1.Isolation and Adherent Culture of Neural Stem Cells from Mouse Embryo

Objective To develop a modified method of adherent culturing NSCs to replace the traditional suspension culturing method.Methods Neural stem cells(NSCs)were isolated from brain of fetal mouse at 15.5-day fetal age.In culture bottle which had been pre-coated with poly-L-ornithine hydrochloride/fibronectin(PO/FN),the cells were adherently cultured and passaged.The cell morphology was observed under inverted microscope.NSCs and their differentiated cells were identified by immunofluorescence.Results NSCs obtained in this study were successfully adherently cultured and expressed specific markers.Conclusion NSCs are successfully adherently cultured in this study.As compared with the traditional suspension culturing method,adherent culturing is simpler and has lower wastage of passage.It is easy to observe cell morphology and differentiation by this method.It is also helpful to culturing and storage of NSCs.
2.Evaluation of Intervention on AIDS High-risk Sexual Behavior among Young Students in Jilin MSM Population

Objective To evaluate effect of intervention on acquired immune deficiency syndrome(AIDS) high-risk sexual behavior of males who have sex with males(MSM)population in Jilin,and to provide a basis for establishing apropriate intervention strategy for this population.Methods A cross-sectional survey was carried out on the MSM population of young students with the method of classification.Comprehensive intervention strategy included health education via new media,behavioral intervention,voluntary conseling and testing,peer education,condom promotion and standardized services for clinic of sexually transmitted diseases(STDs)and other comprehensive interventions.Before and after intervention,300 MSM were enrolled,and their demographic characteristics,knowledge of AIDS prevention and control,high risk behaviors and intervention measures were investigated and analyzed.Results Before and after the intervention,the awareness rate of AIDS related knowledge was 67.67% and 94.33%,respectively,the condom usage rate was 23.57% and 11.79% in the most recent and the last 6 months before the intervention,and it was 76.21% and 62.10 % after the intervention,respectively.The proportion for those who used condom every time in heterosexual sex behavior was increased from 23.53% (before intervention) to 69.60 % (after intervention),and the condom usage rate in the last 6 months was increased from 15.44% to 66.40%.Proportion of the MSM who had STDs related symptoms was decreased from 21.33% to 7.67% after the intervention.Conclusion After the comprehensive intervention,the awareness rate of AIDS related knowledge among MSM in Jilin is significantly increased,the incidence of highrisk sexual behavior is decreased and the condom usage rate is improved.Intervention by use of new media is low-cost,easy to operate,and can be used as a good supplementary strategy for traditional behavioral intervention.
3.Effect of Treatment of a Non-surgical Spinal Decompression System on Neck Muscle Surface Electromyography of Patients with Cervical Spondylotic Radiculopathy

Objective To explore effect of treatment of a non-surgical spinal decompression system(SDS) on neck muscle surface electromyography(EMG)of patients with cervical spondylotic radiculopathy(CSR).Methods Sixty patients with CSR in Department of Rehabilitation Medicine at The First Affiliated Hospital of Sun Yat-sen University in China between February 2014 and February 2016 were recruited and randomly divided into SDS group and control group,with 30 patients for each group.The SDS group and control group were treated with the SDS and general traction system for cervical traction respectively.Surface EMG telemeter was used to measure the surface EMG signals of the affected side cervical erector spinae muscle and sternocleidomastoid with the score of averaged EMG value(AEMG)and the median frequency(MF)as surface electromyogram signal evaluation indexes,and VAS and NDI as therapeutic effect evaluation indexes after a course of treatment.The data before and after the treatment were statistiacaly analyzed.Results During the first traction with tradtional traction or SDS,AEMG values of the cervical paraspinal muscle and sternocleidomastoi were significantly decreased and MF values significantly increased as compared with those before the traction (all P<0.05).After a course of treatment,AEMG values of affected side cervical erector spinae muscle and sternocleidomastoi of SDS group were significantly decreased as compared with those before the treatment,and they were significantly lower in SDS group than in ordinary group (all P<0.05);and MF values of affected side cervical erector spinae muscle and sternocleidomastoi in SDS group were significantly increased as compared with those before the treatment,and they were significantly higher in the SDS group than in the ordinary group (all P<0.05).VAS and NDI scores of the two groups after a course of treatment were significantly lower than those before the treatment (P<0.05),and VAS and NDI scores were significantly lower in the SDS group than in the ordinary group (P<0.05).Conclusion SDS traction is superior to ordinary traction in relieving pain,improving function,relaxing neck muscles and alleviating muscle fatigue.
4.Expression of PP2CE in Human Hepatocellular Carcinoma Tissues and the Clinical Significance

