Human adenovirus (HAdV) is one of the most important pathogens in infants and young children with acute respiratory infections and other diseases. This article reviews the literature on HAdV, including its molecular biological characteristics, detection and typing, and pathogenic mechanism, the clinical features and epidemiological characteristics of HAdV-related diseases, and the prevention and control of HAdV infections. So far, 67 types of HAdV have been identified, including recombinant variants discovered in recent years. The major epidemic strains that cause acute respiratory infections are HAdV-3 and HAdV-7, both of which belong to the subgroup B. HAdV often leads to acute respiratory infections, but it also causes diseases of other systems. HAdV-related diseases have similar clinical manifestations as those caused by other respiratory viruses, but often accompanied by gastrointestinal symptoms. The pathogenic mechanism of HAdV remains unclear, especially for the new recombinant variants, due to few studies on their association with diseases. Because there are no prospective, large randomized controlled trials of HAdV infections, the treatment of HAdV infections is controversial. Vaccine is the most effective measure to reduce respiratory HAdV infections, but it is still not commercially available.

Adenoviruses are double stranded DNA viruses that cause an array of diseases. More than 60 types of human adenovirus have been reported so far. The discovery and division of human adenovirus types 52-67 are based on the genomic sequencing and bioinformatic analysisy and different from the pre-existing 51 serotypes identified by traditional serological methods in composition and pathogenicity. The majority of new types of human adenovirus are homologous recombination within the same subgenus, and as a result, certain new serotypes acquire different pathogenicity. Recombination is a common evolutionary way for human adenovirus, however, the mechanism of recombination and potenital danger to human remain unknown. As new types of human adenovirus have been receiving increasing attention, in this review, the latest progress in new types of human adenovirus is summarized.

As a single-center retrospective study, we analyzed the results of rotavirus and human adenovirus antigens in stool samples with colloidal gold immunochromatography method in children with acute gastroenteritis under the age of five who were treated in our hospital from 2019 to 2022. After excluding nonconforming cases and duplicate cases, 2 896 cases were included, of which 559 cases were detected with at least one viral antigen. According to the test results, they were divided into RV positive group, HAdV positive group and RV & HAdV double positive group. The gender, age, seasonal distribution, clinical symptoms and related laboratory tests were compared and analyzed with χ² test, analysis of variance and nonparametric test. Among the single samples from 2 896 children, the positive rate of RV antigen was 6.21% (180/2 896), the positive rate of HAdV antigen was 10.91% (316/2 896), and the double positive rate of RV & HAdV was 2.18% (63/2 896). The positive rate of HAdV antigen in 2021 was 16.11%, a significant increase compared with 6.20% in 2020. RV infection has obvious seasonality, and spring and winter are the seasons with high incidence of infection (χ²=74.018, P<0.001), while HAdV infection has no obvious seasonality (χ²=2.110, P=0.550), showing sporadic infection throughout the year. The proportions of fever and vomiting symptoms in children with RV infection were significantly higher than those in the HAdV infection group (χ²=40.401, P<0.001; χ²=32.593, P<0.001), but the positive rate of white blood cells in the stool was significantly lower than that in the HAdV infection group (χ²=13.741,P<0.01). In summary, paying attention to the epidemiological changes of RV and HAdV is of great significance for clinical diagnosis and treatment and disease prevention and control.

Obesity is connected with numerous diseases, such as type 2 diabetes, atherosclerosis, cancer, and nervous system dysfunctions. Obesity is affected by genetic, environmental, and cultural factors. However, numerous studies indicate that several pathogens might cause obesity. This review discusses recent data and the characteristics of pathogens that are implicated in obesity. In particular, human adenovirus 36 (Ad36) is the most clearly implicated virus in human obesity. It was recently shown that obese groups from the USA, Korea, and Italy have a higher prevalence of serum antibodies against Ad36. The mechanisms of Ad36-induced obesity remain unclear. However, glucose uptake and inflammation are possible mechanisms of Ad36-induced obesity. Overall, this new understanding of causes of obesity has developed into the concept of 'infectobesity' and the possibility of developing a 'vaccine' or 'therapeutic agents' for obesity.

