1.RNAi silencing of XIAP gene induces apoptosis and susceptibility to chemotherapy of osteosarcoma cell line MG63

Objective To explore the effect RNAi-mediated inhibition of X-linked inhibit apoptosis protein (XIAP) gene on apoptosis and susceptibility to chemical therapy drugs in osteosarcoma cells and its underlying mechanism.Methods shRNA plasmid of XIAP gene were constructed. MG63 cells were conventionally cultured and divided into untreated group (group A,non-transfection),blank plasmid group (group B,psiRNA-Con transfected),the experimental group (group C,psiRNA-XIAP transfected),and 2 chemical drug interfered groups,non-transfection+0.3 ?mol Dox group (A+Dox group) and psiRNA-XIAP transfection +0.3 ?mol Dox group (C+Dox group). The XIAP expression in MG63 cells was detected by Western blotting. The proliferation of above-mentioned MG63 cells was examined by MTT assay. 2.Explorative Eye Tracking Movement of Patients with Depression

Objective: To explore eye tracking movements of patients with depression.Methods:Thirty-one patients with depression were tested by tracking of exploratory eye movement (Type-Ⅳ) before and after treatment (mean interval 34?8 days), at the same time HAMD was applied. Thirty-one normal controls were tested by tracking of exploratory eye movement either.Results:There were significant differences of discriminant analysis between the results of patients and controls (0.52?1.58,0.79?1.44, vs.-1.00?0.85,t=4.40, 5.80, P0.05);Conclusion:There is statistical difference in exploratory eye tracking movements of depressive patients and that of controls. After depression improved, no significant changes were noted between the main indexes of eye tracking movements of the first test and that of the second test.
3.Comparative analysis of nasal NK/T-cell lymphoma with nasal polyps and inverted papilloma by CT

Objective To analyze CT findings of nasal NK/T-cell lymphoma.Methods The CT findings of 11 nasal NK/T-cell lymphomas, 134 nasal polyps and 24 nasal inverted papillomas were analyzed retrospectively.Fisher exact test was used to compare the differences of the three nasal lesions.Results Compared with the nasal polyps, nasal NK/T-cell lymphoma had a higher occurrence in the unilateral nasal cavity, more presented as nodular mass or mold growth,and more easily to invade nasal vestibule,nasal ala,nasal mucosal,nasopharyngeal wall and causebone destruction.Compared with the inverted papilloma,nasal NK/T-cell lymphoma had a higher incidence of tumor invasion to nasal vestibule, nasal ala, nasopharyngeal cavity, nasal mucosal.Conclusion Nasal NK/T-cell lymphoma is a homogenous tumor and often occurres in the unilateral nasal cavity, which creeps along the lateral wall of the nasal cavity, and causes the damage of nasal mucosal and surrounding soft tissue and slight destruction of bone in the early satge.
4.The synergistic effect of endothelin- 1 and basic fibroblast growth factor on rat vascular smooth muscle cell proliferation

AIM: The synergistic effect of basic fibroblast growth factor (bFGF) and endothelin- 1 (ET-1 ) on rat aortic vascular smooth muscle cells(VSMC) proliferation was observed. The possible mechanism of the synergism was also investigated. METHODS: BrdU incorporation and cell counting method were adopted to value the pro - proliferative effect of VSMC. Western- blotting was used to observe the variation of bFGF and FGFR - 1 isoforms expression. RESULTS: bFGF and ET- 1 could promote VSMC proliferation separately, and synergistically in combination. The synergism was dose - and time - dependent. ET- 1 increased all the three bFGF isoforms and FGFR - 1 protein level in dose - and time- dependent manner. In addition, after exhaustion of intracellular PKC, the upregulation effects of ET- 1 on bFGF and FGFR - 1 expressions in VSMC both inclined. CONCLUSION: bFGF and ET- 1 had synergistic effect on VSMC proliferation. ET- 1 may increase the responsiveness of VSMC to bFGF through modulation of bFGF isoforms together with FGFR - 1, which was PKC - dependent.