Objective To investigate expression of PP2CE in human hepatocellular carcinoma(HCC)tissues and the clinical pathological significance,and test its sub-cellular localization in liver cells and HCC cells.Methods A total of 50 samples of HCC tissues and their para-carcinoma tissues were collected in Tongji Hospital(Wuhan).PP2CE mRNA and protein levels were detected in HCC tissues and their para-carcinoma tissues by qRT-PCR and immunohistochemistry method.Immunofluorescent assay was performed to test the sub-cellular localization of PP2CE in the cells.Results PP2CE mRNA and protein expression levels were significantly decreased in HCC tissues as compared with those in para-carcinoma tissues(both P<0.05).Immunohistochemistry analysis found that PP2CE expression was associated with tumor size,differentiated degree,portal vein invasion and clinical stage(both P<0.05).Immunofluorescent assay suggested that PP2CE was mainly located in the cytoplasm and perinuclear area.Conclusion PP2CE is reduced in HCC and may be implicated in the development and progression of HCC.
5.Correlation between Cholecystectomy and Colorectal Adenoma

Objective To explore the correlation between cholecystectomy and colorectal adenoma.Methods A total of 981 patients who underwent colonoscopy between January 2007 to December 2012 at Wuhan Central Hospital were enrolled in the study according to the inclusion criteria.The patients were divided into cholecystectomy group (n=216) and control group(n=765) in terms of cholecystectomy or not.The incidence of colorectal adenoma ,gender ,age ,location and pathological type of ade-noma were analyzed in the two groups.Results Significant differences were noted in the incidence and location of colorectal ad-enoma ,and gender between the two groups (P< 0.05).The incidence of colorectal adenoma was higher in cholecystectomy group than in control group ;the number of female patients with colorectal adenoma was greater than that of male patients ;the incidence of proximal colorectal adenoma was higher than that of distal adenoma.But the average age of patients with adenoma was not significantly associated with the pathological types of adenoma in both groups (P>0.05).Conclusion Cholecystecto-my is closely associated with the occurrence of colorectal adenoma.Screening for colorectal adenoma should be considered in the patients with gall bladder removal.
6.Diffusion Tensor Imaging of Changes of White Matter in Patients with Motor Neuron Disease

Objective To quantitatively assess the changes of white matter in 15 patients with motor neuron disease by dif-fusion tensor imaging(DTI)and to explore the possible pathogenesis.Methods Fifteen patients with a diagnosis of motor neu-ron disease and sixteen age- and sex-matched controls without disorders affecting the central nervous system received 3.0T DTI;whole-brain white matter damage was examined using tract-based spatial statistics(TBSS),and fractional anisotropy(FA), mean diffusivity(MD),λ1,λ2,λ3were extracted from the damage region.The data were analyzed by t-test to disclose the differ-ences of white matter between patients and controls.Results As compared with the control group,FA was significantly de-creased(P=0.034)in the left corticospinal tract(CST),while λ2(P=0.124)and λ3(P=0.064)had a trend to increase,but FA(P=0.050)in the right CST had a trend to decrease in the experimental group.However,λ1and MD were not significantly different between the two groups.Conclusion DTI can quantitatively evaluate CST degeneration in patients with motor neuron disease,which may be caused by demyelination.
7.Comparison of PET/CT vs.EBUS-TBNA in Evaluating Lymph Node Metastasis in NSCLC

Objective To compare positron emission tomography/computed tomography(PET/CT)vs.endobronchial ultra-sound-guided transbronchial needle aspiration(EBUS-TBNA)in evaluating lymph node metastasis in NSCLC.Methods We col-lected 79 NSCLC patients with enlarged mediastinal lymph nodes(diameter >1 cm by CT).The diagnostic values of PET/CT scanning and EBUS-TBNA for mediastinal staging were evaluated.Subgroup analysis according to histologic type was per-formed.Results There were 22 patients of N1 stage and 28 patients of N2 in these 79 cases.In the N1 patients,PET/CT's sen-sitivity and specificity was 59.1% and 75.4%,respectively.EBUS-TBNA's sensitivity and specificity was 86.4% and 100.0%,respectively.In the N2 patients,PET/CT's sensitivity and specificity was 67.9% and 76.5% and EBUS-TBNA's sensitivity and specificity was 89.3% and 100.0%.Conclusion EBUS-TBNA is more accurate than PET/CT in evaluating the metastatic condition of patients,and EBUS-TBNA can also benefit PET/CT(-)patients with adenocarcinoma.
8.Protective Effect of Probucol on Rats with Renal Ischemia-reperfusion Injury and the Mechanism

