1.Determination of Tetramethylpyrazine in Ligusticum Chuanxiong Hort by RP-HPLC Coupled with Countercurrent Extraction

An analytical method for determination of tetramethylpyrazine (TMP) in ligusticum chuanxiong hort by RP-HPLC coupled with countercurrent extraction used for the preconcentration of trace TMP was established. The column was packed with Zorbax ODS (250 mm×4 mm,5 μm). The mobile phase consisted of methanol water (1 % acetic acid)(35∶65),the flow rate was 0.8 ml/min and detection was performed at 300 nm. The calibration curves showed good linearity over the range of 0.41 μg/ml to 4.1 μg/ml,γ=0.9993,the recoveries were 92.3 %. The method was simple,fast,sensitive and accurate and could be applied to the analysis of ligusticum chuanxiong hort. The results showed that the content of TMP in ligusticum chuanxionghort were different with different sources.
2.Clinical Implication of Microvessel Density and Vascular Endothelial Growth Factor Expression in Breast Cancer

The clinical implication of microvessel density (MVD) and vascular endothelial growth factor (VEGF) expression in breast cancer and the relationship between them were studied. Specimens of 53 cases of breast cancer were examined for MVD in tumor and VEGF expression by using immunohistochemical staining. The results showed that the mean MVD in node-positive breast cancers and in the patients with postoperative distant metastasis was higher than that in node-negative breast cancers and those without metastasis, respectively (both P<0.01). The expression rate of VEGF in breast cancer was 67.92 % and was closely relative to MVD. It was suggested that MVD was closely correlated with metastasis of the tumors and might be a useful prognostic indicator for breast cancer. VEGF might be an important factor responsible for the induction of angiogenesis in breast cancer.
3.Studies on the Confirmation of the Structure of Triplidine Hydrochloride

The chemical structure of Triplidine Hydroloride was confirmed by using EA, IR, UV-Vis, 1NMR, 13NMR, MS after it was synthesized. It was concluded that the synthetic Triplidine Hydrochloride was the same as the standard.
4.Clinical Characters of Rhegmatogenous Retinal Detachment

The clinical data of 109 patients with rhegmatogenous retinal detachment were retrospectively analyzed and the relationships among retinal detachment,refraction and the shapes of hole in different age groups discussed. The results showed that myopia was more commonly seen in the patients less than 40 years old and the occurrence of myopia was gradually decreased in the patients over 40 years old. Round hole was more commonly seen in the patients less than 40 years old and horseshoe-shaped tears were more often found in the age group of over 40 years old.
5.The Clinical Use of Double-paper Synergy Test in the Detection of Bacteria Producing Extended-spectrum β-lactamase

The optimal condition of double-paper synergy test in the detection of bacteria producing extended-spectrum β-lactamase (ESBLS) was studied. In 244 isolates of Enteric bacilli,Klebsiella pneumonia,Enteric cloacae,there were 49 ESBLS-producing strains with the positive rate being 19.67 %. Five ESBLS-producing strains were detected in Pseudomonas maltophilla. One ESBLS-producing strains was detected in Pseudomonas aeruginose,Enteric aerogenes and Citrobacter,respectively. The optimal substrates of double-paper synergy test were cefotaxime,aztreonam and cefepime,and the interspace was 25 mm. This method was still a simple,economical and appropriate method in the detection of ESBLS-producing isolates.
6.Study on the Cardiac Damage in the Patients with Polymyositis/Dermatomyositis

To investigate the significance of cardiac damage in the patients with polymyositis (PM)/dermatomyositis (DM),45 cases of PM/DM in our hospital from 1992 to 1998 were retrospectively analyzed in the aspects of the clinical data,muscle zymogram,X-ray plain films,electrocardiograph,ultrasonic cardiogram. It was found that cardiac damage (51.1 %) occurred in 23 out of 45 cases of PM/DM. Pericardium,cardiac muscle,endocardium and coronary artery were involved. Clinical manifestations were various. Among the cases of cardiac damage,3 died of heart failure. It was indicated that cardiac damage is the most common complication and also one of the common death causes of PM/DM.
7.Experimental Study of Lipid-Regulating Capsule and Simvastatin for the Dislipidemic SD Rats