Human adenovirus type 5 (hAd5) vectors have been demonstrated to be useful vehicles for gene expressions in animals. However, it has not been reported whether hAd5 transduction might be hampered in the sera of livestock animals in Republic of Korea. We collected 205 samples of livestock animals, such as pig (n=84), cattle (n=84), and goat (n=37) in Korea. The neutralizing antibody (NAb) titers to hAd5 virus were less than 15 in most of samples. Only 8% of goat samples had a NAb titer of 15 or 30. Thus, we showed that hAd5 virus was not neutralized in sera from cattle, pig, and goat, and suggest that the hAd5 vector could be used for the effective delivery of vaccines or proteins in livestock animals in the field.

OBJECTIVEAdvances in tumor biology and clinical trials indicate that p53 transfer is an alternative therapy for head and neck squamous cell carcinoma. The aim of this study is to evaluate the biologic activity of multiple intraepithelial injections of Ad-p53 in patients with dysplastic oral leukoplakia, which is the most common premalignant lesion of the oral squamous cell carcinoma.

METHODSFrom 2006 to 2007, 18 Chinese patients clinically and histopathologically diagnosed as dysplastic oral leukoplakia were recruited for this study. On a 15-day cycle, intraepithelial injections of Ad-p53 were administered once every three days at dose levels dependent upon lesion size/dose escalation sequence (1 x 10(8) vp). 24-48 h after the last injection, incisional biopsy were performed, and immunohistochemistry was used to examine the protein expression of P53 and P21(CIP/WAF).

RESULTSIn the postreatment patients, P53 protein and P21(CIP/WAF) protein expression were significantly enhanced (100%, 89.9%, respectively) and statistical analysis revealed the expression of P53 protein had a positive correlation with that of P21(CIP/WAF) protein (r=0.598, P<0.01).

CONCLUSIONIntraepithelial injections of gendicine is biologically active in patients with dysplastic oral leukoplakia. It may be a promising treatment for oral leukoplakia.

OBJECTIVETo construct a recombinant adenovirus vector expressing hCD40L gene and explore it in the use of anti-tumor gene therapy.

METHODS1,900 bp gene fragment was obtained form plasmid pORF-hCD40L by Xho I/Swa I cutting and then cloned directionally into the pShuttle plasmid, finally, the resultant plasmid was digested by restriction endonnuclease PmeI and subsequently cotransformtion into BJ5183 cells with the adenoviral backbone pAdEasy-1 to obtain the homologous recombinant and then the recombinant was packaged in the 293 cells. Some methods such as PCR and endonulease digestion were employed to identify the recombinant adenovirus.

RESULTSThe evidences of endonulease digestion and PCR analysis confirmed that recombinant hCD40L gene was correctly inserted into adenovirus vector.

CONCLUSIONThe adenoviral vector which expressed hCD40L gene was constructed. It provides an experimental basis for studies on it expression in the mammalian cells and in tumor gene therapy.

OBJECTIVETo study the prevalence, clinic features and epidemiologic characteristics of human adenovirus diarrhea in Nanjing.

METHODS730 stool specimens were collected from children with diarrhea in Nanjing Children's Hospital of Nanjing Medical University from June 2009 to June 2011. Polymerase chain reaction (PCR) was employed to detect human adenovirus. The total positive PCR products were typed by nest-PCR or multiple PCR.

RESULTS21 samples (21/730) were positive for human adenovirus of all 730 samples from June 2009 to June 2011 and enteric HAdV-41 is the predominant stain.

CONCLUSIONEnteric HAdV-41 and non-enteric adenovirus were the major etiological agents of viral diarrhea among infants and children in Nanjing from 2009 to 2011. We should take the long-term systematic surveillance seriously.
