1.Detection of Mesentery Circumferential Margin Invasion of Middle and Low Rectal Cancer with Large Slices and CK20

Objective To study the rules of circumferential margin invasion(CMI)in middle and(or)lower rectal cancers by pathological large slices stained with HE and labeled with CK20,and provide a pathological basis for clinical therapy of rectal cancer.Methods Forty-one patients with middle or low rectal cancer were randomly selected in 8 months.The surgically resected specimens were detected after they were made into the pathological large slices stained with HE and labeled with CK20.Results The mesentery CMI positive rate in the pathological slices with HE staining and CK20 labeling was 21.95%,and 29.27% respectively.The mesentery CMI positive rate in moderately-and well-differentiated group was lower than in poorly-differentiated group(P<0.05).Moreover,the mesentery CMI rate was higher in the specimens with lower edge less than 5 cm away from the dentate line than that in those with lower edge more than 5 cm away from the dentate line(P<0.05).There was no significant correlation between CMI with the factors such as gender,age,pathological general types,tumor infiltration depth,lymph node metastasis,and surgical procedures(all P>0.05).Conclusion There were risk factors for the CMI such as low location,poor differentiation and so on.The CMI patients should be treated with standardized adjunctive therapy after operation.
2.Completely Thoracoscopic Resection of Benign Neurogenic Tumors on Apical Chest

Objective To discuss the feasibility,safety,technical points and clinical effects of completely thoracoscopic resection of benign neurogenic tumors on apical chest.Methods From January 2004 to June 2009,11 patients underwent surgical resection of benign neurogenic tumours on apical chest.A complete thoracoscopy was used in 5 cases,and the remaining 6 cases received traditional open thoracotomy.By analysis on the clinical symptoms,tumor types,complications,operative time,blood loss and drainage time after operation,the advantages and disadvantages of complete thoracoscopy were compared to traditional open thoracotomy for resection of benign neurogenic tumors on apical chest.Results There was one patient in each group that suffered from light transient Horner's syndrome,who recovered spontaneously.The group of complete thoracoscopy was superior to the group of traditional open thoracotomy in operative time,blood loss during the operation,drainage time and postoperative hospital stay.Conclusion For benign neurogenic tumors on apical chest,a resection with complete thoracoscopy is as safe and effective as the traditional open thoracotomy,and the former is characterized by less operative trauma and quicker recovery.
3.Combined Use of Traditional Chinese and Western Medicine for Chronic Pelvic Inflammatory Diseases: A Meta-analysis

Objective To evaluate the clinical effectiveness of combined use of traditional Chinese and Western medicine for chronic pelvic inflammatory diseases.Methods By searching in the major databases of CNKI,ISI Web of Knowledge,VIP Information and Pubmed,we collected data of randomized controlled trials pertaining to combined use of traditional Chinese and Western medicine in the treatment of chronic pelvic inflammatory diseases.Results Twenty-five randomized controlled trials involving totally 3081 patients were collected according to the inclusion criteria,and meta-analysis of the data indicated that combined use of traditional Chinese and Western medicine can be of great value in the management of chronic pelvic inflammatory diseases in comparison with exclusive use of Western medicine [combined odds ratio(OR)was 3.20 with 95% confidence interval(CI)(2.74,3.73),Z=14.85,P<0.01].Conclusion The clinical evidence derived from the analysis suggests that the combination of traditional Chinese and Western medicine for chronic pelvic inflammatory diseases can be effective with good security.
4.Enhancement of Drug Sensitivity to 5-FU in Human Colorectal Cancer Cell Caco-2 by EGF Stimulation