The role of lipid-regulating capsule in the regulation of experimental dislipidemia was studied in SD rats. The SD rats were divided into 6 groups: group A (normal control),group B to F (experimental hyperlipidemia). The rats in the groups C,D and E received the capsule in a daily dose of 2.5 g,5 g and 10 g/kg respectively for 3 weeks,while the rats in the group F received Simvastatin in a daily dose of 7.5 mg/kg for 3 weeks. By comparison with the group B after 3 weeks,the levels of plasma total cholesterol (TC) and low density lipoprotein (LDL-C) were very significantly decreased in the groups C,D,E and F(P<0.01),the value of plasma total triglyceride (TG) was very significantly decreased in the groups D,E and F (<0.01),and the level of high density lipoprotein was significantly increased in the groups C,D and E (P<0.05),but very significantly increased in the group F (P<0.01). It was suggested that the therapeutic efficiency of the lipid-regulating capsule in low,middle and high dosage was the same as that in high dose of simvastatin for the SD rats with high plasma TC and LDL-C,and middle and high doses of the capsule had the same effect of simvastatin on the plasma TG.
8.Mutations of P16 Gene in 8 Human Malignant Lymphoma Cell Lines

In order to determine the relationship between P16 and the tumorigenesis of malignant lymphoma,silver-stained PCR-SSCP analysis and direct sequencing were used to evaluate the alterations of P16 gene in 8 human malignant lymphoma cell lines. A homolygous loss was found in SU-DHL-9 while mutations of P16 exon 2 in SU-DHL-1 and Daudi. Sequencing of the PCR-product showed three point mutations in SU-DHL-1,which were GAC to AAC in codon 72,GOT to ACT in codon 77,and a insertion of C between codon 82 and 83,respectively. This study suggested the mutations of P16 gene are highly frequent in B-cell lymphoma cell lines and may play an important role in its tumorigenesis.
9.Effect of Erigeron Breviscapini on Chronic Hypoxic Pulmonary Hypertension and Serum Nitric Oxide and Endothelin-1 in Rats

To study the effect of Erigeron breviscapini on chronic hypoxic pulmonary hypertension and mechanism,the hypoxic rats were injected intraperitoneally with Erigeron breviscapini. The mPAP,the content of serum nitric oxide (NO) and endothelin-1 (ET-1) were measured 5,14 and 28 days after intraperitoneal injection of Erigeron breviscapini in hypoxia respectively,and were compared with those in the single hypoxic group. The results showed that the contents of serum mPAP and ET-1 were higher,and serum NO was lower in single hypoxic group than in the normal rats (both P<0.01). The contents of serum mPAP and ET-1 were lower and NO was higher in the Erigeron breviscapini-treated group than in the single hypoxic group (P<0.01). It was suggested that Erigeron breviscapini could reduce the hypoxic pulmonary hypertension by increasing NO and decreasing ET-1 of serum. Erigeron breviscapini could be used to treat hypoxic pulmonary hypertension.
10.Experimental Study on the Effects of Immediate Administration of Inderal on Postinfarction Left Ventricular Remodeling of the Rabbit

The effects of immediate administration of inderal following acute myocardial infarction on left ventricular remodeling were studied in acute myocardial infarction model of the rabbit. The results showed that there was no significant difference in the infarct size and expansion index between the infarct-inderal group and the infarct-placebo group. As compared with sham surgery-inderal group,infarct-inderal group had a higher ventricular weight,left ventricular volume index and left ventricular wall stress [(2.65±0.17) g/kg vs (2.32±0.11) g/kg,P<0.05;(2.22±0.23) ml/kg vs (1.70±0.12) ml/kg,P<0.001;(0.84±0.07) vs (0.75±0.052),P<0.05,respectively],but there was no significant difference between the infarct-inderal group and infarct-placebo group. Heart rate in the infarct-inderal group was significantly decreased as compared with that of infarct-placebo group [(153±6.36) min-1 vs (225±4.08) min-1,P<0.001],but there was no significant difference in left ventricular end-diastolic pressure,+dp/dt max and cardiac index between the two groups. It was suggested that immediate administration of inderal postinfarction had no preventive effect on left ventricular remodeling.
