages in the development of pulmonary fibrosis in rats. METHODS: The number of macrophages, apoptotic cells, the proliferation index (PI) and MTT activity of macrophages were assayed on the day 14 and the day 30 after intratracheal adminstration BLMA_5 in rats.RESULTS: (1) The number of alveolar macrophages was increased in BLMA_5 14 d group and in BLMA_5 30 d group, compared with sham 14 d group and sham 30 d group, respectively. The number of macrophages in BLMA_5 14 d group was higher than that in BLMA_5 30 d group. (2) The PI of macrophages increased in BLMA_5 14 d group, and decreased in BLMA_5 30 d group. (3) The number of apoptotic cells increased both in BLMA_5 14 d group and BLMA_5 30 d group.The number of apoptotic cells in BLMA_5 14 d group was lower than that in BLMA_5 30 d group. (4) The MTT activity of macrophages was higher in BLMA_5 14 d group and in BLMA_5 30 d group than that in sham 14 d group and sham 30 d group, respectively. CONCLUSIONS: The ability of proliferation increased at first, and then decreased, but the apoptosis of macrophages increased all the time, in the development of pulmonary fibrosis. This might be partly contributed to the changes of the number and function of macrophages in lung.
4.Clinical study of Ludloff osteotomy and artificial metatarsophalangeal arthroplasty in the treatment for severe hallux valgus

Objective To analyze the clinical outcomes of Ludloff osteotomy and artificial metatarsophalangeal arthroplasty in the patients with severe hallux valgus and metatarsophalangeal joint osteoarthritis.Methods From Apr.2013 to Aug.2015,120 feet with severe hallux valgus were treated by operation in Beijing Friendship Hospital,Capital Medical University.Before operation,measured some angles,such as hallux valgus angle,intermetatarsal angle,proximal articular surficial intrinsic angle and so on in the two groups.And evaluated the patients condition by Maryland scale at the same time.Through the date,we make sure that there was no significant difference between the two groups before operation.According to the order of hospitalization,all patients were randomly divided into experimental group (n =60) and control group (n =60).The cases of experimental group were treated by Ludloff osteotomy and artificial joint replacement.The cases of control group were treated by Ludloff osteotomy and Akin osteotomy.We used the visual analogue scale,the core quality of life scale-36 and Maryland scale to record the date,such as pain,the range of motion and appearance of the metatarsophalangeal joint and the ability of daily life and sports before and after operation.Compared above categorical variables using paired T-test between before and after operation and using T-test between the two groups.Compared the rate of operative complications by Fisher's exact test between the two groups.Results Through the Maryland scale,the total excellent rate of the experimental group (91.7%) was higher than the total excellent rate of the control group (75.0%),with statistically significant (P < 0.05).The operative complication rate of experimental group (1.7%) was lower than the control group (8.3%),but without statistically significant (P >0.05).The scores of quality of life of the experimental group after treatment were significantly higher than those of the control group,with statistically significant (P < 0.05).The visual analogue scale pain score (1.61 ± 0.12) of the experimental group was significantly lower than the control group (3.68 ± 0.58),with statistically significant (P < 0.05).Conclusion The clinical outcomes of Ludloff osteotomy and artificial metatarsophalangeal arthroplasty in patients with severe hallux valgus and metatarsophalangeal joint osteoarthritis is better,high safety,less trauma and faster recovery.It is worthy of promotion and use.
5.Effect of Sodium Valproate on Weight,Body Mass Index,Serum Glucose,Serum Insulin in Children with Epilepsy

Objective To explore the changes of weight,body mass index(MBI),serum glucose,serum insulin in sodium valproate(VPA)-treated children with epilepsy.Methods The study was carried out in 30 epileptic children treated with VPA for 3 months.Weight,height,serum glucose and insulin levels were conducted before and after VPA treatment.Serum insulin levels were detected by radioimmunoassay.Results After 3 months' treatment,body weight [(15.68?3.82)kg vs(19.64?4.75)kg,t=3.56 P0.05].Conclusions VPA therapy can make insuline weight,BMI,serum insulin increased and may lead to insulin resistance.Insulin resistance may be one of the reasons for weight gain in VPA-treated patients.
6.Effects of Morroniside on Rats Neurons in Focal Cerebral Ischemia/Reperfusion Injury