Objective By observing the effectiveness of epidermal growth factor(EGF)combined with 5-flauouracil(5-FU)in the treatment of human colorectal cancer,to explore the anti-tumor effects of EGF.Methods MTT assay was used to evaluate the effects of EGF and 5-FU alone or combined use on the growth of tumor cells.The effects of EGF on PCNA expression were detected by Western blot,and flow cytometry was done to measure the changes in cell cycle.Results EGF could induce proliferation of tumor cells.With the increase in concentration of EGF,the number of cells in G0 phase was significantly reduced,and the expression of PCNA obviously increased.There was significant difference between 100 or 1 000 ng/mL EGF groups and control group(P<0.05).The combined use of EGF and 5-FU could more significantly inhibited the growth of tumor cells than 5-FU alone(P<0.05).Conclusion EGF can enhance the sensitivity of colorectal cancer cell Caco-2 to 5-FU,which might be related to the induction of proliferation of quiescent colorectal cancer cells.
5.Chitosan Promoting the Growth of in vitro Cultured Retinal Nerve Cells of SD Baby Rats

Objective To study the effect of chitosan on the growth of the in vitro cultured retinal nerve cells. Methods The retinal nerve cells of SD baby rats were cultured in vitro. The effects of chitosan at 0.065%,1.25%,2.5%,5%,10%,15% on the growth of the retinal nerve cells were measured by MTT method. The blank control group was created. Results 1.25%,2.5%,5%,and 10% chitosan could obviously promote the growth of retinal nerve cells,and 10% chitosan was the optimal concentration for the growth of nerve cells. Conclusion Chitosan can effectively promote the growth of retinal nerve cells,and it is necessary to undertake the further study of Chitosan.
6.The Effect of RNA Interference Suppressing the Expression of AKT2 on Sensitivity of Human Pancreatic Cancer Cell Line Xenograft in Nude Mice to Gemcitabine

Objective To study the effect of RNA interference suppressing the expression of AKT2 on sensitivity of human pancreatic cancer cell line xenograft in nude mice to gemcitabine.Methods The human pancreatic cancer cell implanted tumor mode1 in the nude mice was established.By abdominal cavity administration and intratumoral injection,the mice bearing tumor were treated with gemcitabine in combination with vector of pAKT2-siRNA.Tumor growth of tumor tissues was observed,the AKT2 mRNA and protein expression levels in tumor tissues detected by RT-PCR and immunohistochemistry respectively,and tumor apoptosis measured by Tdt-mediated dUTP nick end labeling(TUNEL).Results In chemotherapy+AKT2-siRNA group the expression of AKT2 mRNA and protein was significantly lower than in control group,chemotherapy group and chemotherapy+blank plasmid group.The tumor weight and tumor volume in chemotherapy+AKT2-siRNA group were significantly reduced as compared with those in other three groups.The inhibition rate and apoptosis rate in chemotherapy+AKT2-siRNA group were significantly higher than those in other three groups.Conclusion Sensitivity of human pancreatic cancer cell line to gemcitabine could be significantly improved by RNA interference suppressing the expression of AKT2.
7.Involvement of Heme Oxygenase-1 in the Protective Actions of Curcumin against Ethanol-induced Oxidative Damage in Rat Primary Hepatocytes

Objective To investigate the role of heme oxygenase-1(HO-1)in the protective actions of curcumin against ethanol-induced oxidative damage.Methods Microsomal HO-1 enzyme activities were determined in rat primary hepatocytes pretreated by curcumin.AST,LDH release and hepatic oxidative/antioxidant status were measured with or without ZnPPⅨ/Hemin as a classic HO-1 inhibitor/inducer,respectively.Results Curcumin could induce the HO-1 expression and enzyme activity,which was correlated with antioxidant levels in hepatocytes.HO-1 induction reached a peak under administration of 15 μmol/L curcumin for 1 h.Conclusion HO-1 induction by curcumin is contributed to the heptoprotective effects against ethanol-induced oxidative damage.
8.Application of Bivariate Analysis of Sensitivity and Specificity in Meta-Analysis

