1.Influence of cytarabine on proliferation and Apoptosis of K562 cells

ObjectiveTo explore the mechanisms of proliferation and apoptosis of K562 cells induced by cytarabine. MethodsDifferent doses of cytarabine on the proliferotion at K562 cell 24h,48h,72h were detected by MTT. Cell cycle And Apoptosis rate were detected by FCM. ResultsEach concentration of cytarabine obvious inhibited the growth of K562 cells. In the same concentrations,function of proliferation inhibition with cytarabine increased by the extension of time,which showed dose and time effect relations. Cytarabine on K562 cell,G0/G1 phase cell proportion of role as cytarabine increased when extension of time, S period and the G2/M period cells were decreased.Apoptosis of K562 cell in proportion as drug concentration and effect time increases, which present Cytarabine timeconcentration dependence. CondusionCytarabine was apparent inhibition to proliferation of K562 cell leukemia cell, which could promote K562 cell apoptosis by time and concentration dependence way.
2.Performance and dynamical characteristics of three-dimensional stent materials in tendon tissue engineering

OBJECTIVE: To summarize research progress of tissue-engineered tendon stent material and its application for repairing tendon defect. METHODS: A computer-based online search was conducted with the key words of "tissue engineering, tendon, scaffold" in both Chinese and English from January 1993 to October 2009. Articles about dynamical performance of tissue-engineered tendon,experimental research and clinical application of tissue-engineered tendon were included, but duplicated studies and Meta analysis were excluded. Among 38 articles, 4 major viewpoints were discussed: research progress of tendon tissue engineering; biodynamical analysis of tissue-engineered tendon stent materials; application of biomaterials for tendon tissue engineering; clinical application of tissue engineering for repairing tendon defect. RESULTS: Natural polymer and synthetic macromolecule were major materials in tendon tissue engineering. The preparation and selection of stent materials depended on producing imitated materials with advanced performance and structure based on artificial synthetic materials, preparing natural derivative stent materials based on natural extracellular matrix, and constructing tissue-engineered tendon based on original bio-structure and bio-performance of biological tissue. Construction of tissue-engineered tendon must simulate three-dimensional physical environment in vivo. Microcomputer-controlled step motor was used to force tendon-matrix compound stent in order to increase proliferation velocity of tendon cells and secretory volume of collagen and develop into stress direction. CONCLUSION: Physical stimulation is simple, thus technique and instrument for constructing artificial tendon need to be further studied. Additionally, living implants which may completely replace body tissue and organ function still need to be further studied and solved.

Objective:To explore clinical significance of the further method dealt with in ASCUS(atypical squamous cells undetermined significance) of cervical smear.Methods:118 patients with ASCUS were detected by colposcopy and high risk HPV test,simultaneously make biopsy of cervical guided by colposcopy.Result:By diagnosed with pathology,chronic cervical inflammation is 72 cases,46 cases were CIN(cervical intraepithelial neoplasia).In this study 40 cases were positive in HPV test which 33 cases are CIN(?2=48.18,P0.05).Conclusion:There are high CIN csses in Women with ASCUS whose should be paid attention to and strengthen management.Colposcopy and high risk HPV test are efficiency method to ASCUS,especially high risk HPV test is positive that need detective by colposcopy.The combination of colposcopy and high risk HPV test can improve detective rate and accuracy of cervical intraepithelial neoplasia(CIN).

6.Cisplatin therapy for in vivo enrichment of gastric cancer stem cells

BACKGROUND:Tumor stem cels have self-renewal, drug resistance and metastasis tumorigenicity, which play an important role in occurrence, development and metastasis of tumors. Currently, there are two methods to identify tumor stem cels, namely, in vitro tumor sphere culture experiments and in vivo mouse tumorigenic experiments. However, there ia a lack of reports regarding clinicaly enriched gastric cancer cels by chemotherapy. OBJECTIVE:To investigate the enrichment of rat gastric cancer stem cels by cisplatin, and to explore the screening methods for their surface marker proteins. METHODS: BCG-823 gastric cancer model was established in rats, and then rat models were randomized into two groups: rats in experimental groups were subjected to intravenous injection of 0.1, 0.2, 0.25, 0.3 g/L cisplatin via the tail vein; those in control group were injected with normal salinevia the tail vein. After three courses of chemotherapy, gastric stem cels-enriched tissues were colected. Tumor surface proteins were extracted using high-throughput protein microarray and identified by western blot assay. Effects of cisplatin on enrichment of rat gastric cancer stem cels and screening methods for surface marker proteins were compared. RESULTS AND CONCLUSION:Cisplatin at a dose of 0.3 g/L×200μL exhibited the best therapeutic effects, and moreover, with the dose increasing, the tolerance became worse and the incidence of adverse reaction became higher. Transplantation tumors were verified by hematoxylin-eosin staining. Western blot test results were similar to the findings of protein microarray method, that is, HLA-DQ, PMP22 and Claudin7 protein expressions increased in gastric tissues, but HLA-DR, CD14, CD16 and CD56 protein expression decreased. These findings suggest that cisplatin can be used to enrich gastric cancer stem cels in rats, and to successfuly screen the corresponding surface marker proteins.

