1.Immunohistochemical Study of the Expression of p53, Pan-ras, c-erbB-2 and PCNA in N-Nitrosomorpholine(NNM)-Induced Hepatocellular Carcinoma of Rats.

The focus of this study was o aialyze the morphologic expression of p53, Pan-ras, c-erbB-2, and PCNA in preneoplastic and neoplastic liver lesions induced with NNM of rats. The development of hepatocellular tumors was investigated by histology and electron microscope in 65 Splague-Dawley rats administered with NNM in drinking water at low dose(5 mg/100 ml) and high dose(20 mg/100 ml). Three types of hepatocytic degeneration glycogenotic, eosinophilic and basophilic changes were followed by the appearance of hepatocellullar carcinoma. Hepatocellular carcinoma was increased in number and size according to NNM dosage and to duration of exposure. The histological classifications of hepatocelular carcinoma wer trabecular type, which was which was the most common, large eosinophilic, small cell, adenocarcinomatous and clear cell type. The expression of p53, Pan-ras, c-erbB-2 PCNA was examined by immunohistochemical stains. Eosinophilic degeneration revealed mild positivity at 18-26 weeks for expression of all oncogenic proteins studied and PCNA, whereas precancerous lesions showed variable expression from negative to moderate positivity on PCNA. Hepatocellular carcinoma lesions showed strong positivity for all stains and increased intensity during experimental period. These may indicate that chemical carcinogen produce hepatic eosinophilic degeneration and preCancerOus lesions by genetic mutation, resulting in hepatocellular carcinoma.

2.A literature review on the health status of Korean workers under the Japanese colonialism.

The history of occupational health in Korea has covered the era of the Republic of Korea after the Liberation from Japanese colonialism. But the number of Korean workers exceeded about 2 millions at the times of liberation in 1945, so that it is expected that many occupational health problems inflicted Korean workers under the Japanese colonialism. The authors reviewed medical literatures, administrative documents, and other available data which were published under the colonial state, and collected things which had reference to the health status of Korean workers. The results were as follows; 1. Nutritional status of Korean workers was supposed to be inferior to of general population, some students, and poor inhabitants in a remote moutain villages. 2. It was supposed that the constitution of Korean workers was near lower limit of average build of contemporary Koreans. 3. The accidents rate in mines was significantly high but decreasing year after year, and the most importance cause of accidents was the fall of roof in the mine. The medical facilities and equipments for miners were supposed to be not sufficient in the mines and workshops. 4. Some occupational disease including silicosis, noise-induced hearing impairment, and decompression disease were known. But, overall incidence or prevalence of these diseases could not be identified. 5. On the whole, the fatalities of acute infectious diseases of Korean workers were higher than those of Japanese inhabitants in Korea and Korean inhabitants. The prevalence of pulmonary tuberculosis of Korean workers was increasing with every passing year. 6. The medical personnels and facilities were so deficient that most Korean workers were out of adequate medical use. We discussed only a part of the health status of Korean workers under the Japanese colonialism, so it would be necessary to have a better grasp of details of occupational health policy and health status in the era of afflicting.

3.The Current Status and Prospects of Community Nutrition Services: I. A Study on the Perception of Nutrition Services for the Health Center-Directors in Areas Not Employing Public Health Nutritionists.

The purposes of this study were to investigate the perception of community nutrition programs and the conditions to carry them out, for 32 directors of health centers where public health nutritionists are not employed. The results of this study can be summarized as follows. Nutrition intervention programs were not carried out actively except those for infants but the health center directors strongly felt the necessity of carrying out various nutrition programs. The factors that affect the selecting and priotizing of nutrition programs were the need of community residents, the perception of the local congressmen, and the head of the local government. The most urgent and important problem to be solved in order to intervene nutrition programs was employing public health nutritionists on a tenure basis. Other were securing necessary revenue, precise guidance, political support, hardwares, etc. These results showed the importance of employing public health nutritionists for intervening appropriate nutrition programs and improving the perception of nutrition services for health center directors, local congressmen and personnels in charge of formulating nutrition policies.

4.Morphological Change and Distribution of Laminin and Fibronectin in Early Chick Embryos.

The distribution of fibronectin and laminin in early chick embryos has been studied by immunohistochemical method. Fibronectin was detected not only at the basement membranes of neural tube, dorsal aortae and somites but also at the entire mesenchyme area. But laminin only appeared at the basement membranes. During the development of chick embryo, the distribution pattern of these glycoproteins was not changed but showed strong intensity of staining. The dorsal aorta of a 2-day-old chick embryo had several layer of fibronectin-producing cells, but was devoid of laminin. Laminin expression commenced faintly after Day 3. In the brain capillary wall, fibronectin was distributed but laminin was not at Day 10. In the cardiac jelly, fibronectin was detected as thin dotted strands but laminin was not expressed at Day 2. Along the endocardium, fibronectin was detected but laminin was not found. After Day 3, fibronectin was found as increased in endocardium, myocardium and cardiac jelly, whereas laminin expression commenced. Cushion mesenchymal cell migration and development of myocardial trabeculae were inhibited by antifibronectin injected in the cardiac jelly but not anti-laminin. Conclusively, this study suggests that fibronectin and laminin are glycoproteins in the extracellular matrix but there is a difference between their distributions. Cushion mesenchymal cell migration from endocardium and development of trabeculae in chick embryonic heart are inhibited by anti-fibronectin. This result suggests that fibronectin is a `stepping stone` for the embryonic migrating cells and triggers cell migration. But laminin may not serve as a matrix for cell migration.