Objective To illustrate the evaluation effect of bivariate analysis of sensitivity and specificity meta-analysis model in diagnosis test to provide basis for selecting better evaluation method of diagnostic test.Methods Bivariate model was presented by reanalyzing the data from a published meta-analysis of two diagnostic techniques in diagnosis of schistosomiasis japonica.Results The bivariate model could directly provide summary estimates of(logit)sensitivity,specificity and DOR with corresponding 95% CI for two diagnostic tests(IHA and ELISA).Also,it could elicit any significant difference that existed among sensitivity,specificity and DOR between the two diagnostic methods,and incorporate any correlation that existed between sensitivity;specificity.Conclusion The bivariate model preserves the two dimensional nature of the original data,and separates effects of sensitivity and specificity,which is more rational than a net effect on diagnostic odds ratio scale as in SROC approach.The bivariate model is appropriate and agile,and can be used as an extension and improvement of the traditional SROC method.
9.Effect of Recombinant Interleukin-23 on Systemic Candidasis in Mice

Objective To investigate the effect of recombinant murine interleukin-23(rIL-23)on systemic candidiasis in a murine model.Methods A cyclophosphamide-induced immunosuppressed murine model of systemic candidiasis was established.The mice were divided into control group and rIL-23 treatment group.Colony forming units(CFU)of the kidney and spleen were determined by using plating dilution method.The histopathological changes and degree of infection of the kidney and spleen were graded.Meanwhile,the levels of interferon gamma(IFN-γ)in the spleen were measured by enzyme-linked immunosorbent assay.Results On the 2nd,3rd and 7th day after Candida albicans infection the number of CFU of the fungi in the kidney in the control group was significantly greater than that in rIL-23 treatment group(P<0.01).The number of CFU of the fungi on the 2nd,3rd and 7th day after Candida albicans infection in the spleen in control group was also greater than that in rIL-23 treatment group,but without statistically significant difference(P>0.05).The scores of histopathological changes in the kidney in rIL-23 treatment group were lower than those in control group(P<0.01),and the degree of infection was milder in rIL-23 treatment group.The scores of histopathological changes in the spleen in rIL-23 treatment group were also lower than those in control group,but without statistically significant difference(P>0.05).The levels of IFN-γ in the spleen on the 2nd,3rd and 7th day after infection in rIL-23 treatment group were significantly higher than those in control group(P<0.01).Conclusion rIL-23 has protective effect on murine systemic candidiasis.
10.Construction of the Eukaryotic Expression Vectors and the microRNA Expression Plasmids of Human Fas and TNFR1 Gene and Their Biological Effects in vitro

Objective To construct the eukaryotic expression plasmids of human Fas and TNFR1 gene(pcDNA3.0-hFas and pcDNA3.0-hTNFR1)and microRNA(miRNA)expression plasmid of hFas and hTNFR1 named p-hFasmiRNA and p-hTNFR1miRNA,and to investigate their inhibitory effects in vitro.Methods The eukaryotic expression plasmids of human Fas and TNFR1 gene were constructed(pcDNA3.0-hFas and pcDNA3.0-hTNFR1)and have been shown successfully to express hFas and hTNFR1 protein.miRNA expression plasmids of hFas and hTNFR1 named p-hFasmiRNA and p-hTNFR1miRNA complimentary to the sequence responsible for hFas and hTNFR1 respective were constructed,meanwhile irrelevant miRNA plasmid was used as a control.By respective co-transfection of p-hFasmiRNA and pcDNA3.0-hFas,p-hTNFR1 miRNA and pcDNA3.0-hTNFR1 expression construct into 293T cells,the inhibition of hFas and hTNFR1 expression was analyzed by real-time PCR and Western blot.Results The experiments showed the significant inhibitory effect of p-hFasmiRNA on hFas and p-hTNFR1miRNA on hTNFR1 expression at 48 h post-transfection both at RNA level and protein level as well in 293T cell lines with the inhibitory efficiency being as high as 87% for hFas and 80% for hTNFR1,respectively.Conclusion The p-hFasmiRNA and p-hTNFR1miRNA were constructed successfully,and it was verified that they could specifically inhibit the hFas and hTNFR1 expression at the cellular level.