Objective The aim of the study was to investigate the protective effect of probucol on oxidative stress injury in rats with ischemia-reperfusion injury(IRI)and its mechanism. Methods Thirty male Sprague-Dawley rats were randomly di-vided into sham operation group(S group),ischemia-reperfusion group(IR group),and probucol treatment group(probucol+IR group,P+IR group).Rats in the S and IR groups were fed with warm water every day,and rats in P+IR group were treated with probucol.After 1 week,rat model of renal ischemia and reperfusion was established.Right kidneys of the rats were re-moved.In IR and P+ IR groups,the left renal artery was clamped with a non-invasive artery clamp,and after 30 minutes the blood vessels restored patency by loosening the clamp.After reperfusion,the S and IR groups were perfused with warm water every day,and the P+IR group was treated with probucol for 1 week.After 1 week,all the rats were sacrificed and the blood and kidney tissue specimens were taken.The blood biochemical indexes of the rats were measured,and the pathological changes of the kidneys were observed.Results The levels of blood urea nitrogen(BUN),serum creatinine(SCr)and blood lipids in P+ IR group were significantly lower than those in IR group(all P<0.05);in P+IR group,the level of superoxide dismutase(SOD)in serum and renal tissue was increased significantly(all P<0.05),meanwhile,the levels of malondialdehyde(MDA)were significantly decreased (P<0.05).In P+IR group,the pathological damage of kidney tissue was reduced,and renal index was significantly improved(P<0.05).Conclusion Probucol can increase the activity of SOD in blood and kidney of rats with IRI,improve renal function,reduce the level of blood lipids,reduce the pathological injury of renal tissue,and play a role in renal protection.
9.Experiment Analysis of Antineutrophil Cytoplasmic Antibodies(ANCA)-associated Vasculitis Combined with Pulmonary Fibrosis

Objective To explore the clinical features of antineutrophil cytoplasmic antibodies-associated vasculitis com-bined with pulmonary fibrosis and the relationship of laboratory results.Methods Totally,33 patients with antineutrophil cyto-plasmic antibodies-associated vasculitis were divided into pulmonary fibrosis group(group A)and non-pulmonary fibrosis group (group B).Fourteen patients with connective tissue diseases-related pulmonary fibrosis served as disease control group,and 12 healthy people as normal control group.The levels of ANCA,plasma creatinine,high-sensitivity C-reactive protein,fibrinogen and D-dimer were detected.Results The creatinine level was significantly higher in group B than in disease control group and normal control group.The creatinine level was higher in group A than in disease control group and normal control group,with-out significant differences.The levels of FIB,D-D,and hsCRP were significantly higher in the group A,group B and disease con-trol group(all P< 0.05).In the group A,MPO expression was positively associated with FIB concentration(r= 0.64,P<0.05).Conclusion In pulmonary fibrosis disease,early detection of ANCA has clinical value in the diagnosis of AASV.And the higher concentration of M PO may hint thrombus formation.
10.Up-regulation of TRPC3 in the Cortex of Rats with Kainic Acid-induced Seizures

Objective To observe the expression change of TRPC3 in cortex of rats with Kainic acid(KA)-induced sei-zures,and to explore the role of TRPC3 in epileptogenesis.Methods Fifty-four adult Sprague Dawley(SD)rats were randomly divided into two groups(18 for control group,36 for epilepsy group).The behavior of rats was observed and recorded.The epi-lepsy group was treated with KA(2 μg/kg,about 7 μL,lateral intracerebroventricular injection),and control group was treated with isotonic Nachloride(equivalent volume with KA,lateral intracerebroventricular injection).For epilepsy group,samples were taken 48 and 72 h after seizure burst.For control group,samples were taken 48 h after isotonic Nachloride injection.The mRNA level of TRPC3 in cortex was detected and recorded by RT-PCR,and the protein level of TRPC3 in cortex was measured by Western blotting,and immunohistochemistry was also used to display the TRPC3 expression in cortex.Results The control group showed no seizure activity,and the epilepsy group showed classical seizure activity(Ⅳ-Ⅴ level)about 5 min after injection and last for several hours.The mRNA and protein levels of TRPC3 in epilepsy group were both higher than those in control group with obvious distinction(P< 0.05).Conclusion The TRPC3 level increases in cortex of rats with KA-induced sei-zures.These results point out that TRPC3 is possibly involved in epilepsy etiopathogenesis and development.