AIM: The role of protein kinase C(PKC) in the effect of Interleukin-6(IL-6) on basic fibroblast growth factor(bFGF) expression was investigated in rat vascular smooth muscle cells(VSMC). METHODS: Western-blotting was adopted to observe the variation of bFGF and its receptor type I isoforms expression. RESULTS: IL-6 increased all the three basic fibroblast growth factor isoforms in a dose-dependent (0-10.0 ?g/L) manner. The upregulatory activities peaked at 24 h as demonstrated . Objective To investigate the effect of Yishen-Daotan decoction on TCM syndrome elements of infertility caused by kidney deficiency and phlegm stagnant type polycystic ovary syndrome.Methods 60 patients of kidney deficiency and phlegm stagnant type polycystic ovarian syndrome infertilities were administrated with modified Yishen-Daotan decoction for 3 months and the changes of TCM syndrome elements,menstrual cycle and serum sex hormone were compared before and after the treatment.Results ① TCM syndrome elements had a statistical changes after the treatment compared with those before the treatment(t=62.79,P<0.01).②menstrual cycle showed statistical difference after the treatment compared with that before the treatment,(x2=21.64,P<0.01).③Serum FSH,E2 were increased,while LH,LH/FSH and T were decreased after the treatment,showing a significant difference compared with those before the treatment (t=2.67,P<0.05).④The total effective rate was 91.67% and pregnancy rate was 30%.Conclusion Yishen-Daotan decoction significantly improve clinical symptoms in patients with kidney deficiency and phlegm stagnant type PCOS infertility.
8.Effect of Shoutaiwan on Improving Clinical Pregnancy Rate of In Vitro Fertilization-Embryo Transfer

Objective To observe the effect of Shoutaiwan on improving clinical pregnancy rate of in vitro fertilization-embryo transfer (IVF-ET). Method Eighty people, who were easy to be followed up and pregnant with the assistant of IVF-ET by the tube-baby centre of Shenzhen Urology Surgical Hospital of Zhongshan Medical University from 2001 to 2003, were divided into two groups randomly with 40 people for each. Treatment group, after embryo transfer, were normally injected progesterone and auxiliary treated of traditional Chinese Medicine according to syndrome differentiation. The comparing group were only injected progesterone. The curative effect was observed and compared. Result In 40 cases of treatment group, 23 cases were pregnant, clinical pregnant rate was 57.5%, the average level of progesterone was (22.45?1.31)?g/L, pregnant abortion rate was 13.0%. In 40 cases of comparing group, 12 cases were pregnant, clinical pregnant rate was 30.0%, the average level of progesterone was (10.18?0.89)?g/L, pregnant abortion rate was 25%. There were significant differences in the clinical pregnant statistical rate, the average level of progesterone and pregnant abortion rate between the two groups. Conclusion It’s more effective that the syndrome differentiation method based on Shoutaiwan than progesterone treatment only in improving clinical pregnant rate and reducing pregnant abortion rate of IVF-ET.
9.Construction of safety culture evaluation system for intravenous treatment

Objective To construct safety culture evaluation system for intravenous treatment. Method On the basis of literature review and in-depth interview of two rounds using Delphi method , consensus was achieved to build an evaluation system for safety culture of intravenous treatment . Results The recovery rate from expert consultation questionnaires of two rounds was 90 . 00% and 94 . 40%, respectively . The expert′s authority coefficient was greater than 0 . 80 . The expert′s coordination coefficient was 0 . 308 , with significant difference statistically ( P < 0 . 05 ) . After two rounds of expert consultation , an evaluation index system for safety culture of intravenous treatment was formed , including five 5 primary indexes and 25 secondary indicators . The experts had consensus on the indexes . Conclusion The evaluation system can help us to understand the status quo of intravenous treatment′s safety culture so that we can make targeted improvement . It is of importance for guaranteeing the safety of patients with intravenous therapy .