5.The Role of Endoscopic Balloon Dilation in the Treatment of Esophageal Strictures.

We evaluated the role of endoseopic balloon dilatation for esophageal strictures caused by postoperative anastomosis in 5 cases, sclerotherapy in 4 cases, caustic injury in 6 cases, and malignancy in 16 cases. The success rate of balloon dilatation was 100% in postoperative anastomotic and post-scle- rotherapy strictures, 16.7% in caustic stricture, and 62.5% in malignant stricture. The addition of bougienage increased the final success rate of dilatation therapy from 16.7 to 66.7% in caustic stricture, and 62.5 to 93.8% in malignant stricture. The overall one-year recurrence rate in benign stricture was 34.5%. All of the recurrent strictures were successfully retreated by balloon dilatation. In conclusion, endoscopic balloon dilatation is a safe, effective, and easy method for the management of benign esophageal strictures except longstanding caustic stricture, and can be used as an auxiliary therapy prior to more definitive treatments such as esophageal intubation in malignant stricture.

6.Four Cases of Retinitis Punctata Albescens Occurred in a Family.

The authors observed 4 cases of retinitis punctata albescens presented in a family, namely, a sister and 3 brothers of 6 siblings. Typical fundus findings were found in all four patients. Under ordinary illumination, the perripheral visual field was within the limit of normal, but concentrically contracted in 5 to 30 degrees under the reduced illumination.

8.Taxonomic Studies on Alternaria in Korea (1).

The taxonomy of the Alternaria spp. has been a subject, of controversy because of their high variability in conidial morphology and polymorphism displayed even in pure cultures. The published Korean literature on the genus Alternaria is scattered and fragmentary, and pertains to about 25 species with special emphasis on occurrence and pathogenicity, but mycological studies on this group of fungi are insufficient. This is the first series of detailed and consolidated account of Korean species of Alternaria, which includes 11 species; viz., Alternaria brassicae, A. brassicicola, A. cinerariae, A. dauci, A. dianthi, A. dianthicola, A. helianthi, A. helianthinffciens, A. iridicola, A. japonica and A. protenta. Detailed diagnostic descriptions, specific characterizations, taxonomic discussions and illustrations for each species are presented.

9.Inhibition of Melanoma Cell Lines Using Antisense Sequence Expressing Adenovirus and Cisplatin.

PURPOSE: Telomerase is known to play a role of adding repetitive parts to chromosomal ending and to be involved in carcinogenic process through cell immortalization. The purpose of this study is to evaluate that restraining of telomerase activation can have killing effect on cancer cell and enhance susceptibility of cancer cells to anticancer substance. METHODS: The killing effect on melanoma cells was studied by using recombinant adenovirus that makes it possible to inhibit telomerase from getting activated, with such targets as two types of melanoma cell lines. This recombinant adenovirus was used combined with cisplatin, one of the most representative anticancer medicine to evaluate enhancement in susceptibility of cancer cells to anticancer substance. RESULTS: From the result of cytotoxic assay, it is found that melanoma cells have much resistance to cisplatin on the whole. In the case of using Ad-OA of recombinant virus alone, killing effect on melanoma cells was insignificant. On the other hand, when Ad-OA was used in combination with cisplatin, susceptibility of melanoma cell lines to cisplatin was enhanced. CONCLUSIONS: Ad-OA, recombinant adenovirus, could be used as a supplementary medicine in the targeted cancer gene therapy against cancer cell lines resistant to cisplatin.

10.The Comparison of the Effect of Therapeutic Lrens, Collagen Shield, and Tarsorrhaphy on Stromal Inflammation after Photorefractive Keratectomy in a Rabbit model.

Reducing inflammation after photo refractive keratectomy may decrease postoperative pain and reduce corneal haze intensity. We evaluated the effect of the therapeutic lens(T-lens) and collagen shield on stromal inflammation. Adult New Zealand white rabbits were subjected to excimer laser PRK. Polymorphonuclear leukocytes(PMNs) of ablated corneal stroma were counted 10 hours after surgery with a microscope. The number of PMNs in the central cornea was lower in the T-lens group (29.77 +/- 11.6) and collagen shield group(36.77 +/- 5.9) than in the tarsorrhaphy group (75.33 +/- 14.6). Of all the groups, the eyes which were left unpatched showed the most marked infiltration of PMNs into the wound(226.83 +/- 18.2). The T-lens and collagen shield may reduce postoperative corneal haze intensity better than a pressure patch by suppression of stromal inflammation.