9.The influence of 3 % hypertonic saline and 20 % mannitol on cerebral oxygen metabolism in patients undergoing neurosurgery for supratentorial glioma

Objective To study the effect of 3% hypertonic saline(HS) and 20% mannitol on cerebral oxygen metabolism in patients undergoing neurosurgery for supratentorial glioma.Methods 56 patients undergoing neurosurgery for supratentorial glioma were evenly divided into two groups.Group A(n =28) was treated by 3% hypertonic saline(5.35ml/kg).Group B (n =28) was treated by 20% mannitol(1 g/kg).CSFP was tested before hypertonic saline or mannitol infusion (T0),immediately (T1),15,30,60,120min after infusion (T2 ~5).Various blood-gas indexes were observed at To,T3 ~5.CaO2,CjvO2,Da-jvO2 and CERO2 were calculated.Results CSFP significantly dropped from T2 to T5 in both groups (P < 0.05).Da-jvO2 and CERO2 were significantly reduced at T4~5 in both groups (P < 0.05).There was no significant difference between the two groups.Conclusion 3 % HS is as efficient as 20% mannitol in reducing intracranial pressure and improving cerebral oxygen metabolism in patients undergoing neurosurgery for supratentorial glioma.
10.Effects of different inhalation anesthetic pretreatment on serum S100β protein and neurone specific enolase in infants during open-heart surgery under extracorporeal circulation

Objective To determine the brain protective effects of inhalation anesthetic pretreatment in infants undergoing open-heart surgery under extracorporeal circulation (ECC).Methods Forty-five infants (no more than 3 years old) with congenital heart diseases who received surgical treatment were divided into 3 groups by table of random digit,sevoflurane pretreatment group (group S,15 cases),isoflurane pretreatment group (group Ⅰ,15 cases) and control group (group C,15 cases).In pretreatment group,all infants had been inhaled 1.0-1.5 MAC sevoflurane or isoflurane continually for over 60 min after intubation,while group C did not receive pretreatment.Blood samples were taken before (T1) and in 5 min after ECC (T2),occlusion of aorta (T3),30 min after ECC (T4),end ofECC (T4),and 6 and 24 h after the end of ECC (T6 and T7).Serum levels of S 100 β protein and neurone specific enolase (NSE) were measured by enzyme-linked immunosorbent assay.Results There was no significant difference in the serum levels of S100β protein and NSE among the 3 groups in T1 (P>0.05).There was no significant difference in the serum levels of S100β protein and NSE in group S and group Ⅰ between T1 and T2-7 (P> 0.05).There was no significant difference in the serum levels of S100 β protein and NSE in T2-7 between group S and group Ⅰ (P > 0.05).In group C,there were significant differences in the serum levels of S100 β protein in T4-6 and NSE in T4-7 than those in T1 (P< 0.05).In T4-6,the serum levels of S100 β protein and NSE in group S and group Ⅰ were significantly lower than those in group C [S100 β protein:(0.45 ± 0.31),(0.47 ± 0.35) μ g/L vs.(0.73 ± 0.31) μ g/L,(0.53 ± 0.33),(0.54 ± 0.36) μ g/L vs.(0.91 ± 0.30) μ g/L,(0.47 ± 0.34),(0.49 ±0.35) μ g/L vs.(0.79 ± 0.40) μ g/L,P < 0.05; NSE:(3.26 ± 1.34),(3.32 ± 1.34) μ g/L vs.(7.20 ± 2.71)μ g/L,(5.57 ± 1.49),(5.61 ± 1.52) μ g/L vs.(7.68 ± 3.68) μ g/L,(2.29 ± 1.21),(2.34 ± 1.22) μ g/L vs.(9.46 ± 4.61) μg/L,P < 0.05].Conclusion Sevoflurane or isoflurane pretreatment significantly decreases the serum levels of S100 β protein and NSE during open-heart operation in infants with ECC.