ObjectiveTo investigate the effects of morroniside on super oxide dismutase (SOD) and neurons in rats cortex with focal cerebral ischemia/reperfusion. MethodsThe animal model was induced by middle cerebral artery occlusion with suture embolus, cerebral ischemia 30 min and reperfusion 3 d or 7 d. Vitamin E for the positive control. The content of SOD was detected with spectrophotometry and the nerve cells was observed with immunohistochemistry. ResultsCompared with model group, morroniside (270 mg/kg)increased the activity of SOD and the number of neurons (30 mg/kg, 90 mg/kg, 270 mg/kg) significantly. ConclusionMorroniside may have neuroprotective effect and increasing the activity of SOD in rats cortex.
7.Effects of Morroniside on Apoptosis in Rats with Focal Cerebral Ischemia/Reperfusion Injury

Objective To investigate the effects of morroniside on glutathione (GSH) and Caspase-3 in rats with focal cerebral ischemia/reperfusion. Methods The animal model was induced by middle cerebral artery occlusion with suture embolus with 30 min for cerebral ischemia and 7 d for reperfusion. The content of GSH was detected with spectrophotometry and Caspase-3 expression was observed by Western blot.Results Compared with model group, the GSH increased and Caspase-3 expression reduced significantly at 270 mg/kg of morroniside.Conclusion Morroniside may have neuroprotective effect by increasing GSH in rats cortex and reduce the apoptosis in focal cerebral ischemia/reperfusion injury.
8.Preliminary study of odor change mechanism in Crataegi fructus stir-fried process based on correlation analysis.

In order to investigate the mechanism, the correlation between the odor change in Crataegi Fructus stir-fried process and 5-HMF were studied. Required samples were retrieved from Crataegi Fructus stir-fried process. Statistical quality control (SQC) was used to analyze the response values acquired by the electronic nose. At the same time, the content of 5-HMF was detected by high performance liquid chromatography (HPLC). Correlation analysis was used to analyze the relationship between the above two. Experimental results showed that SQC model established by response values of all samples could show the change law of odor in Crataegi Fructus stir-fried process and changes of 5-HMF content was dropped after the first increase. Correlation analysis showed that the odor change in Crataegi Fructus stir-fried process and 5-HMF were significantly correlated (P < 0.05). Sugar degradation reaction and the Maillard reaction may be one of the mechanisms of the odor change in Crataegi Fructus stir-fried process.

AIM: To study the role of high level of endogenous nitric oxide (NO) in apoptosis of alveolar epithelial cells in the development of pulmonary fibrosis in rats. METHODS: The content of nitrite/nitrate (NO 2-/NO 3-) in out-flowing pulmonary blood (OPB) was assayed by nitric acid reduction method. The apoptosis of alveolar epithelial cells was observed by TdT-mediated dUTP nick-end labeling (TUNEL) and electron microscopy, respectively. 10.Effect of cerebral ischemic preconditioning on NOS activity and NO content in the CA1 region of the hippocampus in rats

AIM: To explore the role of NO in the induction of brain ischemic tolerance (BIT) by observing changes of NOS activity and NO_2-/NO_3- content following a transient cerebral ischemia. METHODS: The rat 4-vessel occluding brain ischemic model was used. 140 male Wistar rats were divided into sham, cerebral ischemic preconditioning (CIP), ischemic insult and CIP+ischemic insult groups. An occlusion of the 4 vessels for 3 min was normally used as CIP, and a relative long one for 10 min was used as ischemic insult. When CIP was followed by ischemic insult, the interval between them was 3 d. The CA1 region of the hippocampus of rats was dissected out at 0 h, 2 h, 16 h, 24 h, 36 h, 72 h and 7 d after the last time of ischemia to assay its NOS activity and NO_2-/NO_3- content. RESULTS: The NOS activity and NO_2-/NO_3- content began to increase at 16 h, peaked at 24 h and decreased to basal level at 36 h of reperfusion after CIP. The duration of the up-regulation of NOS activity and NO_2-/NO_3- content was much shorter than that of BIT, which usually takes place 1-7 d after CIP.
